A spatial transcriptomics comparison of the adult versus metamorphosed axolotl brain.

The Mexican axolotl (Ambystoma mexicanum) is an established model for studying tetrapod regeneration and development. Notably, axolotls exhibit remarkable brain regeneration as adults, a trait rarely observed in other adult vertebrates. Adult axolotls can undergo metamorphosis, a process that induces dramatic remodeling of multiple organs and is accompanied by a gradual decline in regenerative capacity and lifespan. However, systematic studies on whole-brain cellular dynamics and molecular mechanisms in both adult and metamorphosed individuals remain lacking. Here, we profiled five representat

Characterization of the developing axolotl nasal cavity supports multiple evolution of the vertebrate choana.

All tetrapods (mammals, birds, reptiles, and amphibians) share the ability to breathe with their mouths closed due to the formation of choanae, which are openings that allow communication between the nasal and oral cavities. In most fishes, the nasal cavities serve a strictly olfactory function, possessing incurrent and excurrent nares that lie outside of the mouth and therefore, never communicate with the respiratory system. It is not until the evolution of tetrapods, in which the nasal cavities consistently open into the mouth, that they are used both for olfaction and for respiration. Howev

Osteoclast-mediated resorption primes the skeleton for successful integration during axolotl limb regeneration.

Early events during axolotl limb regeneration include an immune response and the formation of a wound epithelium. These events are linked to a clearance of damaged tissue prior to blastema formation and regeneration of the missing structures. Here, we report the resorption of calcified skeletal tissue as an active, cell-driven, and highly regulated event. This process, carried out by osteoclasts, is essential for a successful integration of the newly formed skeleton. Indeed, the extent of resorption is directly correlated with the integration efficiency, and treatment with zoledronic acid resu

Distribution and innervation of taste buds in the axolotl.

Adult axolotls have approximately 1,400 taste buds in the epithelium of the pharyngeal roof and floor and the medial surfaces of the visceral bars. These receptors are most dense on the lingual surfaces of the upper and lower jaws, slightly less dense throughout lateral portions of the pharyngeal roof and floor, and more sparse within medial portions of the pharyngeal roof and floor, except for a median oval patch of receptors located rostrally between the vomerine tooth fields. Each taste bud is a pear-shaped organ, situated at the center of a raised hillock and averaging 80 and 87 microm in

CXCL14-like immunoreactivity in growth hormone-containing cells of urodele pituitaries.

Immunohistochemical techniques were employed to investigate the distribution of a chemokine, namely, CXCL14-like immunoreactivity in the axolotl (Ambystoma mexicanum) and Japanese black salamander (Hynobius nigrescens) pituitaries. CXCL14-immunoreactive cells concentrated at an area of the pars distalis adjacent to the pars intermedia. We found that these cells correspond to the cells immunoreactive to an antibody against rat growth hormone (GH). Immunoelectron microscopy indicated that the CXCL14-like substance and GH coexisted on the secretory granules in the axolotl pituitary. Western blot

Axolotls retain fertility throughout lifespan.

BACKGROUND: Salamanders such as axolotls exhibit exceptional regenerative abilities and longevity. While many ectothermic species reproduce into old age, axolotls have been proposed to experience post-maturation fertility decline. RESULTS: We hereby present a large-scale assessment of axolotl reproductive potential across lifespan based on over 15 years of mating records from captive breeding. We show that axolotl egg number, egg quality, and mating success rates peak after sexual maturation and gradually decline up to 4 years of age, with rates stabilising after the early-life maturation per

The Accessory Limb Model Regenerative Assay and Its Derivatives.

When the Accessory Limb Model (ALM) regenerative assay was first published by Endo, Bryant, and Gardiner in 2004, it provided a robust system for testing the cellular and molecular contributions during each of the basic steps of regeneration: the formation of the wound epithelium, neural induction of the apical epithelial cap, and the formation of a positional disparity between blastema cells. The basic ALM procedure was developed in the axolotl and involves deviating a limb nerve into a lateral wound and grafting skin from the opposing side of the limb axis into the site of injury. In this ch

Morphogenesis of the axolotl pronephric duct: a model system for the study of cell migration in vivo.

Pronephric duct (PND) morphogenesis is a critical early event in the development of the vertebrate excretory system. This structure is the exit channel for both pronephric and mesonephric filtrate, forms the ureteric bud of the metanephros and gives rise to the ductus deferens of the testis. In addition, the PND and ureteric bud epithelia induce terminal differentiation of the mesonephric and metanephric mesenchyme, respectively. Elongation of the PND in all vertebrates involves active cell migration of the primordium. In urodele embryos--unlike in some anuran, avian and mammalian embryos--elo

BMP signaling is essential for sustaining proximo-distal progression in regenerating axolotl limbs.

Amputation of a salamander limb triggers a regeneration process that is perfect. A limited number of genes have been studied in this context and even fewer have been analyzed functionally. In this work, we use the BMP signaling inhibitor LDN193189 on Ambystoma mexicanum to explore the role of BMPs in regeneration. We find that BMP signaling is required for proper expression of various patterning genes and that its inhibition causes major defects in the regenerated limbs. Fgf8 is downregulated when BMP signaling is blocked, but ectopic injection of either human or axolotl protein did not rescue

Comparison of protein expression profile of limb regeneration between neotenic and metamorphic axolotl.

The axolotl (Ambystoma mexicanum) salamander, a urodele amphibian, has an exceptional regenerative capacity to fully restore an amputated limb throughout the life-long lasting neoteny. By contrast, when axolotls are experimentally induced to metamorphosis, attenuation of the limb's regenerative competence is noticeable. Here, we sought to discern the proteomic profiles of the early stages of blastema formation of neotenic and metamorphic axolotls after limb amputation by employing LC-MS/MS technology. We quantified a total of 714 proteins and qRT-PCR for selected genes was performed to validat

Continuous anesthesia for 60 days in an isosmotic environment does not impair limb or cardiac regeneration in the axolotl.

