Mapping Plasmodium transitions and interactions in the Anopheles female.

The human malaria parasite, Plasmodium falciparum, relies exclusively on Anopheles mosquitoes for transmission. Once ingested during blood feeding, most parasites die in the mosquito midgut lumen or during epithelium traversal1. How surviving ookinetes interact with midgut cells and form oocysts is poorly known, yet these steps are essential to initiate a remarkable growth process culminating in the production of thousands of infectious sporozoites2. Here, using single-cell RNA sequencing of both parasites and mosquito cells across different developmental stages and metabolic conditions, we un

Experimental Transmission of Plasmodium malariae to Anopheles gambiae.

Our current knowledge of the clinical burden, biology, and transmission of Plasmodium malariae is extremely scarce. To start addressing some of those questions, we experimentally infected Anopheles gambiae mosquitoes with fresh P. malariae isolates obtained from asymptomatic individuals in Lambaréné, Gabon. The proportion of mosquitoes infected via direct membrane feeding assay with either P. malariae monoinfections (16% [19 of 121]) or coinfections (28% [31 of 112]) was higher after serum replacement than in parallel groups without serum replacement (4% [4 of 102] and 4% [2 of 45], respective

Plasmodium development in Anopheles: a tale of shared resources.

Interactions between the Anopheles mosquito vector and Plasmodium parasites shape how malaria is transmitted in endemic regions. The long association of these two organisms has led to evolutionary processes that minimize fitness costs of infection and benefit both players through shared nutrient resources, parasite immune suppression, and mosquito tolerance to infection. In this review we explore recent data describing how Plasmodium falciparum, the deadliest malaria parasite, associates with one of its most important natural mosquito hosts, Anopheles gambiae, and we discuss the implications o

ASDB: A comprehensive omics database for Anopheles sinensis.

Anopheles sinensis is a key disease vector for human malaria and parasitic diseases such as malayan filariasis, and it is considered to be one of the most important malaria vectors in China and Southeast Asia. As high-throughput sequencing and assembly technology are widely used in An. sinensis, a lot of omics data have been generated, and abundant genome, mRNA transcriptome, miRNA transcriptome and resequencing results have been accumulated. In addition, lots of valuable morphological images and publications have been produced with the in-depth studies on An. sinensis. However, the increased

Geographical distribution of Anopheles stephensi in eastern Ethiopia.

BACKGROUND: The recent detection of the South Asian malaria vector Anopheles stephensi in Ethiopia and other regions in the Horn of Africa has raised concerns about its potential impact on malaria transmission. We report here the findings of a survey for this species in eastern Ethiopia using both morphological and molecular methods for species identification. METHODS: Adult and larval/pupal collections were conducted at ten sites in eastern Ethiopia and Anopheles specimens were identified using standard morphological keys and genetic analysis. RESULTS: In total, 2231 morphologically identif

Viral paratransgenesis in the malaria vector Anopheles gambiae.

Paratransgenesis, the genetic manipulation of insect symbiotic microorganisms, is being considered as a potential method to control vector-borne diseases such as malaria. The feasibility of paratransgenic malaria control has been hampered by the lack of candidate symbiotic microorganisms for the major vector Anopheles gambiae. In other systems, densonucleosis viruses (DNVs) are attractive agents for viral paratransgenesis because they infect important vector insects, can be genetically manipulated and are transmitted to subsequent generations. However, An. gambiae has been shown to be refracto

[Anopheles funestus and rice agriculture in the Madagascar highlands].

An exhaustive study of the potential habitats of Anopheles funestus was led during 1992 in Ankazobe on the Plateau of Madagascar, 95 km northwest of the capital Tananarive. The rice fields provide more than 90% of the positive habitats versus less than 10% for the nonhuman biotopes. Larva are especially abundant on the surfaces of the rice during grain head formation and maturation. The dense vegetation coverage provides them with shade and protection against predators. After harvesting, the follows can be filled with water and wild vegetation, and then also provide an important share of the h

Development of resistance to DDT by Anopheles sacharovi in Greece.

The DDT house-spraying programme carried out throughout Greece between 1946 and 1950 proved highly effective against both the local malaria-vectors (Anopheles sacharovi and A. superpictus) and other domestic pests. However, during 1947 the housefly (Musca domestica) began to reappear in large numbers, and in subsequent years the development of a similar resistance to DDT was noticed among other domestic pests, such as Culex molestus, fleas, bed-bugs, and cockroaches. In 1951, signs of a weakening of DDT efficacy against the local malaria-vector A. sacharovi were noted for the first time.A seri

Single nucleotide polymorphism analysis of the ITS2 region of two sympatric malaria mosquito species in Sweden: Anopheles daciae and Anopheles messeae.

Four species of the Anopheles maculipennis complex have previously been recorded in Sweden. A recent addition to the complex is Anopheles daciae, which is considered to be closely related to, but distinct from Anopheles messeae. The original designation of An. daciae was based on five genetic differences (161, 165, 167, 362 and 382) in the second internal transcribed spacer (ITS) 2 of the ribosomal RNA. Further studies have shown that only two nucleotide differences (362 and 382) robustly separate the species. Thirty-three An. maculipennis complex mosquitoes were collected in the province of U

Does extreme asymmetric dominance promote hybridization between Anopheles coluzzii and Anopheles gambiae s.s. in seasonal malaria mosquito communities of West Africa?

