[The influence of selected antibiotics on the central action of aminophyllines--experimental studies].

Methylxanthines and some antibiotics can cause side effects, provoked by their central action, e.g. seizures. The epileptogenic effects of given drugs can be intensified during combined treatment, as a result of pharmacological interactions. In the present study the author investigated the influence of some commonly used antibiotics: benzylpenicillin, cefuroxime, doxycycline and amikacin upon central activity of methylxanthines in mice. The obtained results suggest, that all tested antibiotics, mainly benzylpenicillin, enhanced epileptogenicity of aminophylline in chemical seizures test. benzy

Carotid bodies and ventilatory response to hypoxia in aminophylline-treated piglets.

Peripheral chemoreceptors may be immature in neonatal animals, exhibiting maturational changes in the perinatal period. Even though methylxanthines are respiratory stimulants, many premature neonates do not respond to them. Thus, we hypothesized that carotid body activity is necessary for aminophylline to reverse hypoxia-induced respiratory depression. We exposed 16 anesthetized newborn piglets (age 2-7 days) to hypoxia (inhalation of 12% oxygen) for 5 min. Aminophylline (15 mg/kg iv) was administered either prior to (11 piglets) or following (5 piglets) carotid body denervation (CBD). Before

The reversal effect of low dose aminophylline on thiopental-induced sedation.

Fifty-two ASA (American Society of Anesthesiologists) class I-II female patients, from 22 to 66 years of age, undergoing D & C (dilatation and curettage) procedure, were randomly divided into 2 groups in a double-blind and placebo-controlled design to investigate whether aminophylline could reverse the sedative effect of thiopental. Patients of both groups were anesthetized with fentanyl (2 micrograms/kg, i.v.) and thiopental (4 mg/kg, i.v. and 1 mg/kg, i.v. repeated if necessary). Postoperatively, they were intravenously given aminophylline (2 mg/kg) or normal saline in an equal volume soluti

Effect of aminophylline and verapamil upon diaphragmatic force generation in dogs.

The effects on the diaphragm of verapamil (VPM) and aminophylline (AMP) were studied in dogs during stimulation of phrenic nerves at the 5th cervical roots (5th-PS) or transvenously at the trunk (T-PS). Transdiaphragmatic pressure (Pdi)/frequency curves were constructed. Our findings were: 1) AMP increased Pdi at all stimulation frequencies (p less than 0.01) during 5th-PS but only at 10-30 Hz during T-PS; 2) in other dogs infusion of VPM (0.14 mg.kg-1.min-1) decreased Pdi at all frequencies (p less than 0.025) without change in diaphragmatic blood flow; 3) the effects of VPM were completely r

Aminophylline effects on ventilatory response to hypoxia and hyperoxia in normal adults.

In 10 normal young adults, ventilation was evaluated with and without pretreatment with aminophylline, an adenosine blocker, while they breathed pure O2 1) after breathing room air and 2) after 25 min of isocapnic hypoxia (arterial O2 saturation 80%). With and without aminophylline, 5 min of hyperoxia significantly increased inspiratory minute ventilation (VI) from the normoxic base line. In control experiments, with hypoxia, VI initially increased and then declined to levels that were slightly above the normoxic base line. Pretreatment with aminophylline significantly attenuated the hypoxic v

[Effect of aminophylline therapy in mature infants with sleep apnea syndrome].

Pneumograms of 33 fullterm infants (age 1-16 weeks) with idiopathic sleep apnea syndrome (SAS), treated with aminophyllin administered orally, were compared with pneumograms of 12 age-matched infants without aminophyllin treatment. In a one hour oxycardiorespirography (OCRG) all infants had pneumogram abnormalities defined as apneas greater than or equal to 15 s, greater than or equal to 3 apneas lasting 10 s, MA-value (mean duration of all apneas during sleep time) greater than or equal to 7 s/min, and greater than or equal to 3 episodes of periodic breathing. Hypokalaemic and electrocardiographic effects of aminophylline and salbutamol in obstructive airways disease.

The hypokalaemic and electrocardiographic effects of nebulised salbutamol (2.35 mg x 2, 120 min apart) and intravenous aminophylline (6 mg/kg followed by 0.5 mg/kg/hr infusion) alone and in combination, were investigated in eight patients with stable asthma. Both agents produced significant hypokalaemia, mean maximum for salbutamol 0.55 mmol/l, mean maximum for aminophylline 0.29 mmol/l. Salbutamol increased the QTc interval and depressed T-wave amplitude whilst aminophylline decreased PR interval. These effects were increased when they were given in combination but were highly variable betwee

Aminophylline may stimulate respiration in rats by activation of dopaminergic receptors.

