Prolonged biocontrol priming sensitizes Aspergillus flavus to cinnamaldehyde via metabolic trade-offs.

The control of Aspergillus flavus and its highly carcinogenic secondary metabolite, aflatoxin B1 (AFB1), remains a major challenge in global agriculture and food safety. In this study, we propose a novel sequential hurdle strategy that combines priming with the biocontrol bacterium Pantoea agglomerans FL1 followed by low-dose cinnamaldehyde (CA) stress, and we further elucidate its underlying mechanisms. Phenotypic analyses revealed that prolonged FL1 priming for 7 days increased the susceptibility of A. flavus upon subsequent CA exposure. Although macroscopic radial growth remained unaffected

Copper-Chitosan Nanocomposite Hydrogels Against Aflatoxigenic Aspergillus flavus from Dairy Cattle Feed.

The integration of copper nanoparticles as antifungal agents in polymeric matrices to produce copper polymer nanocomposites has shown excellent results in preventing the growth of a wide variety of toxigenic fungi. Copper-chitosan nanocomposite-based chitosan hydrogels (Cu-Chit/NCs hydrogel) were prepared using a metal vapor synthesis (MVS) and the resulting samples were described by transmission electron microscopy (TEM), X-ray fluorescence analysis (XRF), and small-angle X-ray scattering (SAXS). Aflatoxin-producing medium and VICAM aflatoxins tests were applied to evaluate their ability to p

Genotyping of clinical and environmental Aspergillus flavus isolates from Iran using microsatellites.

Aspergillus flavus is the second most important Aspergillus species causing human infections in tropical countries. Despite an increasing number of infections of A. flavus in Iran, the molecular epidemiology of clinical and environmental strains has not been well studied. We used a panel of nine microsatellite markers to analyse the genetic relatedness of A. flavus. Microsatellite typing of 143 (n = 119 clinical and n = 24 environmental) isolates demonstrated 118 different genotypes. A possible outbreak at a pulmonary ward was discovered. The discriminatory power for the individual markers ran

Enzymatic activity of clinical and environmental isolates of Aspergillus flavus section Flavi.

BACKGROUND AND OBJECTIVES: The ability of Aspergillus species to produce various extracellular enzymes has a pathogenic role in humans. These enzymes not only play a role in metabolism but also act as virulence factors. The secretion of hydrolytic enzymes facilitates tissue invasion and is considered a virulence factor for several fungi. The current study aimed to compare the enzymatic activity of A. flavus section Flavi isolated from otomycosis and environmental samples. MATERIALS AND METHODS: In the present study, eighty-one isolates of A. flavus including 41 clinical strains from otomycosi

[Aspergillus flavus/oryzae Arthritis of the Knee Joint: First Case in Türkiye].

Aspergillus species are common hyphal fungi. In addition to allergies and mycotoxicosis, Aspergillus species can cause various infections known as aspergillosis. Aspergillosis of the respiratory tract, central nervous system, skin and soft tissues is well described. However, musculoskeletal infections due to invasive aspergillosis are not well described. Fungal joint infection due to invasive aspergillosis is a rare form of septic arthritis. In this case report, a patient who admitted to our hospital for liver transplantation and developed knee joint arthritis caused by Aspergillus flavus/Aspe

Aneuploidy Formation in the Filamentous Fungus Aspergillus flavus in Response to Azole Stress.

Aspergillus flavus is a mycotoxigenic fungus that contaminates many important agricultural crops with aflatoxin B1, the most toxic and carcinogenic natural compound. This fungus is also the second leading cause of human invasive aspergillosis, after Aspergillus fumigatus, a disease that is particularly prevalent in immunocompromised individuals. Azole drugs are considered the most effective compounds in controlling Aspergillus infections both in clinical and agricultural settings. Emergence of azole resistance in Aspergillus spp. is typically associated with point mutations in cyp51 orthologs

Phosphomannose Isomerase Is Involved in Development, Stress Responses, and Pathogenicity of Aspergillus flavus.

Aspergillus flavus causes invasive aspergillosis in immunocompromised patients and severe contamination of agriculturally important crops by producing aflatoxins. The fungal cell wall is absent in animals and is structurally different from that of plants, which makes it a potential antifungal drug target due to its essentiality for fungal survival. Mannose is one of the important components in the fungal cell wall, which requires GDP-mannose (GDP-Man) as the primary donor. Three consecutive enzymes, namely, phosphomannose isomerase (PMI), phosphomannose mutase (PMM), and GDP-mannose phosphoryl

Immunohistochemical Detection of Aflatoxin in Lesions of Aflatoxin-producing Aspergillus flavus Infection.

UNLABELLED: Aflatoxin produced by Aspergillus flavus is known to be strongly related to liver injury (hepatocellular carcinoma) and immune system damage involving leukocytes. This toxin suppresses both the cell-mediated immune system and macrophage function, and decreases the production of complement and interferon molecules. PURPOSE: To evaluate the presence of aflatoxin in infectious lesions as well as how the toxin is taken up by leukocytes. METHOD: Pathological specimens from a patient who died from aspergillosis caused by aflatoxin-producing A. flavus were used. Anti-aflatoxin B1 antibo

Antifungal effects of ZnO, TiO2 and ZnO-TiO2 nanostructures on Aspergillus flavus.

