Bone Morphogenetic Protein-9 Is a Potent Chondrogenic and Morphogenic Factor for Articular Cartilage Chondroprogenitors.

Articular cartilage contains a subpopulation of tissue-specific progenitors that are an ideal cell type for cell therapies and generating neocartilage for tissue engineering applications. However, it is unclear whether the standard chondrogenic medium using transforming growth factor beta (TGFβ) isoforms is optimal to differentiate these cells. We therefore used pellet culture to screen progenitors from immature bovine articular cartilage with a number of chondrogenic factors and discovered that bone morphogenetic protein-9 (BMP9) precociously induces their differentiation. This difference was

Descriptive Analysis of In Vitro Cutting of Swine Mitral Cusps: Comparison of High-Power Laser and Scalpel Blade Cutting Techniques.

BACKGROUND AND OBJECTIVES: The most common injury to the heart valve with rheumatic involvement is mitral stenosis, which is the reason for a big number of cardiac operations in Brazil. Commissurotomy is the traditional technique that is still widely used for this condition, although late postoperative restenosis is concerning. This study's purpose was to compare the histological findings of porcine cusp mitral valves treated in vitro with commissurotomy with a scalpel blade to those treated with high-power laser (HPL) cutting, using appropriate staining techniques. MATERIALS AND METHODS: Fiv

1,1,1,3,3,3-Hexafluoro-2-propanol and 2,2,2-trifluoroethanol solvents induce self-assembly with different surface morphology in an aromatic dipeptide.

Peptide based self-assembled structures, especially those from smaller peptides, have attracted much research interest due to their potential applications as biomaterials. These structures have been produced using different solvents (one of the methods), including alcohols, except fluorinated alcohols, which are believed to support non-aggregated structures. Herein, we have studied the ability of 2,2,2-trifluoroethanol (TFE) and 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) solvents to induce self-assembly of an aromatic dipeptide, namely Tyr-Phe (YF). SEM images showed that HFIP and TFE can induce

Topical treatment with the opioid antagonist naltrexone accelerates the remodeling phase of full-thickness wound healing in type 1 diabetic rats.

Wound repair involves a series of overlapping phases that include inflammation, proliferation, and tissue remodeling, with the latter phase requiring months for proper healing. Delays in any of these processes can result in infection, chronic ulceration, and possible amputation. Diabetes is a major risk factor for improper wound repair, and impaired wound healing is a major complication for more than 26 million people in the US diagnosed with diabetes. Previous studies have demonstrated that the opioid antagonist naltrexone (NTX) dissolved in moisturizing cream reverses delays in wound closure

Diagnostic Challenge of a Tumor-mimicking Spinal Pseudogout Lesion: Case Report and Literature Review.

BACKGROUND/AIM: Retro-odontoid pseudotumor (ROP) is a non-neoplastic soft-tissue mass occurring around the C2 odontoid process and is most often associated with rheumatoid atlantoaxial instability. In rare instances, calcium pyrophosphate dihydrate (CPPD) crystal deposition (pseudogout) in the C1-C2 ligaments - a presentation known as "crowned dens syndrome" - can produce a periodontoid mass. Such CPPD-induced lesions usually present with acute neck pain and stiffness, with calcifications visible around the odontoid process on imaging. However, these masses can occasionally enlarge and compres

Russell Body Gastritis: Histochemical and Immunohistochemical Findings in a Series of Three Patients.

Russell body gastritis is a discrete morphologic pattern of chronic gastritis, characterized by numerous plasma cells filled with intracytoplasmic Russell body inclusions. It can be mistaken for neoplastic lesions and for extracellular hyaline structures. We report three adult men with Russell body gastritis. The patients were diagnosed and treated for various hematolymphoid disorders. During follow-up, they presented with abdominal pain, gastric erythema and thickening. One patient had Helicobacter pylori infection. The gastric biopsy showed an expanded lamina propria filled with numerous pla

Renal oxalosis in African green monkeys (Chlorocebus sabaeus) in St. Kitts.

Renal oxalosis has been reported in New and Old-World monkeys. Occasional reports describe a low prevalence of subclinical renal oxalosis, but these typically lack supporting evidence of primary oxalosis or toxicity and may be a natural background lesion. In a retrospective cross-sectional postmortem observation study, 12 of 156 (7.7%) African green monkeys (AGMs) (Chlorocebus sabaeus) from the Behavioural Science Foundation (St. Kitts) colony had histological evidence of oxalate-induced nephrosis (renal oxalosis). Histologically, affected tubules from both the cortex and medulla were ectatic;

Cutaneous silica-associated (cat litter) granulomas in the paws of 13 domestic cats.

Silica is a well-known stimulus of granulomatous inflammation in the lungs of humans and other animals. However, it has been poorly studied as a cause of cutaneous inflammation in domestic animals, despite the predominance of silica in traditional (clay-based) cat litter substrates. Here we characterize the clinical and pathologic findings of 13 surgical biopsies from the paws or paw pads of domestic cats submitted to 2 veterinary institutions between 2005 and 2023. Gross lesions often included chronic or intermittent paw pad swelling, ulceration and bleeding, or draining tracts, particularly

Non-tuberculous Mycobacterium Infection Associated with Artificial Stone Silicosis: A Case Series.

