Bioremediation of organophosphorus compounds using highly stable packed bed microreactor developed with OPH (organophosphorus hydrolase) coated nanofiber.

Cellulose nanofibers (CNF) provide a large surface area for enzyme immobilization and facilitate easy recovery from solution, enabling repeated use. The crosslinked coating of organophosphorus hydrolase (OPH) on these nanofibers significantly enhanced enzyme stability compared to the free form. Among the tested systems, diethylaminoethyl cellulose/magnetic particle-OPH (DEAE/MP-OPH) exhibited the highest activity retention, maintaining approximately 90% of its initial activity after 30 days. Similarly, carboxymethyl cellulose/margnetic nanopaticle-OPH (CMC/MP-OPH) and cellulose nanofiber-OPH (

Discovery and validation of serum glycoprotein biomarkers for high grade serous ovarian cancer.

PURPOSE: This study aimed to identify serum glycoprotein biomarkers for early detection of high-grade serous ovarian cancer (HGSOC), the most common and aggressive histotype of ovarian cancer. EXPERIMENTAL DESIGN: The glycoproteomics pipeline lectin magnetic bead array (LeMBA)-mass spectrometry (MS) was used in age-matched case-control serum samples. Clinical samples collected at diagnosis were divided into discovery (n = 30) and validation (n = 98) sets. We also analysed a set of preclinical sera (n = 30) collected prior to HGSOC diagnosis in the UK Collaborative Trial of Ovarian Cancer Scre

Expression of PON2 isoforms varies among brain regions in male and female African green monkeys.

Mitochondrial dysfunction and oxidative stress contribute to the neuropathology of neurodegenerative disorders such as Parkinson's disease (PD). Paraoxonase-2 (PON2) is a mitochondrial protein that mitigates oxidative stress, enhances mitochondrial function and exhibits anti-inflammatory properties. Previously, we have documented sex-based variation in PON2 with higher brain PON2 expression in female (2-fold) as compared to male African green monkeys. This aim of this study is to identify PON2 isoforms and explore the region-based variations in the protein level of PON2 in brain of African gre

The epigenetic regulation of paraoxonase 1 (PON1) as an important enzyme in HDL function: The missing link between environmental and genetic regulation.

BACKGROUND: Paraoxonase 1 (PON1) is an important antiatherogenic and antioxidant enzyme in the circulation that has been associated with adverse health outcomes particularly cardiovascular disease (CVD) and other metabolic disorders. PON1 is a highly promiscuous enzyme and can hydrolyse a large variety of substrates, however, detailed structure/function studies have concluded that the natural substrates for PON1 are lipophilic lactones. The interindividual variability in PON1 activity has been mainly attributed to genetic determinants; however, it appears that the contribution of epigenetics h

Genetics of vascular dementia - review from the ICVD working group.

BACKGROUND: Vascular dementia is a common disorder resulting in considerable morbidity and mortality. Determining the extent to which genes play a role in disease susceptibility and their pathophysiological mechanisms could improve our understanding of vascular dementia, leading to a potential translation of this knowledge to clinical practice. DISCUSSION: In this review, we discuss what is currently known about the genetics of vascular dementia. The identification of causal genes remains limited to monogenic forms of the disease, with findings for sporadic vascular dementia being less robust

Paraoxonase 1 in neurological disorders.

Paroxonase 1 displays multiple physiological activities that position it as a putative player in the pathogenesis of neurological disorders. Here we reviewed the literature focusing on the role of paraoxonase 1 (PON1) as a factor in the risk of stroke and the major neurodegenerative diseases. PON1 activity is reduced in stroke patients, which significantly correlates inversely with carotid and cerebral atherosclerosis. The presence of the R allele of the Q192R PON1 polymorphism seems to potentiate this risk for stroke. PON1 exerts peroxidase activities that may be important in neurodegenerativ

Paraoxonase 1 protects against protein N-homocysteinylation in humans.

Genetic or nutritional disorders in homocysteine (Hcy) or folate metabolism elevate plasma Hcy-thiolactone and lead to vascular and/or brain pathologies. Hcy-thiolactone has the ability to form isopeptide bonds with protein lysine residues, which generates N-Hcy-protein with autoimmunogenic and prothrombotic properties. Paraoxonase (PON1), carried on high-density lipoproteins (HDLs) in the blood, hydrolyzes Hcy-thiolactone and protects against the accumulation of N-Hcy-protein in vitro. To determine its role in vivo, we studied how natural variation in Hcy-thiolactonase activity of PON1 affect

Directed evolution of mammalian paraoxonases PON1 and PON3 for bacterial expression and catalytic specialization.

Serum paraoxonases (PONs) are a group of enzymes that play a key role in organophosphate (OP) detoxification and in prevention of atherosclerosis. However, their structure and mechanism of action are poorly understood. PONs seem like jacks-of-all-trades, acting on a very wide range of substrates, most of which are of no physiological relevance. Family shuffling and screening lead to the first PON variants that express in a soluble and active form in Escherichia coli. We describe variants with kinetic parameters similar to those reported for PONs purified from sera and others that show dramatic

Biomimetic engineering of active Zn-doped macrocyclic polyamine carbon dots as organophosphorus hydrolases-like nanozymes for colorimetric and fluorescence on-site detection of methyl-paraoxon.

