Shared Transcriptional Control and Disparate Gain and Loss of Aphid Parasitism Genes.

Aphids are a diverse group of taxa that contain agronomically important species, which vary in their host range and ability to infest crop plants. The genome evolution underlying agriculturally important aphid traits is not well understood. We generated draft genome assemblies for two aphid species: Myzus cerasi (black cherry aphid) and the cereal specialist Rhopalosiphum padi. Using a de novo gene prediction pipeline on both these, and three additional aphid genome assemblies (Acyrthosiphon pisum, Diuraphis noxia, and Myzus persicae), we show that aphid genomes consistently encode similar gen

Response of wheat germplasm to infestation of English grain aphid (Hemiptera: Aphididae).

The English grain aphid, Sitobion avenae (F.), is a destructive insect pest of wheat. In this study, wheat germplasm was evaluated for resistance and tolerance to English grain aphid infestation. Except for spikelet number/spike, the 1,000-kernel weight, spike weight, grain weight/ spike, and sterile spikelet number/spike of infested plants were all significantly impacted by English grain aphid infestation. Of 292 wheat accessions, only one genotype showed English grain aphid high resistance, and most genotypes showed intermediate resistance. Tolerance index values ranged widely among all acce

Heterodera schachtii nematodes interfere with aphid-plant relations on Brassica oleracea.

Aboveground and belowground herbivore species modify plant defense responses differently. Simultaneous attack can lead to non-additive effects on primary and secondary metabolite composition in roots and shoots. We previously found that aphid (Brevicoryne brassicae) population growth on Brassica oleracea was reduced on plants that were infested with nematodes (Heterodera schachtii) prior (4 weeks) to aphid infestation. Here, we examined how infection with root-feeding nematodes affected primary and secondary metabolites in the host plant and whether this could explain the increase in aphid dou

Host-associated populations in the lettuce root aphid, Pemphigus bursarius (L.).

Pemphigus bursarius is a host-alternating aphid in which annual rounds of sexual reproduction on its primary host, Populus nigra, are interspersed with parthenogenesis on a range of secondary hosts. Evidence was sought for the existence of genetically distinct populations, associated with different secondary hosts, in P. bursarius. Microsatellite markers revealed that genetically distinct populations were present on three different secondary host species. Microsatellites were also used, in conjunction with mitochondrial DNA sequence variation, to investigate the relationships between aphids on

The significance of gut sucrase activity for osmoregulation in the pea aphid, Acyrthosiphon pisum.

The osmotic pressure of the body fluids of aphids is lower than in their diet of plant phloem sap. It is hypothesised that aphids reduce the osmotic pressure of ingested food by sucrase-mediated hydrolysis of dietary sucrose to glucose and fructose, and the polymerisation of glucose into oligosaccharides of low osmotic pressure per hexose unit. To test this hypothesis, the impact of the alpha-glucosidase inhibitor acarbose on the sugar relations and osmoregulation of aphids was explored. Acarbose inhibited sucrase activity in gut homogenates and the production of monosaccharides and oligosacch

Gene expression profiling of Arabidopsis thaliana in compatible plant-aphid interactions.

Phloem feeding involves unique biological interactions between the herbivore and its host plant. The economic importance of aphids, whiteflies, and other phloem-feeding insects as pests has prompted research to isolate sources of resistance to piercing-sucking insects in crops. However, little information exists about the molecular nature of plant sensitivity to phloem feeding. Recent discoveries involving elicitation by plant pathogens and chewing insects and limited studies on phloem feeders suggest that aphids are capable of inducing responses in plants broadly similar to those associated w

Emergent symbiont strains provide thermally robust protection against co-evolved and novel parasitoids of introduced pea aphids.

