Extracellular Vesicles and Their Role in Osteogenesis.

Extracellular vesicles (EVs) have emerged as promising cell-free therapeutic agents for bone regeneration due to their ability to modulate osteogenesis, angiogenesis, and immune responses. This review summarizes the current knowledge on EV biology, including their classification, biogenesis, and molecular cargo, with a particular focus on their role in bone healing. EVs derived from mesenchymal stromal cells and other cell types promote osteoblast differentiation, inhibit osteoclast activity, enhance vascularization, and modulate inflammation through the transfer of bioactive proteins and non-

The Effect of Resveratrol on Cell Viability in the Burkitt's Lymphoma Cell Line Ramos.

Resveratrol is a polyphenolic natural compound produced by a variety of crops. Currently, resveratrol is considered a multi-target anti-cancer agent with pleiotropic activity, including the ability to prevent the proliferation of malignant cells by inhibiting angiogenesis and curtailing invasive and metastatic factors in many cancer models. However, the molecular mechanisms mediating resveratrol-specific effects on lymphoma cells remain unknown. To begin tackling this question, we treated the Burkitt's lymphoma cell line Ramos with resveratrol and assessed cell survival and gene expression. Ou

Exploration of the mechanism of anlotinib in reversing PD-1 immunotherapy resistance: insights from single-cell sequencing.

Immune checkpoint inhibitors (ICIs) targeting the PD-1/PD-L1 axis have revolutionized cancer therapy, yet primary and acquired resistance remain major clinical obstacles. Dysregulated angiogenesis fuels the development of an immunosuppressive tumor microenvironment, while crosstalk between immunity and angiogenesis further propels tumor immune evasion and treatment resistance. The present study aimed to establish a penpulimab-resistant model, delineate anti-PD-1 resistance traits via single-cell RNA sequencing, and unravel the precise mechanisms through which anlotinib-an anti-angiogenic agent

Beyond tamoxifen new endpoints for breast cancer chemoprevention, new drugs for breast cancer prevention.

Although tamoxifen appears to markedly reduce breast cancer risk in women with a prior diagnosis of atypical hyperplasia or in situ carcinoma, it is not clear what other groups of women receive substantial benefit. Major breast chemoprevention priorities are to (1) develop new agents that (a) have fewer side effects, (b) are effective in ER--as well as tamoxifen-resistant precancerous tissue, and (c) are compatible with hormone therapy; and (2) develop efficient clinical strategies including prognostic and predictive morphologic and molecular biomarkers. Breast tissue may be repeatedly sampled

Characterization of a Porcine Model for Von Willebrand Disease Type 1 and 3 Regarding Expression of Angiogenic Mediators in the Nonpregnant Female Reproductive Tract.

Von Willebrand disease (VWD), a blood coagulation disorder, is also known to cause angiodysplasia. Hitherto, no animal model has been found with angiodysplasia that can be studied in vivo. In addition, VWD patients tend to have a higher incidence of miscarriages for reasons unknown. Thus, we aimed to examine the influence of von Willebrand factor (VWF) on the female reproductive tract histology and the expression and distribution of angiogenic factors in a porcine model for VWD types 1 and 3. The disease-causing tandem duplication within the VWF gene occurred naturally in these pigs, making th

The backbone of progress--preclinical studies and innovations with zoledronic acid.

Bisphosphonates (BPs) are antiresorptive agents that block pathologic bone resorption by inhibiting osteoclast function and later inducing osteoclast apoptosis. These agents localize to bone and break the vicious cycle of bone resorption that results from cross-stimulation between cancer cells and the bone remodeling cells, thereby reducing cancer-induced osteolysis and the tumor burden in bone. Thus nitrogen-containing BPs (N-BPs) have well established clinical benefits in the treatment of bone metastases from solid tumors and bone lesions from multiple myeloma. Preclinical data indicate that

Itraconazole in the Treatment of Nonfungal Cutaneous Diseases: A Review.