BACKGROUND: The incidence of non-tuberculous mycobacterium (NTM) infections has been rising in patients with chronic lung diseases. These infections cause significant morbidity, mortality, and elevated healthcare costs due to challenges in recognition, delayed diagnosis, and treatment. While NTM infections in natural stone silicosis are documented, the incidence in artificial stone silicosis remains unexplored despite increasing exposure to silica dust. OBJECTIVES: To describe the clinical, radiological, and pathological features of NTM infections in patients with artificial stone silicosis a

Matrix Metalloproteinase-1 and Acid Phosphatase in the Degradation of the Lamina Propria of Eruptive Pathway of Rat Molars.

The comprehension of dental pathogenesis and disorders derived from eruption failure requires a deep understanding of the molecular mechanisms underlying normal tooth eruption. As intense remodelling is needed during tooth eruption, we hypothesize that matrix metalloproteinase-1 (MMP-1) and acid phosphatase (ACP) play a role in the eruptive pathway degradation. We evaluated MMP-1-immunoexpression and the collagen content in the lamina propria at different eruptive phases. Immunohistochemistry and ultrastructural cytochemistry for detection of ACP were also performed. In the maxillary sections

Crystal identification of synovial fluid aspiration by polarized light microscopy. An online test suggesting that our traditional rheumatologic competence needs renewed attention and training.

Testing a reading exercise for identification of several typical crystal such as the negatively birefringent needle-shaped crystals that are under polarized light microscopy is the gold standard for diagnosing gout. The objective of this study was to assess current performance of crystal identification by professionals involved in examining synovial fluid in routine care. Rheumatologists, trainees, lab technicians, and other physicians with an interest in crystal arthritis completed an online test. The test consisted of 30 images: 8 monosodium urate (MSU) crystals, 5 calcium pyrophosphate (CPP

Systemic AA amyloidosis: epidemiology, diagnosis, and management.

The term "amyloidosis" encompasses the heterogeneous group of diseases caused by the extracellular deposition of autologous fibrillar proteins. The global incidence of amyloidosis is estimated at five to nine cases per million patient-years. While amyloid light-chain (AL) amyloidosis is more frequent in developed countries, amyloid A (AA) amyloidosis is more common in some European regions and in developing countries. The spectrum of AA amyloidosis has changed in recent decades owing to: an increase in the median age at diagnosis; a percent increase in the frequency of primary AL amyloidosis w

Periosteal Sharpey's fibers: a novel bone matrix regulatory system?

Sharpey's "perforating" fibers (SF) are well known skeletally in tooth anchorage. Elsewhere they provide anchorage for the periosteum and are less well documented. Immunohistochemistry has transformed their potential significance by identifying their collagen type III (CIII) content and enabling their mapping in domains as permeating arrays of fibers (5-25 μ thick), protected from osteoclastic resorption by their poor mineralization. As periosteal extensions they are crucial in early skeletal development and central to intramembranous bone healing, providing unique microanatomical avenues for

Photoelastic analysis of stress distribution of surgically assisted rapid maxillary expansion with and without separation of the pterygomaxillary suture.

PURPOSE: The aim of this study was to evaluate in vitro the mechanical effect over teeth and bone structures of surgically assisted rapid maxillary expansion (SARME) in photoelastic analogs by observing stress produced during Hyrax-type and Haas-type appliances activation. MATERIALS AND METHODS: Two photoelastic analogs were fabricated by use of birefringent materials to simulate an adult skull that contained teeth, bone, and maxillary sinus. Hyrax-type and Haas-type appliances were applied to the anchor teeth and incrementally activated. SARME was simulated by subsequent cuts of the lateral

Imaging skin pathology with polarized light.

Linearly polarized light that illuminates skin is backscattered by superficial layers and rapidly depolarized by birefringent collagen fibers. It is possible to distinguish such superficially backscattered light from the total diffusely reflected light that is dominated by light penetrating deeply into the dermis. The method involves acquisition of two images through an analyzing linear polarizer in front of the camera, one image (I(par)) acquired with the analyzer oriented parallel to the polarization of illumination and one image (I(per)) acquired with the analyzer oriented perpendicular to

Hemodynamic study of the anterior communicating artery.

BACKGROUND AND PURPOSE: The anterior communicating artery (ACoA) is a site of predilection for intracranial saccular aneurysms causing subarachnoid hemorrhage. ACoA aneurysms are frequently associated with an asymmetrical circle of Willis. In such cases, the ACoA is probably exposed to high hemodynamic stress caused by a considerable shunt flow across the ACoA to the distal segment of the contralateral anterior cerebral artery (ACA). In the present study, the flow pattern and flow-induced shear stress in the ACoA complex that may initiate aneurysmal lesions were studied under steady and pulsat
