Hidden threats in urban environments: Angiostrongylus cantonensis in Banda Aceh's cityscape.

Angiostrongylus cantonensis is a mollusk-borne parasitic nematode originating in Southeast Asia. Commonly known as the rat lungworm, it uses rats as definitive hosts, though other mammals, including humans, can be infected and typically suffer from neurological disorders. This study focuses on the parasite's presence in its gastropod intermediate hosts in several urban and rural areas in Aceh province, Sumatra, Indonesia. Samples of Achatina (Lissachatina) fulica (161) and Pomacea sp. (90) were collected in eight localities in Banda Aceh. Additionally, 531 edible freshwater snails belonging to

First cases of Angiostrongylus cantonensis infection reported in Martinique, 2002-2017.

Neuroangiostrongyliasis is a parasitic disease caused by the accidental ingestion of the nematode Angiostrongylus cantonensis in its larval form. Human infection can lead to eosinophilic meningitis, sometimes complicated by life-threatening radiculomyelitis or encephalitis. Although some cases have been reported from other Caribbean Islands, no cases have been diagnosed in Martinique so far. Here, we report the first eight laboratory-confirmed cases of neuroangiostrongyliasis on the island of Martinique, French West Indies, between 1 January 2002 and 31 December 2017. One case was fatal and fi

Proteasome serves as pivotal regulator in Angiostrongylus cantonensis-induced eosinophilic meningoencephalitis.

Proteasome primarily degrades the unneeded or damaged proteins by proteolysis. Disruption of the brain barrier and its resulting meningoencephalitis caused by Angiostrongylus cantonensis are important pathological events in non-permissive hosts. In this study, the results showed upregulated proteasome during A. cantonensis infection. Occludin degradation and matrix metalloproteinase-9 (MMP-9) activity were significantly increased in infected mice than in uninfected mice. Moreover, confocal immunoflourescence microscopy showed that occludin was co-localized with MMP-9. The infected-mice were tr

Angiostrongylus vasorum Causing Severe Granulomatous Hepatitis with Concurrent Multiple Acquired PSS.

A 14 mo old female Jack Russell terrier presented with a 12 hr history of vomiting and inappetence. She was subsequently diagnosed with multiple acquired portosystemic shunts during an exploratory celiotomy. Gross and histopathological hepatic abnormalities were consistent with chronic disease, including features suggestive of portal hypertension that was potentially caused by migrating and resident Angiostrongylus vasorum larvae. Fecal analysis and polymerase chain reaction of hepatic tissue confirmed the presence of Angiostrongylus vasorum . The dog recovered clinically following empirical t

Neuroangiostrongyliasis due to Angiostrongylus cantonensis in gang-gang cockatoos (Callocephalon fimbriatum).

CASE REPORT: Four gang-gang cockatoos from an aviary in Sydney displayed severe neurological signs. Three were necropsied and histopathology of the brains and spinal cords revealed migrating nematodes, which were identified as Angiostrongylus cantonensis. The migrating larval nematodes created tracts of malacia in the brain, but elicited little inflammatory cell infiltration. However, adult nematodes that had emerged onto the meningeal surface of the spinal cord evoked a marked non-suppurative reaction. Detailed histological examination of other tissues revealed larvae embedded in arterioles i

Thoracic computed tomography findings in dogs experimentally infected with Angiostrongylus vasorum.

To characterize the computed tomography (CT) features of thoracic lesions caused by infection with Angiostrongylus vasorum, pre- and postcontrast CT was performed in six experimentally infected Beagles 13 weeks postinoculation and in four of these 9 weeks postchemotherapy. Findings were compared with survey radiographs and necropsy findings. A multicentric bronchoalveolar pattern more pronounced at the lung periphery was present radiographically. On CT, the predominant abnormality underlying this alveolar pattern was multiple large nodules merging to areas of consolidation, and containing air

Immunological diagnosis of human angiostrongyliasis due to Angiostrongylus cantonensis (Nematoda: Angiostrongylidae).

Angiostrongylus cantonensis-associated eosinophilic meningitis in humans has been commonly reported worldwide. However parasitologically confirmed cases are not common, as the parasite has been recovered only infrequently from the cerebrospinal fluid of patients. The potential value of immunodiagnosis is therefore self-evident. Immunological tests can also help in the differential diagnosis of parasitic (particularly helminths) infections that cause eosinophilic meningitis. This paper summarizes the state of and advances in the immunological diagnosis of human angiostrongyliasis due to Angiost

Long-term observations on mouse strains experimentally infected with Angiostrongylus costaricensis.

BALB/c and C57BL/6 mice were experimentally infected with Angiostrongylus costaricensis and the parasitic parameters and antibody response during the acute and chronic phases of infection were analyzed. Following administration of six third-stage larvae (L3), there was no significant difference in the mean worm recovery or mean larval output. Coinciding with the maturation of worms in infected animals and with the egg output in mesenteric arteries, a strong increase in the humoral immune response was observed in both mouse strains. This response was characterized by a hypergammaglobulinemia, w

Spermatic artery obstruction caused by Angiostrongylus costaricensis Morera and Céspedes, 1971.

An 8-year-old boy was admitted to hospital with acute testicular pain. A diagnosis of torsion of the testicle was made, and an orchidectomy was performed. Histopathological study showed necrosis of the testicular parenchyma caused by arterial obstruction, and adults of Angiostrongylus costaricensis were found within the spermatic arteries. In addition, heavy eosinophilic inflammatory reactions and unfertilized eggs were observed in the excised tissues.

AcNR1F orchestrates epidermal remodeling to enable host adaptation in Angiostrongylus cantonensis.