In intact young rats anesthetized with halothane, aminophylline produces an increase in respiratory minute ventilation due primarily to an increase in respiratory frequency. Although the simulation of respiration by inhalation of 10% CO2 is augmented only at high doses of aminophylline, the response to CO2 is increased at doses as low as 3 mg/kg of aminophylline. Division of the ninth cranial nerve together with removal of the carotid body did not alter the response, whereas vagotomy changed the response from one of increased frequency to an increase in tidal volume. Both awake and anesthetize

Theophylline toxicity after the use of aminophylline in the treatment of cerebral vasospasm.

We are reporting a case of theophylline toxicity after the initiation of i.v. aminophylline and isoproterenol in the treatment of cerebral vasospasm. The dose of aminophylline (125 mg/hour) previously recommended results in a toxic serum theophylline concentration in a majority of patients if the infusion is continued for more than 24 to 48 hours. The rapid achievement of an optimal serum level is obtained with a standard loading dose of 8.0 mg/kg. The initial infusion rate should be based on calculated pharmacokinetic data. Maintenance dosing adjustments are then based on serum determinations

Aminophylline modulation of the mouse respiratory network changes during postnatal maturation.

Aminophylline is a respiratory stimulant commonly used for the treatment of central apnea. Experiences from clinical practice, however, revealed that aminophylline is not reliably effective in preterm infants, whereas it is normally effective in infants and mature patients. In an established animal model for postnatal development of respiratory control mechanisms, we therefore examined the hypothesis that the clinical observations reflect a developmental change in the sensitivity of the central respiratory network to methylxanthines. The medullary respiratory network was isolated at different

Salbutamol attenuates arrhythmogenic effect of aminophylline in a hPSC-derived cardiac model.

The combination of aminophylline and salbutamol is frequently used in clinical practice in the treatment of obstructive lung diseases. While the side effects (including arrhythmias) of the individual bronchodilator drugs were well described previously, the side effects of combined treatment are almost unknown. We aimed to study the arrhythmogenic potential of combined aminophylline and salbutamol treatment in vitro. For this purpose, we used the established atomic force microscopy (AFM) model coupled with cardiac organoids derived from human pluripotent stem cells (hPSC-CMs). We focused on the

Aminophylline for the treatment of symptomatic bradycardia and asystole secondary to cervical spine injury.

INTRODUCTION: Bradycardia is a common complication of cervical spine damage in the weeks following injury, occurring in up to 100% of patients in some studies. Cardiac arrest and asystole have been reported in as many as 15% of these patients and cardiac events are the main cause of death within the first year. We describe the case of a 25-year-old African-American male involved in a motor vehicle collision who suffered C6-C7 subluxation. METHODS: Following cervical discectomy and spinal fusion the patient began to develop progressive bradycardia culminating on hospital day 20 with two asysto

Aminophylline treatment in meconium-induced acute lung injury in a rabbit model.

Administration of methylxanthines may diminish meconium-induced acute lung injury. Meconium-instilled rabbits intravenously received aminophylline (2.0 mg/kg) at two doses 0.5 h and 2.5 h after meconium instillation or were left without treatment, and were oxygen-ventilated for additional 5 h. At the end of experiment, lungs and trachea were excised. Within 5 h after the first dose of treatment, aminophylline significantly improved gas exchange and decreased right-to-left pulmonary shunts, central venous pressure, and ventilatory pressures. Moreover, aminophylline reduced meconium-induced lung

[Effects of aminophylline on proliferation and apoptosis in Raji lympho-blastoid cell line].

The aim of this study was to investigate whether and how phosphodiesterase (PDE) inhibitors modulate the proliferation, cell cycle and apoptosis in lymphoma cells. The effects of aminophylline (AM), a non-specific PDE inhibitor, on Raji cells were explored in vitro. MTT assay, light and transmission electron microscopy and annexin V staining were used to observe cell proliferation, morphologic changes and apoptosis rate in AM-treated cells, and FCM and RT-PCR techniques were adopted to detect the effect on cell cycle, the expression of cyclin B1 and Bcl-2 and mitochondrial transmembrane potent

Lack of effect of olanzapine on the pharmacokinetics of a single aminophylline dose in healthy men.

STUDY OBJECTIVE: To test whether olanzapine, an atypical antipsychotic, is an inhibitor of cytochrome P450 (CYP) 1A2 activity, we conducted a drug interaction study with theophylline, a known CYP1A2 substrate. DESIGN: Two-way, randomized, crossover study. SETTING: Clinical research laboratory. SUBJECTS: Nineteen healthy males (16 smokers, 3 nonsmokers). INTERVENTIONS: Because the a priori expectation was no effect of olanzapine on theophylline pharmacokinetics, a parallel study using cimetidine was included as a positive control. In group 1, 12 healthy subjects received a 30-minute intrave
