[Effect of dielectric permeability and temperature of culture media on inhibitory action of eosin Y on actomyosine ATPase of the uterine smooth muscles].

Influence of dielectric permeability and incubation medium temperature on sensitivity of ATPase of contractile protein complex to eosin Y was studied on preparations of uterine smooth muscle actomyosin. It was shown, that a decrease of dielectric permeability from 74.1 to 68.8 is accompanied by inhibition of activity of actomyosin ATPase by 30%. Administration of eosin Y to the incubation medium intensifies the inhibition of activity of actomyosin ATPase, that correlates with a decrease of dielectric permeability and increase of eosin Y concentration. Catalytic titration of actomyosin ATPase b

Expressing functional domains of mouse calponin: involvement of the region around alanine 145 in the actomyosin ATPase inhibitory activity of calponin.

Previously, we attributed the binding of F-actin to the 38-residue stretch of gizzard calponin encompassing the sequence A145-Y182 and postulated the hexapeptide motif VKYAEK, representing residues 142-147, as a putative actin-binding site [Mezgueldi, M., Fattoum, A., Derancourt, J. & Kassab, R. (1992) J. Biol. Chem. 267, 15943-15951]. Herein, the nature of the ATPase inhibitory amino acids of calponin and their relative position within the actin binding domain was investigated by expressing the following fragments of mouse calponin with or without substitution or deletion of the hexapeptide V

Repression of Wasp by JAK/STAT signalling inhibits medial actomyosin network assembly and apical cell constriction in intercalating epithelial cells.

Tissue morphogenesis requires stereotyped cell shape changes, such as apical cell constriction in the mesoderm and cell intercalation in the ventrolateral ectoderm of Drosophila. Both processes require force generation by an actomyosin network. The subcellular localization of Myosin-II (Myo-II) dictates these different morphogenetic processes. In the intercalating ectoderm Myo-II is mostly cortical, but in the mesoderm Myo-II is concentrated in a medial meshwork. We report that apical constriction is repressed by JAK/STAT signalling in the lateral ectoderm independently of Twist. Inactivation

Effects of temperature, adenosine triphosphate and magnesium concentrations on the contraction of actomyosin isolated from halothane-sensitive and -insensitive German Landrace pigs.

The effects of temperature, adenosine triphosphate and magnesium on the calcium sensitivity of actomyosin were investigated on actomyosin obtained from six halothane-sensitive and five halothane-resistant German Landrace pigs. No difference in the contractile properties was found in actomyosin from the two types of pig. However, in both types calcium sensitivity of the actomyosin was lost at temperatures slightly greater than those occurring physiologically. Both ATP and Mg2+ protect against the loss of calcium sensitivity. These results suggest that the basic lesion in halothane-induced malig

Protein kinase C-mediated phosphorylation of troponin I and C-protein in isolated myocardial cells is associated with inhibition of myofibrillar actomyosin MgATPase.

Phosphorylation of cardiac myofibrillar proteins by protein kinase C (PKC) in isolated adult rat cardiomyocytes has been compared with that mediated by the cAMP-dependent protein kinase (PKA). PKA activation by beta-adrenoreceptor (isoproterenol) stimulation results in stoichiometric phosphorylation of troponin I (TnI) and C-protein. PKC activation by either 12-O-tetradecanoylphorbol-13-acetate (TPA) or by alpha-adrenoreceptor (phenylephrine plus propranolol) stimulation results in phosphorylation of the same two proteins to similar extents. Two-dimensional phosphopeptide mapping shows that th

DEVELOPMENTALLY REGULATED PLASMA MEMBRANE PROTEIN of Nicotiana benthamiana contributes to potyvirus movement and transports to plasmodesmata via the early secretory pathway and the actomyosin system.

The intercellular movement of plant viruses requires both viral and host proteins. Previous studies have demonstrated that the frame-shift protein P3N-PIPO (for the protein encoded by the open reading frame [ORF] containing 5'-terminus of P3 and a +2 frame-shift ORF called Pretty Interesting Potyviridae ORF and embedded in the P3) and CYLINDRICAL INCLUSION (CI) proteins were required for potyvirus cell-to-cell movement. Here, we provide genetic evidence showing that a Tobacco vein banding mosaic virus (TVBMV; genus Potyvirus) mutant carrying a truncated PIPO domain of 58 amino acid residues co

β-catenin-α-catenin and actomyosin signaling differentially regulate growth cone contours and axon undulation and branching of retinal ganglion cells in situ.