Climate change and biological invasions pose synergistic threats; however, organisms may rapidly adapt through microbial symbiosis. We investigated how defensive symbionts in invasive pea aphids, Acyrthosiphon pisum, respond to emerging threats. Previously rare strains of the protective symbiont Hamiltonella defensa increased from <0.5% to 58% in aphid populations over just a few years. Bioassays revealed that these strains confer reciprocal, enemy-specific defences. One strain (C11) protected against Praon pequodorum, a native parasitoid that only began attacking pea aphids post-introduction,

Transgenic plants expressing ω-ACTX-Hv1a and snowdrop lectin (GNA) fusion protein show enhanced resistance to aphids.

Recombinant fusion proteins containing arthropod toxins have been developed as a new class of biopesticides. The recombinant fusion protein Hv1a/GNA, containing the spider venom toxin ω-ACTX-Hv1a linked to snowdrop lectin (GNA) was shown to reduce survival of the peach-potato aphid Myzus persicae when delivered in artificial diet, with survival <10% after 8 days exposure to fusion protein at 1 mg/ml. Although the fusion protein was rapidly degraded by proteases in the insect, Hv1a/GNA oral toxicity to M. persicae was significantly greater than GNA alone. A construct encoding the fusion protein

Using network pharmacology and structural biology to elucidate the molecular mechanism by which Macrolactin A protects against aphids.

Aphids are major agricultural pests that cause severe crop damage. While chemical pesticides remain the dominant control strategy, their associated environmental contamination and resistance issues necessitate the development of eco-friendly biopesticides. This study employed network pharmacology and structural biology to elucidate the molecular mechanism by which Macrolactin A protects against aphids, with validation through molecular docking, 100-ns molecular dynamics (MD) simulations, and enzymatic assays. Target prediction identified five aphid proteins as potential targets of Macrolactin

Efficacy of 2 botanical aphicides, chicoric and 3,5-dicaffeoylquinic acids, on aphids susceptible and resistant to synthetic insecticides.

Dicaffeoyltartaric acid (diCT) and 3,5-dicaffeoylquinic acid (3,5-diCQ) are described for their aphicidal properties on several aphid species. Intending to valorize diCT and 3,5-diCQ as biocontrol products and because of the high adaptive capacities of aphids to xenobiotics, we sought to determine the existence of adaptation first in Myzus persicae (Sulzer) (Hemiptera: Aphididae) and then other aphids. Resistance of aphids to these biopesticides could be promoted by (i) the existence of resistance to synthetic insecticides that may confer cross-resistance and (ii) the presence of these compoun

The lost generation of Pemphigus populiglobuli (Hemiptera, Aphididae): exploring the taxonomy of the Svalbard aphids of genus Pemphigus.

Species identification within the aphid genus Pemphigus Hartig, 1839 poses challenges due to morphological similarities and host-plant associations. Aphids of this genus generally exhibit complex life cycles involving primary hosts (poplars) and secondary (mostly unrelated herbaceous) host-plants, with some species relying solely on root-feeding generation. An example is a representative of the genus Pemphigus, trophically associated with grass roots, found in the High Arctic Svalbard archipelago. Historical records tentatively identify it as Pemphigus groenlandicus (Rübsaamen, 1898), although

Spray application of dsRNA reduces aphid transmitted ZYMV infection in Cucurbita pepo.

Exogenous application of double-stranded RNA (dsRNA) for protection against plant viruses has been demonstrated in a range of pathosystems. Here, we demonstrate the protective efficacy of spray-induced gene silencing (SIGS) in zucchini plants (Cucurbita pepo) against virulent zucchini yellow mosaic virus (ZYMV) upon mechanical inoculation and importantly, aphid mediated transmission under glasshouse environs. We show treatment of plants with 300 ng μL-1 ZYMV_CP-dsRNA (∼240 - 270 μg plant -1) reduced infection rates by ∼73 - 100% when all plants sprayed at day 0 were challenged by mechanical in

Functional assessment of the vascular and translaminar systemicity of insecticides in a cucumber-cotton aphid model.

