Enzymatic and Mutational Analysis of the PruA Pteridine Reductase Required for Pterin-Dependent Control of Biofilm Formation in Agrobacterium tumefaciens.

Pterins are ubiquitous biomolecules with diverse functions including roles as cofactors, pigments, and redox mediators. Recently, a novel pterin-dependent signaling pathway that controls biofilm formation was identified in the plant pathogen, Agrobacterium tumefaciens A key player in this pathway is a pteridine reductase termed PruA, where its enzymatic activity has been shown to control surface attachment and limit biofilm formation. Here, we biochemically characterize PruA to investigate the catalytic properties and substrate specificity of this pteridine reductase. PruA demonstrates maximal

Effect of far-red light exposure on photosynthesis and photoprotection in tomato plants transgenic for the Agrobacterium rhizogenes rolB gene.

Previous work showed in tomato plants harbouring the Agrobacterium rhizogenes rolB gene overexpression of genes involved in chloroplast function and stress response, significant increase in non-photochemical quenching and chlorophyll a and b content, and reduced chlorophyll a/b ratio. The latter condition being typical of plant shade where far-red is dominant, suggested a role for rolB in improving photosynthesis in such condition. To gain a better insight into these results, the photosynthetic performance of transgenic and control plants was compared by means of variable fluorescence kinetics

The Brucella suis homologue of the Agrobacterium tumefaciens chromosomal virulence operon chvE is essential for sugar utilization but not for survival in macrophages.

Brucella strains possess an operon encoding type IV secretion machinery very similar to that coded by the Agrobacterium tumefaciens virB operon. Here we describe cloning of the Brucella suis homologue of the chvE-gguA-gguB operon of A. tumefaciens and characterize the sugar binding protein ChvE (78% identity), which in A. tumefaciens is involved in virulence gene expression. B. suis chvE is upstream of the putative sugar transporter-encoding genes gguA and gguB, also present in A. tumefaciens, but not adjacent to that of a LysR-type transcription regulator. Although results of Southern hybridi

Identification of sequences involved in the polyadenylation of higher plant nuclear transcripts using Agrobacterium T-DNA genes as models.

Sequences in the 3'-untranslated region of two different octopine T-DNA genes were analyzed with regard to their significance in polyadenylation. Poly(A) addition sites were localized precisely by S1 nuclease mapping with T-DNA-derived mRNAs isolated from tobacco. The gene encoding transcript 7' contains two AATAAA hexanucleotides, respectively 119 bp and 170 bp downstream of the TAA stop codon. A single poly(A) site was mapped 24-25 bp downstream of the first AATAAA. Further, we show that a mutant octopine synthase gene, which has lost part of its 3'-untranslated region by deletion, is still

Soybean Embryonic Axis Transformation: Combining Biolistic and Agrobacterium-Mediated Protocols to Overcome Typical Complications of In Vitro Plant Regeneration.

The first successful attempt to generate genetically modified plants expressing a transgene was preformed via T-DNA-based gene transfer employing Agrobacterium tumefaciens-mediated genetic transformation. Limitations over infectivity and in vitro tissue culture led to the development of other DNA delivery systems, such as the biolistic method. Herein, we developed a new one-step protocol for transgenic soybean recovery by combining the two different transformation methods. This protocol comprises the following steps: agrobacterial preparation, seed sterilization, soybean embryo excision, shoot

Efficient Embryogenic Callus Induction and Agrobacterium-Mediated Transformation of the Elite Foxtail Millet (Setaria italica L.) Variety Jingu51.

Foxtail millet (Setaria italica L.) is an important food crop worldwide and serves as a model for studying C4 photosynthesis and drought resistance. However, its application in functional genomics and molecular breeding has been severely constrained by the absence of a reliable genetic transformation system. In this study, 66 varieties were systematically evaluated for embryogenic callus induction capacity, with rates ranging from 0 to 70.22%. Among these, five varieties exhibited high embryogenic potential (>50%). Jingu51, an elite cultivated variety in northern China, was selected as the tra

An Efficient Agrobacterium-Mediated Transformation Method for Hybrid Poplar 84K (Populus alba × P. glandulosa) Using Calli as Explants.

