Novel insights into anti-inflammatory activity-related metabolic markers and biosynthetic regulatory mechanisms between principal and lateral roots of Aconitum carmichaelii Debx. via integrated metabolomics and transcriptomics.

BACKGROUND: The principal roots (CHW) and lateral roots (FZ) of Aconitum carmichaelii Debx. are widely used medicinal herbs in Asia. Although the two roots are derived from the same plant, they exhibit differences in anti-inflammatory activity (AIA). In the market, small FZ is frequently passed off as CHW. Therefore, revealing AIA-related markers and biosynthetic regulatory mechanisms between CHW and FZ are critical for the quality control of these medicinal materials. OBJECTIVE: To identify AIA-related markers and reveal the biosynthetic regulatory mechanisms of these markers. METHODS: A "m

Studies on the stability of diester-diterpenoid alkaloids from the genus Aconitum L. by high performance liquid chromatography combined with electrospray ionisation tandem mass spectrometry (HPLC/ESI/MSn).

The stability of diester-diterpenoid alkaloids (DDA) from plants of the genus Aconitum L. has been studied in different solvents and pH buffers. The HPLC/ESIMS method for analysing the concentration of DDA was established and DDA's decomposition products were elucidated by HPLC/ESI-MS/MS(n). In different solvents, e.g. dichloromethane, ether, methanol and distilled water, the decomposition pathways of DDA are quite different and their difference in stabilities depends on the difference of their structures, in which substituents at the N atom and substituents at C-3 are different. The pyrolytic

Integration of offline two-dimensional chromatography and mass defect filtering-based precursor ion list data acquisition for targeted characterization of diterpenoid alkaloids in the lateral roots of Aconitum carmichaelii.

The hyphenated technique of offline two-dimensional (2D) chromatography with high resolution mass spectrometry (MS) was an efficient tool for separation and characterization of components in complex systems such as herbal medicines, especially those co-eluting components or isomers. In this study, we constructed the ultra-performance convergence chromatography (UPC2) × reversed phase (RP) chromatographic separation system and developed a mass defect filtering (MDF)-based precursor ion list (PIL) acquisition method to improve the selectivity and sensitivity of this technique, and the systematic

Toxicological investigation of 25 aconitine-induced deaths from 2005 to 2023.

Aconitum herbs contain several highly toxic diester-diterpenoid alkaloids, including aconitine, mesaconitine, and hypaconitine. However, finding the cause of death is rather difficult for forensic pathologists during forensic autopsy of aconitine-induced death. Therefore, the ability to determine Aconitum alkaloids is important in these cases. The aim of this study was to review the data for alkaloids in postmortem specimens from 25 aconitine-induced deaths received by the Academy of Forensic Science from 2005 to 2023. Aconitum alkaloids were analyzed using an LC-MS/MS method, which was valida

A case of fatal poisoning with the aconite plant: quantitative analysis in biological fluid.

In recent years recorded cases of plant poisoning have become rare, this may in part be due to the possibility of plant ingestion not being indicated at the beginning of an investigation. Aconitum napellus (aconite, Wolfsbane, Monkshood) is one of the most poisonous plants in the UK. It contains various potent alkaloids such as aconitine, isoaconitine, lycaconitine and napelline. Ingestion of Aconitum plant extracts can result in severe, potentially fatal toxic effects. This paper describes the analytical findings in a recent death in the UK. resulting from deliberate ingestion of Aconitum nap

Aconitine accidental poisoning: two case reports and literature review.

INTRODUCTION: Chinese Herbal Medicine (CHM) is widely integrated into clinical practice across Asia and other regions for disease treatment and health maintenance. Aconitine, a potent diterpenoid alkaloid derived from plants of the Aconitum genus (e.g., Aconitum carmichaelii and Aconitum kusnezoffii), exhibits significant pharmacological activity but also considerable toxicity. While properly processed Aconitum herbs are prescribed for their analgesic and anti-rheumatic properties, inadequate detoxification during processing or improper clinical use can lead to accidental aconitine poisoning.

UPLC-qToF-MSE Metabolomics to Investigate Century-Old Botanicals: A Case Study of Medicinal Aconite.

Despite their importance in botanical research, many herbaria worldwide have closed due to high maintenance costs and limited funding. These collections are especially valuable today, in the era of plant metabolomics, as they allow access to species that are typically inaccessible or otherwise difficult to recollect, enabling analysis through both untargeted and supervised methods. This study highlights this potential by analyzing historic samples of medicinal Aconitum from the New York Botanical Garden Rusby Collection. We hypothesized that applying modern LC-MS metabolomic techniques to hist
