Silymarin induces recovery of pancreatic function after alloxan damage in rats.

Alloxan has been widely used to produce experimental diabetes mellitus syndrome. This compound causes necrosis of pancreatic beta-cells and, as is well known, induces oxidant free radicals which play a relevant role in the etiology and pathogenesis of both experimental and human diabetes mellitus. Previously we have reported hypoglycemic and antilipoperoxidative actions of silymarin in serum and pancreatic tissue respectively. The aim of this study was to test whether silymarin could reduce the hyperglycemia and revert the pancreatic damage in alloxan treated rats, tested with silymarin in two

Ascorbic acid supplementation restores defective leukocyte-endothelial interaction in alloxan-diabetic rats.

BACKGROUND: Defective leukocyte-endothelial interactions are observed in experimental diabetes and may reduce the capacity to mount an adequate inflammatory response. The present study investigated the effect of ascorbic acid, an inhibitor of free radical and glycated protein formation as well as an aldose reductase inhibitor, on leukocyte-endothelial interaction in alloxan-diabetic rats. METHODS: Rats were rendered diabetic by alloxan injection (40 mg/kg; iv). After 30 days, diabetic and nondiabetic controls were supplemented for 12 days with ascorbic acid (50 or 200 mg/kg/day) or received s

Comparison of retinal lesions in alloxan-diabetic rats and galactose-fed rats.

Galactose-fed rats develop a retinal microvascular disease, but retinopathy has not been found to develop reproducibly in diabetic rats. We sought to determine which retinal lesions can be reproducibly produced by long-term diabetes in rats, the extent to which the capillary lesions in diabetic rats and galactosemic rats are similar, and whether the retinopathy induced by 50% galactose can be reproduced satisfactorily by a lower concentration of galactose. Alloxan-diabetic rats and rats fed either a 50% galactose diet or a 30% galactose diet were killed after comparable durations of study (18

Root surface caries and periodontal disease in long-term alloxan diabetic rats.

Caries, periodontal disease, and occlusal wear were studied in rats with short-term (one month) and long-term (12 months) untreated alloxan diabetes and in age-matched control rats. Standard laboratory diet and water were given ad libitum. Streptococcus mutans and lactobacilli were naturally occurring inhabitants of the oral bacterial flora. In short-term diabetic rats, there was less dental plaque in the gingival region of the first mandibular molar than in control rats, but the density of leukocytes in the junctional epithelium was increased. Interdentally, the gingival papillae were intact,

Renin-angiotensin-aldosterone system, electrolyte homeostasis and blood pressure in alloxan diabetes.

The effect of a chronic glucose osmotic diuresis on electrolyte homeostasis was evaluated in alloxan diabetic rats with urine volumes greater than 150 ml/day and glycosuria of 4 to 10 gm/day. Results were compared with control rats for periods up to 84 days. Sodium and potassium intake and urinary losses were significantly higher in diabetic animals throughout the study periods. Negative Na balance, however, persisted for only four days, and negative K balance for only 18 days. Blood volumes were elevated probably secondary to the osmotic effect of hyperglycemia (serum glucose greater than 600

Bone regeneration in cranioplasty and clinical complications in rabbits with alloxan-induced diabetes.

This research evaluated the bone repair process in surgical defects created on the parietal bones of diabetic rabbits using the guided bone regeneration technique to observe the effects of alloxan in the induction of diabetes mellitus. Twenty-four adult rabbits were divided into three study groups: control (C), diabetic (D) and diabetic associated to polytetrafluoroethylene (PTFE) membrane (D-PTFE). For diabetes induction the animals received one dose of monohydrated alloxan (90 mg/kg) by intravenous administration in the auricular or femoral vein. In group D-PTFE the membrane covered both the

The role of exercise on long-term effects of alloxan administered in neonatal rats.

The present study was designed to analyse the effects of aerobic exercise on the metabolic effects of alloxan. Male Wistar newborn rats (2 days old) received alloxan (200 mg (kg body weight)(-1)) intraperitoneally (A rats). Vehicle-injected rats were used as controls (C rats). At 28 days old, some of the A rats were subjected to swimming for 1 h day(-1), 5 day week(-1) (AT rats). At 28, 60 and 90 days old the animals were subjected to glucose (GTTo) and insulin (ITTsc) tolerance tests. All the animals were then killed by decapitation for blood and tissue evaluations. On the 60th day, there was

Metal-mediated DNA damage induced by diabetogenic alloxan in the presence of NADH.

Alloxan is known to induce diabetes in experimental animals through destruction of insulin-producing 3-cells of pancreas. The mechanism of DNA damage induced by alloxan was investigated using 32P-labeled human DNA fragments. Cu(II)-dependent DNA damage increased with the concentration of alloxan and NADH. Alloxan induced DNA cleavage frequently at thymine and cytosine residues in the presence of NADH and Cu(II). Catalase and bathocuproine, a Cu(I)-specific chelator, almost completely inhibited DNA damage, suggesting the involvement of H2O2 and Cu(I). Alloxan induced Cu(II)-dependent production

[Effectiveness of parenteral feeding with various hydrolysates in alloxan diabetes in white rats].

The effect of applying various hydrolysates for parenteral feeding (caseine hydrolysate, hydrolysin L-103, aminosol, moriamine S-2) in albino rats with alloxan-induced diabetes was investigated. The assimilation of the hydrolysates introduced was found in these animals to be down by comparison with intact ones. It was only administration of nitrogenous media containing a sufficient amount of aminic nitrogen (moriamine S-2) that helped establish a positive nitrogenous balance and to prevent a loss of weight and the dry weight of the tissues.
