The role of allogeneic stem cell transplantation in peripheral T-cell lymphoma.

Peripheral T-cell lymphomas (PTCLs) are rare, biologically heterogeneous malignancies associated with poor outcomes following conventional chemotherapy. Despite advances such as brentuximab vedotin-based regimens in CD30-positive disease, relapse rates remain high, particularly in relapsed/refractory (R/R) PTCL. Allogeneic hematopoietic cell transplantation (allo-HCT) represents a potentially curative treatment option through a graft-versus-lymphoma effect capable of overcoming chemoresistance. Retrospective and prospective studies report 5-year overall survival rates of approximately 40-55% i

Donor Cell Leukemia Following Allogeneic Hematopoietic Stem Cell Transplantation.

Approximately 25,000 allogeneic transplants are performed annually worldwide; a figure that has steadily increased over the past three decades. The study of transplant recipient survivorship has become a cogent topic and post-transplant donor cell pathology warrants further study. Donor cell leukemia (DCL) is a rare but serious complication of allogeneic stem cell transplantation (SCT) where the recipient develops a form leukemia originating from the donor cells used for transplantation. Detection of abnormalities predicting donor cell pathology might inform donor selection, and the design of

Allogeneic hematopoietic stem cell transplantation for B-cell lymphoma in Taiwan.

Allogeneic hematopoietic stem cell transplantation (allo-HSCT) is considered for patients with high-risk B-cell lymphoma and relapsed or refractory disease. This study aimed to analyze the long-term follow-up data of patients who underwent allo-HSCT in Taiwan. This was a retrospective observational study using data from the Taiwan Society of Blood and Marrow Transplantation database. A total of 105 patients who underwent allo-HSCT because of high-risk, relapsed, or refractory disease between 2010 and 2019 were included. Forty-five percent of the patients previously underwent autologous stem ce

Allogeneic haematopoietic cell transplantation promotes atherosclerosis in mice via CD8+ T cells.

AIMS: Patients undergoing allogeneic haematopoietic cell transplantation (allo-HCT) are at a risk of developing graft-vs.-host disease (GvHD) and are afflicted with an increased incidence of cardiovascular events. Whether allo-HCT contributes to atherosclerosis progression has not been addressed experimentally. METHODS AND RESULTS: Here, we applied a novel minor histocompatibility antigen mismatch allo-HCT mouse model by transplanting C57BL/6 mice deficient for the LDL receptor (B6.Ldlr-/-) with bone marrow (BM) or BM and T cells (BMT) from BALB/b donors (BALB/b, H-2b → B6.Ldlr-/-, and H-2b),

Unedited allogeneic iNKT cells show extended persistence in MHC-mismatched canine recipients.

Allogeneic invariant natural killer T cells (allo-iNKTs) induce clinical remission in patients with otherwise incurable cancers and COVID-19-related acute respiratory failure. However, their functionality is inconsistent among individuals, and they become rapidly undetectable after infusion, raising concerns over rejection and limited therapeutic potential. We validate a strategy to promote allo-iNKT persistence in dogs, an established large-animal model for novel cellular therapies. We identify donor-specific iNKT biomarkers of survival and sustained functionality, conserved in dogs and human

Targeting T-cell malignancies using allogeneic double-negative CD4-CAR-T cells.

BACKGROUND: Patients with relapsed/refractory T-cell malignancies have limited treatment options. The use of chimeric antigen receptor (CAR)-T cell therapy for T-cell malignancies is challenging due to possible blast contamination of autologous T-cell products and fratricide of CAR-T cells targeting T-lineage antigens. Recently, allogeneic double-negative T cells (DNTs) have been shown to be safe as an off-the-shelf adoptive cell therapy and to be amendable for CAR transduction. Here, we explore the antitumor activity of allogeneic DNTs against T-cell malignancies and the potential of using an

Effect of Allogeneic Oral Mucosa Mesenchymal Stromal Cells on Equine Wound Repair.

OBJECTIVE: To assess the clinical value and safety of the application of allogeneic equine oral mucosa mesenchymal stromal cells (OM-MSCs) to wounds. Animals. 8 healthy adult horses without front limb skin lesions or musculoskeletal disease. Procedures. Stem cells were isolated from the oral mucosa of a donor horse. Horses were subjected to the creation of eight full-thickness cutaneous wounds, two on each distal forelimb (FL) and two on both sides of the thorax (TH). Each wound was subjected to one out of four treatments: no medication (T1), hyaluronic acid- (HA-) gel containing OM-MSC (T2),

Intramuscular Transplantation of Allogeneic Mesenchymal Stromal Cells Derived from Equine Umbilical Cord.

BACKGROUND AND OBJECTIVES: Mesenchymal stromal cells (MSCs) have great therapeutic potential, particularly in the process of tissue repair and immunomodulation through the secretion of biomolecules. Thus, the aim of this study was to evaluate the hypothesis that intramuscular transplantation of allogeneic MSCs obtained from equine umbilical cord (UC-MSCs) is safe, demonstrating that this is a suitable source of stem cells for therapeutic use. METHODS AND RESULTS: For this, UC-MSCs were cultured, characterized and cryopreserved for future transplantation in six healthy mares. On day 0, transpl

Allogeneic guinea pig mesenchymal stem cells ameliorate neurological changes in experimental colitis.

