Staphylococcus aureus produces a wide variety of superantigenic activity Staphylococcal enterotoxins (SE) and they are a major cause of food poisoning. These superantigens are associated with mobile genetic elements such as plasmids, prophages and S. aureus pathogenicity islands (SaPI). The presence of well-known eight SaPI integrase and 13 enterotoxin genes (sea, seb, sec, sed, see, seg, seh, sei, sej, sel, sek, seq, and tst) in 93 S. aureus strains were investigated. All S. aureus isolates were characterized by pulsed-field gel electrophoresis (PFGE), and the genes were detected using five s

Staphylococcal enterotoxin B (SEB) is a potent exotoxin produced by the Staphylococcus aureus. This toxin is classified as a superantigen because of its ability to directly bind with MHC-II class molecules followed by activation of a large proportion of T cells bearing specific Vβ-T cell receptors. Commonly associated with classic food poisoning, SEB has also been shown to induce toxic shock syndrome, and is also considered to be a potential biological warfare agent because it is easily aerosolized. In the present study, we assessed the ability of indole-3-carbinol (I3C) and one of its byprodu

Staphylococcus aureus is a facultative, Gram-positive coccus well known for its disease-causing capabilities. In particular, methicillin and vancomycin resistant strains of S. aureus (MRSA and VRSA, respectively) isolated globally represent daunting medical challenges for the 21(st) Century. This bacterium causes numerous illnesses in humans such as food poisoning, skin infections, osteomyelitis, endocarditis, pneumonia, enterocolitis, toxic shock, and autoimmune disorders. A few of the many virulence factors attributed to S. aureus include antibiotic resistance, capsule, coagulase, lipase, hy

Monitoring data on Staphylococcus (S.) aureus in wild animals are scarce, yet transmission to humans and livestock remains a concern. S. aureus can cause infections in humans and animals, and the production of staphylococcal enterotoxins (SEs) may cause food poisoning. This study monitored the occurrence of S. aureus in wild ungulates intended for human consumption in Brandenburg, Germany. A total of 323 nasal swabs were collected from wild boar, roe, fallow, and red deer during the 2023/24 hunting season. Samples were analyzed by selective enrichment. Isolates were identified using MALDI-TOF

Aqueous phase synthesis of CdTe quantum dots (QDs) with surface functionalization for bioconjugation remains the best approach for biosensing and bioimaging applications. We present a facile aqueous phase method to prepare CdTe QDs by adjusting precursor and ligand concentrations. CdTe QDs had photoluminescence quantum yield up to ≈33% with a narrow spectral distribution. The powder X-ray diffraction profile elucidated characteristic broad peaks of zinc blende cubic CdTe nanoparticles with 2.5-3 nm average crystalline size having regular spherical morphology as revealed by transmission electro

Bacterial superantigens are a family of exotoxins that are the most potent T-cell activators known. Because of their ability to induce strong immune activation, superantigens have been implicated in a variety of diseases ranging from self-limiting food poisoning to more severe toxic shock syndrome (TSS) and have the potential to be used as agents of bioterrorism. Nonetheless, the precise molecular mechanisms by which T-cell activation by superantigens lead to acute systemic inflammatory response, multiple organ dysfunction, and ultimately death are unclear. Inadequate understanding of the path

Features of serological shifts in patients with alimentary toxicoinfections (ATI) caused by one infective agent (E. coli, staphilococci, Proteus, enterococci) and by two infective agents in various combinations have been studied. The suppression of immunogenesis has been found to be most pronounced in patients having ATI mixed according to agglutinin titers to the autostrains of infective agents. The features of serological shifts in mixed ATI should be taken into consideration in evaluating the diagnostic significance of this reaction in the time course of the disease.

The effect of pH on the thermal inactivation of staphylococcal enterotoxin A was investigated. Analysis of heated toxin by immunodiffusion in gel indicated that enterotoxin A in beef bouillon was inactivated faster at pH 5.3 than at pH 6.2. The z values (slopes) for the heat inactivation curves at pH 6.2 and 5.3 were 49.5 and 55 F (about 27 and 30 C), respectively. Enterotoxin produced and heated in dialyzed Casamino Acids medium and assayed by monkey feeding was more easily inactivated by heat at pH 5.3 than at pH 7.8. Thermal inactivation curves for enterotoxin A in beef bouillon (5 mug/ml,

Staphylococcus aureus enterotoxin A (SEA) is a leading cause of food poisoning. The current test for functional activity of SEA requires monkeys or kittens. The major drawbacks of animal assays are lack of quantitation, poor reproducibility, low sensitivity, and high cost. In this report we describe and evaluate an alternative assay using T-cell proliferation to measure SEA activity in food. Human and rat lymphocytes proliferate in response to concentrations of SEA as low as 1 pg/ml, well below the pathogenic dose of 100 ng. This proliferation assay is highly sensitive, quantitative, and simpl

This paper is a comparative study on epidemiological data compiled from papers of intestinal infections and intoxications in Poland during the years 45-89. The following criteria were applied: kind of food as vehicle of infection/intoxication in outbreaks; place of appearance of outbreaks; bacterial aetiological agents of cases and serotypes of Salmonella as causative agent in outbreaks. The number and percentage of outbreaks and cases in outbreaks were analyzed. Most cases and outbreaks were connected with dishes from meat in 1945-1979 and with dishes from uncooked eggs in 1985-1989. Most out

Staphylococcal enterotoxins (SEs) are a family of 17 major serological types of heat-stable enterotoxins that are one of the leading causes of gastroenteritis resulting from consumption of contaminated food. SEs are considered potential bioweapons. Many Staphylococcus aureus isolates contain multiple SEs. Because of the large number of SEs, serological typing and PCR typing are laborious and time-consuming. Furthermore, serological typing may not always be practical because of antigenic similarities among enterotoxins. We report on a microarray-based one-tube assay for the simultaneous detecti

A total of 224 Staphylococcus aureus strains from human carriers (110 strains) and manually handled foods (114 strains) collected in the Principality of Asturias, Spain over 1995-1999 were analysed for the production of enterotoxins (SEs) A, B, C, and D by a reversed passive latex agglutination test and by amplification of ent genes (A, B, C, D, E, and J) using PCR. Sixty-two strains were enterotoxigenic and a good relation between detection of SEs and their ent genes was found. No strain carried entE and all strains producing SED carried entD and entJ genes. Among the enterotoxigenic strains

Staphylococcus aureus is responsible for a wide range of pyogenic infections, food poisoning, and allergic inflammation. S. aureus produces a very wide variety of proteinaceous exotoxins, which are classified by structure as β-barrel pore-forming toxin, oligonucleotide/oligosaccharide-binding (OB)-fold/β-grasp proteins, and triple-helix bundle-containing proteins. We previously reported that three staphylococcal exotoxins belonging to the first two groups activate basophils and mast cells, which are innate immune cells that play a central role in allergic inflammation. In this study, we focuse

Exposure to Staphylococcal enterotoxin B (SEB) causes food poisoning, acute inflammatory lung injury, toxic shock syndrome, and often death. In this study, we investigated whether microRNA (miRNA) play a role in regulating SEB-driven inflammation in the lungs. Exposure to SEB caused immune cell infiltration, robust cytokine and chemokine production, compromised lung function, and 100% mortality in mice. We assessed miRNA and mRNA expression in lung infiltrating mononuclear cells following exposure to SEB and found 89 miRNA that were dysregulated (>2-fold) compared with vehicle controls. In sil
