Fermentation of Sargassum ilicifolium with Probiotics (Lactobacillus plantarum and Saccharomyces cerevisiae) Enhances Growth, Immunity, and Gene Expression in Rainbow Trout Before and After Challenge with Streptococcus iniae.

This study investigates the effect of Sargassum ilicifolium algae fermentation using Lactobacillus plantarum and Saccharomyces cerevisiae probiotics on growth, immunity, gene expression, and digestive and antioxidant enzymes before and after challenge with Streptococcus iniae in trout. Trout (50.69 ± 1.33 g) were fed with different percentages of S. ilicifolium (SI) powder over an 8-week trial. The supplements included both unfermented (SI 0.5%, SI 1%, SI 2%) and fermented (FSI 0.5%, FSI 1%, FSI 2%) forms. A significant increase in white blood cells was observed in the group receiving 2% FSI c

The impact of beauvericin on rainbow trout intestinal epithelial cells at different temperatures and dosing methods.

Mycotoxins in aquatic feeds and their effects on fish are becoming more important in aquaculture, as fishmeal and fish oil in feeds are being replaced with more sustainable plant protein. Here, we investigated the potential of the mycotoxin, beauvericin (BEA), to impact the rainbow trout (RT) intestine by using cultures of the epithelial cell line, RTgutGC. BEA was dosed in different ways and exposed at temperatures ranging from 4 to 26 °C before being evaluated for cell viability by the metabolic reduction of Alamar Blue, by the accumulation of Neutral Red (lysosomal activity), cytotoxicity (

High sensitivity to ocean acidification in wild out-migrating juvenile Pacific salmon is not impacted by feeding success.

Salmon populations are declining worldwide, with high mortality rates during juvenile marine migration presenting a bottleneck to recruitment. The ocean conditions along the main migratory route of juvenile salmon in British Columbia are characterized by high variability in CO2, with the amplitude, duration, and frequency of ocean acidification events exacerbated by climate change. Similarly, the variability in ocean conditions affects the abundance and diversity of plankton prey, leading to areas of food paucity for juvenile salmon. We investigated the combined effects of ocean acidification

Microparticles in Wild and Caged Biota, Sediments, and Water Relative to Large Municipal Wastewater Treatment Plant Discharges.

Anthropogenically modified microparticles including microplastics are present in municipal wastewater treatment plant (WWTP) effluents; however, it is unclear whether biotic exposures are elevated downstream of these outfalls. In the fall of 2019, the present study examined whether microparticle levels in resident fish, environmental samples, and caged organisms were elevated near the Waterloo and Kitchener WWTP outfalls along the Grand River, Ontario, Canada. Wild rainbow darters (Etheostoma caeruleum) were collected from a total of 10 sites upstream and downstream of both WWTPs, along with s

Traditional Chinese medicine bufalin inhibits infectious hematopoietic necrosis virus infection in vitro and in vivo.

UNLABELLED: Infectious hematopoietic necrosis virus (IHNV) causes infectious hematopoietic necrosis and severe economic losses to salmon and trout aquaculture worldwide. Currently, the only commercial vaccine against IHNV is a DNA vaccine with some biosafety concerns. Hence, more effective vaccines and antiviral drugs are needed to prevent IHNV infection. In this study, 1,483 compounds were screened from a traditional Chinese medicine monomer library, and bufalin showed potential antiviral activity against IHNV. The 50% cytotoxic concentration of bufalin was >20 µM, and the 50% inhibitory conc

Dietary supplementation with dandelion (Taraxacum officinale) flower extract provides immunostimulation and resistance against Streptococcus iniae infection in rainbow trout (Oncorhynchusmykiss).

Dandelion flower is a well-known phytomedicine due to its therapeutic effects on dyspepsia, bile duct disorders, spleen and liver complaints, and inflammatory diseases. This study aimed to assess the effect of dietary dandelion flower extract (DFE) on skin mucosal and humoral immunity, immune-related gene expression, and disease resistance in rainbow trout. The fingerlings (N = 300, 20.1 ± 0.9 g) were randomly assigned to 15 tanks and fed with different levels of DEF including 0 (control), 1, 2, 3, and 4 g/kg for 56 days. IgT Plays a Predominant Role in the Antibacterial Immunity of Rainbow Trout Olfactory Organs.