This study aimed to synthesis ZnO, TiO2, and ZnO-TiO2 (ratio weight of 1/1 for Zn/Ti) nanoparticles using zinc acetate and titanium isopropoxide through the sol-gel method. Physicochemical and morphological characterization and antifungal properties evaluation like minimum inhibition concentration (MIC) and minimum fungicide concentration (MFC) of nanopowders were investigated against A.flavus in vitro. All synthesized nanoparticles (50 μg/ml) showed fungal growth inhibition, while ZnO-TiO2 showed higher antifungal activity against A. flavus than pure TiO2 and ZnO. TiO2 and ZnO-TiO2 (300 μg/ml

Aspergillus flavus squalene synthase as an antifungal target: Expression, activity, and inhibition.

Invasive aspergillosis (IA) is a life-threatening disease impacting immunocompromised individuals. Standard treatments of IA, including polyenes and azoles, suffer from high toxicity and emerging resistance, leading to the need to develop new antifungal agents with novel mechanisms of action. Ergosterol biosynthesis is a classic target for antifungals, and squalene synthase (SQS) catalyzes the first committed step in ergosterol biosynthesis in Aspergillus spp. making SQS of interest in the context of antifungal development. Here, we cloned, expressed, purified and characterized SQS from the pa

Microsatellite Typing of Aspergillus flavus Strains in a Tunisian Onco-hematology Unit.

Aspergillus flavus is the most common species associated with invasive aspergillosis in Tunisia. The molecular epidemiology of the species is poorly documented. We used five highly discriminative microsatellite markers for the genotyping of clinical and hospital environmental A. flavus strains to assess whether IA could be hospital-acquired in the onco-hematology unit of the Farhat Hached teaching hospital of Sousse, Tunisia. The genotyping of 18 clinical isolates, collected from sputa of 17 acute leukemia patients, and 81 isolates, collected in these patients' hospital environment and food, i

A Novel Y319H Substitution in CYP51C Associated with Azole Resistance in Aspergillus flavus.

This study aimed to explore any mutation in the CYP51 gene conferring azole resistance in Aspergillus flavus. Two voriconazole-resistant and 45 voriconazole-susceptible isolates were included in the study. Sequence analysis demonstrated a T1025C nucleotide change in CYP51C, resulting in the Y319H amino acid substitution in one resistant isolate. However, the earlier described T788G mutation in CYP51C conferring voriconazole resistance in A. flavus isolates was present in all isolates, irrespective of their susceptibility status.

Isolation and characterization of a novel elastase inhibitor, AFLEI from Aspergillus flavus.

A novel elastase inhibitor from Aspergillus flavus (AFLEI) was isolated, and biochemical properties of AFLEI were examined. Column chromatography using diethylaminoethyl (DE) 52-Cellulose and Sephadex G-75 was used to purify the inhibitor. The final preparation was found to be homogeneous as indicated by a single band after disc polyacrylamide gel (PAGE) and isoelectric focusing electrophoreses. AFLEI had a molecular weight of 7,525.8 as determined by TOF-MS (time of flight mass spectrometry). The elastolytic activity of elastases from A. flavus, A. fumigatus and human leukocytes were inhibite

Cyclopiazonic acid production by Aspergillus flavus and its effects on broiler chickens.

Cyclopiazonic acid (CPA) was purified from cultures of Aspergillus flavus, and ca. 14 g of the toxin was collected for use in feeding studies. Chicken rations were artificially contaminated with purified CPA at concentrations of 10, 50, and 100 ppm (microgram/g) and fed ad libitum to eight groups of chickens for 7 weeks. Chickens receiving feed with 100 ppm of CPA had high mortality, decreased weight gain, and poor feed conversion when compared with birds receiving other doses. Postmortem examination showed that chickens fed the two greatest doses of CPA had proventricular lesions characterize

Aspergillus flavus Infections in Children With Leukemia Despite Liposomal Amphotericin-B Prophylaxis.

Liposomal amphotericin-B (L-AmB) prophylaxis is used in children with leukemia when azoles are contraindicated, but its effect is debated. We reviewed cases of invasive aspergillosis despite L-AmB 2.5 mg/kg twice weekly in children with high-risk leukemia during 2012-2019. Ten (16%) of 62 children had proven or probable aspergillosis. Thus, L-AmB prophylaxis offered insufficient protection for Aspergillus, in particular for Aspergillus flavus.

Harmful Effects and Control Strategies of Aflatoxin B₁ Produced by Aspergillus flavus and Aspergillus parasiticus Strains on Poultry: Review.

The presence of aflatoxin B₁ (AFB₁) in poultry diets decreases the hatchability, hatchling weight, growth rate, meat and egg production, meat and egg quality, vaccination efficiency, as well as impairing the feed conversion ratio and increasing the susceptibility of birds to disease and mortality. AFB₁ is transferred from poultry feed to eggs, meat, and other edible parts, representing a threat to the health of consumers because AFB₁ is carcinogenic and implicated in human liver cancer. This review considers how AFB₁ produced by Aspergillus flavus and Aspergillus parasiticus strains can affect

Population-specific transcriptomic shifts underlie secondary metabolic diversification in Aspergillus flavus Link and the domestication of Aspergillus oryzae (Ahlb.).

The genomic signatures of microbial domestication remain poorly understood within the context of natural population variation. Here, we demonstrate that Aspergillus oryzae, the filamentous fungus used in soy sauce and sake production, shares more recent ancestry with a predominantly northern, largely non-aflatoxigenic population of Aspergillus flavus (population C). A. oryzae isolates also overlap with the recently described, clinically enriched A. flavus population D, suggesting multiple domestication events. All isolates tested retained pathogenicity in a zebrafish infection model. At the tr
