Endocrine disruption by environmental chemicals continues to be a concern for human safety. The rat, a widely used model organism in toxicology, is very sensitive to chemical-induced thyroid perturbation, e.g., histopathological alterations in thyroid tissue. Species differences in the susceptibility to thyroid perturbation lead to uncertainty in human safety risk assessments. Hazard identification and characterization of chemically induced thyroid perturbation would therefore benefit from in vitro models addressing different mechanisms of action in a single functional assay, ideally across sp

Amino acid derivative reactivity assay (ADRA) is an in chemico assay for assessing the skin sensitization potential of chemicals by evaluating the reactivity of nucleophilic reagents that mimic skin proteins. N-(2-(1-Naphthyl)acetyl)-l-cysteine (NAC) and α-N-(2-(1-naphthyl)acetyl)-l-lysine (NAL), used as nucleophilic reagents, are small-molecule derivatives of two different amino acids, each with a naphthalene ring attached. The rate of decrease in the amount of NAC or NAL in the reaction solution is evaluated in this assay as an indicator of the test substance's skin sensitization ability. Ho

For hazard classifications of chemicals, continuous data from animal- or nonanimal testing methods are often dichotomized into binary positive/negative outcomes by defining classification thresholds (CT). Experimental data are, however, subject to biological and technical variability. Each test method's precision is limited resulting in uncertainty of the positive/negative outcome if the experimental result is close to the CT. Borderline ranges (BR) around the CT were suggested, which represent ranges in which the study result is ambiguous, that is, positive or negative results are equally lik

The whole blood pyrogen test was first described in this journal exactly thirty years ago. Its variant based on cryopreserved blood followed one year later. Together with other monocyte activation tests (MATs), it has fundamentally changed the landscape of pyrogen testing. In the five years since the 25th anniversary article in this series, progress has been remarkable: The European Pharmaco-poeia deleted the rabbit pyrogen test (RPT) effective January 2026, ending a 55-year era; ISO 10993-1:2025 removed material-mediated pyrogenicity from mandatory evaluation endpoints for medical devices; th

In the cosmetics sector, many products such as shampoos have a probability of accidental ocular exposure during their routine use. One very specific safety parameter is the residence time of the substance on the corneal surface, as prolonged exposure may cause injury. In this study, we developed a system that simulates corneal exposure to blinking and tear flow, for comparing the corneal clearance times of viscous detergent formulations. The Ex Vivo Eye Irritation Test (EVEIT), which uses corneal explants from discarded rabbit eyes from an abattoir, was used as the basis for the new system. To

The scientific and ethical issues associated with the use of animal-derived antibodies in research can be overcome by the use of animal-free, sequence-defined recombinant antibodies, whose benefits are well documented. Here, we describe progress made following a 2019 expert meeting focused on improving the quality and reproducibility of biomedical research by accelerating the production and use of animal-free recombinant antibodies in the USA. In the five intervening years since the meeting, participants have established multifaceted initiatives to tackle the next steps outlined during the mee

We developed an alternative approach to teach diabetes mellitus in our practical classes, replacing laboratory animals. We used custom rats made of cloth, which have a ventral zipper that allows stuffing with glass marbles to reach different weights. Three mock rats per group were placed into metabolic cages with real food and water and with test tubes containing artificial urine, simulating a sample collection of 24 h. For each cage, we also provided other test tubes with artificial blood and urine, simulating different levels of hyperglycemia. The artificial "diabetic" urine contained differ

Current preclinical models for cervical dysplasia rely on animal systems that poorly mimic human anatomy. To address this and align with emerging initiatives to reduce animal experimentation, we developed a human-sized, 3D in vitro model of high-grade cervical dysplasia. The construct integrates normal human fibroblasts, keratinocytes, and SiHa cancer cells within a gelatin methacrylate (GelMA) hydrogel, engineered with a cervical "os" and asymmetric lesion to replicate native human cervical architecture. We utilized this platform to evaluate the efficacy and safety of ethyl cellulose-ethanol

Predictive testing to characterise substances for their skin sensitisation potential has historically been based on animal models such as the Local Lymph Node Assay (LLNA) and the Guinea Pig Maximisation Test (GPMT). In recent years, EU regulations, have provided a strong incentive to develop non-animal alternatives, such as expert systems software. Here we selected three different types of expert systems: VEGA (statistical), Derek Nexus (knowledge-based) and TIMES-SS (hybrid), and evaluated their performance using two large sets of animal data: one set of 1249 substances from eChemportal and

Bioprinted skin has emerged as a disruptive technology at the intersection of tissue engineering and plastic surgery. Through controlled deposition of bioinks containing living cells and biomaterials, it is now possible to create human skin constructs with unprecedented physiological fidelity. These constructs are already transforming the evaluation of drugs and cosmetics by providing reproducible, human-relevant alternatives to animal testing. In reconstructive and aesthetic surgery, bioprinted skin holds the promise of patient-specific grafts that integrate seamlessly, accelerate healing, an

