Epizootic lymphangitis (EL) is a highly prevalent and contagious infectious disease affecting horses in many parts of Ethiopia caused by Histoplasma capsulatum sensu lato ('var. farciminosum'). In this study, 12 suspected isolates of H. capsulatum sensu lato or yeasts unidentified by conventional biochemical tests isolated from Ethiopian horses with EL were characterised by internal transcribed spacer sequencing. Six of the 12 isolates were identified to be members of H. capsulatum sensu lato and the other six were Pichia kudriavzevii (synonym: Candida krusei) (n = 3), Trichosporon asahii (n =

Largemouth bass ranavirus (LMBV) is highly contagious and lethal to largemouth bass, causing significant economic losses to the aquaculture industry. Oral vaccination is generally considered the most ideal strategy for protecting fish from viral infection. In this study, the fusion protein MCP-FlaC, consisting of the main capsid protein (MCP) as the antigen and flagellin C (FlaC) as the adjuvant, was intracellularly expressed in Pichia pastoris. Subsequently, the recombinant P. pastoris was freeze-dried to prepare the oral vaccine P-MCP-FlaC. Transmission electron microscopy and scanning elect

Supplementing chicken feed with antibiotics can improve survival and prevent disease outbreaks. However, overuse of antibiotics may promote the development of antibiotic-resistant bacteria. Recently, antimicrobial peptides have been proposed as alternatives to antibiotics in animal husbandry. Here, we evaluate the effects of antimicrobial peptide, Epinephelus lanceolatus piscidin (EP), in Gallus gallus domesticus. The gene encoding EP was isolated, sequenced, codon-optimized and cloned into a Pichia pastoris recombinant protein expression system. The expressed recombinant EP (rEP) was then use

BACKGROUND: Human hookworms (Necator americanus, Ancylostoma duodenale, and Ancylostoma ceylanicum) are intestinal blood-feeding parasites that infect ~500 million people worldwide and are among the leading causes of iron-deficiency anemia in the developing world. Drugs are useful against hookworm infections, but hookworms rapidly reinfect people, and the parasites can develop drug resistance. Therefore, having a hookworm vaccine would be of tremendous benefit.
METHODOLOGY/PRINCIPAL FINDINGS: We investigated the vaccine efficacy in outbred Syrian hamsters of three A. ceylanicum hookworm antig

In this study, fungemia cases from four tertiary hospitals located in Shanghai and Anhui province in China from March 2012 to December 2013 were enrolled to investigate clinical features, species distribution, antifungal susceptibility and strain relatedness. During the study period, 137 non-duplicate cases and their corresponding isolates were collected. Six different genera of fungi were identified, of which Candida spp. were the most common (126/137, 91.97 %), with C. albicans predominating (48/137, 35.03 %). The non-Candida fungi rate reached 8.03 % (11/137), and Pichia spp. was the most c

In this study, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) was used as a rapid method to identify yeasts isolated from patients in Tunisian hospitals. When identification could not be exstablished with this procedure, sequencing of the internal transcribed spacer with 5.8S ribosomal DNA (rDNA) (ITS1-5.8S-ITS2) and D1/D2 domain of large-subunit (LSU rDNA) were employed as a molecular approach for species differentiation. Candida albicans was the dominant species (43.37% of all cases), followed by C. glabrata (16.55%), C. parapsilosis (13.23%), C.

BACKGROUND: Epstein-Barr virus (EBV) associated malignancies with a Type II latency gene expression pattern, such as Hodgkin's disease, and nasopharyngeal carcinoma (NPC), frequently express the EBV antigen latent membrane protein 2A (LMP2A). We expected to establish a highly expressing LMP2A yeast cell strain and get the high quality LMP2A protein, which was used for detection, analysis and characterization of its antibodies in various patients' sera of EBV associated malignancies.
METHODS: The plasmid pPICZalphaA-LMP2A containing the full length of LMP2A cDNA was constructed and transformed

Defensins are a superfamily of antimicrobial peptides, present in vertebrates, invertebrates, fungi and plants, suggesting that they appeared prior to the divergence in eukaryotes. The destitution of toxicity to mammalian cells of plant defensins has led to a new research ground, i.e., their potential medical use against human infectious diseases. Isolating defensins from natural sources, like plant tissues, can be time-consuming, labor intensive and usually present low yields. Strategies for large-scale production of purified active defensins have been employed using heterologous expression s

MD-2, a eukaryotic accessory protein, is an essential component for the molecular pattern recognition of bacterial endotoxins. MD-2 interacts with lipid A of endotoxins [lipopolysaccharide (LPS) or lipooligosaccharide (LOS)] to activate human toll-like receptor (TLR) 4. The structure of lipid A influences the subsequent activation of human TLR4 and the immune response, but the basis for the discrimination of lipid A structures is unclear. A recombinant human MD-2 (rMD-2) protein was produced in the Pichia pastoris yeast expression system. Human embryonic kidney (HEK293) cells were transfected

