Animal models with enhanced erythropoiesis and iron absorption.

The regulation of iron absorption is of considerable interest in mammals since excretion is minimal. Recent advances in iron metabolism have expounded the molecular mechanisms by which iron absorption is attuned to the physiological demands of the body. The pinnacle was the discovery and identification of hepcidin, a hepatic antimicrobial peptide that regulates absorption to maintain iron homeostasis. While the intricacies of its expression and regulation by HFE, transferrin receptor 2 and hemojuvelin are still speculative, hepcidin responsiveness has correlated negatively with iron absorption

Physicochemical characteristics and gastrointestinal absorption behaviors of S-propargyl-cysteine, a potential new drug candidate for cardiovascular protection and antitumor treatment.

1. As a potential new drug candidate for cardiovascular protection and antitumor treatment, the physicochemical properties, gastrointestinal (GI) absorption behaviors and mechanisms of S-propargyl-cysteine (SPRC) were investigated in this study. 2. SPRC exhibited favorable solubility in aqueous media. The log P and log D values were low (≤1.93 ± 0.08). The pKa in the acidic and basic regions was 2.08 ± 0.02 and 8.72 ± 0.03, respectively. The isoelectric point was 5.40 ± 0.02. SPRC was stable in the rat GI fluids, and showed no obvious adsorption and metabolism in the rat GI tract. 3. SPRC disp

Understanding Mechanisms of Food Effect and Developing Reliable PBPK Models Using a Middle-out Approach.

Over the last 10 years, 40% of approved oral drugs exhibited a significant effect of food on their pharmacokinetics (PK) and currently the only method to characterize the effect of food on drug absorption, which is recognized by the authorities, is to conduct a clinical evaluation. Within the pharmaceutical industry, there is a significant effort to predict the mechanism and clinical relevance of a food effect. Physiologically based pharmacokinetic (PBPK) models combining both drug-specific and physiology-specific data have been used to predict the effect of food on absorption and to reveal th

Pharmacokinetics of antimicrobial agents in the elderly.

Many elderly individuals are treated with antibiotics, but knowledge of the pharmacokinetic performance of the compounds in these patients is limited. Advanced age is accompanied by many physiologic alterations that may have secondary effects on absorption, distribution, metabolism, and excretion of drugs. Information on absorption and metabolism in the elderly is scarce and conflicting. Because renal excretion of penicillins and cephalosporins decreases with the physiologic signs of the kidneys, dose reduction is advisable in elderly patients. Monitoring of serum drug levels is recommended fo

Challenges in nasal drug absorption: how far have we come?

The nasal route is commonly used for local delivery of drugs to treat inflammatory conditions. It is also an attractive route for systemic delivery of some drugs. Irrespective of intended use, administered drugs must permeate the epithelial or olfactory membrane to be effective. The enthusiasm for potential use of the nasal route for systemic drug delivery has not been met by comparable success. In this paper, the anatomical and physiological attributes of the nasal cavity and paranasal sinuses important for drug delivery and challenges limiting drug absorption are discussed. Efforts made so f

Influence of rhein on rat colonic Na+,K+-ATPase and permeability in vitro.

Rhein, an aglucone of a laxative-acting anthraquinone, which induces net secretion in the human jejunum and colon, does not induce net secretion under open circuit current in in vitro conditions but reduces net absorption of electrolytes and water. In the stripped rat descending colon at a concentration of 2.10(-4) mol/l, the Na+, K+-ATPase is almost completely inhibited resulting in a decreased net absorption of electrolytes and water. There are no changes of the diffusional resistance of the paracellular pathway. Results point to a cellular mechanism of rhein by which absorption is inhibited

Mechanisms and regulation of epithelial Ca2+ absorption in health and disease.

Ca2+ is essential for numerous physiological functions in our bodies. Therefore, its homeostasis is finely maintained through the coordination of intestinal absorption, renal reabsorption, and bone resorption. The Ca2+-selective epithelial channels TRPV5 and TRPV6 have been identified, and their physiological roles have been revealed: TRPV5 is important in final renal Ca2+ reabsorption, and TRPV6 has a key role in intestinal Ca2+ absorption. The TRPV5 knockout mice exhibit renal leak hypercalciuria and accordingly upregulate their intestinal TRPV6 expression to compensate for their negative Ca

An in vivo study of the gastrointestinal absorption site for zinc chloride in mice.

The experimental model presented below enables quantitation of the uptake of zinc (Zn++) into gastrointestinal mucosal cells in vivo using gamma-counting of 65Zn. Experiments were performed in mice fed their normal diet under natural physiological conditions. The in vivo site(s) of significant zinc absorption may thereby be identified. Absorption of zinc was extensive during the first hour after administration of a single oral dose of ZnCl2. Apparently, absorption continued during at least eight hours postdosage, and probably continued for 48 hours. The intestinal mucosal labelling profile for

Glucose absorption in small intestinal diseases.