A highly efficient Agrobacterium-mediated transformation method is needed for the molecular study of model tree species such as hybrid poplar 84K (Populus alba × P. glandulosa cv. '84K'). In this study, we report a callus-based transformation method that exhibits high efficiency and reproducibility. The optimized callus induction medium (CIM1) induced the development of calli from leaves with high efficiency, and multiple shoots were induced from calli growing on the optimized shoot induction medium (SIM1). Factors affecting the transformation frequency of calli were optimized as follows: Agro

MDC-Analyzer-facilitated combinatorial strategy for improving the activity and stability of halohydrin dehalogenase from Agrobacterium radiobacter AD1.

Halohydrin dehalogenase from Agrobacterium radiobacter AD1 (HheC) displays a broad substrate range with high regio- and enantioselectivity of both ring-closure and ring-opening reactions, making the enzyme a useful catalyst for the production of optically pure epoxides and β-substituted alcohols. In this study, we report a novel method using an MDC-Analyzer-facilitated combinatorial strategy to improve the activity and stability of HheC by simultaneously randomizing multiple contiguous residues. Six contiguous active-site residues, which are the hotspots for improving the activity of HheC, wer

Agrobacterium rhizogenes-mediated transformation of opium poppy, Papaver somniferum l., and California poppy, Eschscholzia californica cham., root cultures.

An efficient protocol for the establishment of transgenic opium poppy (Papaver somniferum L.) and California poppy (Eschscholzia californica Cham.) root cultures using A. grobacterium rhizogenes is reported. Five strains of A. rhizogenes were tested for their ability to produce hairy roots on wounded opium poppy seedlings and California poppy embryogenic calli. Three of the strains induced hairy root formation on both species, whereas two others either caused the growth of tumorigenic calli or produced no response. To characterize the putative transgenic roots further, explant tissues were co-

Interactions between VirB9 and VirB10 membrane proteins involved in movement of DNA from Agrobacterium tumefaciens into plant cells.

The 11 VirB proteins from Agrobacterium tumefaciens are predicted to form a membrane-bound complex that mediates the movement of DNA from the bacterium into plant cells. The studies reported here on the possible VirB protein interactions in such a complex demonstrate that VirB9 and VirB10 can each form high-molecular-weight complexes after treatment with a chemical cross-linker. Analysis of nonpolar virB mutants showed that the formation of the VirB10 complexes does not occur in a virB9 mutant and that VirB9 and VirB10 are not components of the same cross-linked complex. VirB9, when stabilized

Climate change and the potential global distribution of Agrobacterium tumefaciens: a MaxEnt modeling approach for current and future climate scenarios.

Climate change is fundamentally altering the geographic ranges of plant-pathogenic microorganisms, with profound implications for global food security and agricultural sustainability. Agrobacterium tumefaciens, the gram-negative soil bacterium responsible for crown gall disease in over 140 genera of dicotyledonous plants, is among the most economically significant phytopathogens worldwide, yet its macroecological distribution under current and projected future climates has never been rigorously characterised. Here, we apply the Maximum Entropy algorithm (MaxEnt) to 415 quality-filtered georefe

Establishment of a High-Efficiency In Vitro Regeneration and Agrobacterium-Mediated Genetic Transformation System for Ajania achilleoides (Turcz.) Poljakov.

Ajania achilleoides, a wild species with strong environmental adaptability, is a valuable germplasm resource for genetic improvement and functional gene studies. However, efficient in vitro regeneration and genetic transformation systems for this species remain undeveloped, limiting functional gene characterization and genetic improvement of this valuable germplasm resource. In this study, leaf explants were used to establish an efficient regeneration system by evaluating different combinations of plant growth regulators and to develop an Agrobacterium tumefaciens-mediated genetic transformati

The introduction of a fungus-originated lip H8 confers higher gene transformation efficiency of Agrobacterium fabrum in Nicotiana benthamiana.

Agrobacterium is a genus of bacteria commonly used for transgenic plant production. Transgenic efficiency of Agrobacterium is influenced by virulence (vir) gene expression, host immune responses, etc. Here, we introduced a lignin peroxidase gene lip H8 (derived from Phanerodontia chrysosporium), into Agrobacterium strain C58 - a parental strain of widely used transgenic tools. When fused with an A. fabrum signal peptide, LiP H8 was successfully synthesized and secreted extracellularly by the recombinant A. fabrum C58 strain. Lip H8 not only slightly promoted the growth of A. fabrum C58 in a nu

Establishment of an efficient Agrobacterium-mediated genetic transformation protocol for Saccharum officinarum using Black Cheribon as a model genotype.

