Advanced nitrogen removal from secondary effluent using the SAD/A process under the toxicity of PFOA.

In response to increasingly stringent wastewater discharge standards, the need for advanced nitrogen removal from secondary effluent has become urgent. The sulfur autotrophic denitrification (SAD) coupled with anaerobic ammonium oxidation (Anammox) process (SAD/A), holds significant promise for this application due to its advantage of requiring no external organic carbon. However, its resilience to emerging contaminants, such as per- and polyfluoroalkyl substances (PFAS), which may persist in secondary effluent, remains unknown. This study investigated the long-term impact of perfluorooctanoic

Iron sulphides mediated autotrophic denitrification: An emerging bioprocess for nitrate pollution mitigation and sustainable wastewater treatment.

Iron sulphides, mainly in the form of mackinawite (FeS), pyrrhotite (Fe1-xS, x = 0-0.125) and pyrite (FeS2), are the most abundant sulphide minerals and can be oxidized under anoxic and circumneutral pH conditions by chemoautotrophic denitrifying bacteria to reduce nitrate to N2. Iron sulphides mediated autotrophic denitrification (ISAD) represents an important natural attenuation process of nitrate pollution and plays a pivotal role in linking nitrogen, sulphur and iron cycles in a variety of anoxic environments. Recently, it has emerged as a promising bioprocess for nutrient removal from var

Analysis of the intermediary metabolism of a reductive chemoautotroph.

All extant life forms depend, directly or indirectly, on the autotrophic fixation of the dominant elements of the biosphere: carbon, hydrogen, nitrogen, oxygen, phosphorus, and sulfur. We have earlier presented the canonical network of reactions that constitute the anabolism of a reductive chemoautotroph. Separating this network into subgraphs reveals several empirical generalizations: (1) acetate (acetyl-CoA), pyruvate, phosphoenol pyruvate, oxaloacetate, and 2-oxoglutarate serve as universal starting points for all pathways leading to the universal building blocks-20 amino acids and 4 ribonu

Autotrophic isopropanol production by Cupriavidus necator using industrial biogas and incinerator flue gases as carbon sources.

Gas fermentation is a promising approach to reduce carbon emissions of production processes by using microorganisms to convert industrial gas streams into valuable carbon-based products. Here, isopropanol production by engineered Cupriavidus necator from CO2-rich gas streams is presented. Four different engineered strains were tested first on synthetic gas mixture, and one was selected, namely Re2133/pEG7d, for further evaluation on two different industrial CO2-rich gas streams. Up to 1.6 g L-1 of isopropanol was obtained in autotrophic gas flasks with this strain. Then, raw biogas from landfi

Autotrophy to Heterotrophy: Shift in Bacterial Functions During the Melt Season in Antarctic Cryoconite Holes.

Microbes residing in cryoconite holes (debris, water, and nutrient-rich ecosystems) on the glacier surface actively participate in carbon and nutrient cycling. Not much is known about how these communities and their functions change during the summer melt-season when intense ablation and runoff alter the influx and outflux of nutrients and microbes. Here, we use high-throughput-amplicon sequencing, predictive metabolic tools and Phenotype MicroArray techniques to track changes in bacterial communities and functions in cryoconite holes in a coastal Antarctic site and the surrounding fjord, duri

Effective carbon and nitrogen removal with reduced sulfur oxidation in an anaerobic baffled reactor for fresh leachate treatment.

The application of an anaerobic baffled reactor (ABR) with four compartments was investigated for the simultaneous removal of carbon and nitrogen from leachate. The nitrified effluent was recycled to compartment 3 of the ABR, thereby avoiding the adverse influence of nitrogen oxides on anaerobic methanogenesis in compartment 1. Nitrified effluent recirculation not only enhanced chemical oxygen demand removal (>95.6%) but also improved the total nitrogen removal efficiency from 12.7% to 67.4% with increasing recirculation ratio from 0.25 to 2. The challenge of insufficient carbon sources for he

The impact of dissolved inorganic nitrogen and phosphorous on responses of microbial plankton to the Texas City "Y" oil spill in Galveston Bay, Texas (USA).

