The LORF5 Gene Is Non-essential for Replication but Important for Duck Plague Virus Cell-to-Cell Spread Efficiently in Host Cells.

Duck plague virus (DPV) can cause high morbidity and mortality in many waterfowl species within the order Anseriformes. The DPV genome contains 78 open reading frames (ORFs), among which the LORF2, LORF3, LORF4, LORF5, and SORF3 genes are unique genes of avian herpesvirus. In this study, to investigate the role of this unique LORF5 gene in DPV proliferation, we generated a recombinant virus that lacks the LORF5 gene by a two-step red recombination system, which cloned the DPV Chinese virulent strain (DPV CHv) genome into a bacterial artificial chromosome (DPV CHv-BAC); the proliferation law of

Molecular Characterization of Avulaviruses Isolated from Mallard Ducks in Moscow in 2008-2024.

Species of the orders Charadriiformes and Anseriformes serve as the primary long-distance disseminators of various avulaviruses. The most economically significant among them is Newcastle disease virus (NDV), or Avian orthoavulavirus 1 (AOAV-1), which causes diseases of varying severity in both domestic and wild birds. Other avulaviruses have been studied to a much lesser extent, and for most of them, only single isolates are known, which does not allow a comprehensive assessment of their potential threat. To evaluate the biological diversity and potential risks posed by avian paramyxoviruses s

Cutaneous, respiratory and hepatic aspergillosis in Brazilian white Pekin mallards (Anas platyrhynchos).

Aspergillosis is one of the most frequent mycosis affecting avian species. Here is reported an outbreak of aspergillosis affecting 60-day-old white Pekin mallards (Anas platyrhynchos). About 10% of animals in a lot of 200 mallards from a commercial husbandry presented respiratory disorders and skin lesions at slaughter. Three out of 13 animals sent to diagnosis showed, simultaneously, airsacculitis, lung and liver presenting white nodules with variable diameters and elevated, yellowish brown, crusted, multifocal skin lesions located at the base of the feather follicles in the breast. Histopath

Seroconversion in wild birds and local circulation of West Nile virus, Spain.

A serosurvey for neutralizing antibodies against West Nile virus (WNV) in common coots (Fulica atra) was conducted in Doñana, Spain. Antibody prevalence was highest in 2003, intermediate in 2004, and lowest in 2005. Some birds seroreverted <1 year after first capture. Seroconversion of birds suggests local circulation of the virus.

Obliterative endophlebitis in mute swans (Cygnus olor) caused by Trichobilharzia sp. (Digenea: Schistosomatidae) infection.

Schistosome infections in mammals cause chronic proliferative vascular lesions associated with the presence of adult parasites in the lumen of mesenteric and portal veins. In birds, however, this has never been reported. In this study, we found obliterative endophlebitis associated with the presence of adult schistosomes (Trichobilharzia sp., probably Trichobilharzia filiformis) as the main pathologic finding in five of eight mute swans (Cygnus olor). On histologic examination, the intestinal and portal veins of these swans showed moderate to severe, diffuse, hyperplastic endophlebitis, charac

Gene Flow Accompanies Divergence in Beringian Birds.

The generation and maintenance of biodiversity are driven by population divergence and speciation. We investigated divergence, gene flow, and speciation in Beringia, a region at the top of the North Pacific Ocean with a history of dramatic landscape alteration through Pleistocene glacial cycles. These cycles repeatedly split and connected the Asian and North American continents, separating and reconnecting avian populations. Glacial refugia within Beringia also isolated some populations for a time before potentially enabling them to reunite during interglacial periods. Prior work suggests gene

Serosurvey of Toxoplasma gondii, Sarcocystis sp. and Neospora caninum in geese (Anser sp.) from urban parks and captivity.

Geese, ducks, mallards, and swans are birds of the order Anseriformes, which are found in the wild, in zoos and parks, and raised for meat consumption. Toxoplasma gondii, Sarcocystis sp., and Neospora caninum are protozoans of several species of animals. Wild and domestic birds can serve as intermediate hosts, disseminators and potential sources of infection of these protozoa to humans through contaminated meat. The aims of this study were: (i) to perform a serological survey of T. gondii, Sarcocystis sp. and N. caninum in geese (Anser sp.) from public parks and from captivity and (ii) to comp

Pioneering discovery: US3-phosphorylated sites on VP22 orchestrate duck plague virus release and pathogenicity.

Duck plague virus (DPV), a member of the Alphaherpesvirinae subfamily, causes severe infectious disease in waterfowl and Anseriformes birds. Although the VP22 protein (encoded by the UL49 gene) is known to play a pivotal role in secondary viral envelopment, its post-translational modifications and regulatory mechanisms during the viral life cycle remain poorly understood. In this study, we found that the conserved serine/threonine kinase US3 of DPV phosphorylates VP22. We identified eight serine residues (Ser37, Ser66, Ser68, Ser82, Ser114, Ser198, Ser226, and Ser227) as phosphorylation target

Characterization of avian influenza virus attachment patterns to human and pig tissues.