Longitudinal animal experiments in the field of regenerative biology often require repeated use of short-term anesthesia (minutes to a few hours). Regain of consciousness limits the level of acceptable invasiveness of procedures, and it makes it difficult to untangle behavioral changes caused by injury to physiological processes involved in the regenerative response. Therefore, a method to keep a regenerative research animal in a comatose state under continuous anesthesia during regenerative experiments often spanning months, would be ethically and experimentally desirable. Here we report on a

Axolotl mandible regeneration following complete transverse amputation involves a blastema formation and exhibits a limit along the proximodistal axis.

Current therapeutic strategies aimed at promoting the regeneration of craniofacial tissues do not fully address all physiological and aesthetic needs. This highlights the need to investigate the regenerative response of craniofacial tissues in regenerative species like Ambystoma mexicanum (A. mexicanum), particularly concerning the mandible. However, although previous studies have demonstrated the regenerative response after lateral cuts of the mandible in A. mexicanum, it is unknown whether it can regenerate the mandible after complete transverse amputation, a type of injury used to remove tu

Behavioural evidence of a humidistat: a temperature-compensating mechanism of hydroregulation in spotted salamanders.

The ability to control hydration state is essential for terrestrial species, especially amphibians, which are highly susceptible to dehydration. Here, we examined how temperature (17°C versus 22°C) influenced behavioural hydroregulation in spotted salamanders (Ambystoma maculatum) using a laboratory humidity gradient. Salamanders defended a constant vapour pressure deficit (VPD) between temperatures by targeting higher relative humidity at 22°C than at 17°C, possibly to compensate for increased evaporative demand at warmer temperatures. Individuals selecting higher VPDs experienced greater eva

Molecular basis for de novo thymus regeneration in a vertebrate, the axolotl.

The thymus is the primary site of T cell development, central to the establishment of self-tolerance and adaptive immune function. In mammals, the thymus undergoes age-related involution, resulting in a global decline in immune function. The thymus has some regenerative ability that relies on pre-existing thymic remnants but is insufficient to prevent involution. Here, we show that the juvenile axolotl (Ambystoma mexicanum) is able to regenerate its thymus de novo after complete removal, constituting an exception among vertebrates. Using single-cell transcriptomics and genetic and transplantat

Axolotl mandible regeneration occurs through mechanical gap closure and a shared regenerative program with the limb.

The mandible plays an essential part in human life and, thus, defects in this structure can dramatically impair the quality of life in patients. Axolotls, unlike humans, are capable of regenerating their lower jaws; however, the underlying mechanisms and their similarities to those in limb regeneration are unknown. In this work, we used morphological, histological and transcriptomic approaches to analyze the regeneration of lateral resection defects in the axolotl mandible. We found that this structure can regenerate all missing tissues in 90 days through gap minimization, blastema formation a

The role of innervation in the development of taste buds: insights from studies of amphibian embryos.

Amphibian embryos have long been model organisms for studies of development because of their hardiness and large size, as well as the ease with which they can be experimentally manipulated. These particular advantages have allowed us recently to test the role of innervation in the development of vertebrate taste buds using embryos of an aquatic salamander, the axolotl. The predominant model of taste bud genesis has been one of neural induction, in which ingrowing sensory neurites induce taste bud differentiation in the epithelium that lines the mouth and pharynx. However, when we prevented emb

Artificial Insemination in Axolotl.

In axolotls (Ambystoma mexicanum), fertilization takes place internally. After courtship, the male axolotl deposits spermatophores, which the female takes up into her cloaca in order to fertilize eggs internally. The success of axolotl breedings is subject to several poorly understood factors including age, pairing, and genotype. In some cases, individuals are unable to breed naturally despite having significant scientific value. Assisted reproductive technologies represent one approach to maintaining stocks of such individuals, as well as supplementing natural breedings of laboratory stocks.H

Excitable properties of olfactory receptor neurons.

Action potential-generating properties of olfactory receptor neurons in the olfactory epithelium of the salamander, Ambystoma tigrinum, were studied in control animals, and 2 and 4 weeks after olfactory nerve transection. The threshold for impulse generation in response to injected current was extremely low (74 +/- 46 pA). In addition, the discharge frequencies of the receptor neurons were exquisitely sensitive to small increments of injected current. These high sensitivities may be characteristic of small neurons and stand in contrast to the much lower sensitivities reported for large neurons

[Histochemical study of the cholinesterases of various formations of the vertebrate endbrain].

Histochemical studies have been made on true (AChE) and false (BChE) cholinesterases in different parts of the telencephalon of amphibians, reptiles and mammals. The data obtained indicate that the enzymic localization depends on both the level of the development of the brain and phylogenetic age of its individual formations. In the telencephalon of Ambystoma mexicanum, high concentration of AChE was found in vascular-capillary network, in other investigated amphibians (Triturus vulgaris, Rana temporaria) diffuse distribution of AChE of different intensity was observed in ancient formations of

A natural heterohaemagglutinin in Xenopus laevis serum.

In most adult Xenopus laevis the serum contains a 'natural' factor capable of lysing the erythrocytes from a wide variety of amniote species. The factor has no effect on the erythrocytes of another amphibian, Ambystoma mexicanum, nor will serum from one animal lyse red cells from another Xenopus individual. No lysing factor was present in the serum of larval (tadpole) Xenopus. Heating of Xenopus serum to 56 degrees for 30 min, absorption of the serum with zymosan or inulin, or removal of calcium and magnesium ions results in loss of lytic activity, although haemagglutinating activity remains,