INTRODUCTION: Cadherin adhesive and actomyosin signaling are key cytomechanical cues required for neuronal circuit formation, but whether they function together to sculpt developing neurons is not known. Previously, we demonstrated that a β-catenin mutant (β-catNTERM) that disrupts binding of endogenous, full length β-catenin to α-catenin in the Cadherin adhesion complex, and a pharmacological inhibitor for actin regulator, non-muscle Myosin II (Blebbistatin), resulted in growth cones with fewer and more filopodia or filopodia-like protrusions than control growth cones of retinal ganglion cell

Cardiomyocyte binucleation is associated with aberrant mitotic microtubule distribution, mislocalization of RhoA and IQGAP3, as well as defective actomyosin ring anchorage and cleavage furrow ingression.

AIMS: After birth mammalian cardiomyocytes initiate a last cell cycle which results in binucleation due to cytokinesis failure. Despite its importance for cardiac regenerative therapies, this process is poorly understood. Here, we aimed at a better understanding of the difference between cardiomyocyte proliferation and binucleation and providing a new tool to distinguish these two processes. METHODS AND RESULTS: Monitoring of cell division by time-lapse imaging revealed that rat cardiomyocyte binucleation stems from a failure to properly ingress the cleavage furrow. Astral microtubule require

Molecular movements in the actomyosin complex: F-actin-promoted internal cross-linking of the 25- and 20-kDa heavy chain fragments of skeletal myosin subfragment.

We describe, for the first time, the F-actin-promoted changes in the spatial relationship of strands in the NH2-terminal 25-kDa and COOH-terminal 20-kDa heavy chain fragments of the skeletal myosin subfragment 1 (S-1), detected by their exclusive chemical cross-linking in the rigor F-actin-S-1 complex with m-maleimidobenzoic acid N-hydroxysuccinimide ester (MBS). Quantitative electrophoretic analysis of the reaction products showed extensive conversion of the 95-kDa heavy chain of the actin-bound S-1 into a new species with an apparent mass of 135 kDa (yield = 50-60%), whereas the heavy chain

Controlling Intracellular Machinery via Polymer Pen Lithography Molecular Patterning.

The plasma membrane and the actomyosin cytoskeleton play key roles in controlling how cells sense and interact with their surrounding environment. Myosin, a force-generating actin network-associated protein, is a major regulator of plasma membrane tension, which helps control endocytosis. Despite the important link between plasma membranes and actomyosin (the actin-myosin complex), little is known about how the actomyosin arrangement regulates endocytosis. Here, nanoscopic ligand arrangements defined by polymer pen lithography (PPL) are used to control actomyosin contractility and examine cell

Actin and myosin dynamics during epithelial remodeling in avian gastrulation.

Epithelial remodeling is powered by contractile forces exerted by the actomyosin cytoskeleton. In invertebrates, pulsatile contractile flows of the medio-apical actomyosin cortex have been shown to be crucial in promoting junction contraction, ultimately driving cell rearrangements, apical constriction and cell extrusion. However, how actomyosin dynamics drive epithelial remodeling in vertebrates, especially amniotes, remains poorly understood. In this study, we generated transgenic quail lines reporting actin and myosin, and investigated their dynamics in gastrulating embryos. We show that, d

Reprint of: Comparative Studies of Rabbit Cardiac and Skeletal Myosins.

Rabbit cardiac myosin contains fewer cysteine residues than the skeletal myosin (7 and 8.8 moles/105 gm. of myosin, respectively). A similar difference is found between the cysteine content of rabbit cardiac and skeletal heavy meromyosins; the cardiac heavy meromyosin contains 8.9 moles/105 gm. of protein as compared to 11 moles in the skeletal heavy meromyosin. In skeletal myosin, actomyosin, and myofibrils the Ca++-ATPase, Ca++-ITPase, and EDTA-ATPase activities are about three times higher than those of cardiac myosin, actomyosin, and myofibrils; whereas the skeletal to cardiac actomyosin-A

Nonmuscle myosin II regulates presynaptic actin and neuronal mechanobiology in Drosophila.

