Enamelin/ameloblastin gene polymorphisms in autosomal amelogenesis imperfecta among Syrian families.

AIM: This study was undertaken to investigate whether a single G deletion within a series of seven G residues (codon 196) at the exon 9-intron 9 boundary of the enamelin gene ENAM and a tri-nucleotide deletion at codon 180 in exon 7 (GGA vs deletion) of ameloblastin gene AMBN could have a role in autosomal amelogenesis imperfecta among affected Syrian families. METHODS: A new technique - size-dependent, deletion screening - was developed to detect nucleotide deletion in ENAM and AMBN genes. Twelve Syrian families with autosomal-dominant or -recessive amelogenesis imperfecta were included. RE

The Evolutionary Young Actin Nucleator Cobl Is Important for Proper Amelogenesis.

The actin cytoskeleton plays an important role in morphological changes of ameloblasts during the formation of enamel, which is indispensable for teeth to withstand wear, fracture and caries progression. This study reveals that the actin nucleator Cobl is expressed in ameloblasts of mandibular molars during amelogenesis. Cobl expression was particularly pronounced during the secretory phase of the enamel-forming cells. Cobl colocalized with actin filaments at the cell cortex. Importantly, our analyses show an influence of Cobl on both ameloblast morphology and cytoskeletal organization as well

Autoimmune amelogenesis imperfecta in patients with APS-1 and coeliac disease.

Ameloblasts are specialized epithelial cells in the jaw that have an indispensable role in tooth enamel formation-amelogenesis1. Amelogenesis depends on multiple ameloblast-derived proteins that function as a scaffold for hydroxyapatite crystals. The loss of function of ameloblast-derived proteins results in a group of rare congenital disorders called amelogenesis imperfecta2. Defects in enamel formation are also found in patients with autoimmune polyglandular syndrome type-1 (APS-1), caused by AIRE deficiency3,4, and in patients diagnosed with coeliac disease5-7. However, the underlying mecha

Dental age estimation by different methods in patients with amelogenesis imperfecta.

Amelogenesis imperfecta (AI) is one of the developmental enamel defects. In patients with AI, as well as enamel defects, some dental anomalies are encountered and may affect tooth development. For children whose enamel structure is not normal, whether an accurate result can be obtained by dental age determination methods is not clear. The aims of this study are to determine the dental age for the assessment of tooth development in patients with AI by 3 different methods, to decide a method that will present the most accurate result to the chronological age, and to build a basis in terms of pos

Deletion of amelotin exons 3-6 is associated with amelogenesis imperfecta.

Amelogenesis imperfecta (AI) is a heterogeneous group of genetic conditions that result in defective dental enamel formation. Amelotin (AMTN) is a secreted protein thought to act as a promoter of matrix mineralization in the final stage of enamel development, and is strongly expressed, almost exclusively, in maturation stage ameloblasts. Amtn overexpression and Amtn knockout mouse models have defective enamel with no other associated phenotypes, highlighting AMTN as an excellent candidate gene for human AI. However, no AMTN mutations have yet been associated with human AI. Using whole exome se

Multidisciplinary management of a child with severe open bite and amelogenesis imperfecta.

AIM: To present a case of multidisciplinary management and fixed rehabilitation of a young girl with amelogenesis imperfecta (AI), a severe open bite and occlusal instability. BACKGROUND: AI is a genetic disorder characterized by enamel malformations, disturbances in tooth eruption and significant attrition. Early diagnosis is essential, since rapid breakdown of tooth structure may occur, giving rise to acute symptoms and complicated treatment. As AI is frequently accompanied by unesthetic appearance, open bite deformity and malocclusion, a multidisciplinary approach is often required. CASE

Possible linkage of SP6 transcriptional activity with amelogenesis by protein stabilization.

Ameloblasts produce enamel matrix proteins such as amelogenin, ameloblastin, and amelotin during tooth development. The molecular mechanisms of ameloblast differentiation (amelogenesis) are currently not well understood. SP6 is a transcription factor of the Sp/KLF family that was recently found to regulate cell proliferation in a cell-type-specific manner. Sp6-deficient mice demonstrate characteristic tooth anomalies such as delayed eruption of the incisors and supernumerary teeth with disorganized amelogenesis. However, it remains unclear how Sp6 controls amelogenesis. In this study, we used

Restoring function and aesthetics in a patient previously treated for amelogenesis imperfecta.

A 26-year-old male patient presented with generalised gingival recession related to the margins of crowns fitted between the ages of 12 and 15 years. The patient, previously treated for Amelogenesis Imperfecta, suffered a mandibular fracture at the age of 22 years. He had an anterior open bite and in the retruded contact position the 28 and 38 were the only teeth in contact. This case demonstrates the management of compromised fixed prosthodontic care following further growth and trauma. On completion of treatment a good functional and aesthetic result was achieved.

Immunochemical and biochemical characteristics of enamel proteins in hypocalcified amelogenesis imperfecta.

Amelogenesis imperfecta is a hereditary disease of the enamel that is unassociated with generalized defects. Cases of the condition are clinically classified into three groups: hypoplastic, hypomaturation, and hypocalcified. In this study, soluble protein fractions of the enamel from three patients with hypocalcified amelogenesis imperfecta were examined through the use of immunochemical and biochemical techniques. In immunochemical analyses done with a polyclonal anti-amelogenin antibody, all samples from enamel in which there was amelogenesis imperfecta were found to contain considerable amo

Mutations in CNNM4 cause recessive cone-rod dystrophy with amelogenesis imperfecta.

Cone-rod dystrophies are inherited dystrophies of the retina characterized by the accumulation of deposits mainly localized to the cone-rich macular region of the eye. Dystrophy can be limited to the retina or be part of a syndrome. Unlike nonsyndromic cone-rod dystrophies, syndromic cone-rod dystrophies are genetically heterogeneous with mutations in genes encoding structural, cell-adhesion, and transporter proteins. Using a genome-wide single-nucleotide polymorphism (SNP) haplotype analysis to fine map the locus and a gene-candidate approach, we identified homozygous mutations in the ancient

Transformative comprehensive functional and esthetic rehabilitation of amelogenesis imperfecta: An interdisciplinary approach.

This clinical report describes the interdisciplinary treatment of a patient with hypoplastic amelogenesis imperfecta (AI) and an anterior open occlusal relationship. Prior to referral to the prosthodontics department, she had undergone active orthodontic treatment for 5 years. As she presented with skeletal, functional, and esthetic concerns, orthodontic treatment alone was insufficient to address her complex treatment goals. Given the complexity, optimal comprehensive management necessitated an interdisciplinary approach involving orthodontics, endodontics, periodontal therapy, orthognathic s
