HIF-1α is essential for the augmentation of myometrial contractility during labor†.

Uterine contraction is crucial for a successful labor and the prevention of postpartum hemorrhage. It is enhanced by hypoxia; however, its underlying mechanisms are yet to be elucidated. In this study, transcriptomes revealed that hypoxia-inducible factor-1alpha was upregulated in laboring myometrial biopsies, while blockade of hypoxia-inducible factor-1alpha decreased the contractility of the myometrium and myocytes in vitro via small interfering RNA and the inhibitor, 2-methoxyestradiol. Chromatin immunoprecipitation sequencing revealed that hypoxia-inducible factor-1alpha directly binds to

The reduction in circulating levels of estrogen and progesterone in women with preeclampsia.

Abnormalities in the development of placental vasculature in early pregnancy and the failure of transformation of the spiral arteries are associated with the pathogenesis of preeclampsia. Sex hormones influence neovascularisation during pregnancy. However the profiling of estrogen and progesterone in preeclampsia is controversial. In this study we investigated the serum levels of estrogen and progesterone in women with preeclampsia. Blood samples were collected from 86 preeclamptic and 97 gestation-matched normotensive pregnancies. The levels of 17β-estradiol (E2), progesterone and 2-methoxyes

HIF-1alpha inhibition ameliorates neonatal brain injury in a rat pup hypoxic-ischemic model.

Источник пометил публикацию как отозванную

Hypoxia-inducible factor-1alpha (HIF-1alpha) has been considered as a regulator of both prosurvival and prodeath pathways in the nervous system. The present study was designed to elucidate the role of HIF-1alpha in neonatal hypoxic-ischemic (HI) brain injury. Rice-Vannucci model of neonatal hypoxic-ischemic brain injury was used in seven-day-old rats, by subjecting unilateral carotid artery ligation followed by 2 h of hypoxia (8% O2 at 37 degrees C). HIF-1alpha activity was inhibited by 2-methoxyestradiol (2ME2) and enhanced by dimethyloxalylglycine (DMOG). Results showed that 2ME2 exhibited d

Development of an HPLC-FLD Method for Estradiol and Metabolites: Application of Solid-Phase Microextraction.

Estrogens are potent hormones involved in numerous physiological and pathological processes. Their typically low concentrations in biological samples necessitate highly sensitive analytical methods for accurate quantification. This study presents a high-performance liquid chromatography with fluorescence detection (HPLC-FLD) method for quantifying estradiol and its metabolites in blood serum and saliva. Analytes were extracted using solid-phase microextraction with a divinylbenzene sorbent and methanol as the desorption agent. FLD was performed after the derivatization of the analytes with dan

Invasive metastatic tumor-camouflaged ROS responsive nanosystem for targeting therapeutic brain injury after cardiac arrest.

Drug transmission through the blood-brain barrier (BBB) is considered an arduous challenge for brain injury treatment following the return of spontaneous circulation after cardiac arrest (CA-ROSC). Inspired by the propensity of melanoma metastasis to the brain, B16F10 cell membranes are camouflaged on 2-methoxyestradiol (2ME2)-loaded reactive oxygen species (ROS)-triggered "Padlock" nanoparticles that are constructed by phenylboronic acid pinacol esters conjugated D-a-tocopheryl polyethylene glycol succinate (TPGS-PBAP). The biomimetic nanoparticles (BM@TP/2ME2) can be internalized, mainly med

Combination therapy with arsenic trioxide for hematological malignancies.

Arsenic trioxide (ATO) has shown great promise in the treatment of patients with relapsed or refractory acute promyelocytic leukemia (APL). However, the risk/benefit ratios of ATO in hematologic malignancies other than APL are still unclear. In this review, the author attempts to provide current experimental and clinical challenges to gain more knowledge of the effects of ATO by examining combination therapies with other agents, especially for non-APL hematologic malignancies, such as acute myeloid leukemia (AML), acute lymphoid leukemia (ALL), chronic myeloid leukemia (CML), chronic lymphoid

Transcriptional profiling uncovers microglial HIF-1α signaling as a therapeutic target of radiation-induced brain injury.