Neuromuscular junctions (NMJs) are evolutionarily ancient, specialized contacts between neurons and muscles. They experience lifelong strain, yet the mechanism preserving their integrity under mechanical load remains unclear. Here, we identify a novel actomyosin structure at Drosophila larval NMJs, consisting of a long-lived, low-turnover presynaptic actin core that colocalizes with nonmuscle myosin II (NMII) and becomes disorganized upon manipulating neuronal NMII levels or activity. Intriguingly, neuronal NMII depletion altered postsynaptic NMII levels and organization near synapses, suggest

Scar/WAVE has Rac GTPase-independent functions during cell wound repair.

Rho family GTPases regulate both linear and branched actin dynamics by activating downstream effectors to facilitate the assembly and function of complex cellular structures such as lamellipodia and contractile actomyosin rings. Wiskott-Aldrich Syndrome (WAS) family proteins are downstream effectors of Rho family GTPases that usually function in a one-to-one correspondence to regulate branched actin nucleation. In particular, the WAS protein Scar/WAVE has been shown to exhibit one-to-one correspondence with Rac GTPase. Here we show that Rac and SCAR are recruited to cell wounds in the Drosophi

Consequences of impeding in mitochondrial function in Physarum polycephalum. I. Reversible effects of anoxia, KCN, and influences of the Ca2+ ionophore A-23187.

5 mM KCN as well as anoxia have corresponding effects on Physarum plasmodia: Both induce a disintegration of the plasmalemma invaginations, an increase in cytoplasmic vacuoles, formation of a thick cortical actomyosin layer and an increase of cytoplasmic actomyosin fibrils. Both KCN treatment and anoxia cause a temporary increase in the level of the force oscillations and a reversible prolongation of the periods of the contraction-relaxation cycle of cytoplasmic actomyosin. The normal pattern of oscillation can be restored by the addition of 10 mM alpha-ketoglutarate +5 mM AMP to the solution

Tensions on the actin cytoskeleton and apical cell junctions in the C. elegans spermatheca are influenced by spermathecal anatomy, ovulation state and activation of myosin.

Cells generate mechanical forces mainly through myosin motor activity on the actin cytoskeleton. In C. elegans, actomyosin stress fibers drive contractility of the smooth muscle-like cells of the spermatheca, a distensible, tube-shaped tissue in the hermaphrodite reproductive system and the site of oocyte fertilization. Stretching of the spermathecal cells by oocyte entry triggers activation of the small GTPase Rho. In this study, we asked how forces are distributed in vivo using the spermatheca, and explored how this tissue responds to alterations in myosin activity. Using laser ablation, we

Structural and protein interaction effects of hypertrophic and dilated cardiomyopathic mutations in alpha-tropomyosin.

The potential alterations to structure and associations with thin filament proteins caused by the dilated cardiomyopathy (DCM) associated tropomyosin (Tm) mutants E40K and E54K, and the hypertrophic cardiomyopathy (HCM) associated Tm mutants E62Q and L185R, were investigated. In order to ascertain what the cause of the known functional effects may be, structural and protein-protein interaction studies were conducted utilizing actomyosin ATPase activity measurements and spectroscopy. In actomyosin ATPase measurements, both HCM mutants and the DCM mutant E54K caused increases in Ca(2+)-induced m

Extracellular hyaluronate pressure shaped by cellular tethers drives tissue morphogenesis.

How tissues acquire complex shapes is a fundamental question in biology and regenerative medicine. Zebrafish semicircular canals form from invaginations in the otic epithelium (buds) that extend and fuse to form the hubs of each canal. We find that conventional actomyosin-driven behaviors are not required. Instead, local secretion of hyaluronan, made by the enzymes uridine 5'-diphosphate dehydrogenase (ugdh) and hyaluronan synthase 3 (has3), drives canal morphogenesis. Charged hyaluronate polymers osmotically swell with water and generate isotropic extracellular pressure to deform the overlyin

A multicellular star-shaped actin network underpins epithelial organization and connectivity.

Epithelial tissues withstand external stresses while maintaining structural stability. Bicellular junctions and the actomyosin network support epithelial integrity, packing and remodelling. While their role in development and disease are well studied, their synergistic impact on maintaining tissue organization remains unclear. Here, we identify a tissue-scale actomyosin network in adult murine intestinal villi, as well as in an ex vivo organoid-based epithelium model. This actomyosin network consists of repeated units of actin stars - radial actin structures at the base of hexagonal cells - li
