Spacelab experiments on space motion sickness.

Recent research results from ground and flight experiments on motion sickness and space sickness conducted by the Man Vehicle Laboratory are reviewed. New tools developed include a mathematical model for motion sickness, a method for quantitative measurements of skin pallor and blush in ambulatory subjects, and a magnitude estimation technique for ratio scaling of nausea or discomfort. These have been used to experimentally study the time course of skin pallor and subjective symptoms in laboratory motion sickness. In prolonged sickness, subjects become hypersensitive to nauseogenic stimuli. Re

Hypertension and orthostatic hypotension in applicants for spaceflight training and spacecrews: a review of medical standards.

The inauguration of NASA of the position of Payload Specialists for SHUTTLE-SPACELAB flights has broken the tradition of restrictive medical physical standards in several ways: by reducing physical requirements and extensive training; by permitting the selection of older individuals and women; by selecting individuals who may fly only one or several missions and do not spend an entire career in space activities. Experience with Payload Specialists to be gained during the forthcoming SPACELAB missions, observing man in spaceflight step by step on an incremental basis, will provide valuable data

RTMS Mitigates Simulated Spaceflight-Induced Emotional Dysfunction Through VEGFR-2-Mediated Neuroprotection.

PURPOSE: Repetitive transcranial magnetic stimulation (rTMS) has shown potential in mitigating emotional deficits induced by extreme environments, such as those encountered during spaceflight. However, its underlying molecular mechanisms remain poorly understood. This study investigates the role of vascular endothelial growth factor receptor 2 (VEGFR-2) in mediating the therapeutic effects of rTMS in a mouse model of simulated space composite environment (SSCE). METHOD: 10 Hz rTMS treatment was initiated after the last two weeks of SSCE exposure and continued for the remaining 2 weeks. Throug

Brain plasticity and sensorimotor deterioration as a function of 70 days head down tilt bed rest.

BACKGROUND: Adverse effects of spaceflight on sensorimotor function have been linked to altered somatosensory and vestibular inputs in the microgravity environment. Whether these spaceflight sequelae have a central nervous system component is unknown. However, experimental studies have shown spaceflight-induced brain structural changes in rodents' sensorimotor brain regions. Understanding the neural correlates of spaceflight-related motor performance changes is important to ultimately develop tailored countermeasures that ensure mission success and astronauts' health. METHOD: Head down-tilt b

Frequent premature ventricular contractions in an orbital spaceflight participant.

BACKGROUND: Commercial spaceflight participants on orbital flights typically are older than career astronauts and they often have medical conditions that have not been studied at high g or in microgravity. This is a case report of a 56-yr-old orbital spaceflight participant with essential tremor and frequent premature ventricular contractions that occurred at rates up to 7000 per day. Before training and spaceflight, he was required to complete extensive clinical investigations to demonstrate normal cardiac structures and the absence of cardiac pathology. The evaluation included signal average

The effect of head turn velocity on cross-coupled stimulation during centrifugation.

Short-radius centrifugation (SRC) provides a practical means of producing artificial gravity for long duration space flights, though perceptual side-effects could limit its operational feasibility. Head turns (HT) during SRC, other than those about the centrifugation axis, produce Cross-Coupled Stimulation (CCS), perceived as a tumbling sensation. CCS can be nauseagenic, though adaptation can minimize this detrimental effect over time. The force environment of CCS suggests that the head turn velocity plays a role in determining the stimulus magnitude, though its degree has not been characteriz

Preflight virtual reality training as a countermeasure for space motion sickness and disorientation.

INTRODUCTION: Research suggests that preflight training in virtual reality devices can simulate certain aspects of microgravity and may prove to be an effective countermeasure for space motion sickness (SMS) and spatial disorientation (SD). It is hypothesized that exposing subjects preflight to variable virtual orientations, similar to those encountered during spaceflight, will reduce the incidence and/or severity of SMS and SD. METHODS: Subjects were assigned to either a variable training (VT) or nonvariable training (NVT) condition to perform a simple navigation and switch activation task i

Blood volume regulating hormones, fluid and electrolyte modifications during 21 and 198-day space flights (Altair-MIR 1993).

During the Altair MIR '93 mission we studied several parameters involved in blood volume regulation. The experiment was done on two cosmonauts before (B-60, B-30), during (D6, D12, D18 for French and D7, D12, D17 for Russian) and after the flight (R+1, R+3 and R+7). Space flight durations were different for two cosmonauts: for the Russian the flight duration was 198 days and for the French 21 days. On board the MIR station only urinary (volume and electrolytes, atrial natriuretic peptide (ANP), cyclic guanosine monophosphate (cGMP) and catecholamines) and salivary (cGMP and cortisol) samples w

Establishment of a 3D-dynamic osteoblasts-osteoclasts co-culture model to simulate the jawbone microenvironment in vitro.