Cranial irradiation is a widely used therapeutic modality for brain tumors but is frequently associated with long-term neurological complications, including cognitive deficits and mood disturbances. Despite its clinical relevance, the cellular and molecular mechanisms underlying radiation-induced brain injury (RIBI) remain poorly understood. In this study, we employed single-cell RNA sequencing to elucidate cell-type-specific transcriptional changes following cranial irradiation in mice. Irradiated animals exhibited marked microglial alterations, including a dynamic shift toward a pathological

The Role of Nanomedicine in Benign Gynecologic Disorders.

Nanomedicine has revolutionized drug delivery in the last two decades. Nanoparticles appear to be a promising drug delivery platform in the treatment of various gynecological disorders including uterine leiomyoma, endometriosis, polycystic ovarian syndrome (PCOS), and menopause. Nanoparticles are tiny (mean size < 1000 nm), biodegradable, biocompatible, non-toxic, safe, and relatively inexpensive materials commonly used in imaging and the drug delivery of various therapeutics, such as chemotherapeutics, small molecule inhibitors, immune mediators, protein peptides and non-coding RNA. HIF-1α-ASS1 axis drives reprogramming of arginine metabolism controls hepatic stellate cell activation in hepatic fibrosis.

Dysregulated arginine metabolism promotes hepatic stellate cell (HSC) activation and liver fibrosis. However, the mechanisms underlying arginine hosmeostasis in hepatic fibrogenesis and therapeutic targeting remain unclear. Using Multicellular hepatic spheroid (MCHS) models recapitulating in vivo liver fibrosis, we investigated the effects of arginine deprivation or supplementation. Genetic and pharmacological (ABRL8301, a 2-methoxyestradiol derivative) inhibition of hypoxia-inducible factor 1 alpha (HIF-1α) were assessed for effects on argininosuccinate synthetase 1 (ASS1) expression and anti

Disulfidptosis induced by intermittent fasting and metformin enhances the efficacy of anti-PD-1 therapy in renal cancer.

PBRM1-mutant ccRCC is a subtype of ccRCC with limited treatment options. Currently, there were no effective drugs specifically targeting this mutation. Here, we discovered that PBRM1-mutant ccRCC patients with comorbid type 2 diabetes mellitus (T2DM) show significantly prolonged OS and PFS. We further investigated the antitumor effects and mechanisms of metformin using mouse models and cell models. In vivo, the combination of intermittent fasting and metformin significantly inhibits tumor growth and in vitro, glucose deprivation combined with metformin treatment induces marked cell death in PB

Dexmedetomidine alleviates cerebral ischemia-reperfusion injury in rats via inhibition of hypoxia-inducible factor-1α.

Dexmedetomidine (Dex) was reported to reduce ischemia-reperfusion (I/R) injury in kidney and brain tissues. Thus, we aimed to study the role and mechanism of Dex in cerebral I/R injury by inhibiting hypoxia-inducible factor-1α (HIF-1α) and apoptosis. First, I/R injury models were established. Six groups were assigned after different treatments: sham, I/R, I/R+Dex, I/R+2-methoxyestradiol (2ME2) (HIF-1α inhibitor), I/R+CoCl 2 (HIF-1α activator), and I/R+Dex+CoCl 2 groups. Neurological function, cerebral infarction volume, survival, and apoptosis of brain cells were then analyzed. Besides, immuno

Medroxyprogesterone abrogates the inhibitory effects of estradiol on vascular smooth muscle cells by preventing estradiol metabolism.

Sequential conversion of estradiol (E) to 2/4-hydroxyestradiols and 2-/4-methoxyestradiols (MEs) by CYP450s and catechol-O-methyltransferase, respectively, contributes to the inhibitory effects of E on smooth muscle cells (SMCs) via estrogen receptor-independent mechanisms. Because medroxyprogesterone (MPA) is a substrate for CYP450s, we hypothesized that MPA may abrogate the inhibitory effects of E by competing for CYP450s and inhibiting the formation of 2/4-hydroxyestradiols and MEs. To test this hypothesis, we investigated the effects of E on SMC number, DNA and collagen synthesis, and migr

Synthesis of 2-aryl/alkylaminomethyl 17-sulfamate/17-methyl estratrienes and their in vitro cytotoxicity in human cancer cell cultures.