BACKGROUND: The use of mesenchymal stem cells (MSCs) to treat inflammatory bowel disease (IBD) is of great interest because of their immunomodulatory properties. Damage to the enteric nervous system (ENS) is implicated in IBD pathophysiology and disease progression. The most commonly used model to study inflammation-induced changes to the ENS is 2,4,6-trinitrobenzene-sulfonate acid (TNBS)-induced colitis in guinea pigs; however, no studies using guinea pig MSCs in colitis have been performed. This study aims to isolate and characterise guinea pig MSCs and then test their therapeutic potential

The Management of Membranous Glomerulopathy in Allogeneic Stem Cells Transplantation: Updated Literature.

BACKGROUND: membranous glomerulopathy (MG) is an immunomediated disorder which accounts for the most common cause of nephrotic syndrome (NS) following allogeneic hematopoietic stem cell transplantation (HSCT). OBJECTIVE AND METHODS: to provide an update on the issue by reviewing pertinent literature on the MEDLINE database. RESULTS: sixty-nine post allogenic HSCT patients (42 male) with MG were identified. The median age was 43 (5 to 68) years. Time interval from allogenic HSCT to MG diagnosis ranged from 3 to 134 months (median 17). Most MG patients had a history of acute (70%) or chronic (

Allogeneic transplantation of CD34+ selected hematopoietic cells--clinical problems and current challenges.

Since G-CSF mobilized peripheral blood has become the major source of hematopoietic cells (PBSC) for allogeneic transplantation of patients with hematological malignancies, new technologies of T-cell depletion were developed to reduce the risk of graft-versus-host disease. CD34+ selection by immunomagnetic separation has been shown to be the most effective method to achieve a 3-4 log depletion of T-cells while preserving the high number of hematopoietic progenitors in the graft. The high number of CD34+ cells contained in the positive fraction have allowed to facilitate engraftment from siblin

Autologous and allogeneic transplantation with peripheral blood CD34+ cells: a pediatric experience.

BACKGROUND AND OBJECTIVE: Peripheral blood stem cells (PBSC) have replaced bone marrow (BM) as the primary form for autologous hematopoietic stem cell transplantation. Furthermore, the use of allogeneic PBSC transplantation is now rapidly expanding and several centers have adopted this procedure. A new strategy in the use of PBSC is positive selection of CD34+ hematopoietic progenitor (CD34+) cells, and indeed large-scale devices for the clinical exploitation of CD34+ cell selection are now commercially available. In the autologous setting, the primary advantage of using CD34+ selected PBSC is

Interleukin-2 gene-modified allogeneic tumor cells for treatment of relapsed neuroblastoma.

Tumor cells that have been genetically modified to express immunostimulatory genes will induce effective antitumor responses in a range of syngeneic animal models. For human applications, transduced autologous tumor cell lines are often difficult or impossible to prepare, so that there are strong incentives for substituting a standardized allogeneic tumor cell line. However, such lines may be inferior immunogens if they differ from host tumors in the antigens they express. We have evaluated the safety, immunostimulatory, and antitumor activity of an interleukin-2-secreting allogeneic neuroblas

Monotypic immunoglobulin E plasma cells in an allogeneic bone marrow transplant recipient.

A patient with acute lymphoblastic leukaemia in first remission received a bone marrow transplant from his HLA-identical brother. The patient had a remote history of asthma and the bone marrow donor had allergic asthma. The patient developed acute graft-versus-host disease and died 2 months after transplantation. At autopsy there were high numbers of plasma cells in lymphoid tissues. The majority of this cell population was of polytypic IgG, IgM or IgA origin, but there was a significant contribution by monotypic IgE-lambda-containing cells, varying from 10% in the appendix to 35% in lymph nod

Stimulatory ability of human leukaemic cells in the allogeneic mixed leucocyte reaction.

Cryopreserved mononuclear cells from 34 patients with a high proportion of blasts in the peripheral blood were tested for their ability to stimulate normal allogeneic lymphocytes in the one-way mixed leucocyte reaction (MLR). Of these, more than half failed to stimulate allogeneic responders, despite the fact that many of them carried Ia-like antigens. In general, acute myeloid leukaemia cells were non-stimulatory whereas acute myelomonocytic leukaemia cells were effective stimulators. Thus, MLR stimulatory ability appears to be a differentiation marker in acute non-lymphocytic leukaemias. Acu

Regeneration of transplanted allogeneic hemopoietic progenitor cells in mouse spleen organ cultures.

The capacity of CBA, C57BL/6 and BALB/c mouse bone marrow cells to repopulate lethally-irradiated syngeneic or allogeneic recipients was monitored by measuring hemopoietic progenitor cell (colony forming cell, CFC) levels in the bone marrow and spleen from 0-7 days after transplantation. Bone marrow cells (10(7)/mouse) injected into allogeneic recipients failed to repopulate the spleen in vivo whereas regeneration was possible in spleen organ cultures. In contrast, the repopulation of the bone marrow by injected hemopoietic cells was successful in both allogeneic and syngeneic recipients. The

Allogeneic Mature Human Dendritic Cells Generate Superior Alloreactive Regulatory T Cells in the Presence of IL-15.

Expansion of Ag-specific naturally occurring regulatory T cells (nTregs) is required to obtain sufficient numbers of cells for cellular immunotherapy. In this study, different allogeneic stimuli were studied for their capacity to generate functional alloantigen-specific nTregs. A highly enriched nTreg fraction (CD4(+)CD25(bright)CD127(-) T cells) was alloantigen-specific expanded using HLA-mismatched immature, mature monocyte-derived dendritic cells (moDCs), or PBMCs. The allogeneic mature moDC-expanded nTregs were fully characterized by analysis of the demethylation status within the Treg-spe