During a 5-month period, Hansenula anomala (H. anomala), an opportunistic fungus, caused an outbreak of infections in eight adult patients treated at a surgical intensive care unit (ICU). The source of the infections and route of transmission could not be identified. A case-control study included 32 patients treated simultaneously at the surgical ICU. Univariate analysis pointed to the following significant risk factors: blood alkalosis, reduced urea, duration of hospitalization, bacteremia and colonization with Pseudomonas aeruginosa, and an APACHE II score >17 (during bacteremia or fungemia)

Bioprospecting for extremozymes from unique ecological niches is crucial for developing robust biocatalysts for green chemistry. Here, we report the de novo hybrid genome assembly of Penicillium crustosum CTM10622, isolated from the humid montane forest of El Feïdja National Park, Tunisia. Using Illumina NextSeq™ 500 and Nanopore PromethION 2 Solo, a highly contiguous 31.38 Mb assembly (N50 = 1.94 Mb; 98.3% BUSCOs) was achieved. This robust genomic foundation enabled the identification of an extensive hydrolase repertoire, leading to the discovery of a novel alkaline serine lipase, PCLIP, subs

H9N2 subtype avian influenza virus (AIV) facilitates viral adaptative evolution and cross-species transmission, result in inflicting severe economic losses on the poultry industry and posing significant public health threats. Currently, conventional vaccine-based prophylactic strategies exhibit certain limitations, highlighting an urgent need for the development of novel prevention and control measures. In this study, using phage display technology, we screened 37 potential binding sequences from the PBMCs of an alpaca immunized with the H9N2 virus and its HA protein. AlphaFold3 further predic

The efficient production of food and biochemicals using microorganisms that utilize single-carbon feedstocks presents a promising approach for advancing a circular bioeconomy. Komagataella phaffii (formerly Pichia pastoris) is a methylotrophic yeast already widely used in industry, making it an attractive host for such applications. Recently, K. phaffii was converted into an autotrophic strain capable of assimilating CO2 into both biomass and secreted organic acids, using energy derived from dissimilation of methanol to CO2. In these strains, methanol oxidation is catalysed by an alcohol oxida

Wild and synanthropic birds harbour a diverse range of yeasts, including species of recognised clinical relevance. Given their close interaction with human activities, these birds represent a valuable source for investigating environmental yeasts and assessing the performance of identification methods. We investigated yeasts recovered from cloacal and crop samples of birds from Gran Canaria and compared routine identification methods with molecular sequencing. Twenty-four isolates were examined by biochemical profiling (API ID32C) and MALDI-TOF MS. Molecular identification based on ITS sequenc

Engineered yeast-living drugs offer significant potential for oral protein therapy but remain limited by poor intestinal retention and lack of precise spatiotemporal control over therapeutic delivery. Here, we developed an ingestible hydrogel-encapsulated optogenetic yeast system controlled by a wearable electronic device for programmable interleukin-10 (IL-10) delivery in inflammatory bowel disease (IBD). We engineered Pichia pastoris with an EL222-based blue-light-inducible circuit achieving rapid, stringent IL-10 secretion (50% saturated concentration within just 1 h upon induction). The ye

Cysticercosis, caused by the larval stage of T. solium, remains a major neglected tropical disease with severe clinical and socioeconomic consequences in endemic regions. Although cyclic AMP-dependent protein kinase (PKA) signaling is fundamental to parasite development and survival, its therapeutic relevance in T. solium has not been comprehensively explored. This study provides an integrated molecular and functional characterization of the T. solium PKA catalytic subunit (TsPKA-c) and evaluates its potential as a novel therapeutic target for cysticercosis control. Recombinant TsPKA-c was suc

Vulvovaginal candidiasis (VVC) is usually attributed to Candida albicans (C. albicans), but non-albicans Candida species and rare yeasts are increasingly encountered in recurrent, complicated, or treatment-refractory disease. Their clinical significance is not uniform: some isolates represent causative infection, whereas others reflect colonization, mixed vaginitis, or detection without causality. This Mini Review addresses three linked clinical and biological questions. First, pathogen attribution should move from detection to causative diagnosis. Recurrent or refractory symptoms warrant stru

Drosophila suzukii is a pest of soft and stone fruits that is attracted to yeast volatile metabolites. Drosophila suzukii has distinct summer and winter morphs which are found in different habitats. Complex communities of yeasts likely differ between habitats and thus we hypothesized morphs differ in their attraction to yeast volatiles from different species and combinations of species. We presented D. suzukii with yeast species in isolation and in combinations, as either post-culture mixes or co-cultured, and measured activity of the flies in laboratory choice tests with a Locomotor Activity