BACKGROUND: Fusobacterium nucleatum is an oncobacterium capable of promoting the growth and chemotherapy resistance of colonised tumours. Although F. nucleatum is usually susceptible to a range of antibiotics in vitro, these have been associated with worse outcomes when administered with anti-neoplastic chemotherapy. Bacteriophages are viewed as natural alternatives to antibiotics that provide bacterial-specific targeting.
METHODS: In this study, we have employed an F. nucleatum specific bacteriophage, FNU1, to limit the effects of this oncobacteria in colon cancer and gastric cancer cell mod

Collagen vitrigel membranes (CVMs) comprising high-density collagen fibrils equivalent to in vivo connective tissues have been widely used in cell culture applications. A human corneal epithelium (hCE) model was previously developed by the Takezawa group, by culturing HCE-T cells (derived from hCE cells) on a CVM scaffold in a chamber that provided an air-liquid interface culture system. This hCE model was used to establish a new test method, known as the Vitrigel-Eye Irritancy Test (Vitrigel-EIT) method, which can be used to estimate the ocular irritation potential of test chemicals by analys

Microcystins (MC) are a group of cyanobacterial toxins that comprises MC-LF and other cyclic heptapeptides, best known as potent hepatotoxicants. Cell culture and epidemiological studies suggest that MC might also affect the nervous system when there is systemic exposure, e.g., via drinking water or food. We asked whether in vitro studies with human neurons could provide estimates on the neurotoxicity hazard of MC-LF. First, we used LUHMES neurons, a well-established test system for neurotoxicants and neuropathological processes. These central nervous system cells express OATP1A2, a presumed c

Chemical sensitization remains an important environmental and occupational health issue. A wide range of substances have been shown to possess the ability to induce skin sensitization or respiratory sensitization. As a consequence, there is a need to have appropriate methods to identify sensitizing agents. Although a considerable investment has been made in exploring opportunities to develop methods for hazard identification and characterization, there are, as yet, no validated nonanimal methods available. A state of the art of the different in vitro approaches to identify contact and respirat

Three-dimensional, self-organizing structures derived from stem cells, known as organoids, represent a groundbreaking advancement in preclinical drug development. Organoid-based platforms advance preclinical testing by providing an accurate representation of human tissue architecture and genetics, surpassing traditional two-dimensional cultures and animal models in testing both drug safety and efficacy. Researchers are shifting toward organoid-based systems as primary components of new approach methodologies, as global regulatory bodies increasingly acknowledge animal testing limitations. This

Acute oral toxicity (AOT) testing is widely used for hazard identification in chemical safety assessment, but traditional animal methods are still implemented despite their scientific limitations and ethical implications. Despite the availability of non-animal methods, regulatory uptake has been inconsistent, also due to a lack of structured guidance. To address this, the Animal-Free Safety Assessment Collaboration has developed a science-based Decision Tree (DT) aligned with the Organisation for Economic Co-operation and Development (OECD) Integrated Approaches to Testing and Assessment (IATA

According to ISO 10993-23 (Biological evaluation of medical devices – Part 23: Tests for irritation, Annex D2), no specific alternative test method has yet been established to identify substances that may cause eye irritation or serious eye damage in medical devices. Therefore, the in vivo animal test OECD TG 405 continues to be used. This study adapted the in vitro eye irritation test (EIT) using the MCTT HCE™ Reconstructed Human Corneal Epithelial (RHCE) model, which was originally developed for testing chemicals, to the testing of ophthalmic medical devices such as contact lenses and intrao

Since its adoption as a test guideline (EIT, OECD test guideline (TG) 492) in 2015, the EpiOcular™ eye irritation test has been the in vitro method of choice to identify agrochemical formulations non-irritant to the eye (neither category 1 nor 2, according to the Globally Harmonized System of Classification and Labelling of Chemicals, GHS). For serious eye damage (GHS category 1), however, none of the in vitro test methods including the bovine cornea opacity and permeability test (BCOP, OECD TG 437, and protocol modifications), the isolated chicken eye test (ICE, OECD TG 438), the ocular irrit

For several decades the European Pharmacopoeia monographs Tetanus vaccine (adsorbed) (0452) and Tetanus vaccine for veterinary use (0697) required that Specific toxicity and Absence of toxin and irreversibility of the toxoidof each bulk of tetanus toxoids had to be tested by an in vivo toxicity test in guinea pigs before it could be included in vaccines for human or veterinary use. In line with the 3Rs concept of replacing, reducing and refining animal experiments, an in vitro method for the detection of active tetanus neurotoxin (TeNT) has been developed at the Paul-Ehrlich-Institut (PEI, Ger