2-Methoxyestradiol (2-ME), a natural metabolite of 17β-estradiol, exhibits potent anticancer activity by inhibiting tubulin polymerization and modulating steroid sulfatase (STS). However, its rapid metabolism limits its clinical usefulness. To overcome these issues, we designed a new series of 2-aryl/alkylaminomethyl estratriene analogues with sulfamate groups at C‑3 and/or C‑17, serving as hybrid agents that disrupt microtubules and modulate STS, derived from the 2-ME pharmacophore. We used a stepwise transformation of estrone to access C‑2 aminomethyl frameworks, followed by regioselective s

NMR-based plasma metabolic profiling in patients with unstable angina.

OBJECTIVES: Unstable angina (UA) is a form of the acute coronary syndrome (ACS) that affects more than a third of the population before age 70. Due to the limitations of diagnostic tests, appropriate identification of UA is difficult. In this study, we proceeded to investigate metabolite profiling in UA patients compared with controls to determine potential candidate biomarkers. MATERIALS AND METHODS: Ninety-four plasma samples from UA and 32 samples from controls were analyzed based on 1H NMR spectroscopy. The raw data were processed, analyzed, and subjected to partial least squares-discrimi

Ubiquitin-specific protease 13 regulates FcεRI-mediated mast cell activation and allergic inflammation via SYK protein modulation.

Mast cells (MCs) are therapeutic targets for high-affinity IgE Fc receptors (FcεRI)-mediated allergic responses. Deubiquitinating enzymes (DUBs), including ubiquitin-specific protease 13 (USP13), are involved in multiple inflammatory processes. This study aims to reveal USP13's role in FcεRI-mediated MC activation and its underlying mechanisms. Our results showed USP10/13 inhibitor spautin-1 inhibited IgE-mediated MC activation, as evidenced by a reduction in the release of β-hexosaminidase (β-hex) and histamine and decreased expression and secretion of inflammatory cytokines. Spautin-1 also a

Simultaneous determination of 18 steroids in the hypothalamic pituitary gonadal axis based on UPLC-MS/MS with multimode ionization.

To objectively quantify changes in steroid hormones in organisms caused by adverse environmental loads, we developed a simple and sensitive UPLC-MS/MS (ultra-performance liquid chromatography triple quadrupole mass spectrometry) method for the simultaneous determination of 18 steroid hormones on the HPG axis. This analytical method was based on liquid extraction and a multimode electrospray and atmospheric pressure chemical ionization (ESCi) source, which was optimized by mass spectrometry, liquid phase and pretreatment for the quantification of cholesterol (CH), aldosterone (A), cortisone (E)

HIF1α Signaling in the Endogenous Protective Responses after Neonatal Brain Hypoxia-Ischemia.

Hypoxia-inducible factor 1α (HIF1α) is a key regulator of oxygen homeostasis, and its target genes mediate adaptive, protective, and pathological processes. The role of HIF1α in neuronal survival is controversial and the brain maturation stage is important in determining its function in brain ischemia or hypoxia-ischemia (HI). In this study, we used neuron-specific HIF1α knockout mice at postnatal day 9 (P9), and immature cortical neurons (days 7-8 in vitro) treated with the HIF1α inhibitor 2-methoxyestradiol (2ME2) or stabilizer dimethyloxalylglycine (DMOG), to examine the function of neurona

The Role of Sex in the Pathophysiology of Pulmonary Hypertension.

Pulmonary arterial hypertension (PAH) is a progressive disease characterised by increased pulmonary vascular resistance and pulmonary artery remodelling as result of increased vascular tone and vascular cell proliferation, respectively. Eventually, this leads to right heart failure. Heritable PAH is caused by a mutation in the bone morphogenetic protein receptor-II (BMPR-II). Female susceptibility to PAH has been known for some time, and most recent figures show a female-to-male ratio of 4:1. Variations in the female sex hormone estrogen and estrogen metabolism modify FPAH risk, and penetrance
