Maillard reaction products inhibit the periodontal pathogen Aggregatibacter actinomycetemcomitans by chelating iron.

OBJECTIVE: To determine the mechanism of growth inhibition of Aggregatibacter actinomycetemcomitans by Maillard reaction products (MRP). DESIGN: Growth and cell viabilities in the presence or absence of MRP were measured for both the rough and smooth variants of the bacteria. Effects of addition of ferrous and ferric ions on the inhibition of the bacteria by MRP were determined. RESULTS: MRPs decreased the extent of complex formation of Chrome Azurol S with iron suggesting that MRPs can chelate iron effectively. The chelation causes growth inhibition of both the rough and smooth strains. At

Aggregatibacter actinomycetemcomitans-induced hypercitrullination links periodontal infection to autoimmunity in rheumatoid arthritis.

A bacterial etiology of rheumatoid arthritis (RA) has been suspected since the beginnings of modern germ theory. Recent studies implicate mucosal surfaces as sites of disease initiation. The common occurrence of periodontal dysbiosis in RA suggests that oral pathogens may trigger the production of disease-specific autoantibodies and arthritis in susceptible individuals. We used mass spectrometry to define the microbial composition and antigenic repertoire of gingival crevicular fluid in patients with periodontal disease and healthy controls. Periodontitis was characterized by the presence of c

Signaling transduction analysis in gingival epithelial cells after infection with Aggregatibacter actinomycetemcomitans.

Periodontal diseases result from the interaction of bacterial pathogens with the host's gingival tissue. Gingival epithelial cells are constantly challenged by microbial cells and respond by altering their transcription profiles, inducing the production of inflammatory mediators. Different transcription profiles are induced by oral bacteria and little is known about how the gingival epithelium responds after interaction with the periodontopathogenic organism Aggregatibacter actinomycetemcomitans. In the present study, we examined the transcription of genes involved in signaling transduction pa

Localized Periodontitis in Young Individuals: Aggregatibacter JP2 Clone, Immunological Dysfunctions and Other Stories.

Grade C molar-incisor pattern periodontitis (C-MIP) is the latest in a series of names given to a unique phenotype, characterized by an aggressive and rapid loss of tooth-supporting structures, starting early in life and usually affecting first molars and incisors. Although much less prevalent than the more generalized "chronic" forms of this disease in adults, it affects mostly young individuals of lower socioeconomic status and African descent, or from mixed-race populations, and shows a strong familial aggregation, pointing to a possible genetic contribution, yet not fully elucidated. Even

Pott's puffy tumour based on Aggregatibacter aphrophilus in an adult patient with bilateral cheilognathopalatoschisis.

Pott's puffy tumour (PPT) is a rare condition defined by subperiosteal abscess formation following osteomyelitis of the frontal bone. Complications can be severe and involve intracranial progression. Here, we present a rare case of a PPT due to an Aggregatibacter aphrophilus infection of odontogenic origin in a young adult patient with a history of bilateral cheilognathopalatoschisis. Due to the unusual location of the abscess, the diagnosis was initially missed, leading to a delay of more than 1 month until adequate treatment. Odontogenic treatment was initiated as source control, external dra

Interspecies Interactions of Single- and Mixed-Species Biofilms of Candida albicans and Aggregatibacter actinomycetemcomitans.

Polymicrobial biofilms involving fungal and bacterial species are increasingly recognized as contributors to persistent infections, particularly in the oral cavity. Candida albicans and Aggregatibacter actinomycetemcomitans are two commensals that can turn into opportunistic pathogens and are able to form robust biofilms. Objectives: This study aimed to assess the interaction dynamics between these two microorganisms and to evaluate their susceptibility to fluconazole and azithromycin in single- and mixed-species forms. Methods: Biofilm biomass was quantified using crystal violet assays, while

The Hunger Games: Aggregatibacter actinomycetemcomitans Exploits Human Neutrophils As an Epinephrine Source for Survival.

Aggregatibacter actinomycetemcomitans is a gram-negative facultative anaerobe and an opportunistic oral pathogen, strongly associated with periodontitis and other inflammatory diseases. Periodontitis is a chronic inflammation of the periodontium resulting from the inflammatory response of the host towards the dysbiotic microbial community present at the gingival crevice. Previously, our group identified catecholamines and iron as the signals that activate the QseBC two-component system in A. actinomycetemcomitans, necessary for the organism to acquire iron as a nutrient to survive in the anaer
