A review of bovine anaplasmosis (Anaplasma marginale) with emphasis on epidemiology and diagnostic testing.

Bovine anaplasmosis, caused by the rickettsia Anaplasma marginale, is an economically important tick-borne disease of cattle that is found worldwide. Its clinical effects of severe anemia, decreased growth, weight loss, and death negatively impact cattle welfare and create a significant economic burden for cattle producers. Despite availability of highly sensitive and specific assays for anti-A. marginale antibodies (competitive ELISA) and A. marginale genetic material (real-time PCR), the interpretation of test results for the diagnosis of clinical anaplasmosis in cattle remains challenging.

A rare case of delayed anaplasma phagocytophilum-induced pancytopenia: A diagnostic conundrum.

INTRODUCTION: Human granulocytic anaplasmosis (HGA) is a potentially fatal tick-borne disease caused by the obligate intracellular bacterium Anaplasma phagocytophilum. It is most commonly found in the Northeastern and Midwestern parts of the United States especially during spring and summer months. The clinical picture of anaplasmosis is varied ranging from common symptoms such as fever, headache and myalgia to rarer presentations such as pancytopenia. CASE PRESENTATION: We present a case of a 62 year old male who presented with watery diarrhea, fever, and pancytopenia. Although there is a br

Tick infestation and occurrence of Anaplasma phagocytophilum and piroplasms in cattle in the Republic of Serbia.

Ticks transmit important pathogens affecting cattle such as intracellular bacteria of the genus Anaplasma or protozoa of the order Piroplasmida. This study aimed at assessing tick species present on pastures and cattle and determining occurrence of the tick-borne pathogens Anaplasma phagocytophilum and Babesia spp. in cattle. During May and June 2013, ticks and EDTA blood were collected from 185 dairy cows at six locations in Serbia. Ticks were also collected directly from the pastures at four of these locations. The occurrence of A. phagocytophilum was investigated by serological (immunofluor

Co-infection of Trypanosoma pestanai and Anaplasma phagocytophilum in a dog from Germany.

Infections with arthropod-borne pathogens are an increasing threat world-wide that requires heightened vigilance from veterinary and medical practitioners, especially when they involve new or unusual organisms. A dog was presented to a local veterinary clinic in Germany with malaise, pale mucous membranes and stiff joints. Clinical assessment revealed pyrexia, leukopenia and thrombocytopenia. On suspicion of a tick-borne infection, blood samples were examined for clinical and biochemical parameters and subjected to a Anaplasma phagocytophilum-, Borrelia spp.- and Ehrlichia canis-specific real-

Molecular Epidemiological Survey and Genetic Characterization of Anaplasma Species in Mongolian Livestock.

Anaplasma species are obligate intracellular rickettsial pathogens that cause great economic loss to the animal industry. Few studies on Anaplasma infections in Mongolian livestock have been conducted. This study examined the prevalence of Anaplasma marginale, Anaplasma ovis, Anaplasma phagocytophilum, and Anaplasma bovis by polymerase chain reaction assay in 928 blood samples collected from native cattle and dairy cattle (Bos taurus), yaks (Bos grunniens), sheep (Ovis aries), and goats (Capra aegagrus hircus) in four provinces of Ulaanbaatar city in Mongolia. We genetically characterized posi

Factors associated with Anaplasma spp. seroprevalence among dogs in the United States.

BACKGROUND: Dogs in the United States are hosts to a diverse range of ticks and tick-borne pathogens, including A. phagocytophilum, an important emerging canine and human pathogen. Previously, a Companion Animal Parasite Council (CAPC)-sponsored workshop proposed factors purported to be associated with the infection risk for tick-transmitted pathogens in dogs in the United States, including climate conditions, socioeconomic characteristics, local topography, and vector distribution. METHODS: Approximately four million test results from routine veterinary diagnostic tests from 2011-2013, which

[Lyme borreliosis and co-infections. Place of Anaplasma phagocytophilum and Bartonella henselae].

