Apolipoproteins and amyloid fibril formation in atherosclerosis.

Amyloid fibrils arise from the aggregation of misfolded proteins into highly-ordered structures. The accumulation of these fibrils along with some non-fibrillar constituents within amyloid plaques is associated with the pathogenesis of several human degenerative diseases. A number of plasma apolipoproteins, including apolipoprotein (apo) A-I, apoA-II, apoC-II and apoE are implicated in amyloid formation or influence amyloid formation by other proteins. We review present knowledge of amyloid formation by apolipoproteins in disease, with particular focus on atherosclerosis. Further insights into

Alterations in lipoprotein density classes in infantile visceral leishmaniasis: presence of apolipoprotein SAA.

This study describes the alterations in the plasma lipoproteins from nine young Tunisian children with active visceral Leishmaniasis. The plasma lipid profile from affected patients was characterized by a marked hypertriglyceridaemia associated with reduced levels of total and high density lipoprotein (HDL)-cholesterol and a significant increase in the plasma ratio of unesterified to total cholesterol. Quantitative determination of plasma apolipoproteins revealed significantly decreased levels of all measured apolipoproteins, especially of apolipoproteins A-I and A-II, with the exception of ap

Relation of Serum Lipoprotein(a) Levels to Lipoprotein and Apolipoprotein Profiles and Atherosclerotic Diseases in Japanese Patients with Heterozygous Familial Hypercholesterolemia: Familial Hypercholesterolemia Expert Forum (FAME) Study.

AIMS: Lipoprotein(a) [Lp(a)] is a plasma lipoprotein consisting of a low-density lipoprotein (LDL)-like particle with apolipoprotein (Apo)(a), attached via a disulfide bond to Apo B100. Previous studies have shown that high Lp(a) levels are associated with an increased risk of cardiovascular disease in patients with familial hypercholesterolemia (FH). To date, limited data are available as to distribution of Lp(a) in FH and associations of Lp(a) with other lipid profiles and cardiovascular disease. Our study aimed to investigate serum Lp(a) levels in relation to other lipid profiles and clinic

Factors affecting the lipid and apolipoprotein levels of cord sera.

In studying the human lipid transport system as it changes from the fetal to the adult form, data were collected on cord sera levels of lipids and apolipoproteins (A-I, A-II, ApoB, C-I, C-II, C-III, ApoD, and ApoE) from infants whose lipid metabolism might have been modified by a variety of factors present before, at, or after delivery. The results clearly indicated that many factors affect the levels of both lipids and apolipoproteins at birth. Method of delivery had little effect except in relation to either fetal immaturity (increased total cholesterol) or fetal stress (increased triglyceri

Very-Low-Density Lipoprotein-Associated Apolipoproteins Predict Cardiovascular Events and Are Lowered by Inhibition of APOC-III.

BACKGROUND: Routine apolipoprotein (apo) measurements for cardiovascular disease (CVD) are restricted to apoA-I and apoB. Here, the authors measured an unprecedented range of apolipoproteins in a prospective, population-based study and relate their plasma levels to risk of CVD. OBJECTIVES: This study sought to measure apolipoproteins directly by mass spectrometry and compare their associations with incident CVD and to obtain a system-level understanding of the correlations of apolipoproteins with the plasma lipidome and proteome. METHODS: Associations of 13 apolipoproteins, 135 lipid species

The prognostic significance of HDL-associated apolipoproteins in ascitic fluid from patients with cirrhosis and spontaneous bacterial peritonitis.

Apolipoproteins (Apo) A-I and A-II are major components of high-density lipoproteins (HDL) with immunomodulatory functions. Low serum apoA-I levels indicate complications of cirrhosis. We hypothesized that HDL-associated apolipoproteins in ascitic fluid (AF) are differentially abundant during spontaneous bacterial peritonitis (SBP). Serum and non-infected AF samples from 18 patients with cirrhosis (cohort 1) were analyzed using LC-MS/MS to identify differentially regulated apolipoproteins. In cohort 2, samples from 59 patients with SBP and 59 without were analyzed for AF apoA-II and apoA-I con

Variation in lipoprotein(a) concentration associated with different apolipoprotein(a) alleles.

Plasma lipoprotein(a) (Lp(a)) concentrations vary considerably between individuals. To examine the variation for products of the same and different apolipoprotein(a) (apo(a)) alleles, conditions were established whereby phenotyping immunoblots could be used to estimate the concentration of Lp(a) associated with the constituent apo(a) isoforms. In these studies 28 distinct isoforms were identified, each differing by a single kringle IV unit. Tracking the isoforms through 10 families showed that there could be up to 200-fold difference in the Lp(a) concentration associated with the same-sized is

The role of genetic variation in apolipoprotein response to diet among Brazilians with cardiovascular disease.

BACKGROUND/OBJECTIVES: Data on the interaction between dietary patterns, single nucleotide polymorphisms (SNPs), and cardiometabolic markers remain limited in genetically admixed populations, particularly among individuals with established cardiovascular disease (CVD). This study aimed to evaluate how SNPs modulate the effects of dietary interventions on apolipoprotein concentrations (A-I, A-II, B, C-II, C-III, and E) in Brazilians undergoing secondary cardiovascular prevention. SUBJECTS/METHODS: This sub-study used data from the three-year multicenter BALANCE Trial, which enrolled individual

Apolipoprotein A and prognosis after myocardial infarction in non-diabetic men.