INTRODUCTION: The anti-inflammatory and pro-kinetic properties of antibiotics have been widely reported. However, the non-antifungal properties of antifungal agents are less well known and less explored in clinical practice. The purpose of this review was to survey the literature on the non-antifungal use of itraconazole in dermatological practice and the possible modes of action of this agent. METHODS: The PubMed database was searched for relevant articles published up to January 2017. The references in the articles identified by the search were then hand-searched for additional relevant pub

Current Trends in Anti-Cancer Molecular Targeted Therapies: Renal Complications and Their Histological Features.

Among recent advances in cancer treatment, the emergence of novel drugs targeting specific molecules has considerably modulated therapeutic strategies. Despite the efficacy of these agents, renal complications that are distinct from those of conventional chemotherapeutic drugs have been reported. Targeted therapy drugs include monoclonal antibodies and small-molecule agents. Bevacizumab is a monoclonal antibody that targets vascular endothelial growth factor (VEGF) and blocks tumor angiogenesis. This anti-angiogenic effect causes endothelial injury, resulting in "thrombotic microangiopathy-lik

Thalidomide has limited single-agent activity in relapsed or refractory indolent non-Hodgkin lymphomas: a phase II trial of the Cancer and Leukemia Group B.

Thalidomide is an immunomodulatory agent with demonstrated activity in multiple myeloma, mantle cell lymphoma and lymphoplasmacytic lymphoma. Its activity is believed to be due modulation of the tumour milieu, including downregulation of angiogenesis and inflammatory cytokines. Between July 2001 and April 2004, 24 patients with relapsed/refractory indolent lymphomas received thalidomide 200 mg daily with escalation by 100 mg daily every 1-2 weeks as tolerated, up to a maximum of 800 mg daily. Patients had received a median of 2 (range, 1-4) prior regimens. Of 24 evaluable patients, two achieve

Role of dietary polyphenols in the management of peptic ulcer.

Peptic ulcer disease is a multifactorial and complex disease involving gastric and duodenal ulcers. Despite medical advances, the management of peptic ulcer and its complications remains a challenge, with high morbidity and death rates for the disease. An accumulating body of evidence suggests that, among a broad reach of natural molecules, dietary polyphenols with multiple biological mechanisms of action play a pivotal part in the management of gastric and duodenal ulcers. The current review confirmed that dietary polyphenols possess protective and therapeutic potential in peptic ulcer mediat

The Role of Chemopreventive Agents in the Management of Oral Potentially Malignant Disorders and Oral Cancer: A Narrative Review.

BACKGROUND: Oral squamous cell carcinoma (OSCC) is a common and aggressive cancer sometimes subsequent to oral potentially malignant disorders (OPMDs). Notwithstanding progress in surgical, radiotherapeutic, and chemotherapeutic interventions, survival rates for OSCC continue to be inadequate and are associated with significant functional impairment, underscoring the necessity for preventive and interceptive therapeutic approaches. Chemoprevention has emerged as a viable strategy to inhibit, postpone, or reverse oral carcinogenesis by targeting the molecular and cellular processes implicated i

Cardiotoxicity Induced by Targeted Cancer Therapies: Understanding the Risks and Developing Solutions.

BACKGROUND: Cardiotoxicity has emerged as a major clinical concern associated with targeted cancer therapies. While these agents, including tyrosine kinase inhibitors (TKIs), HER2 inhibitors, angiogenesis inhibitors, and immune checkpoint inhibitors, revolutionize cancer treatment by selectively modulating tumor-associated molecular pathways, they inadvertently disrupt cardiovascular homeostasis due to the overlap of signaling mechanisms between tumorigenesis and cardiac physiology. OBJECTIVES: This review aims to elucidate the mechanisms underlying cardiotoxicity induced by targeted cancer t

Targeting Cancer Hallmarks with Melatonin-based Analogues and Hybrids: From Pleiotropic Hormone to Multitarget Anticancer Agents.