Efficient Agrobacterium-mediated transformation (AMT) is vital for the biotechnological improvement of sugarcane (Saccharum spp.). Saccharum officinarum is the main ancestor of all modern cultivars, yet little research has been conducted on its AMT system. In this work, an efficient AMT protocol for S. officinarum was developed, with Black Cheribon as the model genotype owing to its superior tissue culture performance and regeneration capacity. The optimized agro-infection protocol comprised the following main parameters: concentration of acetosyringone (AS) in Agrobacterium culture, concentra

Development and Optimization of a Non-Tissue Culture Hairy Root Transformation System in Camellia oleifera Mediated by Agrobacterium rhizogenes.

Camellia oleifera is an important woody oil crop species endemic to China. However, gene functional analysis and molecular breeding in oiltea are greatly constrained by the low efficiency of conventional Agrobacterium-mediated transformation systems. In this study, a rapid and efficient Agrobacterium rhizogenes-mediated hairy root transformation system was established using the elite cultivar 'Xianglin 210'. A vector carrying the RUBY visual reporter gene was introduced into leaf and hypocotyl explants, and the transformation efficiencies of three Agrobacterium strains (GV3101, MSU440, and K59

Expression, characterization, and improvement of a newly cloned halohydrin dehalogenase from Agrobacterium tumefaciens and its application in production of epichlorohydrin.

A gene encoding halohydrin dehalogenase (HHDH) from Agrobacterium tumefaciens CCTCC M 87071 was cloned and expressed in Escherichia coli. To increase activity and stability of HHDH, 14 amino acid residues around the active site and substrate-binding pocket based on the structural analysis and molecular docking were selected as targets for site-directed mutagenesis. The studies showed that the mutant HHDH (Mut-HHDH) enzyme had a more accessible substrate-binding pocket than the wild-type HHDH (Wt-HHDH). Molecular docking revealed that the distance between the substrate and active site was close

Agrobacterium tumefaciens-mediated transformation of rice with the spider insecticidal gene conferring resistance to leaffolder and striped stem borer.

Immature embryos of rice varieties "Xiushuill" and "Chunjiang 11" precultured for 4d were infected and transformed by Agrobacterium tumefaciens strain EHA101/pExT7 (containing the spider insecticidal gene). The resistant calli were transferred onto the differentiation medium and plants were regenerated. The transformation frequency reached 56% approximately 72% measured as numbers of Geneticin (G418)-resistant calli produced and 36% approximately 60% measured as numbers of transgenic plants regenerated, respectively. PCR and Southern blot analysis of transgenic plants confirmed that the T-DNA

Evaluation of sequence-based tools to gather more insight into the positioning of rhizogenic agrobacteria within the Agrobacterium tumefaciens species complex.

Rhizogenic Agrobacterium, the causative agent of hairy root disease (HRD), is known for its high phenotypic and genetic diversity. The taxonomy of rhizogenic agrobacteria has undergone several changes in the past and is still somewhat controversial. While the classification of Agrobacterium strains was initially mainly based on phenotypic properties and the symptoms they induced on plants, more and more genetic information has been used along the years to infer Agrobacterium taxonomy. This has led to the definition of the so-called Agrobacterium tumefaciens species complex (Atsc), which compri

Generation of lachrymatory factor synthase-suppressed onion (Allium cepa L.) by Agrobacterium-mediated gene transfer for CRISPR/Cas9 genome editing.

INTRODUCTION: Lachrymatory factor, an irritating volatile compound that induces tearing, is produced when onion bulbs are cut or chopped. Lachrymatory factor synthase (LFS)-specific suppression in onion may reduce the lachrymatory effect and enhance flavor and health-functional properties. Genome editing enables precise modification of target genes in plant genomes. However, no reports targeting genes controlling onion-specific traits have been published. Therefore, we aimed to generate onion plants with reduced LFS activity via clustered regularly interspaced short palindromic repeats (CRISPR

Rapid genetic transformation of herbaceous peony without tissue culture via Agrobacterium rhizogenes: Optimization using rhizomes, stems, roots, and seedlings.

Herbaceous peony is a famous traditional flower worldwide. However, the lack of a robust transgenic system has severely restricted its genetic improvement efforts. In this study, we established a tissue culture-free Agrobacterium rhizogenes-mediated transformation system using the rhizomes, stems, root segments, and seedlings of herbaceous peony. Soaking rhizomes and root segments in a resuspension solution containing acetosyringone and 2-morpholinoethanesulfonic acid induced more GFP-positive hairy roots than that in other tissues.
