Amino acids are important for cellular metabolism. Their uptake across the plasma membrane is mediated by transport proteins. Despite the fact that the organic anion transporting polypeptide 4C1 (OATP4C1, Uniprot: Q6ZQN7) mediates transport of l-arginine and l-arginine derivatives, other members of the OATP family have not been characterized as amino acid transporters. The OATP family member OATP3A1 (gene symbol SLCO3A1, Uniprot: Q9UIG8) is ubiquitously expressed in human cells and highly expressed in many cancer tissues and cell lines. However, only a few substrates are known for OATP3A1. Acc

BACKGROUND: Prostate cancer is one of the most frequent malignancies in the world, with significant morbidity and death rates. Identifying novel biomarkers is critical to reducing morbidity and mortality from the illness today. Although lipids have been linked to an increased risk of prostate cancer, it is unclear if the non-high-density lipoprotein cholesterol (NHDL-C) to high-density lipoprotein cholesterol (HDL-C) ratio (NHHR) is related to prostate cancer. Therefore, we assessed the relationship between NHHR and prostate cancer risk.
METHODS: This study comprised 1,718 individuals who att

Two highly water-soluble amino acids, which derived from beta-CDs, i.e., glutamic acid-beta-cyclodextrin (GluCD) and ethylene-diamine-beta-cyclodextrin (EDCD), were synthesized and were examined for their effect on solubilization of anthracene (ANT), complexation of cadmium (Cd2+), and elution removal of ANT and Cd2+ in soil. The results showed that GluCD and EDCD were powerful complexant for ANT and Cd2+. In the presence of 10 g/L GluCD and EDCD, the solubilization of ANT increased by 47.04 and 23.85 times compared to the control, respectively. GluCD resulted in approximately 90% complexation

Glioblastoma (GBM) displays profound iron dependence and metabolic plasticity, yet how iron deprivation interfaces with stress-response pathways and amino acid metabolism in GBM remains incompletely understood. Deferoxamine (DFO), an iron chelator and hypoxia mimetic, is widely used experimentally, but the integration of autophagy, apoptosis, and ferroptosis under DFO-induced stress is unclear. This study aims to clarify how iron chelation reshapes stress signaling and metabolism in GBM cells and to define the role of aromatic amino acid metabolism in autophagy-mediated adaptation to iron depr

Pyridoxal 5'-phosphate (PLP) is an essential cofactor required for metabolic functions including amino acid biosynthesis. While the pathways of PLP synthesis are well characterized, it is unclear how newly synthesized PLP is delivered to apo PLP-dependent enzymes (PLP-DEs) in the cell. The highly conserved PLP Homeostasis Protein (PLPHP) is hypothesized to act as a PLP carrier that traffics the cofactor to PLP-DEs. Prior studies have shown transfer of PLP between PLPHP homologs and PLP-DEs in vitro. Data herein support a role for this transfer in vivo. This study utilizes a Salmonella enterica

Amino acids represent versatile building blocks for the design of functional supramolecular architectures, combining inherent chirality with a rich palette of non-covalent interactions. In recent years, considerable effort has been devoted to integrating amino acid motifs into π-conjugated aromatic cores such as naphthalene diimides (NDIs), benzene-1,3,5-tricarboxamides (BTAs) and other synthetically accessible scaffolds. These systems exemplify the power of modular molecular design, where subtle variations in side chains or peptide fragments can precisely tune self-assembly pathways, supramol

BACKGROUND: Aromatic l-amino acid decarboxylase (AADC), or dopa decarboxylase (DDC), is a key enzyme in levodopa metabolism. Previous studies suggested that AADC protein levels may be increased in Parkinson's disease (PD), particularly if treated with levodopa in conjunction with a peripheral decarboxylase inhibitor (PDI).
METHODS: Serum AADC enzyme activity was measured by quantifying ex vivo conversion of levodopa to dopamine, and correlated to diagnosis, disease stage and clinical parameters in (i) n = 43 people with probable prodromal PD; (ii) n = 592 people with early PD (≤5 years since

BACKGROUND: Aromatic L-amino acid decarboxylase (AADC) deficiency is an autosomal recessive neurotransmitter metabolism disorder and is clinically characterized by infancy hypotonia, ophthalmic crisis, and developmental delay. With the emergence of gene therapy for AADC deficiency, accurate prediction of AADC deficiency is required. This study aimed to analyze the carrier frequency and expected incidence of AADC deficiency using exome data from the Genome Aggregation Database (gnomAD).
METHODS: We analyzed 125,748 exomes from gnomAD, including 9197 East Asian exomes, for the DDC gene. All ide

Approximately one-third of the human population is infected with the intracellular cosmopolitan protozoan Toxoplasma gondii (Tg), and a specific treatment for this parasite is still needed. Additionally, the increasing resistance of Tg to drugs has become a challenge for numerous research centers. The high selectivity of a compound toward the protozoan, along with low cytotoxicity toward the host cells, form the basis for further research, which aims at determining the molecular targets of the active compounds. Thiosemicarbazide derivatives are biologically active organic compounds. Previous s