AIMS: We aimed to establish a 3D osteoblasts/osteoclasts co-culture system requiring limited amounts of human primary cells and useful as platform to 1. recapitulate an "oral bone microenvironment" in healthy or pathological condition, and 2. produce potential implantable cell constructs for regeneration of jawbone which can be negatively affected by bisphosphonates (BPs). MAIN METHODS: Osteoblasts from normal bone chips (hOBs) or from jawbone of patients taking BPs (hnOBs) were co-cultured with monocytes (hMCs) either in static (3D-C) or dynamic (3D-DyC) condition using the RCCS-4™ bioreacto

Effects of local vibration on bone loss in -tail-suspended rats.

We investigated the effects of vibration (35 Hz, 45 Hz and 55 Hz) as countermeasure locally applied to unloading hind limbs on bone, muscle and Achilles tendon. 40 female Sprague Dawley rats were divided into 5 groups (n=8, each): tail-suspension (TS), TS plus 35 Hz/0.3 g vibration (TSV35), TS plus 45 Hz/0.3 g vibration (TSV45), TS plus 55 Hz/0.3 g vibration (TSV55) and control (CON). After 21 days, bone mineral density (BMD) and the microstructure of the femur and tibia were evaluated by μCT in vivo. The biomechanical properties of the femur and Achilles tendon were determined by a materials

Responses of motor and sensory neurons of rodents to spaceflight.

Spinal motoneurons innervating skeletal muscles comprised predominantly of high oxidative fibers, i.e. slow oxidative and fast oxidative glycolytic, have higher oxidative enzyme activities than motoneurons innervating skeletal muscles comprised primarily of low oxidative fibers, i.e. fast glycolytic. These findings suggest that there is a close relationship between the oxidative phosphorylation capacity of a motoneuron and of the muscle fibers that it innervates. Since some skeletal muscles become faster and less oxidative after 4-14 days of spaceflight, it might be expected that oxidative enz

Critical path plan for food and nutrition research required for planetary exploration missions.

In preparation for future planetary exploration, NASA-Johnson Space Center has developed a critical path plan for food and nutrition research needs. The plan highlights the risk factors pertaining to food and nutrition associated with exposure to the space flight environment as well as the possible consequences if no corrective measures are implemented. Included in the plan are the initiating events such as microgravity, remote environment and mission duration, which obviously impact the risk factors. The plan includes points of intervention where mitigating factors can be implemented to avoid

[Role of growth hormone underproduction and support load deficit in development of muscle atrophy and osteopenia in tail-suspended rats].

In a 20-day experiment with tail-suspended male rats histological and histomorphometric techniques were used to study the effects of growth hormone, thyroxin, and graded support loads on the progress of atrophy in soleus and gastrocnemius m.m., tibial metaphyses spongiosis, and growth of tibiae. Daily injections of growth hormone at a dose of 0.5 mg/kg of the body mass were found to restore the longitudinal growth of tibiae and to suppress osteopenia in the spongiosis of metaphyses; however, they did not have any noteworthy effect on the muscular atrophy in the suspended rats. A spaceflight study of synaptic plasticity in adult rat vestibular maculas.

Behavioral signs of vestibular perturbation in altered gravity have not been well correlated with structural modifications in neurovestibular centers. This ultrastructural research investigated synaptic plasticity in hair cells of adult rat utricular maculas exposed to microgravity for nine days on a space shuttle. The hypothesis was that synaptic plasticity would be more evident in type II hair cells because they are part of a distributed modifying macular circuitry. All rats were shared with other investigators and were subjected to treatments unrelated to this experiment. Maculas were obtai

Electrical vestibular stimulation and space motion sickness.

Electrical vestibular stimulation (EVS) in dynamic balance condition was studied in order to search for a new provocative test of space motion sickness (SMS). SMS is usually attributed to a sensory conflict caused by exposure to microgravity. Vestibular information is conflicting but also unusual and insignificant. EVS is in accordance with this feature because it is not the adequate stimulus of the vestibular receptors. EVS was achieved by means of binaural electrical stimulation. Effects of EVS were potentiated by compelling the subject to maintain dynamic balance on a seesaw. The quantifica

Limited musculoskeletal benefits of artificial gravity combined with cycling during bed rest: Results from the BRACE study.

Prolonged exposure to microgravity, simulated via 6° head-down tilt bed rest (HDT), induces musculoskeletal deconditioning and negatively impacts body composition. This study evaluated whether a combination of aerobic exercise with artificial gravity (AG) offers superior protection in comparison to exercise alone. Twenty-four healthy male participants completed 60 days of HDT, randomized into control (C), exercise-only (EX) and exercise with AG (EX-AG) groups. Muscle volume, intramuscular fat, body composition and isokinetic strength were assessed via whole-body MRI and isokinetic dynamometry.

The Effect of OSM on MC3T3-E1 Osteoblastic Cells in Simulated Microgravity with Radiation.

Bone deterioration is a challenge in long-term spaceflight with significant connections to patients experiencing disuse bone loss. Prolonged unloading and radiation exposure, defining characteristics of space travel, have both been associated with changes in inflammatory signaling via IL-6 class cytokines in bone. While there is also evidence for perturbed IL-6 class signaling in spaceflight, there has been scant examination of the connections between microgravity, radiation, and inflammatory stimuli in bone. Our lab and others have shown that the IL-6 class cytokine oncostatin M (OSM) is an i
