Amino acid metabolism in skeletal cells.

Amino acid metabolism regulates essential cellular functions, not only by fueling protein synthesis, but also by supporting the biogenesis of nucleotides, redox factors and lipids. Amino acids are also involved in tricarboxylic acid cycle anaplerosis, epigenetic modifications, next to synthesis of neurotransmitters and hormones. As such, amino acids contribute to a broad range of cellular processes such as proliferation, matrix synthesis and intercellular communication, which are all critical for skeletal cell functioning. Here we summarize recent work elucidating how amino acid metabolism sup

Amino Acid Metabolism in Apicomplexan Parasites.

Obligate intracellular pathogens have coevolved with their host, leading to clever strategies to access nutrients, to combat the host's immune response, and to establish a safe niche for intracellular replication. The host, on the other hand, has also developed ways to restrict the replication of invaders by limiting access to nutrients required for pathogen survival. In this review, we describe the recent advancements in both computational methods and high-throughput -omics techniques that have been used to study and interrogate metabolic functions in the context of intracellular parasitism.

Amino Acids in Intestinal Physiology and Health.

Dietary protein digestion is an efficient process resulting in the absorption of amino acids by epithelial cells, mainly in the jejunum. Some amino acids are extensively metabolized in enterocytes supporting their high energy demand and/or production of bioactive metabolites such as glutathione or nitric oxide. In contrast, other amino acids are mainly used as building blocks for the intense protein synthesis associated with the rapid epithelium renewal and mucin production. Several amino acids have been shown to support the intestinal barrier function and the intestinal endocrine function. In

Amino Acid Intake in Preterm Infants.

A large proportion of extremely low-birth-weight infants requires parenteral nutrition for variable lengths of time. Amino acids are the key ingredients of parenteral nutrition. The goal of appropriate amino acid administration is to promote anabolism and normal cellular development in order to limit the incidence of postnatal growth restriction, which is associated with neurodevelopmental delays. The benefits of early amino acid commencement soon after birth are compelling, especially on nitrogen balance, while long-term outcome studies are lacking. Amino acid administration at 2.5 g/kg per d

The role of amino acids in hydroxyapatite mineralization.

Polar and charged amino acids (AAs) are heavily expressed in non-collagenous proteins (NCPs), and are involved in hydroxyapatite (HA) mineralization in bone. Here, we review what is known on the effect of single AAs on HA precipitation. Negatively charged AAs, such as aspartic acid, glutamic acid (Glu) and phosphoserine are largely expressed in NCPs and play a critical role in controlling HA nucleation and growth. Positively charged ones such as arginine (Arg) or lysine (Lys) are heavily involved in HA nucleation within extracellular matrix proteins such as collagen. Glu, Arg and Lys intake ca

Functional amino acids in nutrition and health.

The recent years have witnessed growing interest in biochemistry, physiology and nutrition of amino acids (AA) in growth, health and disease of humans and other animals. This results from the discoveries of AA in cell signaling involving protein kinases, G protein-coupled receptors, and gaseous molecules (i.e., NO, CO and H2S). In addition, nutritional studies have shown that dietary supplementation with several AA (e.g., arginine, glutamine, glutamate, leucine, and proline) modulates gene expression, enhances growth of the small intestine and skeletal muscle, or reduces excessive body fat. Use of amino acids in refractive surgery.

PURPOSE: To assess the influence of amino acid supplements on corneal fibroblast activity during corneal healing in patients with chronic ulcers, as well as after refractive surgery when delayed re-epithelialization had been observed in the fellow eye. METHODS: Studies were conducted in vitro on eye bank eyes, and in vivo. In vitro cultures of rabbit corneal fibroblasts were enriched with progressively higher concentrations of amino acids. The human eye bank cornea study involved corneal epithelium removal, with corneas kept for 7 days in standard storage medium, either simple or enriched wit

Intravenous amino acids, cholestasis and kwashiorkor.

Kwashiorkor dermatosis has not been reported in children on parenteral nutrition (PN). We report a case of kwashiorkor developing in a partially PN dependent patient with cholestasis, when amino acids were removed from the PN solution in an effort to control the cholestasis. Although the kwashiorkor dermatosis improved within 24 hours after the addition of amino acids (1 g/kg) to the PN solution, the cholestasis worsened.