In patients with aromatic l-amino acid decarboxylase (AADC) deficiency, a decrease in catecholamines and serotonin levels in the brain leads to developmental delay and movement disorders. The beneficial effects of gene therapy in patients from 1 to 8 years of age with homogeneous severity of disease have been reported from Taiwan. We conducted an open-label phase 1/2 study of population including adolescent patients with different degrees of severity. Six patients were enrolled: four males (ages 4, 10, 15 and 19 years) and one female (age 12 years) with a severe phenotype who were not capable

The Toxoplasma gondii genome contains two aromatic amino acid hydroxylase genes, AAH1 and AAH2 encode proteins that produce L-DOPA, which can serve as a precursor of catecholamine neurotransmitters. It has been suggested that this pathway elevates host dopamine levels thus making infected rodents less fearful of their definitive Felidae hosts. However, L-DOPA is also a structural precursor of melanins, secondary quinones, and dityrosine protein crosslinks, which are produced by many species. For example, dityrosine crosslinks are abundant in the oocyst walls of Eimeria and T. gondii, although

The aromatic amino acid hydroxylase (AAAH) enzyme family includes phenylalanine hydroxylase (PAH), tyrosine hydroxylase (TH) and the tryptophan hydroxylases (TPH1 and TPH2). All four members of the AAAH family require iron, dioxygen and the cofactor (6R)-L-erythro-5,6,7,8-tetrahydrobiopterin (BH4) to hydroxylate their respective substrates. The AAAHs are involved in severe diseases; whereas polymorphisms and variants in the TPH genes are associated to neuropsychiatric disorders, mutations in PAH and TH are responsible for the autosomal recessive disorders phenylketonuria (PKU) and TH deficienc

Toxoplasma gondii infection has been described previously to cause infected mice to lose their fear of cat urine. This behavioral manipulation has been proposed to involve alterations of host dopamine pathways due to parasite-encoded aromatic amino acid hydroxylases. Here, we report successful knockout and complementation of the aromatic amino acid hydroxylase AAH2 gene, with no observable phenotype in parasite growth or differentiation in vitro and in vivo. Additionally, expression levels of the two aromatic amino acid hydroxylases were negligible both in tachyzoites and in bradyzoites. Final

Autoantibodies against aromatic L-amino acid decarboxylase (AADC) are present in about 50 percent of sera from patients with autoimmune polyendocrine syndrome type I (APS I) but absent in sera from patients with different organ-specific autoimmune diseases, such as insulin-dependent diabetes mellitus, Hashimoto's thyroiditis, and Graves' disease. AADC is expressed in the pancreatic beta-cells, the liver, and the nervous system; and the presence of AADC antibodies has been shown to correlate to hepatitis and vitiligo in APS I patients. Among 101 investigated patients with autoimmune Addison's d

Patients with autoimmune polyendocrine syndrome type I (APS I) have autoantibodies against the enzyme aromatic L-amino acid decarboxylase (AADC) of pancreatic beta-cells. The aim of the present study was to investigate the presence of anti-AADC antibodies in a large cohort of patients with APS I, and in patients with isolated insulin-dependent diabetes mellitus (IDDM). We found autoantibodies against AADC in 35 of 69 patients (51%) with APS I but in none of 138 patients with isolated IDDM or 91 healthy controls. Among the patients with APS I, anti-AADC antibodies were more often found in those

The N-terminal portion of the third intracellular loop (i3) of muscarinic acetylcholine and other G protein-coupled receptors has been shown to largely determine the G protein coupling profile of a given receptor subtype. Using the rat m3 muscarinic receptor as a model system, we have recently demonstrated that a tyrosine residue (Tyr-254), located at the beginning of the i3 domain, is critically involved in muscarinic receptor-mediated stimulation of phosphatidylinositol (PI) hydrolysis (Blüml, K., Mutschler, E., and Wess, J. (1994) J. Biol. Chem. 269, 402-405). This study was designed to inv

Glutamate, the major excitatory neurotransmitter, is critical for normal brain development and function. Both extremes of glutamate receptor activity are detrimental for the brain. Glutamate's role in excitotoxicity has driven the search for receptor antagonists as neuroprotective agents, most of which have failed to achieve clinical, i.e. efficacious and safe, neuroprotection. High selectivity and potency provide potential explanations for this failure. For example, targeting individual glutamate receptor subtypes leaves other pathways of glutamatergic excitotoxicity intact. Furthermore, pote

The human microbiota produces a diverse array of bioactive molecules, including classic neurotransmitters (dopamine and serotonin) and trace amines (tryptamine, tyramine, and phenylethylamine). Although long considered products of host metabolism, these aromatic monoamines are now also known to originate in part from the microbiota, where they are synthesized by bacterial aromatic L-amino acid decarboxylases (AADCs). This review explores the distribution, biochemical diversity, and host interactions of microbiota-encoded AADCs, highlighting their roles in gut and skin ecosystems. Bacterial AAD
