Synergistic anticandidal activity of etomidate and azoles against clinical fluconazole-resistant Candida isolates.

Aim: The purpose of this study was to evaluate the effect of etomidate alone and in combination with azoles on resistant strains of Candida spp. in both planktonic cells and biofilms. Materials & methods: The antifungal activity of etomidate was assessed by the broth microdilution test; flow cytometric procedures to measure fungal viability, mitochondrial transmembrane potential, free radical generation and cell death; as well detection of DNA damage using the comet assay. The interaction between etomidate and antifungal drugs (itraconazole and fluconazole) was evaluated by the checkerboard as

Synergistic effects of vorinostat (SAHA) and azoles against Aspergillus species and their biofilms.

BACKGROUND: Invasive aspergillosis is a fungal infection that occurs mainly in immunocompromised patients. It is responsible for a high degree of mortality and is invariably unresponsive to conventional antifungal treatments. Histone deacetylase inhibitors can affect the cell cycle, apoptosis and differentiation. The histone deacetylase inhibitor vorinostat (SAHA) has recently received approval for the treatment of cutaneous T cell lymphoma. Here, we investigated the interactions of SAHA and itraconazole, voriconazole, and posaconazole against Aspergillus spp. in vitro using both planktonic ce

Ctt1 catalase activity potentiates antifungal azoles in the emerging opportunistic pathogen Saccharomyces cerevisiae.

Fungi respond to antifungal drugs by increasing their antioxidant stress response. How this impacts antifungal efficacy remains controversial and not well understood. Here we examine the role of catalase activity in the resistance of Saccharomyces cerevisiae to the common antifungals, fluconazole and miconazole, for which we report minimum inhibitory concentrations (MICs) of 104 and 19 μM, respectively. At sub-MIC concentrations, fluconazole and miconazole stimulate catalase activity 2-3-fold but, unexpectedly, deletion of cytosolic catalase (ctt1) makes cells more resistant to these azoles an

Does fungicide application in vineyards induce resistance to medical azoles in Aspergillus species?

This study assessed if the use of sterol demethylase inhibitor fungicides in vineyard production can induce resistance to azoles in Aspergillus strains and if it can induce selection of resistant species. We also tried to identify the Aspergillus species most prevalent in the vineyards. Two vineyards from northern Portugal were selected from "Vinhos Verdes" and "Douro" regions. The vineyards were divided into plots that were treated or not with penconazole (PEN). In each vineyard, air, soil, and plant samples were collected at three different times. The strains of Aspergillus spp. were isolate

In vitro activity of the antifungal azoles itraconazole and posaconazole against Leishmania amazonensis.

Leishmaniasis, caused by protozoan parasites of the Leishmania genus, is one of the most prevalent neglected tropical diseases. It is endemic in 98 countries, causing considerable morbidity and mortality. Pentavalent antimonials are the first line of treatment for leishmaniasis except in India. In resistant cases, miltefosine, amphotericin B and pentamidine are used. These treatments are unsatisfactory due to toxicity, limited efficacy, high cost and difficult administration. Thus, there is an urgent need to develop drugs that are efficacious, safe, and more accessible to patients. Trypanosoma

Interaction of azoles with rifampin, phenytoin, and carbamazepine: in vitro and clinical observations.

Twelve patients receiving therapy with an azole agent (ketoconazole, itraconazole, and/or fluconazole) for systemic mycoses experienced drug interactions with rifampin, phenytoin, and/or carbamazepine resulting in substantial decreases in azole concentrations in serum. All four patients receiving azoles and concurrent phenytoin and/or carbamazepine failed to respond to treatment or suffered a relapse of their fungal infection. Four of five patients with cryptococcosis who received itraconazole and rifampin responded despite decreases in their serum itraconazole concentrations; synergy between

Salvianolic acid B synergizes with azoles against Candida glabrata cells in vulvovaginal candidiasis.

INTRODUCTION: The rising prevalence of antifungal resistance poses a significant threat to global health, with Candida glabrata (C. glabrata) emerging as a particular concern due to its increasing incidence in clinical settings and the intrinsic low susceptibility to azoles. Salvianolic acid B (SAB), a major bioactive component extracted from Salvia miltiorrhiza, exhibits potent antioxidant properties; however, its potential as an antifungal remains unexplored. OBJECTIVES: This study aimed to evaluate the antifungal efficacy of SAB alone and in combination with azoles against C. glabrata, bot

Directed Evolution Detects Supernumerary Centric Chromosomes Conferring Resistance to Azoles in Candida auris.

Candida auris exhibits resistance to multiple antifungal drug classes and sterilization agents, posing threats to the immunocompromised worldwide. Among the four major geographical clades, the East Asian clade 2 isolates of C. auris are mostly drug susceptible. In this study, we experimentally evolved one such drug-susceptible isolate for multiple generations in the presence of the antifungal compound fluconazole and analyzed changes in the karyotype, DNA sequence, and gene expression profiles in three evolved drug-resistant isolates. Next-generation sequencing and electrophoretic karyotyping

Efficacy of Lamiaceae essential oils with selected azoles against Candida albicans clinical isolates.

A current problem in candidiasis treatment is increasing resistance to azoles, which are often prescribed to patients. The study underlines the high resistance of yeasts to fluconazole, which achieved high MIC (minimal inhibitory concentration) values both alone and in combination with essential oils (EOs). Antifungal activity of Hyssopus officinalis, Thymus vulgaris, Salvia officinalis and Rosmarinus officinalis EOs was determined against 13 clinical isolates of Candida albicans and reference strain Candida albicans ATCC 10231. The synergistic effect was investigated for the combination of it

In vitro interactions of azoles and echinocandins against clinical strains of Aspergillus flavus.

Combinations of an azole (itraconazole, voriconazole, or posaconazole) with an echinocandin (caspofungin, micafungin, or anidulafungin) were tested against 20 clinical isolates of Aspergillus flavus according to EUCAST guidelines. The interactions were determined using two endpoints-minimal effective concentration (MEC) and minimal inhibitory concentration (MIC)-via calculation of the fractional inhibitory concentration (FIC) index. A higher prevalence of synergistic interactions was observed for MIC, whereas indifference was the most frequent outcome according to MEC among the 20 strains. Com

Treatment of experimental zygomycosis in guinea pigs with azoles and with amphotericin B.

Nonpredisposed Albino guinea pigs were infected intravenously with Rhizopus microsporus var. rhizopodiformis or with Rhizopus oryzae. Both strains were highly pathogenic. They killed all control animals between days 4 and 7 and between days 5 and 9 after infection, respectively. All animals presented invasion of almost all internal organs and skin eruptions developing into ulcers. Oral treatment with ketoconazole, itraconazole, fluconazole or saperconazole was inefficacious. Parenteral treatment with amphotericin B prolonged survival and was life-saving in 9 out of 12 guinea pigs infected with

Global epidemiology of azole resistance in Aspergillus fumigatus.

The fungus Aspergillus fumigatus has evolved as an important cause of opportunistic fungal diseases in humans worldwide. It is the most frequent filamentous fungus colonizing the airways of patients with cystic fibrosis and can affect immunocompetent as well as immunocompromised individuals. Aspergillus disease is commonly treated with azoles, which inhibit lanosterol 14α-demethylase, a key component of the ergosterol biosynthesis pathway, essential for membrane integrity and fluidity. Lanosterol 14α-demethylase is encoded by the CYP51A gene. Azole-resistant A. fumigatus isolates often show am
