BACKGROUND: Newborn screening for maple syrup urine disease (MSUD) relies on finding increased concentrations of the branched-chain amino acids (BCAAs) leucine, isoleucine, and valine by tandem mass spectrometry (MS/MS). d-Alloisoleucine (allo-Ile) is the only pathognomonic marker of MSUD, but it cannot be identified by existing screening methods because it is not differentiated from isobaric amino acids. Furthermore, newborns receiving total parenteral nutrition often have increased concentrations of BCAAs. To improve the specificity of newborn screening for MSUD and to reduce the number of d

BACKGROUND: Female carriers of ornithine transcarbamylase deficiency (OTCD) have nearly normal rates of total urea synthesis, but they derive less urea from systemic glutamine amide nitrogen than do healthy persons.
OBJECTIVE: The objective of the study was to investigate whether females with symptomatic OTCD rely on alternative pathways to compensate for the reduced urea synthesis activity observed in this disorder.
DESIGN: The 5-d study involved 6 control subjects (3 males, 3 females) and 6 female OTCD carriers who had a fixed energy intake of 133 kJ. kg(-)(1). d(-)(1) and a protein intake

Major thermal injury is associated with extreme hypermetabolism and catabolism as the principal metabolic manifestations encountered following successful resuscitation from the shock phase of the burn injury. Substrate and hormonal measurements, indirect calorimetry, and nitrogen balance are biochemical metabolic parameters which are useful and more readily available biochemical parameters worthy of serial assessment for the metabolic management of burn patients. However, the application of stable isotopes with gas chromatography/mass spectroscopy and more recently, new immunoassays for growth

Agricultural nutrient loading is associated with variation in dissolved organic matter (DOM) composition and microbial communities across reservoir-agricultural river continuums, yet the covariation among nutrient conditions, DOM molecular heterogeneity, and bacterial metabolic potential remains insufficiently understood. This study integrated optical spectroscopy, ultrahigh-resolution mass spectrometry, nitrate dual-isotope tracing, end-member mixing analysis (EMMA), 16S rRNA sequencing, and metagenomic functional profiling to characterize DOM sources and composition and their associations wi

Atmospheric nitrogen (N) deposition is generally expected to stimulate plant carbon (C) sequestration and promote tree growth, thereby mitigating atmospheric CO2 accumulation. Yet, the magnitude of N deposition contribution to forest productivity remains contentious. While correlative studies suggest substantial plant growth enhancement, controlled fertilization experiments typically demonstrate a limited impact. This discrepancy may arise from whether or not to consider canopy N uptake processes. Here, we conducted a 10-y field experiment comparing canopy addition of N (CAN) with understory a

Typhoons and rainstorms (>250 mm/day) are extreme weather events changing hydrological characteristics and thus nitrogen (N) cycle in coastal waters. However, responses of N cycle to rainstorms and typhoons and their underlying mechanisms need to be elucidated. In this study, we conducted an analysis of a comparative dataset encompassing concentrations of nitrate (NO3-), ammonium (NH4+), dissolved oxygen (DO), chlorophyll a (Chl a), hydrological parameters, dual isotopic composition of NO3- (δ15N-NO3- and δ18O-NO3-) in Zhanjiang Bay during three distinct periods: the normal wet season, rainsto

We present an interdisciplinary analysis of finds from the Trypillia settlement of Kosenivka, Ukraine (ca. 3700-3600 BCE, Trypillia C1-2), that links information on human, faunal, and botanical remains with archaeological data to provide exceptionally detailed insights into life and death at a giant Trypillia settlement. We obtained osteological, palaeopathological, morphological and histotaphonomic data from human bone fragments; performed carbon and nitrogen stable isotopic analysis of human and animal bone to calculate food webs; modelled newly generated radiocarbon dates to refine the site

Mercury (Hg) concentrations and their associated toxicological effects in terrestrial ecosystems of the Gulf of Mexico are largely unknown. Compounding this uncertainty, a large input of organic matter from the 2010 Deepwater Horizon oil spill may have altered Hg cycling and bioaccumulation dynamics. To test this idea, we quantified blood concentrations of total mercury (THg) in Seaside Sparrows (Ammospiza maritima) and Marsh Rice Rats (Oryzomys palustris) in marshes west and east of the Mississippi River in 2015 and 2016. We also tested for a difference in THg concentrations between oiled and

Landfill operations and waste processing facilities are important and highly heterogeneous sources of both greenhouse gases (GHGs) and non-GHG air pollutants in the atmosphere. This arises the need for detailed apportionment of waste sources in order to locate and subsequently reduce emissions from landfills. Here, a time series of in situ measurements of atmospheric trace gases and spatial allocation of specific emission source types under different processing phases and environmental conditions were conducted in and in the surroundings of a Municipal Solid Waste Treatment Plant (MSWTP) in so

