Excessive Cu2+ deteriorates arsenite-induced apoptosis in chicken brain and resulting in immunosuppression, not in homeostasis.

Trace elements such as copper (Cu) and arsenic (As) are two of the major contaminants and well-known inducers of cognitive deficits and neurobehavioral changes. This study evaluated the immunotoxicity of their individual or combined exposure on different brain regions in chickens. Consequently, nuclear damage and organelle lesions, especially mitochondria were observed under Cu or/and As stress, in which positive regulation of key proteins, dynamin-related protein 1 (Drp1), Cytochrome C (Cyt c), BCL2-associated X (Bax), Caspases 3 and P53 was detected by qRCR and Western blot analyses, indicat

Evaluation of the toxic effects of arsenite, chromate, cadmium, and copper using a battery of four bioassays.

The sensitivities of four different kinds of bioassays to the toxicities of arsenite, chromate, cadmium, and copper were compared. The different bioassays exhibited different sensitivities, i.e., they responded to different levels of toxicity of each of the different metals. However, with the exception of the α-glucosidase enzyme activity, arsenite was the most toxic compound towards all the tested organisms, exhibiting the highest toxic effect on the seeds of Lactuca, with an EC(50) value of 0.63 mg/L. The sensitivities of Lactuca and Raphanus were greater than the sensitivities of two other

Arsenite suppression of involucrin transcription through AP1 promoter sites in cultured human keratinocytes.

While preserving keratinocyte proliferative ability, arsenite suppresses cellular differentiation markers by preventing utilization of AP1 transcriptional response elements. In present experiments, arsenite had a dramatic effect in electrophoretic mobility supershift analysis of proteins binding to an involucrin promoter AP1 response element. Without arsenite treatment, binding of JunB and Fra1 was readily detected in nuclear extracts from preconfluent cultures and was not detected a week after confluence, while c-Fos was detected only after confluence. By contrast, band shift of nuclear extra

Preparation and evaluation of a novel Fe-Mn binary oxide adsorbent for effective arsenite removal.

Arsenite (As(III)) is more toxic and more difficult to remove from water than arsenate (As(V)). As there is no simple treatment for the efficient removal of As(III), an oxidation step is always necessary to achieve higher removal. However, this leads to a complicated operation and is not cost-effective. To overcome these disadvantage, a novel Fe-Mn binary oxide material which combined the oxidation property of manganese dioxide and the high adsorption features to As(V) of iron oxides, were developed from low cost materials using a simultaneous oxidation and coprecipitation method. The adsorben

Embryotoxicity of arsenite and arsenate: distribution in pregnant mice and monkeys and effects on embryonic cells in vitro.

The distribution of 74As-labelled arsenate and arsenite in pregnant mice and a monkey has been studied by autoradiography and gamma counting of isolated tissues, and their in vitro toxicity to a chondrogenic system has been investigated. With both arsenic forms, given as single intravenous injections to the mother, the 74As-arsenic appeared to pass the mouse placenta relatively free and approximately to the same extent. The retention time in maternal tissues including the placenta was, however, around three times longer with arsenite than with arsenate. In early gestation, high activity was re

Sonochemically synthesized CuO-NPs: individual and synergistic toxicity assessment with sodium arsenite in adult zebrafish.

Sodium arsenite (NaAsO2) and sonochemically synthesized copper oxide nanoparticles (CuO-NPs) pose escalating risks to aquatic animals due to their bio-accumulative potential, persistence, and interaction; when co-occurring, their combined effects represent a new class of emerging contaminants (ECs) with heightened ecological relevance. We synthesized CuO-NPs and characterized them using powder X-ray diffraction, scanning electron microscopy, Fourier transform infrared spectroscopy, energy dispersive X-ray spectroscopy and UV-visible spectroscopy. Zebrafish (Danio rerio) were exposed to NaAsO2,

Aquaporin-3 amplifies arsenite genotoxicity as a dose-dependent gene-by-environment interaction in human cells.

Chronic exposure to inorganic arsenic affects more than 200 million people, and disease outcome varies widely among individuals with comparable intake. Explanations have centered on arsenic metabolism, which acts on arsenite that is already inside the cell. Uptake sits upstream of metabolism and sets how much arsenite reaches the genome, yet whether it acts as a gene-by-environment modifier of genotoxicity has been asserted far more often than it has been measured. We expressed human aquaporin-3 (AQP3), its point mutants and the paralogues AQP7, AQP9 and AQP10 in three human cell lines, and me

Neuroprotection by Nauclea latifolia extract in arsenite & high-fat diet-induced brain stress.

