Persistent Anhedonia After Intermittent Long-Access Nicotine Self-Administration in Rats.

Tobacco use disorder is a chronic condition characterized by compulsive nicotine use and withdrawal symptoms after smoking cessation. Smoking is the leading preventable cause of morbidity and mortality worldwide. Smoking cessation leads to anhedonia, which is an inability to experience pleasure from previously enjoyed activities and is caused by dysregulation of the brain's reward and stress systems. It is also a key withdrawal symptom that contributes to relapse to smoking after a period of abstinence. To better understand the development of anhedonia, we investigated its onset and time cours

Neural mapping of anhedonia across psychiatric diagnoses: A transdiagnostic neuroimaging analysis.

Anhedonia has been associated with abnormal reward-related striatal dopamine functioning in patients with different psychiatric disorders. Here, we tested whether anhedonia expression mapped onto striatal volume across several psychiatric diagnoses. T1-weighted images from 313 participants including 89 healthy controls (HC), 22 patients with opioid use disorder (OUD), 50 patients with major depressive disorder (MDD), 45 patients with borderline personality disorder (BPD), 49 patients with first-episode psychosis (FEP), 43 patients with cocaine use disorder (CUD) and 15 patients with schizophre

Stress-induced expression pattern of glutamate signaling genes associated with anhedonia.

Chronic stress can predispose vulnerable individuals to mood disorders, including depression. Glutamate, one of the key participants in this process, may exert both pathological and therapeutic psycho-emotional effects. However, the role of expression of genes encoding proteins that provide glutamatergic signal is still unclear. In this study, we attempted to distinguish changes in expression of glutamatergic genes associated with stress-induced anhedonia, a core symptom of depression, from those related to other stress-related effects. For this, expression of genes was compared between rats a

Adolescent intermittent ethanol exposure diminishes anhedonia during ethanol withdrawal in adulthood.

Adolescent alcohol use may interfere with neurodevelopment, increasing the likelihood of adult alcohol use disorders (AUDs). We investigated whether adolescent intermittent ethanol (AIE) exposure alters the adult reward response to ethanol. Adolescent rats were administered ethanol once (moderate exposure; Cohort 1) or three times per day (severe exposure; Cohort 2) in a 2 days on/2 days off pattern. In adulthood, subjects responded for electrical stimulation directed at the posterior lateral hypothalamus in a discrete-trial intracranial self-stimulation (ICSS) procedure that provides current-

Social anhedonia is associated with neural abnormalities during face emotion processing.

Human beings are social organisms with an intrinsic desire to seek and participate in social interactions. Social anhedonia is a personality trait characterized by a reduced desire for social affiliation and reduced pleasure derived from interpersonal interactions. Abnormally high levels of social anhedonia prospectively predict the development of schizophrenia and contribute to poorer outcomes for schizophrenia patients. Despite the strong association between social anhedonia and schizophrenia, the neural mechanisms that underlie individual differences in social anhedonia have not been studie

Disentangling Anhedonia and Self-Stigma in Schizophrenia Using Bayesian Network Analysis.

BACKGROUND: The interrelationships among the negative symptoms of schizophrenia and factors affecting quality of life remain unclear. To address this gap, the present study explores the complex interrelationships among anhedonia, self-stigma, self-esteem, psychological resilience, and social support in individuals with schizophrenia. METHODS: A comprehensive battery of measures was used to assess 447 patients with schizophrenia. Undirected network analysis was employed to examine the centrality of various factors, and Bayesian network analysis was used to explore the indirect influence of ups

Possible New Symptoms of Tobacco Withdrawal II: Anhedonia-A Systematic Review.

INTRODUCTION: When animals undergo nicotine deprivation, rewards become less rewarding (ie, anhedonia occurs). We searched for tests of whether anhedonia occurs in abstinent smokers. METHODS: The major inclusion criterion was a within-participants comparison of behavioral measures of reward sensitivity or self-reported anhedonia during smoking versus during abstinence among daily smokers. A computerized search of PubMed, PsychInfo, and Cochrane databases and other methods located 13 studies. All but one were laboratory studies. RESULTS: The number of studies and participants were small and t

The New York High-Risk Project: anhedonia, attentional deviance, and psychopathology.

In the New York High-Risk Project (NYHRP) we followed subjects at risk for schizophrenic or affective disorders and low-risk controls from childhood to adulthood, with the goal of identifying early predictors of later schizophrenia-related psychopathology. In this article, we focus on two potential predictors: the Physical Anhedonia Scale administered in adolescence and the Attention Deviance Index obtained in childhood. Subjects of this report are 161 members of the NYHRP's first sample (sample A), who had scores on both attention and anhedonia and had followup clinical assessments in adultho

Identifying distinct trajectories of change in anhedonia during psychological treatment for depression.