Wild birds of Anseriformes and Charadriiformes are natural reservoirs of influenza A viruses (IAVs). Occasionally, IAVs transmit and adapt to mammalian hosts, and are maintained as epidemic strains in their new hosts. Viral adaptions to mammalian hosts include altered receptor preference of host epithelial sialylated oligosaccharides from terminal α2,3-linked sialic acid (SA) towards α2,6-linked SA. However, α2,3-linked SA has been found in human respiratory tract epithelium, and human infections by avian IAVs (AIVs) have been reported. To further explore the attachment properties of AIVs, fou

Prevalence of Chlamydia psittaci and Other Chlamydia Species in Wild Birds in Poland.

Avian chlamydiosis is a zoonotic disease occurring in humans, poultry, and exotic birds. It has been suggested that some wild bird species play an important role as reservoirs for Chlamydia, especially Chlamydia psittaci. Whereas C. psittaci is the predominant chlamydial agent in birds, in the present study we have determined the prevalence of different species of Chlamydia among selected wild bird species in Poland using a rapid and sensitive real-time PCR method. In total, 369 free-living birds from 35 bird species and 15 orders were examined. Samples from 27 birds (7.3%) were positive for c

Embryo toxicity of 2,3,7,8-tetrachlorodibenzo-p-dioxin to the wood duck (Aix sponsa).

We examined the sensitivity of the wood duck (Aix sponsa) embryo to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) by injecting the toxicant into their eggs. Six groups of wood duck eggs (n = 35 to 211 per trial) were injected with 0 to 4600 pg TCDD/g egg between 2003 and 2005. Injections were made into yolk prior to incubation, and eggs were subsequently incubated and assessed weekly for mortality. Significant TCDD-induced mortality was not observed through day 25 (90% of incubation). Liver, heart, eye, and brain histology were generally unremarkable. Direct and indirect causes of sex differences in mercury concentrations and parasitic infections in a marine bird.

In many animal species, males and females differ in their levels of contaminants and/or parasitic infections. Most contaminants and gastro-intestinal parasites are obtained through prey ingestion, and thus the causes of sex differences in the distribution of these factors might follow similar pathways. We studied the northern common eider duck (Somateria molissima borealis) as an avian model, and used directed separation path analysis to explore the causes of sex differences in mercury (Hg) and gastro-intestinal helminths. Two trophically transmitted helminths were examined: a cestode (Laterip

Waterbird Susceptibility to Avian Cholera at Hayward Marsh, California, USA.

We characterized past avian cholera outbreaks in waterbirds at Hayward Marsh, California, US. In 2013, we surveyed populations and determined the presence of disease using several diagnostic methods, including behavioral and physical observations, field necropsy, and bacterial culture. We compiled this information with data from previous outbreaks from 1990-2012 to compare waterbird abundance to various measures of mortality, including percentage of mortality and percentage of difference between abundance and mortality by species. We suggest that Ruddy Duck ( Oxyura jamaicensis ) have consiste

Adaptive amino acid substitutions enhance the virulence of an H7N7 avian influenza virus isolated from wild waterfowl in mice.

Although H7N7 AIVs primarily circulate in wild waterfowl, documented cases of human infection with H7N7 viruses suggest they may pose a pandemic threat. Here, we generated mouse-adapted variants of a wild waterfowl-origin H7N7 virus to identify adaptive changes that confer enhanced virulence in mammals. The mouse lethal doses (MLD50) of the adapted variants were reduced >5000-fold compared to the parental virus. Mouse-adapted variants viruses displayed enhanced replication in vitro and in vivo, and acquired the ability to replicate in extrapulmonary tissues. These observations suggest that enh

Molecular detection and characterization of Trichomonas spp. in wild birds in the Brazilian Pantanal.

Members of the family Trichomonadidae, particularly Trichomonas gallinae, are globally distributed avian parasites that primarily infect birds from the orders Columbiformes, Falconiformes, and Strigiformes. Although infections in Passeriformes are often subclinical, clinical cases have been reported. Transmission occurs through direct contact or indirectly via contaminated food or water, enabling infection across various avian orders, including Anseriformes, Falconiformes, Galliformes, Gruiformes, Passeriformes, Piciformes, Psittaciformes, and Strigiformes. This study aimed to assess the occur

Synthesis of Maternal Transfer of Mercury in Birds: Implications for Altered Toxicity Risk.

Maternal transfer is a predominant route of methylmercury (MeHg) exposure to offspring. We reviewed and synthesized published and unpublished data on maternal transfer of MeHg in birds. Using paired samples of females' blood (n = 564) and their eggs (n = 1814) from 26 bird species in 6 taxonomic orders, we conducted a meta-analysis to evaluate whether maternal transfer of MeHg to eggs differed among species and caused differential toxicity risk to embryos. Total mercury (THg) concentrations in eggs increased with maternal blood THg concentrations; however, the proportion of THg transferred fro

Phorcotabanus cinereus (Wiedemann, 1821) (Diptera, Tabanidae), an ornithophilic species of Tabanid in Central Amazon, Brazil.

In Central Amazon, Brazil, the tabanid Phorcotabanus cinereus (Wiedemann) was recorded attacking the native duck Cairina moschata (Linnaeus) (Anseriformes, Anatidae). The flight and behavior of the tabanid during the attacks and the host's defenses were videotaped and analyzed in slow motion. The tabanid was recorded flying rapidly around the heads of the ducks before landing. Landing always took place on the beak, and then the tabanid walked to the fleshy caruncle on the basal part of the beak to bite and feed. Firstly the duck defends itself through lateral harsh head movements, and then, wh