Sodium arsenite (NaAsO2) and high fat diet (HFD) are already documented to provoke oxidative stress, neuro inflammation and learning and memory deficits. This work aimed to determine the possible neuroprotection of the root extract of Nauclea latifolia (NlREq) against NaAsO2/HFD induced neurotoxicity in Wistar rats. Twenty-five rats were divided into five groups: groups include control; NaAsO2/HFD treated; NaAsO2/HFD + NlREq at 200 mg /kg and 400 mg/kg; and NaAsO2/HFD treated with silymar in at the dose of 50 mg/kg. The miR-125a-5p/IRF4 Axis Mediates Sodium Arsenite-Induced M2 Macrophage Polarization.

Arsenic, a ubiquitous metalloid, is commonly found in surface waters; as well as serious human health issues, it also induces systemic diseases and carcinogenesis upon chronic exposure. To better understand how arsenic potentially alters the immune system, it is important to study its effects on macrophage polarization. Micro-RNA plays an epigenetic regulatory role in organisms. The miR-125 family regulates macrophage polarization and tumorigenesis, yet its role in arsenic-induced macrophage polarization remains unexplored. This study investigated the mechanism of sodium arsenite (NaAsO2)-driv

Butylated hydroxytoluene protects against sodium arsenite-induced hepatorenal toxicities: in vivo and in silico studies.

Arsenic is a global environmental contaminant. Sodium arsenite (NaAsO2), one of the arsenic compounds, is toxic. Prolonged exposure to NaAsO2 causes various detrimental effects on body organs. Butylated hydroxytoluene (BHT), a synthetic antioxidant may retard the deleterious effects of free-radical-induced damage. We hereby investigated the chemo-protective role of BHT on sodium arsenite-induced hepatorenal toxicity in rats. Rats (n = 49, 110 ± 10 g) were grouped into seven with 7 animals each: groups 1 and 2 served as the negative controls and were administered corn oil and distilled water re

Targeted inhibition of MLKL-mediated necroptosis attenuates chronic arsenite exposure-induced lung injury.

Arsenic, a ubiquitous environmental toxicant, poses a global public health concern through drinking water contamination. While chronic arsenic exposure is epidemiologically associated with respiratory diseases, the molecular mechanisms driving its pulmonary toxicity remain incompletely understood. Emerging evidence implicates necroptosis-a regulated cell death pathway mediated by the RIPK1-RIPK3-MLKL axis-is implicated in diverse diseases, though its contribution to arsenic-induced pulmonary injury is uncharacterized. This study aims to elucidate the contribution of necroptosis to arsenic-indu

The role of SLC7A11 in arsenite-induced oncogenic phenotypes of human bronchial epithelial cells: A metabolic perspective.

Chronic arsenic exposure enhances the probability of lung cancer with the underlying mechanisms remain unknown. Glutamine-driven synthetic metabolism, including nucleotide synthesis, amino acid production, TCA cycle replenishment, glutathione synthesis, and lipid biosynthesis, is crucial for both cancer initiation and progression. This study demonstrated that chronic exposure to 0.1 μM arsenite for as long as 36 weeks induced malignant transformation in human bronchial epithelial cells (BEAS-2B). Metabolomics were used to systematically disclose metabolic characteristics in arsenic-transformed

Dual role of arsenite in hydrolysis and post-hydrolysis fluorescence sensing of selective pH-dependent probes.

In comparison to the sensing activity, the reactivity of arsenite (AsO2-) is less explored. Herein, we focused on AsO2- reactivity studies based on its pKa and compared the study with other common anions. All three pKa values of arsenite are >9.0, affording a flexible working pH range to design a probe for reactivity studies. We designed and synthesized six pH dependent benzothiazole-based Schiff bases, namely, 1-(((6-nitrobenzo[d]thiazol-2-yl)imino)methyl)naphthalen-2-ol (1), 5-(diethylamino)-2-(((6-nitrobenzo[d]thiazol-2-yl)imino)methyl)phenol (2), 9-(((6-nitrobenzo[d]thiazol-2-yl)imino)meth

Arsenite removal by using ZnAlFe mixed metal oxides derived from layered double hydroxides.

ZnAlFe mixed metal oxides (ZnAlFe-MMOs) were synthesized from layered double hydroxides (LDHs) prepared by the coprecipitation method at pH 9 using an initial weight composition of Zn2+ = 75%, Al3+ = 15% and Fe3+ = 10%, with or without the addition of citric or oxalic acid. The solids were calcined at 400 °C to obtain the respective MMOs, which exhibited relatively high specific surface areas (165.3-63.8 m2 g-1) and semiconductor properties active in the visible region (bandgap values (Eg) of 2.42-1.77 eV). The synthesized materials were tested for the removal of trivalent arsenic by adsorptio