Organic phosphate esters compounds, especially methyl-paraoxon (MP), pose a serious threat to the environment and human health. The development of robust organophosphorus hydrolases (OPHs)-like nanozymes to mimic natural enzymes is paramount for on-site monitoring MP. Herein, we reported a biomimetic engineering of highly active OPHs-like nanozymes through Zn-doped macrocyclic polyamine carbon dots (DOTA(Zn)-CDs). XPS confirmed ZnN coordination, while FTIR revealed macrocyclic polyamine frameworks with Zn-O/N coordination shifts, together mimicking the histidine‑zinc centers of native OPHs. Th

Laparoscopic Artificial Insemination Affects Stress and Inflammatory Markers in Ewes.

Laparoscopy is the preferred technique for artificial insemination (AI) with frozen semen in sheep, given the ovine cervix's anatomical challenges. This study assessed if and when laparoscopic AI alters stress and inflammation markers. Ewes underwent estrus synchronization and were divided into Control (hormonal protocol only; n=4) and LP (laparoscopic AI 54-60 h post-protocol; n=5) groups. Blood samples were collected at D-1 (pre-fasting), 0 h (during AI), 30 min, 60 min, 24 h, and 48 h post-procedure to measure serum cortisol, total protein (TP), albumin, and paraoxonase 1 (PON1). Anti-biofilm efficiency and substrate specificity of recombinantly produced human paraoxonases.

The human paraoxonase (PON) gene family, comprising PON1, PON2, and PON3, has evolved through parallel evolutionary trajectories leading to distinct substrate selectivity, thereby enabling these enzymes to hydrolyze a range of substrates such as lactones, arylesters, and organophosphates. This study explores the evolutionary divergence of PON family members and their varied anti-biofilm activities through in silico molecular docking and in vitro analysis. The huPON1, huPON2, and huPON3 genes were cloned and expressed in the E. coli expression system. These overexpressed proteins formed inclusi

The Association between Single Nucleotide Polymorphisms, including miR-499a Genetic Variants, and Dyslipidemia in Subjects Treated with Pharmacological or Phytochemical Lipid-Lowering Agents.

Disorders of lipoprotein metabolism are among the major risk factors for cardiovascular disease (CVD) development. Single nucleotide polymorphisms (SNPs) have been associated with the individual variability in blood lipid profile and response to lipid-lowering treatments. Here, we genotyped 34 selected SNPs located in coding genes related to lipid metabolism, inflammation, coagulation, and a polymorphism in the MIR499 gene-a microRNA previously linked to CVD-to evaluate the association with lipid trait in subjects with moderate dyslipidemia not on lipid-lowering treatment (Treatment-naïve (TN)

Paraoxonase-1 as a Regulator of Glucose and Lipid Homeostasis: Impact on the Onset and Progression of Metabolic Disorders.

Metabolic disorders are characterized by an overall state of inflammation and oxidative stress, which highlight the importance of a functional antioxidant system and normal activity of some endogenous enzymes, namely paraoxonase-1 (PON1). PON1 is an antioxidant and anti-inflammatory glycoprotein from the paraoxonases family. It is mainly expressed in the liver and secreted to the bloodstream, where it binds to HDL. Although it was first discovered due to its ability to hydrolyze paraoxon, it is now known to have an antiatherogenic role. Recent studies have shown that PON1 plays a protective ro

Serum paraoxonase-1 activity is inversely related to free thyroxine in euthyroid subjects: The PREVEND Cohort Study.

BACKGROUND: Low-normal thyroid function within the euthyroid range has been suggested to enhance atherosclerosis susceptibility. Paraoxonase-1 (PON-1) may protect against atherosclerotic cardiovascular disease development by attenuating oxidative stress. We evaluated relationships of PON-1 with thyroid stimulating hormone (TSH), free T4 , free T3 , lipids and apolipoprotein (apo)A-I in euthyroid subjects, and assessed whether such relationships are modified in the context of the metabolic syndrome (MetS). MATERIALS AND METHODS: Serum PON-1 activity (arylesterase activity), TSH, free T4 , free

Signature of paraoxonases in the altered redox homeostasis in Alzheimer's disease.

Paraoxonase (PON) enzymes (PON1, PON2 and PON3) exert antioxidant properties through arylesterase, lactonase and paraoxonase activities. Increasing findings suggested their potential involvement, particularly PON1 and PON2, in Alzheimer's disease (AD), a neurodegenerative pathology characterized by early oxidative stress. Specifically, decreased serum PON1-arylesterase and lactonase activities seem to be associated with an increased brain oxidative damage in early AD, leading to hypothesize that PON activity alterations might be an early event in AD. To address this hypothesis, the levels of 4

PGC-1α Inhibits Schwann Cell Dedifferentiation and Delays Peripheral Nerve Degeneration by Targeting PON1.

PPARγ coactivator-1 alpha (PGC-1α) is an essential transcription factor co-activator that regulates gene transcription and neural regeneration. Schwann cells, which are unique glial cells in peripheral nerves that dedifferentiate after peripheral nerve injury (PNI) and are released from degenerative nerves. Wallerian degeneration is a series of stereotypical events that occurs in response to nerve fibers after PNI. The role of PGC-1α in Schwann cell dedifferentiation and Wallerian degeneration is not yet clear. As Wallerian degeneration plays a crucial role in PNI, we conducted a study to dete
