The management of Babesia, amoeba and other zoonotic diseases provoked by protozoa.

INTRODUCTION: There are 12 protozoan genera that provoke zoonotic disease in humans and animals. We discuss the most common ones with a highlight on Babesia spp and Entamoeba histolytica, also mentioning Toxoplasma gondii, Trypanosoma cruzi, and Leishmania spp. AREAS COVERED: The complex life cycle of pathogenic protozoans is deeply understood but this did not contribute to the discovery of new drugs. The clinical armamentarium is poor and includes antiinfectives originally proposed as antibacterial (azithromycin, clindamycin, paromomycin, sulfadrugs), antifungals (amphotericin B), or they ar

An intracellular replication niche for Vibrio cholerae in the amoeba Acanthamoeba castellanii.

Vibrio cholerae is a human pathogen and the causative agent of cholera. The persistence of this bacterium in aquatic environments is a key epidemiological concern, as cholera is transmitted through contaminated water. Predatory protists, such as amoebae, are major regulators of bacterial populations in such environments. Therefore, we investigated the interaction between V. cholerae and the amoeba Acanthamoeba castellanii at the single-cell level. We observed that V. cholerae can resist intracellular killing. The non-digested bacteria were either released or, alternatively, established a repli

Social amoeba farmers carry defensive symbionts to protect and privatize their crops.

Agricultural crops are investments that can be exploited by others. Farmer clones of the social amoeba Dictyostelium discoideum carry bacteria to seed out new food populations but they also carry other non-food bacteria such as Burkholderia spp. Here we demonstrate that these farmer-carried Burkholderia inhibit the growth of non-farmer D. discoideum clones that could exploit the farmers' crops. Using supernatants, we show that inhibition is due to molecules secreted by Burkholderia. When farmer and non-farmer amoebae are mixed together at various frequencies and allowed to complete the social

Serologic prevalence of amoeba-associated microorganisms in intensive care unit pneumonia patients.

BACKGROUND: Patients admitted to intensive care units are frequently exposed to pathogenic microorganisms present in their environment. Exposure to these microbes may lead to the development of hospital-acquired infections that complicate the illness and may be fatal. Amoeba-associated microorganisms (AAMs) are frequently isolated from hospital water networks and are reported to be associated to cases of community and hospital-acquired pneumonia. METHODOLOGY/PRINCIPAL FINDINGS: We used a multiplexed immunofluorescence assay to test for the presence of antibodies against AAMs in sera of intens

Photophobic and phototactic responses of Amoeba proteus in KCN and SHAM solutions.

The effects of the metabolic inhibitors KCN and SHAM on phototaxis and photophobic response in Amoeba proteus have been studied. Both drugs neither change amoebae photophobic response nor the phototactic reaction. The results indicate clearly that the negative phototactic orientation is not impaired by impediment of respiration thus it is not directly coupled to the differences in energy production in different parts of A. proteus body.

Morphological alterations in the mitochondria of Amoeba proteus induced by uncoupling agents.

Ultrastructural changes in mitochondrial morphology were observed in amoebae exposed to the uncoupling agents dinitrophernol (DNP), pentachlorophenol (PCP), and m-chlorocarbonyl cyanide phenylhydrazone (CCCP). These alterations occurred with rapidity and were present before whole-cell activity changes could be detected. They included changes in profile shape and overall dimensions, matrix density changes, and alterations to the cristal membranes, so that distinction between control Type I and Type II conformations was eliminated and a form with intermediate characters generated. It is proposed

Phosphates and Phosphorylated Amino Acids for the AMOEBA-HFC Polarizable Force Field.

Protein phosphorylation plays a crucial role in regulating protein activity, localization, and signaling in many biological processes. While nonpolarizable models for molecular dynamics (MD) simulations include parameters for phosphorylated amino acids, their counterparts for polarizable models are lacking. This limits the ability to understand environment-dependent effects associated with phosphorylation processes such as electric-field-induced polarization and nonadditive many-body energetic contributions. Such effects are expected to contribute substantially due to phosphate's high negative

Computational discovery of potential therapeutic agents against brain-eating amoeba (Naegleria fowleri).

Naegleria fowleri is a human and animal pathogen well-known for its ability to digest neurons and astrocytes of the host's brain, causing a haemorrhagic and necrotizing inflammation called Primary Amoebic Meningoencephalitis. Although infections are rare, the mortality rate is over 97%, due to both the non-specificity of the symptoms and the absence of an effective treatment. In this work we employed bioinformatics tools to evaluate the possibility of treating the infection with tubulin-targeting compounds, which we regard as the most promising approach given the unclear view on the pathogenic

Adaptation of Amoeba Plate Test To Recover Legionella Strains from Clinical Samples.

The isolation of Legionella from respiratory samples is the gold standard for diagnosis of Legionnaires' disease (LD) and enables epidemiological studies and outbreak investigations. The purpose of this work was to adapt and to evaluate the performance of an amoebic coculture procedure (the amoeba plate test [APT]) for the recovery of Legionella strains from respiratory samples, in comparison with axenic culture and liquid-based amoebic coculture (LAC). Axenic culture, LAC, and APT were prospectively performed with 133 respiratory samples from patients with LD. The sensitivities and times to r

Icm/dot-independent entry of Legionella pneumophila into amoeba and macrophage hosts.