Storms, in subtropical regions such as S.E. China, cause major changes in the physical and biogeochemical fluxes of anthropogenic N species through the river-estuary continuum to the coast. Two weeks continuous observations at a sampling station (Station E) in the upper Jiulong River Estuary (S.E. China) were conducted to track the changes of physical and biogeochemical parameters together with genomic identification of nitrogen cycling microbes through a complete storm event in June 2019. In conjunction with previous N flux measurements, it was found that there was greatly increased flux of N

Despite growing concerns over mercury (Hg) accumulation in birds in recent decades, little is known about Hg exposure in nocturnal migratory birds. Here, total mercury (THg) and methylmercury (MeHg) were detected in the feathers of nocturnal migratory birds (n = 286, belonging to 46 species) passing through Mount Ailao in Southwest China. The stable isotope ratios of carbon (δ13C) and nitrogen (δ15N) were also determined to clarify the effects of trophic position, foraging guild, and foraging behavior on Hg bioaccumulation. Our results show that the THg and MeHg concentrations varied by two or

Mercury (Hg) is a toxic metal that accumulates in organisms and biomagnifies along food webs; hence, long-lived predators such as seabirds are at risk as a result of high Hg bioaccumulation. Seabirds have been widely used to monitor the contamination of marine ecosystems. In the present study, we investigated Hg concentrations in blood, muscle, and feathers of 7 procellariform seabirds breeding on the Chatham Islands, New Zealand. Using bulk and compound-specific stable isotope ratios of carbon and nitrogen as a proxy of trophic position and distribution, we also tested whether Hg contaminatio

The most direct test of functional capacity of the liver in nitrogen disposal is to stress the urea cycle with a high protein load. This has been used in the diagnosis of heterozygosity for ornithine carbamoyltransferase deficiency for many years by measuring the subsequent excretion of orotic acid in urine. Reports have shown some ambiguity in both this and the more recent allopurinol test. We investigated the effects of different foods as the protein load and of different analytical methods. A standardized protocol was developed, giving 35 g protein per m2 surface area as steamed fat-free ch

Branched-chain amino acids (BCAA), including valine, alloisoleucine, isoleucine, and leucine, play significant roles in a number of metabolic pathways in the body. Deficiency in branched-chain ketoacid dehydrogenase complex, an enzyme required for metabolism of those amino acids, will lead to elevation and accumulation of BCAA and ketoacids in bodily fluids. This results in maple syrup urine disease (MSUD), a condition estimated to affect 1 in 100,000-300,000 births. If MSUD is not diagnosed in the first few days of life, progression of this disease can lead to intellectual disability, coma, i

Clostridium histolyticum collagenase causes extensive degradation of collagen in connective tissue that results in gas gangrene. The C-terminal collagen-binding domain (CBD) of these enzymes is the minimal segment required to bind to a collagen fibril. CBD binds unidirectionally to the undertwisted C-terminus of triple helical collagen. Here, we examine whether CBD could also target undertwisted regions even in the middle of the triple helix. Collageneous peptides with an additional undertwisted region were synthesized by introducing a Gly → Ala substitution [(POG)(x) POA(POG)(y)]₃, where x +

BACKGROUND: Studies have documented an altered body composition in Turner syndrome (TS). Body fat is increased and muscle mass is decreased. Ovarian failure necessitates substitution with female hormone replacement therapy (HRT), and HRT induces favourable changes in body composition. It is unknown how HRT affects protein metabolism.
AIM: To test whether alterations in body composition before and after HRT in TS are a result of altered protein metabolism.
DESIGN: We performed a randomized crossover study with active treatment (HRT in TS and oral contraceptives in controls) or no treatment.

Human XPA is a 31 kDa protein involved in nucleotide excision repair (NER), a ubiquitous, multi-enzyme pathway responsible for processing multiple types of DNA damage in the eukaryotic genome. A zinc-associated, C4-type motif (C105-X(2)-C108-X(17)-C126-X(2)-C129) located in the minimal DNA-binding region (M98-F219) of XPA (XPA-MBD) is essential for damaged DNA recognition. Cadmium is a known carcinogen and can displace the zinc in many metal-binding proteins. It has been suggested that the carcinogenic properties of cadmium may result from structural changes effected in XPA when Cd(2+) is subs

Structural characterization of the SARS-CoV-2 full length nsp1 protein will be an essential tool for developing new target-directed antiviral drugs against SARS-CoV-2 and for further understanding of intra- and intermolecular interactions of this protein. As a first step in the NMR studies of the protein, we report the 1H, 13C and 15N resonance backbone assignment as well as the Cβ of the apo form of the full-lengthSARS-CoV-2 nsp1 including the folded domain together with the flaking N- and C- terminal intrinsically disordered fragments. The 19.8 kD protein was characterized by high-resolution

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by progressive cognitive and memory impairment. Within the brain, senile plaques, which comprise extracellular deposits of the amyloid-β peptide (Aβ), are the most common pathological feature of AD. A high concentration of Cu(2+) is found within these plaques, which are also areas under oxidative stress. Laboratory work has shown that in vitro Aβ will react with Cu(2+) to induce peptide aggregation and the production of reactive oxygen species. As such, this interaction offers a possible explanation for two of the defining