Lyme borreliosis (LB) is certainly the most common infection transmitted through the bite of Ixodes in Northern Hemisphere. These ticks are also able to transmit other microorganisms such as the bacteria Anaplasma phagocytophilum (Ap) and Bartonella henselae (Bh), with the latter discovered fairly recently, leading to diferent clinical presentations often close to those of LB. The aim of this study was to evaluate the frequency of co-infection by either of these bacteria in patients with LB, particularly when a treatment with beta-lactam antibiotic was only partially effective. Of these patien

Introduction to the alpha-proteobacteria: Wolbachia and Bartonella, Rickettsia, Brucella, Ehrlichia, and Anaplasma.

Wolbachia is an obligate intracellular endosymbiont and likely mutualist living within the heartworm Dirofilaria immitis and a number of other filarial nematodes in the family Onchocercidae. The bacterial infection is passed from worm to worm transovarially; the organisms are in ovarian cells, the developing microfilariae, and multiply and persist in all later developmental stages through the mosquito and into the next host. Besides being present in the ovaries of the adult worms, they also are present in large numbers within the hypodermal tissues of the nematode. It is now know that these ba

Multiplex real-time PCR for detection of anaplasma phagocytophilum and Borrelia burgdorferi.

A multiplex real-time PCR assay was developed for the simultaneous detection of Anaplasma phagocytophilum and Borrelia burgdorferi. The assay was tested on various Anaplasma, Borrelia, Erhlichia, and Rickettsia species, as well as on Bartonella henselae and Escherichia coli, and the assay was found to be highly specific for A. phagocytophilum and the Borrelia species tested (B. burgdorferi, B. parkeri, B. andersonii, and B. bissettii). The analytical sensitivity of the assay is comparable to that of previously described nested PCR assays (A. phagocytophilum, 16S rRNA; B. burgdorferi, fla gene)

Conserved recombinant antigens of Anaplasma marginale and Babesia equi for serologic diagnosis.

The competitive inhibition ELISA (CI-ELISA) format overcomes problems associated with antigen purity since the specificity of the CI-ELISA depends solely on the monoclonal antibody (mAb) used. Therefore, the CI-ELISA format is well suited for use with recombinant antigens. Molecular clones expressing a conserved 19 kDa protein of Anaplasma marginale and a 34 kDa protein of Babesia equi were derived and characterized. The 19 kDa A. marginale protein, conserved in all recognized Anaplasma species, and present in the infected tick salivary gland, was reactive with all bovine immune sera tested. T

A case of transfusion-transmission Anaplasma phagocytophilum from leukoreduced red blood cells.

BACKGROUND: Anaplasma phagocytophilum is a tick-borne bacterium and the cause of human granulocytic anaplasmosis (HGA). Here, we report a case of transfusion-transmitted (TT)-HGA involving a leukoreduced (LR) red blood cell (RBC) unit. CASE REPORT: A 64-year-old woman with gastric adenocarcinoma and multiple myeloma who received weekly blood transfusions developed persistent fevers, hypotension, and shortness of breath 1 week after receiving an RBC transfusion. Persistent fevers, new thrombocytopenia, and transaminitis suggested a tick-borne infection. RESULTS: The absence of blood parasites

Adaptation of Anaplasma phagocytophilum to the tick vector is controlled by the transcriptional regulator Tr1.

Rickettsial pathogens are strictly dependent on the cellular biology of their hosts for survival and replication. Predominantly transmitted by blood-feeding arthropods, these vector-borne pathogens are forced to adapt between the disparate environments of their mammalian host and arthropod vector. To achieve this, the Rickettsial bacteria Anaplasma phagocytophilum undergo extensive transcriptional reprogramming, with over 41% of its genes differentially transcribed between mammals and Ixodes scapularis ticks. How the bacterium orchestrates this dramatic transcriptional reprogramming is not und

Prevalence and genetic diversity of Theileria and Anaplasma species infecting cattle in Paraguay.

Bovine theileriosis and anaplasmosis are major tick-borne diseases affecting cattle health and productivity worldwide. Although cattle farming significantly contributes to Paraguay's economy, limited epidemiological data on the causative agents hinder the formulation of disease control measures. To address this challenge, the present study investigated the prevalence, risk factors, and genetic diversity of Theileria annulata, Theileria orientalis, and Anaplasma marginale among cattle populations in Paraguay. Blood DNA samples from 326 cattle in nine departments of Paraguay were screened with t