The implications of apolipoproteins A-I and A-II for the prognosis of 178 non-diabetic men after acute myocardial infarction were studied. During a mean follow-up period of 4 years, one or more "coronary events" (nonfatal myocardial infarction, fatal coronary heart disease, coronary artery bypass graft surgery, deterioration of exercise ECG) were recorded in 37 patients. Serum levels of apolipoproteins A-I and A-II did not discriminate between patients with and without coronary events. This applied to the entire sample as much as to subgroups defined by presence or absence of interventions (co

Plasma high-density lipoprotein cargo is altered in Alzheimer's disease and is associated with regional brain volume.

Cholesterol levels have been repeatedly linked to Alzheimer's Disease (AD), suggesting that high levels could be detrimental, but this effect is likely attributed to Low-Density Lipoprotein (LDL) cholesterol. On the other hand, High-Density Lipoproteins (HDL) cholesterol levels have been associated with reduced brain amyloidosis and improved cognitive function. However, recent findings have suggested that HDL-functionality, which depends upon the HDL-cargo proteins associated with HDL, rather than HDL levels, appears to be the key factor, suggesting a quality over quantity status. In this repo

The distribution of apo C-II and apo C-III in very low density lipoproteins of normal and type IV subjects.

The apoC-II/apoC-III ratio and the distribution of C apoproteins as percentage of the total protein in VLDL obtained from normolipidemic and type IV patients was studied by analytical isoelectrofocusing. While the percentage concentration of apoCI-III was unchanged that of apoC-II was 9.2% of total protein in normal VLDL and only 7.4% in type IV VLDL (P less than 0.05). The apoC-II/apo C-III ratio significantly decreased in type IV patients. The analysis of VLDL subfractions obtained by density gradient ultracentrifugation gave essentially the same results. These data confirm the decrease of a

Mechanism of action of gemfibrozil on lipoprotein metabolism.

Gemfibrozil is a potent lipid regulating drug whose major effects are to increase plasma high density lipoproteins (HDL) and to decrease plasma triglycerides (TG) in a wide variety of primary and secondary dyslipoproteinemias. Its mechanism of action is not clear. Six patients with primary familial endogenous hypertriglyceridemia with fasting chylomicronemia (type V lipoprotein phenotype) with concurrent subnormal HDL cholesterol levels (HDL deficiency) were treated initially by diet and once stabilized, were given gemfibrozil (1,200 mg/d). Each patient was admitted to the Clinical Research Ce

Lipids and apolipoproteins in patients treated with major tranquilizers.

The concentrations of lipids and apolipoproteins in serum from men with chronic schizophrenia who were receiving major tranquilizers (17 receiving phenothiazines and 14 receiving a butyrophenone) were quantitated and compared with serum from male controls (n = 14). Concentrations of high-density lipoprotein cholesterol and apolipoproteins A-I and A-II were significantly lower in the patients than in the controls. Mean apolipoproteins C-II and C-III and very low-density lipoprotein cholesterol levels in the patients receiving phenothiazines were higher than levels in the patients receiving buty

Changes in apolipoproteins following ingestion of a beverage delivering β-hydroxybutyrate: results from a randomized placebo-controlled trial.

BACKGROUND: Apolipoproteins play important roles in the metabolism of triglyceride-rich lipoproteins. Ketone monoester β-hydroxybutyrate (KEβHB) has been shown to reduce the circulating levels of remnant cholesterol and triglycerides. However, the mechanisms behind this action remain unknown. AIM: To investigate the effect of KEβHB supplementation on apolipoproteins and to study whether circulating levels of triglycerides play a role in this effect. METHODS: The study was a randomized placebo-controlled trial, registered at https://www.clinicaltrials.gov/ (NCT03889210). It included 18 adults

Apolipoprotein status in type 2 diabetes mellitus and its complications (Review).

Dyslipidaemia in type 2 diabetes mellitus (T2DM) is characterized by high plasma triglyceride concentrations, reduced high‑density lipoprotein concentrations and increased small density low‑density lipoprotein concentrations. Dyslipidaemia may lead to cardiovascular disease (CVD) and other complications. Apolipoproteins mainly comprise six species, apolipoprotein (apo)A, apoB, apoC, apoD, apoE and apoM, which are important components of plasma lipoproteins that carry lipids and stabilize the structure of lipoproteins. Complex metabolic disorders of apolipoproteins are present in T2DM, such as

Plasma and lipoprotein lipids and apolipoproteins AI, AII and B in patients with chronic airflow limitation.

Plasma and lipoprotein cholesterol and triglycerides, and plasma apolipoproteins AI, AII and B were compared in patients with chronic airflow limitation, and normal controls matched for body mass index. The controls were non-smokers, and free from respiratory disease. High-density lipoprotein (HDL) cholesterol concentration was significantly elevated in the patients, due mainly to a raised HDL2 cholesterol level. HDL triglyceride was significantly lower in the patients. All other lipids were not different from normal. Apolipoprotein AI levels were significantly raised in the patients but other

[The apolipoproteins in man ].

Twelve different apolipoproteins have been described in human serum. Apo A-I and apo A-II are essential for the structure of the HDL particles and for the function of LCAT activity. Apo B is the main protein in LDL but does also occur in the triglyceride-rich particles. Apo B represents the binding protein for the LDL-receptor pathway. The C-apolipoproteins are located on the surface of VLDL. They are transferred to HDL throughout the catabolism of VLDL and affect lipoprotein lipase activity. This enzyme is also affected by the E-apolipoproteins which occur in the triglyceride-rich particles a