Recent developments in the field of diabetes and obesity management have established the central role of the gut in glucose homeostasis; not only is the gut the primary absorptive site, but it also triggers neurohumoral feedback responses that regulate the pre- and post-absorptive phases of glucose metabolism. Structural and/or functional disorders of the intestine have the capacity to enhance (e.g.: diabetes) or inhibit (e.g.: short-gut syndrome, critical illness) glucose absorption, with potentially detrimental outcomes. In this review, we first describe the normal physiology of glucose abso

Factors Affecting the Absorption of Subcutaneously Administered Insulin: Effect on Variability.

Variability in the effect of subcutaneously administered insulin represents a major challenge in insulin therapy where precise dosing is required in order to achieve targeted glucose levels. Since this variability is largely influenced by the absorption of insulin, a deeper understanding of the factors affecting the absorption of insulin from the subcutaneous tissue is necessary in order to improve glycaemic control and the long-term prognosis in people with diabetes. These factors can be related to either the insulin preparation, the injection site/patient, or the injection technique. This re

Intranasal delivery of insulin: An update on status quo and challenges for diabetes treatment.

Diabetes mellitus is a chronic metabolic disorder characterised by either autoimmune-mediated destruction of pancreatic β-cells or impaired physiological responses to circulating insulin. Conventional subcutaneous insulin administration does not achieve optimal pharmacokinetics due to delayed systemic absorption and its inability to replicate the prandial, pulsatile secretion pattern of endogenous insulin. In contrast, intranasal insulin delivery offers a promising non-invasive alternative, with a pharmacokinetic profile that more closely mirrors physiological insulin release following meals,

Multi-phase multi-layer mechanistic dermal absorption (MPML MechDermA) model to predict local and systemic exposure of drug products applied on skin.

Physiologically-based pharmacokinetic models combine knowledge about physiology, drug product properties, such as physicochemical parameters, absorption, distribution, metabolism, excretion characteristics, formulation attributes, and trial design or dosing regimen to mechanistically simulate drug pharmacokinetics (PK). The current work describes the development of a multiphase, multilayer mechanistic dermal absorption (MPML MechDermA) model within the Simcyp Simulator capable of simulating uptake and permeation of drugs through human skin following application of drug products to the skin. Zonulin, regulation of tight junctions, and autoimmune diseases.

Recent studies indicate that besides digestion and absorption of nutrients and water and electrolytes homeostasis, another key function of the intestine is to regulate the trafficking of environmental antigens across the host mucosal barrier. Intestinal tight junctions (TJs) create gradients for the optimal absorption and transport of nutrients and control the balance between tolerance and immunity to nonself antigens. To meet diverse physiological challenges, intestinal epithelial TJs must be modified rapidly and in a coordinated fashion by regulatory systems that orchestrate the state of ass

Modulation of jejunal ion and water absorption by endogenous angiotensin after dehydration.

In the pentobarbital sodium-anesthetized rat, dehydration for 24 h increased ion and water absorption from the jejunum. Dehydration also elevated plasma concentrations of angiotensin peptides and plasma renin activity but did not significantly alter plasma aldosterone concentrations. Infusion of tyramine, norepinephrine, and angiotensin II (AII) also stimulated jejunal absorption in a manner similar to dehydration. The elevation of jejunal absorption in response to dehydration is totally inhibited by the converting enzyme inhibitor captopril and the angiotensin receptor antagonist [lle7]AIII.

pH-Responsive Glycosylated PC Nanoparticles for Oral Insulin Delivery.

Oral insulin delivery represents a compelling alternative to conventional injections because of its enhanced patient compliance and administration convenience. However, this approach faces substantial challenges stemming from physiological barriers that include the gastrointestinal environment, mucus layer obstruction, and poor intestinal absorption. To overcome these limitations, we designed glycosylated proanthocyanidin (PC) nanoparticles using a sacrificial ZIF-8 template for insulin encapsulation. The resulting nanoparticles achieved high loading efficiency and combined two critical functi

A cycle of cerebrospinal fluid: supporting evidence and theoretical considerations.

An entrenched theory of cerebrospinal fluid (CSF) absorption by the arachnoid villi fails to explain observations regarding the movement of substances in the fluid. Experiments that demonstrated the arachnoid villi as the site of absorption were based on non-physiologic methods. CSF does not flow through the arachnoid villi, because villi require bulk flow and bulk flow of CSF does not exist. CSF is transported through the choroid fissure and recycled through the tela choroidae by the choroid plexus, with reentry into the ventricular system. Observed failures of the effete arachnoid villus the

Study of Magnesium Formulations on Intestinal Cells to Influence Myometrium Cell Relaxation.

Background: Magnesium is involved in a wide variety of physiological processes including direct relaxation of smooth muscle. A magnesium imbalance can be considered the primary cause or consequence of many pathophysiological conditions. The smooth muscle tissue of the uterus, i.e., the myometrium, undergoes numerous physiological changes during life, fundamental for uterine activities, and it receives proven benefits from magnesium supplementation. However, magnesium supplements have poor absorption and bioavailability. Furthermore, no data are available on the direct interaction between intes
