Endemicity and seasonality of transmission of human schistosomiasis in Ile-Ife, south western Nigeria.

Endemicity and seasonality of transmission of Schistosoma mansoni and S. haematobium were studied in Ile-Ife, south western Nigeria. Transmission of S. mansoni was demonstrated throughout the year in a man-made dam, whereas transmission of S. haematobium in a stream only occurred in the dry season. This difference in the transmission patterns should be considered when incorporating community-based focal plant molluscicide application in the integrated strategy for the control of schistosomiasis morbidity.

Chemotactic attraction of Schistosoma mansoni miracidia to acidic carboxyl-containing solutions in vitro.

The life cycle of Schistosoma mansoni includes a short-lived miracidial stage, during which miracidia must locate and penetrate suitable Biomphalaria snail hosts. Although several snail-derived attractants have been reported, the physicochemical properties underlying miracidial chemotaxis remain poorly defined. To examine the contribution of acidity and carboxyl groups to miracidial attraction, we first evaluated chemotactic responses to a previously reported snail-derived peptide (P12) and its amino acid-substituted derivatives under acidic and neutral conditions. Under acidic conditions, mir

Simultaneous genotyping of snails and infecting trematode parasites using high-throughput amplicon sequencing.

Several methodological issues currently hamper the study of entire trematode communities within populations of their intermediate snail hosts. Here we develop a new workflow using high-throughput amplicon sequencing to simultaneously genotype snail hosts and their infecting trematode parasites. We designed primers to amplify four snail and five trematode markers in a single multiplex PCR. While also applicable to other genera, we focused on medically and economically important snail genera within the superorder Hygrophila and targeted a broad taxonomic range of parasites within the class Trema

Endogenous growth factor stimulation of hemocyte proliferation induces resistance to Schistosoma mansoni challenge in the snail host.

Digenean trematodes are a large, complex group of parasitic flatworms that infect an incredible diversity of organisms, including humans. Larval development of most digeneans takes place within a snail (Gastropoda). Compatibility between snails and digeneans is often very specific, such that suitable snail hosts define the geographical ranges of diseases caused by these worms. The immune cells (hemocytes) of a snail are sentinels that act as a crucial barrier to infection by larval digeneans. Hemocytes coordinate a robust and specific immunological response, participating directly in parasite

Schistosomes and snails: a molecular encounter.

Biomphalaria glabrata snails play an integral role in the transmission of Schistosoma mansoni, the causative agent for human schistosomiasis in the Western hemisphere. For the past two decades, tremendous advances have been made in research aimed at elucidating the molecular basis of the snail/parasite interaction. The growing concern that there is no vaccine to prevent schistosomiasis and only one effective drug in existence provides the impetus to develop new control strategies based on eliminating schistosomes at the snail-stage of the life cycle. To elucidate why a given snail is not alway

An ecological model for quantitative risk assessment for schistosomiasis: the case of a patchy environment in the coastal tropical area of northeastern Brazil.

We developed a stochastic model for quantitative risk assessment for the Schistosoma mansoni (SM) parasite, which causes an endemic disease of public concern. The model provides answers in a useful format for public health decisions, uses data and expert opinion, and can be applied to any landscape where the snail Biomphalaria glabrata is the main intermediate host (South and Central America, the Caribbean, and Africa). It incorporates several realistic and case-specific features: stage-structured parasite populations, periodic praziquantel (PZQ) drug treatment for humans, density dependence,

The epidemiology of schistosomiasis in Burundi and its consequences for control.

This paper summarizes the results of a series of studies on the epidemiology, morbidity and transmission of Schistosoma mansoni in Burundi, and discusses their consequences for control. The main endemic area is the Imbo lowland, consisting of the Rusizi plain, the urban focus of Bujumbura, and the shores of lake Tanganyika; a small, new focus was discovered in the highlands, around lake Cohoha. Distribution studies on 5-10% population samples with duplicate 28 mg Kato smears in these 4 foci showed prevalences of 33%, 26%, 17%, 19% and mean (positive) egg loads of 110, 105, 92, 144 eggs/g, resp

African schistosomes in small mammal communities: Perspectives from a spatio-temporal survey in the vicinity of Lake Guiers, Senegal.

Schistosomiasis is a neglected tropical disease of public health significance. In view of its elimination as a public health problem by 2030, adopting a One Health approach is necessary, considering its multidimensional nature. Animal reservoirs, in particular, pose a significant threat to schistosomiasis control in Africa and beyond. In this study, we conducted a spatio-temporal survey of Schistosoma infections in small mammal communities and intermediate snail hosts in the vicinity of Lake Guiers in northern Senegal. Sampling campaigns were undertaken four times between April 2021 and August

Predicting the effects of climate change on Schistosoma mansoni transmission in eastern Africa.