Ongoing bioremediation research seeks to promote naturally occurring microbial polycyclic aromatic hydrocarbon (PAH) degradation during and after oil spill events. However, complex relationships among functionally different microbial groups, nutrients and PAHs remain unconstrained. We conducted a surface water survey and corresponding nutrient amendment bioassays following the Texas City "Y" oil spill in Galveston Bay, Texas. Resident microbial groups, defined as either heterotrophic or autotrophic were enumerated by flow cytometry. Heterotrophic abundance was increased by oil regardless of nu

Phosphoglycolate salvage in a chemolithoautotroph using the Calvin cycle.

Carbon fixation via the Calvin cycle is constrained by the side activity of Rubisco with dioxygen, generating 2-phosphoglycolate. The metabolic recycling of phosphoglycolate was extensively studied in photoautotrophic organisms, including plants, algae, and cyanobacteria, where it is referred to as photorespiration. While receiving little attention so far, aerobic chemolithoautotrophic bacteria that operate the Calvin cycle independent of light must also recycle phosphoglycolate. As the term photorespiration is inappropriate for describing phosphoglycolate recycling in these nonphotosynthetic

Differential diversity and structure of autotrophs in agricultural soils of Qinghai Province.

UNLABELLED: The biodiversity of CO2-assimilating bacterial communities is pivotal for carbon sequestration in agricultural systems. Changes in the diversity, structure, and activity of the soil chemolithoautotrophic bacteria were examined in four agricultural areas, Dulan (DL), Gonghe (GH), Huzhu (HZ), and Datong (DT) counties in Qinghai Province, where wheat, oilseed rape, and barley were planted. This process was performed using Illumina amplicon sequencing of the ribulose-1,5-bisphosphatecarboxylase/oxygenase (RubisCO) gene (cbbL Form I) and activity data. The diversity, community, and acti

Exploring the Diversity and Metabolic Potential of CO2 fixation Mediated by RubisCO in Prokaryotes in the Japan Collection of Microorganisms.

A genome anal-ysis is essential for identifying valuable microbial resources for future applications. In the present study, we exami-ned potential CO2-fixing microorganisms based on the presence of the Calvin-Benson-Bassham (CBB) cycle using 6,262 bacterial and 487 archaeal genomes from available cultures in the Japan Collection of Microorganisms (JCM), a well-established culture collection, in October 2023. A total of 306 strains (147 genera, eight phyla) carried CBB cycle genes, and a literature survey showed that 74 genera had experimental evidence of autotrophic growth while 73 lacked supp

Metal removal capability of two cyanobacterial species in autotrophic and mixotrophic mode of nutrition.

BACKGROUND: Cyanobacteria are ecologically significant prokaryotes that can be found in heavy metals contaminated environments. As their photosynthetic machinery imposes high demands for metals, homeostasis of these micronutrients has been extensively considered in cyanobacteria. Recently, most studies have been focused on different habitats using microalgae leads to a remarkable reduction of an array of organic and inorganic nutrients, but what takes place in the extracellular environment when cells are exposed to external supplementation with heavy metals remains largely unknown. METHODS: H

Effects of HRT and water temperature on nitrogen removal in autotrophic gravel filter.

Organic Carbon added to low ratio of carbon to nitrogen (C/N ratio) wastewater to enhance heterotrophic denitrification performance might lead to higher operating costs and secondary pollution. In this study, sodium thiosulfate (Na2S2O3) was applied as an electron donor for a gravel filter (one kind of constructed wetland) to investigate effects of hydraulic retention time (HRT) and water temperature on the nitrate removal efficiency. The results show that with an HRT of 12 h, the average total nitrogen (TN) removal efficiencies were 91% at 15-20 °C and 18% at 3-6 °C, respectively. When HRT in

Genomic and Transcriptomic Insights Into the Autotrophic Metabolism on H2 + CO2 or CO of the Thermophilic Acetogenic Model Organism Moorella thermoacetica.

