Nitrogen Sources Reprogram Carbon and Nitrogen Metabolism to Promote Andrographolide Biosynthesis in Andrographis paniculata (Burm.f.) Nees Seedlings.

Carbon (C) and nitrogen (N) metabolisms participate in N source-regulated secondary metabolism in medicinal plants, but the specific mechanisms involved remain to be investigated. By using nitrate (NN), ammonium (AN), urea (UN), and glycine (GN), respectively, as sole N sources, we found that N sources remarkably affected the contents of diterpenoid lactone components along with C and N metabolisms reprograming in Andrographis paniculata, as compared to NN, the other three N sources raised the levels of 14-deoxyandrographolide, andrographolide, dehydroandrographolide (except UN), and neoandrog

Dealkenylation of neoandrographolide, a phytochemical from Andrographis paniculata stimulates FXR (Farnesoid X Receptor) and enhances gallstone dissolution.

FXR (Farnesoid X Receptor) is one of the nuclear receptors expressed in the liver performing a significant role in the maintenance of bile acid concentration. An imbalance of cholesterol and bile acid ratio due to any undefined reason could cause gallstone formation. Hence, this paper aims to screen phytochemicals that could maintain a requisite balance of cholesterol and bile acid by targeting FXR and thereby contributing to the dissolution of gallstone. Nineteen phytochemicals were selected and queried for Pa and Pi in the way2drug online server for hepatoprotective property, cholesterol syn

Mixed nitrogen forms coordinate growth and andrographolide accumulation in Andrographis paniculata via sulphur metabolism and abscisic acid signalling.

BACKGROUND: Mixed nitrogen (N) sources, comprising nitrate (NO3-) and ammonium (NH4+), have been shown to enhance plant growth and secondary metabolite accumulation in medicinal plants; however, the underlying mechanisms remain largely unclear. METHODS: This study investigated the effects of different nitrogen treatments on Andrographis paniculata, including sole NO3- (NN), a 1:1 ratio of NO3- to NH4+ (NA1), a 1:2 ratio of NO3- to NH4+ (NA2) and sole NH4+ (AN). KEY RESULTS: Treatments containing NH4+ significantly increased the levels of andrographolide, 14-deoxyandrographolide and dehydroan

Andrograpalides A-N, structurally diverse ent-labdane diterpenoids with anti-inflammatory activities from the leaves of Andrographis paniculata.

Andrograpalides A-N (1-14), fourteen previously undescribed ent-labdane-type diterpenoids, together with a previously undescribed naturally occurring diterpenoid (15) and two known analogues (16-17), were isolated from the leaves of Andrographis paniculata. Their structures were unambiguously elucidated via extensive spectroscopic analysis, electronic circular dichroism analysis, and X-ray crystallographic studies. Structurally, compounds 1-8 and 16 are rare cases of 19-nor labdane-type diterpenoids with an unusual C19 carbon skeleton. Notably, compounds 1 and 2 feature an unusual hydroperoxy

In silico molecular docking study of Andrographis paniculata phytochemicals against TNF-α as a potent anti-rheumatoid drug.

Tumor necrosis factor-α (TNF-α) is a proinflammatory cytokine which plays a crucial role in controlling inflammatory responses. The pathway of Rheumatoid arthritis (RA) leading to TNF-alpha is activated by macrophages and quite often by natural killer cells and lymphocytes. In the inflammatory phase, it is believed to be the main mediator and to be anchored with the progression of different diseases such as ankylosing spondylitis, Crohn's disease, and Rheumatoid arthritis (RA). The major goal of this study is to use in silico docking studies to investigate the anti-inflammatory potential of a

Источник пометил публикацию как отозванную

The present investigation looked into the various biomedical potentials of Andrographis paniculata shoot extracts. The results showed that the methanol extract (Met-E) of A. paniculata contains more phytochemicals than the acetone and petroleum ether extracts, including alkaloids, saponins, tannins, phenolics, flavonoids, glycosides, terpenoids, phytosterol, steroids, and protein. Accordingly, the Met-E alone showed considerable bactericidal activity (through agar well diffusion method) against the bacterial pathogens namely Shigella dysenteriae, Bacillus cereus, Salmonella typhi, Enterococcus

Evaluations of Andrographolide-Rich Fractions of Andrographis paniculata with Enhanced Potential Antioxidant, Anticancer, Antihypertensive, and Anti-Inflammatory Activities.

Andrographis paniculata is widely used as a traditional medicine in Asian countries. It has been classified as a safe and non-toxic medicine by traditional Chinese medicine. The investigation of the biological activities of A. paniculata is still focused on the crude extract and isolation of its main active compound, andrographolide, and its derivatives. However, the use of andrographolide alone has been shown to exacerbate unwanted effects. This highlights the importance of developing a fraction of A. paniculata with enhanced efficacy as an herbal-based medicine. In this study, the extraction

Andrographis-mediated chemosensitization through activation of ferroptosis and suppression of β-catenin/Wnt-signaling pathways in colorectal cancer.

Colorectal cancer (CRC) remains one of the leading causes of cancer-related mortality in the USA. As much as 50-60% of CRC patients develop resistance to 5-fluorouracil (5FU)-based chemotherapeutic regimens, attributing the increased overall morbidity and mortality. In view of the growing evidence that active principles in various naturally occurring botanicals can facilitate chemosensitization in cancer cells, herein, we undertook a comprehensive effort in interrogating the activity of one such botanical-andrographis-by analyzing its activity in CRC cell lines [both sensitive and 5FU resistan

Whole transcriptome analysis identifies full-length genes for neoandrographolide biosynthesis from Andrographis alata, an alternate source for antiviral compounds.

Andrographolide and related compounds are effective against several viral diseases, including dengue, COVID-19, influenza, and chikungunya. Andrographis paniculata is the primary source for these compounds, but its availability is limited. A. alata is a potential alternative source, and neoandrographolide (NAG) is the major antiviral compound in this species. Since molecular studies in A. alata are scarce, we sequenced its leaf transcriptome to identify the full-length genes involved in neoandrographolide biosynthesis. We assembled 13.6 Gb RNA-Seq data and generated 81,361 transcripts with 100