BACKGROUND: Anopheles gambiae s.s. and An. coluzzii are two of the most important malaria vector species in sub-Saharan Africa. These recently-diverged sibling species do not exhibit intrinsic post-zygotic barriers to reproduction and are thought to be separated by strong assortative mating combined with selection against hybrids. At present, little is known about the ecological conditions that determine hybridization and introgression between these cryptic taxa. METHODS: Swarm segregation and assortative mating were studied in Western Burkina Faso in the villages of Vallée du Kou (VK7) and S

Evaluation of the effectiveness of Aquatain, Bacillus thuringiensis var. israelensis, and Temephos on Anopheles arabiensis and Anopheles stephensi larvae in the laboratory and field settings.

BACKGROUND: The main tools to control malaria vectors in sub-Saharan Africa are long-lasting insecticidal nets and indoor residual spraying. However, their sustainability is threatened by the emergence of insecticide resistances, behavioral avoidance and presence of outdoor biting mosquito populations. Thus, complementary interventions such as larval source management, which includes larviciding, are required to achieve better results in malaria vector control. This study aimed to evaluate the effectiveness of three larvicides (Aquatain AMF®, Temephos and Bacillus thuringiensis var. israelensi

Description and bionomics of Anopheles (Cellia) ovengensis (Diptera: Culicidae), a new malaria vector species of the Anopheles nili group from south Cameroon.

Mosquito species of the Anopheles nili group (Diptera: Culicidae) transmit malaria to humans along rivers in Africa. To date, the An. nili group includes the species Anopheles nili s.s. and its pale-winged variant known as the "Congo form," Anopheles somalicus and Anopheles carnevalei. Larval and adult mosquito collections in the forest region of Campo, in southern Cameroon, uncovered an additional morphological variant provisionally called "Oveng form" that was subsequently found to be genetically distinct from the other members of the An. nili group. In this study, we provide further biologi

Structural Insights into Pyrethroid Recognition by Anopheles Chemosensory Proteins.

Mosquito-borne diseases, particularly malaria, remain a major global health challenge. Among different mosquitoes, Anopheles are solely responsible for malaria. Insecticide-based interventions such as insecticide-treated nets (ITNs) and indoor residual spraying (IRS) are central to mosquito control. However, the widespread emergence of insecticide resistance, especially to pyrethroids, the most commonly used insecticides for mosquito worldwide, threatens their effectiveness. While genetic and metabolic mechanisms of resistance are well characterized, behavioral and sequestration-based mechanis

Sickle Trait Hemoglobin Does Not Influence Anopheles Biting Rates.

Children with sickle cell trait (HbAS) are protected against severe and symptomatic Plasmodium falciparum (P. falciparum) malaria. Although several within-host resistance mechanisms have been investigated, it is unknown whether this protection is attributable in part to reduced exposure to P. falciparum parasites via mosquito bites. In a 15-month cohort in Western Kenya, mosquito blood meals were matched to human hosts on the basis of short tandem repeat genotypes to determine individual mosquito biting rates. Using a multilevel multivariable model, mosquito biting behavior was assessed with r

Future global distribution and climatic suitability of Anopheles stephensi.

Anopheles stephensi, an urban malaria vector, is expanding into new regions and poses a growing global health threat. Using a robust multi-model framework with eight algorithms and high-resolution climate data, this study assessed and mapped current and future global climate suitability for the species, incorporating demographic data to estimate populations at risk in different scenarios. Currently, An. stephensi can thrive in 13% of the Earth's surface, affecting nearly 40% of the global population. Projections indicate that suitable areas could exceed 30% by 2100, exposing up to 56% of the w

Malaria: influence of Anopheles mosquito saliva on Plasmodium infection.

Malaria is caused by Plasmodium protozoa that are transmitted by anopheline mosquitoes. Plasmodium sporozoites are released with saliva when an infected female mosquito takes a blood meal on a vertebrate host. Sporozoites deposited into the skin must enter a blood vessel to start their journey towards the liver. After migration out of the mosquito, sporozoites are associated with, or in proximity to, many components of vector saliva in the skin. Recent work has elucidated how Anopheles saliva, and components of saliva, can influence host-pathogen interactions during the early stage of Plasmodi

Anopheles ecology, genetics and malaria transmission in northern Cambodia.

In the Greater Mekong Subregion, malaria cases have significantly decreased but little is known about the vectors or mechanisms responsible for residual malaria transmission. We analysed a total of 3920 Anopheles mosquitoes collected during the rainy and dry seasons from four ecological settings in Cambodia (villages, forested areas near villages, rubber tree plantations and forest sites). Using odor-baited traps, 81% of the total samples across all sites were collected in cow baited traps, although 67% of the samples attracted by human baited traps were collected in forest sites. Overall, 20%