Although the Wood-Ljungdahl pathway, a wide-spread pathway for CO2 fixation in anaerobic microorganisms, was elucidated in the thermophilic acetogenic bacterium Moorella thermoacetica, still relatively little is known about the enzymes involved in hydrogen oxidation, CO2 fixation, energy conservation and the role of quinones and cytochromes. Here, we have used transcriptomics, enzyme assays and genome analyses to identify missing links. NADPH, generated by a [FeFe] hydrogenase, is the reductant for CO2 reduction to formate, a key reaction in CO2 fixation. This reaction is slightly endergonic u

Differential fates of antibiotics in sulfur-driven autotrophic denitrification: Ciprofloxacin transformation linked to sulfide oxidation.

The coexistence of residual antibiotics in wastewater raises concerns about the functional stability of sulfur-driven autotrophic denitrification (SDAD) mediated by sulfur-oxidizing bacteria (SOB) and the fate of antibiotics during treatment. This study investigated the functional tolerance of SOB to three representative antibiotics-ciprofloxacin (CIP), oxytetracycline (OTC), and sulfamonomethoxine (SMM)-under 360-min batch exposure and elucidated their removal behaviors during SOB-mediated sulfide (S2-) oxidation and NO3-/NO2- reduction. Batch experiments revealed distinct fates among the thr

Leptothrix ochracea genomes reveal potential for mixotrophic growth on Fe(II) and organic carbon.

UNLABELLED: Leptothrix ochracea creates distinctive iron-mineralized mats that carpet streams and wetlands. Easily recognized by its iron-mineralized sheaths, L. ochracea was one of the first microorganisms described in the 1800s. Yet it has never been isolated and does not have a complete genome sequence available, so key questions about its physiology remain unresolved. It is debated whether iron oxidation can be used for energy or growth and if L. ochracea is an autotroph, heterotroph, or mixotroph. To address these issues, we sampled L. ochracea-rich mats from three of its typical environm

Metabolic Performance and Fate of Electrons during Nitrate-Reducing Fe(II) Oxidation by the Autotrophic Enrichment Culture KS Grown at Different Initial Fe/N Ratios.

Autotrophic nitrate-reducing Fe(II)-oxidizing (NRFeOx) microorganisms fix CO2 and oxidize Fe(II) coupled to denitrification, influencing carbon, iron, and nitrogen cycles in pH-neutral, anoxic environments. However, the distribution of electrons from Fe(II) oxidation to either biomass production (CO2 fixation) or energy generation (nitrate reduction) in autotrophic NRFeOx microorganisms has not been quantified. We therefore cultivated the autotrophic NRFeOx culture KS at different initial Fe/N ratios, followed geochemical parameters, identified minerals, analyzed N isotopes, and applied numeri

Autotrophic nitrogen removal after ureolytic phosphate precipitation to remove both endogenous and exogenous nitrogen.

Anaerobic digestion yields effluents rich in ammonium and phosphate and poor in biodegradable organic carbon, thereby making them less suitable for conventional biological nitrogen and phosphorus removal. In addition, the demand for fertilizers is increasing, energy prices are rising and global phosphate reserves are declining. This requires both changes in wastewater treatment technologies and implementation of new processes. In this contribution a description is given of the combination of a ureolytic phosphate precipitation (UPP) and an autotrophic nitrogen removal (ANR) process on the anae

Close spatial and metabolic association between heterotrophic and ammonia-oxidizing marine Nitrososphaerota.

Following the ubiquitous autotrophic ammonia-oxidizing archaea (AOA), heterotrophic representatives of the marine Nitrososphaerota (HMN) form the second most abundant group within this archaeal phylum. However, their eco-evolutionary strategies remain poorly understood. Previous studies have reported a consistent co-occurrence of HMN with marine AOA (MAOA), prompting a detailed investigation into their potential interaction. Through large-scale (meta)genomic and metatranscriptomic analyses, we reveal that HMN possess ultra-streamlined genomes and globally co-occur with marine AOA. The absence