Nuclear receptor NR1F regulates circadian rhythms in mammals, metamorphosis in insects, and molting in free-living Caenorhabditis elegans; however, its role in parasitic nematode development remains elusive. Angiostrongylus cantonensis, a zoonotic pathogen that causes eosinophilic meningitis in humans, exhibits distinct host adaptation, with mice and most mammals serving as non-permissive hosts and rats as the permissive host. We combined multi-omic analyses, lentivirus-mediated RNAi, LC-MS/MS, and dual-luciferase assays to elucidate AcNR1F functions in development and host adaptation. Acnr1f

IL-25 Enhances Eosinophil-Associated Inflammation in Angiostrongylus cantonensis-Infected Mice.

This study aimed to investigate the role of interleukin (IL)-25 in the pathogenesis of angiostrongylosis. We used ICR mice as non-permissive hosts, divided them into groups and experimentally infected them with infective larvae of the Angiostrongylus cantonensis. Two groups of infected mice were injected intraperitoneally with either mouse IL-25 or an anti-IL-25 monoclonal antibody (mAb) at 3 days post-infection (dpi), followed by booster injections at the same dose every 5 days. Serum samples and brain tissues were collected weekly from each group for immunological and pathological examinatio

Predictive Approach to Mapping Angiostrongylus cantonensis Nematode Distribution, Canary Islands, Spain.

The invasive nematode Angiostrongylus cantonensis (rat lungworm) can cause eosinophilic meningitis in humans. Once restricted to Southeast Asia, A. cantonensis nematodes are now widespread across the tropics and have been reported in Europe. Tenerife, in the Canary Islands, and the Mediterranean region are emerging hotspots. We surveyed gastropods, rats, and lizards across Tenerife and detected the parasite in all host groups at 2.4%-41.6% prevalence. Using species distribution models, we identified precipitation seasonality as the main driver of habitat suitability; tree cover and climatic va

Upregulation of Nrf2 attenuates Angiostrongylus cantonensis-induced parasitic meningitis in mice.

BACKGROUND: Angiostrongylus cantonensis is a food-borne parasite that can infect mammals, including humans, causing angiostrongyliasis. The nuclear factor E2-related factor 2 (Nrf2) is a transcription factor that plays a crucial role in the host's antioxidant defense and inflammation mechanisms. Herein, this study investigates the anti-inflammatory effects of Nrf2 in A. cantonensis-induced parasitic meningitis in mice. METHODS: We used animal infection and treatment, larvae collection, western blotting, enzyme-linked immunosorbent assay (ELISA), hematoxylin and eosin (H&E) stain, blood-brain

Release of Angiostrongylus cantonensis larvae from live intermediate hosts under stress.

The metastrongyloid nematode Angiostrongylus cantonensis causes eosinophilic meningitis in a variety of homeothermic hosts including humans. Third-stage infectious larvae develop in gastropods as intermediate hosts. Humans are usually infected by intentional or incidental ingestion of an infected mollusk or paratenic host (poikilothermic vertebrates and invertebrates). The infection may also hypothetically occur through ingestion of food or water contaminated by third-stage larvae spontaneously released from gastropods. Larvae are thought to be released in greater numbers from the intermediate

Rapid Single-Step Immunochromatographic Assay for Angiostrongylus cantonensis Specific Antigen Detection.

Angiostrongylus cantonensis is the major etiological nematode parasite causing eosinophilic meningitis and/or eosinophilic meningoencephalitis in humans. The rapid global spread of Angiostrongylus cantonensis and the emerging occurrence of the infection have exposed the shortcomings of traditional/conventional diagnostics. This has spurred efforts to develop faster, simpler and more scalable platforms that can be decentralized for point-of-need laboratory testing. By far, the point-of-care immunoassays such as the lateral flow assay (LFA) are the best-placed. In this work, a LFA in the form of

RPAcan3990: an Ultrasensitive Recombinase Polymerase Assay To Detect Angiostrongylus cantonensis DNA.

Angiostrongylus cantonensis is one of the leading causes of eosinophilic meningitis worldwide. A field-deployable molecular detection method could enhance both environmental surveillance and clinical diagnosis of this emerging pathogen. Accordingly, RPAcan3990, a recombinase polymerase assay (RPA), was developed to target a region predicted to be highly repeated in the A. cantonensis genome. The assay was then adapted to produce a visually interpretable fluorescent readout using an orange camera lens filter and a blue light. Using A. cantonensis genomic DNA, the limit of detection was found to

Angiostrongylus costaricensis infection in Martinique, Lesser Antilles, from 2000 to 2017.

Human abdominal angiostrongyliasis (HAA) is a parasitic disease caused by the accidental ingestion of the nematode Angiostrongylus costaricensis in its larval form. Human infection can lead to severe ischemic and inflammatory intestinal lesions, sometimes complicated by life-threatening ileal perforations. Only one case had been reported in Martinique, an Island in the French Antilles, in 1988. We retrospectively reviewed the medical charts of patients diagnosed with abdominal angiostrongyliasis at the University Hospital of Martinique between 2000 and 2017. The objectives of this study were t

Angiostrongylus vasorum infection in red foxes (Vulpes vulpes) in southern Italy.

Angiostrongylus vasorum (Nematoda: Angiostrongylidae) infection was detected at post-mortem examination in the pulmonary arteries and hearts of 34/102 (33,3%) of red foxes (Vulpes vulpes) from the Campania Region in southern Italy. Pathological changes consisted of granulomatous interstitial pneumonia caused by larvae and intravascular pulmonary adult nematodes. These changes confirm that angiostrongylosis infection in red foxes has a mainly chronic course, in which the infected host may disperse parasite larvae in the environment over its lifetime. Results suggest that the life cycle of A. va