The olfactory organs (OOs) of vertebrates play important roles in their extraordinary chemosensory capacity, a process during which they are continuously exposed to environmental pathogens. Nasopharynx-associated lymphoid tissue (NALT) contains B cells and immunoglobulins (Igs), which function as the first defense line against antigens in mammals and also exist in teleosts. However, the immune responses of teleost NALT B cells and Igs during bacterial infection remain largely uncharacterized. In this study, rainbow trout were infected with Flavobacterium columnare via continuous immersion, aft

Establishment of time-resolved fluoroimmunoassays for detection of growth hormone and insulin-like growth factor I in rainbow trout plasma.

The GH/IGF-I axis influences many aspects of salmonid life history and is involved in a variety of physiological processes that are related to somatic growth (e.g., reproduction, smoltification, and the response to fasting and stress). As such, fisheries studies utilize GH/IGF-I axis components as indicators of growth and metabolic status. This study established time-resolved fluoroimmunoassays (TR-FIAs) for rainbow trout plasma GH and IGF-I using commercially available reagents. For the GH TR-FIA, the ED80 and ED20 were 0.6 and 28.1 ng/mL, the minimum detection limit was 0.2 ng/mL, and the in

Negligible cytotoxicity induced by different titanium dioxide nanoparticles in fish cell lines.

Titanium dioxide nanoparticles (TiO2-NPs) have a wide number of applications in cosmetic, solar and paint industries due to their photocatalyst and ultraviolet blocking properties. The continuous increase in the production of TiO2-NPs enhances the risk for this manufactured nanomaterial to enter water bodies through treated effluents or agricultural amendments. TiO2-NPs have shown very low toxicity in a number of aquatic organisms. However, there are no conclusive data about their deleterious effects and on their possible mechanisms of toxic action. At this level, in vitro cell culture systems

Tissue-specific cadmium and metallothionein levels in rainbow trout chronically acclimated to waterborne or dietary cadmium.

Rainbow trout were exposed to a sublethal concentration of waterborne Cd (0 or 3 microg/L) or dietary Cd (0 or 500 mg/kg dry wt) for 30 days to induce acclimation, and tissue Cd and metallothionein (MT) levels were examined. The greatest Cd concentrations were observed in the kidney followed by the gills and liver of the fish exposed to Cd via water, but in the gut tissues followed by the kidney, liver, and gills for dietary-exposed fish, reflecting a variation depending on the route of Cd exposure. Some MT was found in the nonacclimated naïve fish with no experience of elevated Cd exposure, a

Acute and chronic influence of temperature on red blood cell anion exchange.

Unidirectional (36)Cl(-) efflux via the red blood cell anion exchanger was measured under Cl(-) self-exchange conditions (i.e. no net flow of anions) in rainbow trout Oncorhynchus mykiss and red-eared freshwater turtle Trachemys scripta to examine the effects of acute temperature changes and acclimation temperature on this process. We also evaluated the possible adaptation of anion exchange to different temperature regimes by including our previously published data on other animals. An acute temperature increase caused a significant increase in the rate constant (k) for unidirectional Cl(-) ef

Renal responses of trout to chronic respiratory and metabolic acidoses and metabolic alkalosis.

Exposure to hyperoxia (500-600 torr) or low pH (4.5) for 72 h or NaHCO(3) infusion for 48 h were used to create chronic respiratory (RA) or metabolic acidosis (MA) or metabolic alkalosis in freshwater rainbow trout. During alkalosis, urine pH increased, and [titratable acidity (TA) - HCO(-)(3)] and net H(+) excretion became negative (net base excretion) with unchanged NH(+)(4) efflux. During RA, urine pH did not change, but net H(+) excretion increased as a result of a modest rise in NH(+)(4) and substantial elevation in [TA - HCO(-)(3)] efflux accompanied by a large increase in inorganic phos

Modulation of ganglion cell activity in the pineal gland of the rainbow trout: effects of cholinergic, catecholaminergic, and GABAergic receptor agonists.

Second order neurons within intact isolated pineal glands of the rainbow trout were explored by extracellular recordings to investigate modulatory effects of putative intrapineal neurotransmitters. Acetylcholine, dopamine, and norepinephrine were found to increase ganglion cell activity in a majority of cells tested. The excitatory effects of acetylcholine, dopamine, and norepinephrine were mimicked by muscarinic, dopamine D2, and beta-adrenergic receptor agonists and significantly increased with the applied light intensity, resulting in an attenuation of the ganglion cell response to light. Fish models for environmental carcinogenesis: the rainbow trout.