BACKGROUND: Survival and fitness attributes of free-living and sporocyst schistosome life-stages and their intermediate host snails are sensitive to water temperature. Climate change may alter the geographical distribution of schistosomiasis by affecting the suitability of freshwater bodies for hosting parasite and snail populations. METHODS: We have developed an agent-based model of the temperature-sensitive stages of the Schistosoma mansoni and intermediate host snail lifecycles. The model was run using low, moderate and high warming climate projections over eastern Africa. For each climate

Coevolution and compatibility in the snail-schistosome system.

In stark contrast to the huge body of theoretical work on the importance of hosts and parasites as selective agents acting on each other, until recently, little systematic empirical investigation of this issue has been attempted. Research on snail-schistosome interactions have, therefore, the potential for making an important contribution to the study of coevolution or reciprocal adaptation. This may be particularly pertinent since snail-schistosomes represent an indirectly transmitted macroparasite system, so often overlooked amongst both theoretical and empirical studies. Here we review idea

Effect of anti-schistosomal treatment on schistosomal hepatic pathology: an experimental study in Syrian golden hamsters.

Thirty Syrian golden hamsters were infected with Schistosoma mansoni and 10 were used as negative controls. Hamsters were infected by 100 cercariae; 15 were treated by praziquantel in doses of 100 mg/kg at 12, 13, 14 and 15 weeks postinfection, and 15 hamsters were left as positive control. Five from each subgroup were sacrificed at 24, 28 and 32 weeks after infection. Animals were subjected to weekly analysis for total plasma protein, serum albumin and urinary total protein excretion. At the end point, animals were sacrificed and the mesenteric venous plexus was explored for adult worms. Live

Long-term dynamics of natural populations of Schistosoma mansoni among Rattus rattus in patchy environment.

Dynamics of natural populations of Schistosoma mansoni were studied during 8 consecutive years among Rattus rattus populations from 8 transmission sites of the marshy forest focus of Guadeloupe (French West Indies). The schistosome population is over-dispersed (k = 0.119) within the murine hosts and ecological factors linked to the patchy environment may be responsible for such aggregated distribution. Analysis of the spatio-temporal variations in prevalences, intensities and abundances showed limited variations of the infection during the 8 years at the level of the whole parasite population

Ecological studies on the intermediate host snails and the relevance to schistosomiasis control.

A detailed knowledge of distribution patterns of schistosome intermediate hosts and their population dynamics and factors affecting these patterns will provide useful information about the possibilities and desirability of conducting snail control measures in various transmission situations. On the basis of various case studies the association between the occurrence of human water contacts and the presence of schistosome intermediate hosts or infections in the intermediate hosts is illustrated. Other parameters affecting snail distribution patterns and density fluctuations are discussed. It is

Schistosomiasis Control in Nigeria: Moving Round the Circle?

BACKGROUND: Schistosomiasis has continued to plague low-resource areas of the Nigerian population. Mass drug administration (MDA) has been the only adopted interventional program for decades. However, it appears this effort does not culminate in transmission and morbidity reduction. PURPOSE: To highlight the current situation of schistosomiasis in Nigeria, why MDA alone cannot achieve the expected result, identify research needs, and promotion of integrated control approach for schistosomiasis. METHOD: A viewpoint based on practices, research findings, and personal and professional experienc

DNA vaccination using recombinant Schistosoma mansoni fatty acid binding protein (smFABP) gene.

Schistosomiasis is a fatal disease that has a negative impact on health and economics. Praziquantel (PZQ) is the drug of choice for schistosomiasis treatment, but it has no prophylactic effect; therefore, vaccination is an essential requirement in schistosomiasis control. This work was carried out to investigate the possible effect of DNA vaccination against Schistosoma mansoni infection using recombinant S. mansoni fatty acid binding protein (rsmFABP). The smFABP gene was cloned into the eukaryotic expression vector pcDNAI/Amp in order to obtain an smFABP-pcDNAI recombinant plasmid (DNA vacci

The potential effects of silver and gold nanoparticles as molluscicides and cercaricides on Schistosoma mansoni.

Schistosomiasis seriously affects human health in tropical regions. Its prevention is more important than treatment, raising the need for effective control methods. Recently, the role of nanomaterials in medical science has been growing. The present study aimed to evaluate the potential effects of silver (Ag) and gold (Au) nanoparticles (NPs) on Biomphalaria alexandrina snails and Schistosoma mansoni cercariae in vitro and to assess their effects on the infectivity of cercariae in vivo. The in vitro study proved that Ag and Au NPs were effective in killing B. alexandrina snails, with 30 μg/ml