It is difficult to control insect pests with piercing-sucking mouthparts such as aphids due to their concealed feeding behavior and limited exposure to foliar-applied insecticides. Although systemic insecticides with internal translocation through plant tissues provide a practical alternative to contact-based strategies, they often suffer from uneven deposition and behavioral avoidance by pests. Furthermore, conventional assays used for investigating systemicity provide useful information on insecticide performance; however, their outcomes can reflect a combination of systemic translocation an

Traits analysis of an aphid-killing Pantoea agglomerans strain for use in biological control.

INTRODUCTION: In crop pest control, microbial bioinsecticides are considered promising for overcoming the critical limitations of traditional chemical pesticides regarding environmental and health impact. In this study, we report a strain of Pantoea agglomerans (ELIV) lethal to black bean aphid Aphis fabae: we investigated the effects of this bacterium on aphids and host plants to test its potential as a natural insecticide. METHODS AND RESULTS: We found that the entomopathogenic properties of P. agglomerans ELIV occur only through ingestion via oral infection assays, with no pathogenicity by

Multiple toxins and a protease contribute to the aphid-killing ability of Pseudomonas fluorescens PpR24.

Aphids are globally important pests causing damage to a broad range of crops. Due to insecticide resistance, there is an urgent need to develop alternative control strategies. In our previous work, we found Pseudomonas fluorescens PpR24 can orally infect and kill the insecticide-resistant green-peach aphid (Myzus persicae). However, the genetic basis of the insecticidal capability of PpR24 remains unclear. Genome sequencing of PpR24 confirmed the presence of various insecticidal toxins such as Tc (toxin complexes), Rhs (rearrangement hotspot) elements, and other insect-killing proteases. Upon

A novel gene REPTOR2 activates the autophagic degradation of wing disc in pea aphid.

Wing dimorphism in insects is an evolutionarily adaptive trait to maximize insect fitness under various environments, by which the population could be balanced between dispersing and reproduction. Most studies concern the regulatory mechanisms underlying the stimulation of wing morph in aphids, but relatively little research addresses the molecular basis of wing loss. Here, we found that, while developing normally in winged-destined pea aphids, the wing disc in wingless-destined aphids degenerated 30-hr postbirth and that this degeneration was due to autophagy rather than apoptosis. Activation

Transcriptomic and volatile signatures associated with maize defense against corn leaf aphid.

BACKGROUND: Maize (Zea mays L.) is a major cereal crop, with the United States accounting for over 40% of the worldwide production. Corn leaf aphid [CLA; Rhopalosiphum maidis (Fitch)] is an economically important pest of maize and several other monocot crops. In addition to feeding damage, CLA acts as a vector for viruses that cause devastating diseases in maize. We have shown previously that the maize inbred line Mp708, which was developed by classical plant breeding, provides heightened resistance to CLA. However, the transcriptomic variation conferring CLA resistance to Mp708 has not been i

Poplar Tree Response to Feeding by the Petiole Gall Aphid Pemphigus spyrothecae Pass.

Pemphigus spyrothecae Pass. which is a member of the subfamily Pemphiginae is one of the gall-inducing aphids that occurs on poplar trees. Phloem feeding of a founding mother on leaf petiole results in the formation of a new organ, i.e., the spiral gall. This study documents aphid development inside the galls during the growing season and the effect of their feeding on leaf architecture and physiology of the host plant. In particular, leaf length, width, and area were measured, as well as hydrogen peroxide (H2O2) content, electrolyte leakage (EL), malondialdehyde (MDA) concentration, and the a

Spatiotemporal fluctuations in natural selection acting on the gall-parasitic aphid Tetraneura sorini.

The measurement of the selection gradient is crucial for understanding the magnitude of selection acting directly on a trait and predicting the evolutionary trajectory of that trait. This study evaluated the selection gradient acting on the morphology of the gall-parasitic aphid Tetraneura sorini during the galling process and compared the strength among populations. Gall formers (first instars) frequently fight with conspecifics or heterospecifics for usurping incipient galls using their well-developed hind legs. First instars that successfully acquired galls were found within galls, whereas