Progress over the past 30 years has revealed many strengths of the rainbow trout as an alternative model for environmental carcinogenesis research. These include low rearing costs, an early life-stage ultrasensitive bioassay, sensitivity to many classes of carcinogen, a well-described tumor pathology, responsiveness to tumor promoters and inhibitors, and a mechanistically informative nonmammalian comparative status. Low-cost husbandry, for example, has permitted statistically challenging tumor study designs with up to 10,000 trout to investigate the quantitative interrelationships among carcin

Chlorine dioxide suppresses Aeromonas through thiol oxidation and modulates hepatic redox-immune responses in Aeromonas salmonicida-infected rainbow trout (Oncorhynchusmykiss).

Chlorine dioxide (ClO2) is a potent oxidative disinfectant with potential use in bacterial disease control; however, its antibacterial mechanism and its influence on host redox-immune responses during Aeromonas salmonicida infection in rainbow trout (Oncorhynchus mykiss) remain unclear. Here, we evaluated the antibacterial activity of ClO2 against common bacterial pathogens, examined its thiol-targeting antibacterial mechanism, and characterized host toxicity, innate immune responses, oxidative stress, and hepatic transcriptomic regulation. ClO2 markedly inhibited Aeromonas hydrophila and A. s

Characterization of Gastrointestinal Absorption of Salmon Milt-Derived Oligodeoxyribonucleic Acids in Mice.

Dietary nucleic acids exert physiological effects beyond serving as substrates for nucleic acid synthesis; however, their exact molecular forms that are absorbed in the small intestine remain unclear. In this study, we investigated the absorption forms of food-derived DNA by comprehensively quantifying 31 nucleic acid monomers that permeated the intestinal membrane after oral administration, using LC-MS/MS. Hydrolyzed Oncorhynchus milt DNA (HD-omDNA; mainly 1-4-mer nucleotides), deoxyribonucleotides (dNTs), and deoxyribonucleosides (dNSs) were orally administered to mice, and blood samples wer

Differential Effects of Adult Salmon Lice Lepeophtheirus salmonis on Physiological Responses of Sockeye Salmon and Atlantic Salmon.

The salmon louse Lepeophtheirus salmonis, a type of sea lice (family Caligidae), is enzootic in marine waters of British Columbia and poses a health risk to both farmed Atlantic Salmon Salmo salar and wild Pacific salmon Oncorhynchus spp. At the adult stage, sea lice infections can often result in severe cutaneous lesions in their salmonid hosts. To evaluate and compare the physiological consequences of adult L. salmonis infections, smolts of Atlantic Salmon and Sockeye Salmon O. nerka were exposed to 2 (low), 6 (medium), or 10 (high) adult female lice/fish. Mean lice abundance decreased over

Copper toxicity in Bristol Bay headwaters: Part 1-Acute mortality and ambient water quality criteria in low-hardness water.

The world-class Alaskan Bristol Bay salmon fishery and vast deposits of copper (Cu) and other metals in the watershed warrant further investigation into the potential toxicity of Cu to salmonids under the low water-hardness conditions that occur in the watershed. Therefore we investigated the acute toxicity of Cu to rainbow trout (Oncorhynchus mykiss) and fathead minnows (Pimephales promelas) in low-hardness water (∼ 30 mg/L as CaCO3 ) formulated in the laboratory and collected from the Bristol Bay watershed. The median lethal concentration (LC50) for rainbow trout exposed to Cu in low-hardnes

Transmission potential of infectious hematopoietic necrosis virus in APEX-IHN®-vaccinated Atlantic salmon.

Infectious hematopoietic necrosis virus (IHNV) outbreaks have had a significant negative impact on Atlantic salmon Salmo salar production in British Columbia, Canada, since the first outbreak was reported in 1992. In 2005, the APEX-IHN® vaccine was approved for use in Canada for prevention of IHN. The vaccine was proven to be safe and efficacious prior to approval; however, it is unknown as to whether APEX-IHN®-vaccinated Atlantic salmon infected with IHNV can support replication and virus shedding in sufficient quantities to provide an infectious dose to a nearby susceptible host. To determin

Aquaporin family genes exhibit developmentally-regulated and host-dependent transcription patterns in the sea louse Caligus rogercresseyi.

Aquaporins are small integral membrane proteins that function as pore channels for the transport of water and other small solutes across the cell membrane. Considering the important roles of these proteins in several biological processes, including host-parasite interactions, there has been increased research on aquaporin proteins recently. The present study expands on the knowledge of aquaporin family genes in parasitic copepods, examining diversity and expression during the ontogeny of the sea louse Caligus rogercresseyi. Furthermore, aquaporin expression was evaluated during the early infes