BACKGROUND: While initial anhedonia predicts poor psychotherapy outcomes, little is known about its trajectory during treatment. This study aimed to: (1) identify distinct anhedonia trajectories during high-intensity depression treatment; (2) examine patient and treatment predictors; and (3) compare outcomes across treatment types. METHODS: Sessional anhedonia scores (PHQ-9 item-1) from 22,605 patients in NHS talking therapies (primarily receiving either cognitive-behavioral therapy [CBT] or counseling for depression [CfD]) were analyzed using latent growth curve (LGC) and growth mixture mode

The impact of anhedonia on physical health and somatic diseases: an exploratory literature review.

Anhedonia, or the lack of interest and pleasure, is a transdiagnostic feature of mental health disorders with emerging links to physical health. This exploratory literature review investigated evidence of anhedonia's association with physical health and somatic diseases. A citation chaining approach was applied to 20 papers identified through targeted searches conducted from September to October 2024 on PubMed and Google/Google Scholar. Studies published in English and reporting outcomes in adults with anhedonia were included. Quality assessments were performed using the Centre for Evidence-Ba

The Role of Chronic Anhedonia in the Relationship Between Treatment-Resistant Depression and Suicidality.

Objective: Treatment-resistant depression (TRD) is a severe form of major depressive disorder (MDD) associated with increased suicidality. Anhedonia, the reduced ability to experience pleasure, is recognized across various psychiatric conditions, including TRD. This study aims to investigate the relationship between chronic anhedonia, or low hedonic tone, and suicidal behavior in patients with TRD. Methods: A cohort of 160 participants from the START Clinic for Mood and Anxiety Disorders in Toronto, Canada, was assessed. Diagnosis of TRD followed criteria established by McIntyre et al (2014) a

Examining the effects of psilocybin-assisted psychotherapy on anhedonia in treatment-resistant depression.

Anhedonia, a core symptom of depression, is often resistant to conventional treatments and significantly impacts quality of life. This secondary analysis aimed to evaluate the effects of psilocybin-assisted psychotherapy (PAP) on anhedonia severity in individuals with treatment-resistant depression (TRD). Participants (n = 30) with TRD and a primary diagnosis of Major Depressive Disorder or Bipolar II Disorder received at least one 25 mg dose of oral psilocybin with psychotherapy as part of a randomized, waitlist-controlled trial (NCT05029466). The primary outcome of the present secondary anal

The association between anhedonia and speech features in depression: A cross-sectional study.

BACKGROUND: Anhedonia is a core symptom of depression. Most anhedonia assessments rely on self-reporting, which does not accurately reflect hedonic capacity and is biased by individual subjectivity. Therefore, objective indicators are needed. Anhedonia may result in different speech features among depressive patients. Thus, speech features have become an emerging objective indicator in depression assessment. This study aims to investigate the relationship between anhedonia and speech features in individuals with depression by comparing the speech features of patients with and without anhedonia

Transcriptional substrates of cortical thickness alterations in anhedonia of major depressive disorder.

BACKGROUND: Anhedonia is a core symptom of major depressive disorder (MDD), which has been shown to be associated with abnormalities in cortical morphology. However, the correlation between cortical thickness (CT) changes with anhedonia in MDD and gene expression remains unclear. METHODS: We investigated the link between brain-wide gene expression and CT correlates of anhedonia in individuals with MDD, using 7 Tesla neuroimaging and a publicly available transcriptomic dataset. The interest-activity score was used to evaluation MDD with high anhedonia (HA) and low anhedonia (LA). Nineteen pati

Exploring mechanisms of anhedonia in depression through neuroimaging and data-driven approaches.

BACKGROUND: Anhedonia is a core symptom of depression that is closely related to prognosis and treatment outcomes. However, accurate and efficient treatments for anhedonia are lacking, mandating a deeper understanding of the underlying mechanisms. METHODS: A total of 303 patients diagnosed with depression and anhedonia were assessed by the Snaith-Hamilton Pleasure Scale (SHAPS) and magnetic resonance imaging (MRI). The patients were categorized into a low-anhedonia group and a high-anhedonia group using the K-means algorithm. A data-driven approach was used to explore the differences in brain

Insulin Resistance, Temperament and Personality Traits Are Associated with Anhedonia in a Transdiagnostic Sample.

Anhedonia constitutes a core symptom of major depressive disorder (MDD) mediating the ultimate goal of MDD treatment: functional remission. Anhedonia is also present in other clinical populations, including patients with chronic pain. Recent data links anhedonia to insulin resistance (IR). Some researchers have underlined a different dimension of anhedonia as a temperament/personality trait. The objective of this post-hoc analysis was to explore the links between anhedonia (main outcome) and (1) IR, (2) temperamental, personality, and schizotypy traits (exposures). The study population include