Legionella pneumophila, the causative agent of Legionnaires' disease, expresses a type IVB secretion apparatus that translocates bacterial proteins into amoeba and macrophage hosts. When stationary-phase cultures are used to infect hosts, the type IVB apparatus encoded by the icm/dot genes is required for entry, delay of phagosome-lysosome fusion, and intracellular multiplication within host cells. Null mutants with mutations in icm/dot genes are defective in these phenotypes. Here a new model is described in which hosts are infected with stationary-phase cultures that have been incubated over

Amoeba angeitic lesions of the central nervous system in Balamuthia mandrilaris amoebiasis.

Balamuthia mandrilaris amoebiasis is a fatal disease. It primarily affects the nasal pyramid or the skin, producing granulomatous amoebic lesions. The amoeba spread from the primary nasal lesion to the meninges where they infiltrate vessels. Thrombotic amoebic angitis produce infarcts of the central nervous system substance which then become infiltrated by amoeba. The primary cutaneous lesion can be present for weeks or even months. However, the appearance of neurological disease predicts a poor prognosis, in which death usually occurs within a few days or weeks.

Relationship between free amoeba and Legionella: studies in vitro and in vivo.

In 1980, Robowtham demonstrated that Legionella multiplies in free amoeba cytoplasm and hypothesized that the amoeba could act as a reservoir of virulent bacteria. In this paper we report various aspects of the relationship between amoeba and Legionella. A liquid medium co-culture method was applied to Acanthamoeba sp. and Legionella pneumophila serogroup 1. Within 4 days, Legionella growth increased by 2 log s CFU/ml. Using a direct immunofluorescence assay and electron microscopy, Legionella was shown to grow abundantly inside phagosomes, and bacteria and/or antigen were present on the cytop

Amoebic meningo-encephalitis due to a free-living amoeba. A case report.

A 47-year-old man developed an undiagnosed encephalitic illness and died within 12 days. Autopsy showed focal granulomatous lesions associated with a necrotizing vasculitis. Light microscopy revealed the presence of free-living amoebae morphologically resembling Acanthamoeba species. The pathology of this form of meningo-encephalitis is contrasted with that caused by Naegleria fowleri infection, and attention is drawn to its existence within the RSA.

Bacterial microbiota management in free-living amoebae (Heterolobosea lineage) isolated from water: The impact of amoebae identity, grazing conditions, and passage number.

Free-living amoebae (FLA) are ubiquitous protozoa mainly found in aquatic environments. They are well-known reservoirs and vectors for the transmission of amoeba-resistant bacteria (ARB), most of which are pathogenic to humans. Yet, the natural bacterial microbiota associated with FLA remains largely unknown. Herein, we characterized the natural bacterial microbiota of different FLA species isolated from recreational waters in Guadeloupe. Monoxenic cultures of Naegleria australiensis, Naegleria sp. WTP3, Paravahlkampfia ustiana and Vahlkampfia sp. AK-2007 (Heterolobosea lineage) were cultivate

Transgenic approaches in the pathogenic free-living amoebae: what are the hurdles?

Naegleria fowleri, Acanthamoeba spp., Balamuthia mandrillaris and Sappinia spp. are free-living amoebae that can infect humans and cause serious disease; therefore, these organisms are commonly referred to as pathogenic free-living amoebae (pFLA). Diagnosis and treatment of pFLA infections have historically been very challenging. If a diagnosis is made, treatment regimens currently include a combination of antifungals, antimicrobials and anticancer agents that have, to date, proven to be of little help in resolving the disease. Discovery of new therapies is critical to reduce the >90% mortalit

Emerging therapies against infections due to free-living amoebae.

INTRODUCTION: Pathogenic free-living amoebae, including Naegleria fowleri, Acanthamoeba spp. and Balamuthia mandrillaris, are rare but frequently fatal opportunistic pathogens associated with severe central nervous system and ocular infections, and cutaneous infections. Despite advances in understanding their biology and pathogenesis, therapeutic options remain limited, mortality rates remain high, and delayed diagnosis continues to compromise clinical outcomes and public health responses. Neuroinvasive Free-Living Amoebae Pathogenesis, Neuroinflammation and Therapeutic Challenges.

Neuroinvasive free-living amoebae (FLA), particularly Naegleria fowleri and Acanthamoeba spp., are responsible for rare but devastating infections of the central nervous system (CNS). Approximately 480 cases of primary amoebic meningoencephalitis (PAM) and fewer than 200 well-documented cases of Acanthamoeba-associated granulomatous amoebic encephalitis (GAE) have been reported worldwide. Mortality rates frequently exceed 90%. PAM typically develops following exposure to warm freshwater contaminated with N. fowleri and progresses rapidly in otherwise healthy individuals. In contrast, GAE usual

[The fascinating world of free-living amoebae, these remarkably versatile protists].

Free-living amoebae, microorganisms with variable forms, are essential to the ecology. However, some species, such as Naegleria fowleri, Acanthamoeba spp., and Balamuthia mandrillaris, cause severe, though rare, infections including meningitis and keratitis, which are often linked to water. Free-living amoebae are also reservoirs and vectors of various pathogens (bacteria, viruses, and fungi), which facilitates their dissemination and the potential emergence of diseases. Free-living amoebae can also be used as models to study interactions between eukaryotes and prokaryotes. This review explore
