Suspected Plasmodium falciparum resistance to artesunate in Cameroon: A case report.

Malaria in pregnancy is associated with adverse maternal and perinatal outcomes. The first-line treatment for severe malaria in the second and third trimesters of pregnancy is parenteral artesunate, according to WHO recommendations. Resistance of Plasmodium falciparum to artesunate has not yet been noted in our country. We report a case highly suspicious of such. A pregnant woman presented with the clinical signs of malaria. After paraclinical confirmation of the diagnosis, she was admitted and injectable artesunate was given for 72 h at the recommended dosage, with antipyretic without any imp

Efficacy of artesunate in the treatment of urinary schistosomiasis, in an endemic community in Nigeria.

The efficacy and tolerability of oral artesunate for the treatment of urinary schistosomiasis was assessed among schoolchildren aged 5-18 years in Adim community, Nigeria. Overall, 500 children, randomly selected from those attending the Presbyterian primary school, were each invited to provide two consecutive urine samples. Using standard parasitological procedures, Schistosoma haematobium ova were found in the samples from 145 (29.0%) of the subjects. Most (87) of the infected subjects were then treated orally with artesunate, using two doses, each of 6 mg/kg, given 2 weeks apart. When the t

Artesunate. A review of its pharmacology and therapeutic efficacy in the treatment of malaria.

Artesunate is an antimalarial agent, available in oral, rectal and parenteral formulations, that provides a rapid clinical effect in patients with Plasmodium falciparum malaria. The rapidity of effect, availability of an intravenous and intramuscular formulation and convenient dosage regimen make artesunate an ideal candidate for the treatment of severe malaria, including cerebral disease. While some results have been promising, there is no clear evidence to date that artesunate reduces mortality in patients with cerebral malaria to any greater extent than standard quinine therapy. When given

Artesunate nanoliposome-hydrogel: a dual-modal therapy for post-surgical melanoma.

Background: Melanoma management faces the dual challenge of preventing tumor recurrence while ensuring optimal post-surgical wound healing, particularly problematic given melanoma's high recurrence rates and therapeutic resistance. Artesunate (ARS) emerges as a promising multimodal agent with concurrent anticancer, anti-inflammatory, and tissue-regenerative properties. However, its anti-melanoma mechanisms remain incompletely characterized, and clinical translation has been limited by suboptimal pharmacokinetics. Methods: We employed transcriptomic profiling (RNA-seq) to identify novel ARS-reg

Artesunate attenuates ACLT-induced osteoarthritis by suppressing osteoclastogenesis and aberrant angiogenesis.

Artesunate (ART) is a semi-synthetic derivative of artemisinin and used preferentially in treatment of malaria in China. ART has strong anti-inflammatory, anti-tumor, and anti-angiogenic properties. Although the pharmacological effect of ART in osteoarthritis (OA) is unknown, evidence suggests that ART might regulate osteoclastogenesis through NF-κB signaling. In this study we explored the therapeutic effect of ART in an anterior cruciate ligament transection (ACLT)-induced OA model. We found that ART effectively relieved ACLT-induced osteoarthritis, as demonstrated by the improvement expressi

In vitro action of antiparasitic drugs, especially artesunate, against Toxoplasma gondii.

INTRODUCTION: Toxoplasmosis is usually a benign infection, except in the event of ocular, central nervous system (CNS), or congenital disease and particularly when the patient is immunocompromised. Treatment consists of drugs that frequently cause adverse effects; thus, newer, more effective drugs are needed. In this study, the possible activity of artesunate, a drug successfully being used for the treatment of malaria, on Toxoplasma gondii growth in cell culture is evaluated and compared with the action of drugs that are already being used against this parasite. METHODS: LLC-MK2 cells were c

Anthelmintic activity of artesunate against Fasciola hepatica in naturally infected sheep.

In light of rapidly spreading triclabendazole resistance alternative fasciocidal drugs are urgently needed. Following up on promising results obtained with artemether in Fasciola hepatica infected sheep, we here report the efficacy and safety of artesunate in sheep with a natural F. hepatica infection. Artesunate was administered intravenously and intramuscularly, adverse events were monitored and drug efficacy was elucidated by means of faecal egg and worm burden reductions. A single 40 mg/kg intravenous dose of artesunate induced an egg count reduction of 68.9% and a worm burden reduction of

Artesunate-resistant Plasmodium falciparum at a Tertiary Care Hospital in North India.

The emergence of artemisinin resistance poses a serious global challenge to malaria elimination programs. Artemisinin and its derivatives, especially artesunate, are highly effective against Plasmodium falciparum, particularly in severe and uncomplicated cases. However, increasing reports of treatment failure, particularly with artesunate monotherapy, raise concerns regarding emerging resistance. We present a clinically significant case of suspected artesunate-resistant P. falciparum in an Indian male residing in Nigeria. Despite standard therapy, parasite clearance was delayed, necessitating

Artesunate reduces oxidative stress and inflammation in NAFLD by activating the Nrf2 pathway.

Non-alcoholic fatty liver disease (NAFLD) has emerged as the most prevalent chronic liver disorder worldwide, characterized by progression from simple steatosis to inflammatory liver injury. Artesunate (AS), an antimalarial agent derived from Artemisia annua, possesses well-documented metabolic regulatory, antioxidant, and anti-inflammatory properties. This study comprehensively evaluated the therapeutic effects of AS on NAFLD pathogenesis and elucidated its underlying molecular mechanisms using integrated experimental approaches. AS administration significantly mitigated NAFLD progression, as

Effects of artesunate-loaded nanoparticles on Plasmodium berghei treatment in a mouse model.

Malaria is still one of the most important parasitic diseases with millions of cases reported globally every year. Combination therapies of artemisinin or its derivatives, with a partner drug, are the first-and second-line treatments for malaria. However, recently, artemisinin partial resistance or tolerance has emerged and emphasizes the need for new therapeutic approaches to malaria. For example, the encapsulation of artemisinin and its derivatives within nanoparticles offers a highly promising potential approach to improved therapeutic efficacy. In this study, we investigated the effects of