D-amino acids in animal peptides.

D-amino acids have been detected in a variety of peptides synthesized by animal cells. These include opiate and antimicrobial peptides from amphibian skin, neuropeptides from snail ganglia, a hormone from crustaceans, and a constituent of a spider venom. cDNA cloning has shown that at those positions where a D-amino acid is found in the end-product, a normal codon for the corresponding L-amino acid is present. This implies that the D-residues are formed from L-amino acids by a posttranslational reaction. A prototype enzyme catalyzing such a reaction has recently been isolated from the venom of

Compartmentation of amino acids in the rat kidney.

Amino acid concentrations ([AA]) were determined in cortical, outer and inner medullary (OM and IM), and papillary tissue of rat kidney (Cti, mmol/kg wet wt), in plasma (Cpl), and in urine. In all regions, Cti values were highest for Tau, Gly, and Glu-, making up 54-65% of the total [AA]:27, 21, and 11 mmol/kg wet wt in cortex, OM, and IM and papilla, respectively. Cortical cell water [AA] values (CcH2O, mmol/kgH2O) were between 12.4 (Tau) and 0.09 (Orn+), representing cell water-to-plasma water ratios (CcH2O/CpH2O) between 134 (Asp-) and 0.9 (Thr and Cit). Oesophagus carcinoma amino acids control.

Serum amino acids level examination of the oesophagus carcinoma patients admitted to I Surgical Clinic of Teaching Hospital in Olomouc from 1/1/1990 to 31/7/1991 were carried out by the authors. The standard values were found with the group of patients with resectionable tumor or with a tumor in non-advanced stage on the one hand and on the other hand there were some lower values with the patients with a tumor in an advanced stage. In this way two groups of the sick were examined--the first one with benign oesophagus stenosis accompanied by nutritional disorder, the second with resectionable l

Plasma amino acid levels in cataract.

Fasting plasma amino-acid profiles were determined in 32 subjects with either cataractous lenses (24 subjects) or normal lenses (eight subjects). The findings were compared with established laboratory normal values. The amino-acid levels of all subjects with normal lenses were within the normal range for the subjects' age groups. In contrast, subjects with presenile or senile cataracts had markedly elevated levels of glutamine, with similar elevations of alanine in presenile cataract and of histidine in senile cataract. To the best of our knowledge, this is the first report of such a plasma am

Plasma amino acids in rheumatoid arthritis.

Plasma amino acid concentrations have been investigated in 12 female patients with rheumatoid arthritis (RA), who were hospitalized for two 14-day periods, one of which included 7 days of total fasting, whereas the other served as control period with normal food intake. All medical treatment was stopped on admission to the hospital. Plasma amino acid levels were repeatedly determined during both periods. Another group, consisting of 8 healthy volunteers, also underwent total fasting, for 6 days. The response to food deprivation with regard to plasma amino acid levels was compared with that in

Amino acid-specific ADP-ribosylation.

[adenine-U-14C]ADP-ribose-agmatine and [adenine-U-14C ))ADP-ribose-histone were synthesized by an NAD:arginine ADP-ribosyltransferase from [14C]NAD and agmatine and histone, respectively. The pseudo-first order rate constants for breakdown of the two components either in 0.4 N NaOH or in 0.4 M neutral hydroxylamine were identical. Hydroxylamine treatment of [14C]ADP-ribose-agmatine or [32P]ADP-ribose-histone yielded a single radioactive product which was separated by high pressure liquid chromatography and identified as ADP-ribose-hydroxamate by the formation of a ferric chloride complex. Amino acid requirements of Legionella pneumophila.

The amino acids required for growth and as energy sources by 10 strains of Legionella pneumophila were determined by using a chemically defined medium. All strains required arginine, cysteine, isoleucine, leucine, threonine, valine, methionine, and phenylalanine or tyrosine. Most strains (7 of 10) required serine, and two strains had to be supplied proline before growth could be established. All 10 strains used serine and, to a lesser extent, threonine as the sole sources of carbon and energy. The Y serine calculated was 94.9 +/- 8.5 g (dry weight) of cells/mol of serine.
