Intravenous paracetamol as an antipyretic and analgesic medication: the significance of drug metabolism.

One prospective, open-label, non-randomized study was conducted in 100 patients to define the antipyretic and analgesic effect of a new intravenous formulation of 1 g of paracetamol; 71 received paracetamol for the management of fever and 29 received paracetamol for pain relief after abdominal surgery or for neoplastic pain. Serial follow-up measurements of core temperature and of pain intensity were done for 6 h. Additional rescue medications were recorded for 5 days. Blood was sampled for the measurement of free paracetamol (APAP) and of glucuronide-APAP and N-sulfate-APAP by an HPLC assay.

Developmental effects of propyphenazone in analgesic and antipyretic combination with caffeine or paracetamol.

The aim of the study was to determine the influence of an over-the-counter (OTC) mixture of propyphenazone with caffeine or paracetamol on prenatal development. Propyphenazone:caffeine and propyphenazone:paracetamol mixtures were prepared with constant 3:1 and 3:5 ratios, respectively. Three dose levels of each of the mixtures were administered separately in Tween-80 water suspension once a day to pregnant Wistar rats on gestation days 8-14. The low dose was similar to the OTC preparations, 2.1 mg/kg of propyphenazone, 0.7 mg/kg of caffeine or 3.5 mg/kg of paracetamol. The middle dose was 21.0

Possible involvement of brain oxytocin in modulating vasopressin antipyretic action.

Experiments were undertaken to test whether oxytocin (OT) may modulate the antipyretic action of arginine vasopressin (AVP) and to determine whether the action of endogenously released OT and/or AVP evoked by fever may modulate the motor actions of exogenous AVP. Intracerebroventricular (icv) injection of interleukin-1 alpha (IL-1 alpha, 40 ng) elicited a significantly attenuated rise in body temperature during the 2nd h of the febrile responses in OT-pretreated (0.1-10 pmol icv, 24 h earlier) rats. At the end of the 2nd h, administration of AVP (1 pmol icv), but not OT (10 pmol icv), signific

Analysis of the antipyretic action of alpha-melanocyte-stimulating hormone in rabbits.

alpha-Melanocyte-stimulating hormone (alpha-MSH) or paracetamol was injected into a lateral cerebral ventricle (I.C.V.) of rabbits with elevations in rectal temperature induced by sodium arachidonate (I.C.V.), prostaglandin E2 (I.C.V.) or leucocytic pyrogen (I.V.). alpha-MSH (200 ng) was more effective than paracetamol (0.5 mg) in reducing fever caused by leucocytic pyrogen, but it did not alter hyperthermia induced by sodium arachidonate. In contrast, paracetamol reduced hyperthermic responses to arachidonate by about 70%. Neither alpha-MSH nor paracetamol affected hyperthermic responses to p

Antipyretic use among children during the 1983 influenza season.

We conducted a telephone survey in Houston in March 1983 to assess the level of aspirin use among children during the influenza season. We completed interviews of 200 households with 346 children aged 12 years or younger. Fifty-two percent of the 346 children experienced at least one acute illness in the preceding three months. Fever was measured with a thermometer in 114 ill children, 103 of whom had at least one measured temperature of 37.7 degrees C or greater. Fourteen percent of these 103 children received aspirin only, 61% received acetaminophen only, and 20% received both. Among a subgr

Changes in body temperature after administration of antipyretics, LSD, delta 9-THC and related agents: II.

Antipyretics, in particular acetaminophen, aspirin and ibuprofen, constitute the single most important class of drugs used therapeutically for an effect on body temperature. Hallucinogens exert prominent actions on the central nervous system, and it is not surprising that, like so many other centrally-acting agents, they too often affect temperature. This compilation primarily covers the considerable amount of data published from 1981 through 1985 on the interactions of these drugs and thermoregulation, but data from many earlier papers not included in a previous compilation are also tabulated

Antipyretic Mechanism Exploration of HuanglianShangqing Pill Based on Metabolomics and Network Pharmacology.

BACKGROUND AND OBJECTIVE: HuanglianShangqing pill (HLSQ), a well-known traditional Chinese medicine (TCM), has been used to treat fever in China for a long time. Our previous study had demonstrated that a total of 45 prototype components of HLSQ could be absorbed into the plasma of rats after intragastric administration. However, the detailed mechanisms related to the antipyretic effects of HLSQ were still unclear. METHODS: In the present work, urinary metabolomics coupled with network pharmacology were employed to evaluate the mechanisms of HLSQ in the treatment of fever compared with ibupro

The antipyretic effect of dipyrone is unrelated to inhibition of PGE(2) synthesis in the hypothalamus.

BACKGROUND AND PURPOSE: Bacterial lipopolysaccharide (LPS) induces fever through two parallel pathways; one, prostaglandin (PG)-dependent and the other, PG-independent and involving endothelin-1 (ET-1). For a better understanding of the mechanisms by which dipyrone exerts antipyresis, we have investigated its effects on fever and changes in PGE(2) content in plasma, CSF and hypothalamus induced by either LPS or ET-1. EXPERIMENTAL APPROACH: Rats were given (i.p.) dipyrone (120 mg·kg(-1)) or indomethacin (2 mg·kg(-1)) 30 min before injection of LPS (5 µg·kg(-1), i.v.) or ET-1 (1 pmol, i.c.v.).

Phenacetin isolated from Bursera grandifolia, a herbal remedy with antipyretic properties.

Bursera grandifolia and other related species have been used in traditional herbal medicine in Mexico and other Latin American countries for their analgesic, antipyretic and anti-inflammatory properties. From the chloroform extract of leaves of B. grandifolia, a substance was isolated and identified as phenacetin, a well known compound with widely tested analgesic and antipyretic properties. The structural identity of the compound was elucidated on the basis of chemical and spectroscopic evidence and by comparison with an authentic sample.

Hepatitis during measles in young adults: possible role of antipyretic drugs.

Fifty-six per cent of 118 young adults observed during a recent measles epidemic had some disturbance on liver function tests. Five per cent developed overt jaundice. Patients treated for fever with paracetamol were found to have significantly higher rates of transaminase impairment compared to those treated with dipyrone. Sixty-five and 58% of patients given paracetamol had elevated ALT and AST levels, respectively. Only 15% of patients given dipyrone had elevated levels of these two enzymes (p less than 0.01 for ALT and p less than 0.02 for AST). The mean levels of transaminases and bilirubi

Anthropogenic and temporal components in a complex trigger of type 1 diabetes suggest the active participation of antipyretics.

Tremendous efforts in research without a conclusion on the cause of type 1 diabetes allow the presumption that there is still a blind spot in the development of T1D that is not covered by current hypotheses. The review of geographical knowledge suggests that there is a well-expressed anthropogenic element within the complex environmental trigger of T1D. On the other hand, the initiation of T1D's directed autoimmunity is temporally related to the organism's immune response, induced by entero-viruses, most expectedly. Consequently, the searched for anthropogenic environmental factor is a player

Effects of antipyretics on mortality due to influenza B virus in a mouse model of Reye's syndrome.

OBJECTIVES: To determine the effects of acetylsalicylic acid (ASA) and acetaminophen on mortality due to influenza B infection in neonatal and weanling mice, as well as any synergistic, antagonistic or indifferent effects of the combined antipyretic and virus on mortality in mice pretreated with low doses of an industrial surfactant, Toximul MP8, which has been shown to reproduce many of the features of Reye's syndrome. In vitro studies were done to determine whether ASA or acetaminophen altered the normal, interferon-mediated antiviral responses of mammalian cells. The involvement of ASA or o