Cancer remains one of the leading causes of mortality worldwide and represents a multifactorial disease characterized by the dysregulation of multiple interconnected biological processes, including uncontrolled cell proliferation, resistance to apoptosis, metabolic reprogramming, angiogenesis, chronic inflammation, immune evasion, and metastatic dissemination. Within this complex scenario, melatonin has attracted increasing interest as a pleiotropic oncostatic molecule capable of modulating several cancer hallmarks through both receptor-dependent (MT1/MT2) and receptor-independent mechanisms,

Targeted Metabolic Therapy in Cancer: A Comprehensive Metabolic Treatment Strategy.

Cancer progression arises from the convergence of three domains: intrinsic metabolic and signaling rewiring within tumor cells, microenvironmental conditions that support survival and dissemination, and systemic metabolic dysfunction in the host. These layers collectively drive treatment resistance, immune evasion, and recurrence.  Targeted metabolic therapy (TMT) is proposed as a comprehensive therapeutic framework designed to simultaneously target all three domains of cancer progression. TMT targets key metabolic pathways implicated in tumor growth and survival such as glycolysis, glutaminol

Recent Advances in Natural Polymers-based Hydrogels for Periodontal Regeneration.

Conventional therapies still provide only limited true tissue regeneration for periodontal diseases, particularly periodontitis, which are highly prevalent chronic inflammatory conditions associated with irreversible loss of tooth-supporting tissues and relevant systemic consequences. In this context, hydrogels based on natural polymers have been widely explored in periodontal tissue engineering as biomimetic matrices capable of modulating the inflammatory response, enabling localized delivery of bioactive agents, and offering temporary structural support. This review summarizes recent advance

NSC23766 effectively inhibited retinal neovascularization by disrupting the positive feedback loop between Rac1 and VEGFR2.

Retinal neovascularization (RNV) is a leading cause of blindness. Although anti-VEGF therapy remains the first-line treatment for RNV, a subset of patients exhibits poor or no response to anti-VEGF agents. Therefore, exploring alternative therapeutic strategies is imperative. Rac1, a small GTPase, has been reported to regulate angiogenesis through multiple signaling pathways. Nevertheless, the precise role of Rac1 in RNV progression remains unclear. Our study demonstrates that NSC23766, a Rac1 activation inhibitor, effectively attenuates VEGF-induced angiogenic responses in human retinal micro

Recent advances in engineered exosome-based therapies for ocular vascular disease.

Ocular neovascular diseases (ONDs), including corneal neovascularization (CoNV), age-related macular degeneration (AMD) and diabetic retinopathy (DR), are among the leading causes of visual impairment worldwide. Current therapeutic strategies predominantly involve intravitreal injection of anti-vascular endothelial growth factor (anti-VEGF) agents, which, despite their efficacy, present significant limitations such as drug resistance, frequent intravitreal injections, and insufficient addressing of underlying pathological mechanisms. This review critically examines recent advancements in the a

Pharmacologic therapies in endometriosis: a systematic review.

To assess the literature on preclinical and clinical efficacy and safety data of pharmacologic groups proposed in the treatment of endometriosis, we performed a systematic review of publications from March 2002 to January 2012 via PubMed search. Additional relevant articles were identified from citations within these publications. A high number of medications were tested in preclinical models of endometriosis due to their theoretic capacity of disrupting important pathophysiologic pathways of the disease, such as inflammatory response, angiogenesis and cell survival, proliferation, migration,

Lactylation and targeted therapy resistance in hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) persists as a leading cause of cancer-related mortality globally. While targeted therapies, including sorafenib and lenvatinib, serve as standard first-line treatments, their efficacy is frequently compromised by low response rates and the rapid development of resistance, posing a significant clinical challenge. Lactylation (Kla), a metabolism-driven post-translational modification stemming from lactate accumulation, has recently emerged as a pivotal regulator of HCC progression and therapeutic resistance. The extent of lactylation is governed by lactate availabi
