GPR4 Knockout Attenuates Intestinal Inflammation and Forestalls the Development of Colitis-Associated Colorectal Cancer in Murine Models.

GPR4 is a proton-sensing G protein-coupled receptor highly expressed in vascular endothelial cells and has been shown to potentiate intestinal inflammation in murine colitis models. Herein, we evaluated the proinflammatory role of GPR4 in the development of colitis-associated colorectal cancer (CAC) using the dextran sulfate sodium (DSS) and azoxymethane (AOM) mouse models in wild-type and GPR4 knockout mice. We found that GPR4 contributed to chronic intestinal inflammation and heightened DSS/AOM-induced intestinal tumor burden. Tumor blood vessel density was markedly reduced in mice deficient

Distal transposition of rat caecum does not render it susceptible to carcinogenesis.

As the relative resistance of rat caecum to chemical carcinogens could reflect its luminal environment, caecal mucosa was exposed to the distal faecal stream in male Sprague-Dawley rats (n = 50) previously treated with azoxymethane (total dose 90 mg/kg sc). After colonic transection at the pelvic brim, the caecum was inserted isoperistaltically between colocaecal and caecorectal anastomoses (n = 30); an ileocolic anastomosis restored intestinal continuity. Controls (n = 20) had transection and reanastomosis at equivalent points of the bowel, plus caecotomy and resuture. Caecal crypt cell produ

TRIM27 mediates STAT3 activation at retromer-positive structures to promote colitis and colitis-associated carcinogenesis.

STAT3 is a transcription factor that plays central roles in various physiological processes and its deregulation results in serious diseases including cancer. The mechanisms on how STAT3 activity is regulated remains enigmatic. Here we identify TRIM27 as a positive regulator of II-6-induced STAT3 activation and downstream gene expression. TRIM27 localizes to retromer-positive punctate structures and serves as a critical link for recruiting gp130, JAK1, and STAT3 to and subsequent phosphorylation of STAT3 at the retromer-positive structures. Overexpression of TRIM27 promotes cancer cell growth

Flaxseed supplementation and early markers of colon carcinogenesis.

Since flaxseed ingestion produces potentially anticarcinogenic lignans in the colon, this study determined whether flaxseed decreases the risk for colon carcinogenesis. Following a single injection of azoxymethane (15 mg/kg body wt.), five groups of male Sprague-Dawley rats were fed a high-fat (20% corn oil) basal diet with or without supplementation with 5% or 10% flaxseed meal (FM) or flaxseed flour (FF) for four weeks. Upon sacrifice, colons were examined for aberrant morphology and cell proliferation. In the descending colon of supplemented groups, the total number of aberrant crypts and f

LRMDA promotes intestinal mucosal barrier integrity and limits colitis and colon cancer.

Intestinal homeostasis relies on the integrity of the mucosal barrier, whose disruption contributes to inflammatory bowel disease (IBD), enteric infection, and colorectal cancer (CRC). Here, we report that leucine-rich melanocyte differentiation-associated protein (LRMDA) is markedly downregulated in inflamed mucosal tissues from patients with IBD and in colorectal tumor tissues. Genetic ablation of LRMDA in mice exacerbates dextran sulfate sodium (DSS)-induced colitis, enhances colorectal tumorigenesis in both azoxymethane (AOM)/DSS and AOM/Vil-Cre;Trp53 fl/fl models, and increases susceptibi

Effect of concomitant polyethylene glycol and celecoxib on colonic aberrant crypt foci and tumors in F344 rats.

We investigated whether celecoxib augments the protective effect of polyethylene glycol (PEG) on colonic aberrant crypt foci (ACF) and tumor formation in F344 rats treated with azoxymethane (AOM). Three groups of rats received AOM: I (AOM alone), II (PEG), and III (PEG/celecoxib). PEG reduced the mean number of total ACF per colon from 190 to 141 (P < 0.05; 26% reduction) and > or = 4-crypt ACF from 95 to 58 (P < 0.01; 39%). Group III rats had a greater proportion of their ACF distally; whereas transverse colon ACF were reduced approximately 50%, distal ACF were reduced by only approximately 8

Interleukin-34 deficiency aggravates development of colitis and colitis-associated cancer in mice.

BACKGROUND: Although expression of interleukin (IL)-34 is upregulated in active ulcerative colitis (UC), the molecular function and underlying mechanism are largely unclear. AIM: To investigate the function of IL-34 in acute colitis, in a wound healing model and in colitis-associated cancer in IL-34-deficient mice. METHODS: Colitis was induced by administration of dextran sodium sulfate (DSS), and carcinogenesis was induced by azoxymethane (AOM). Whether the impact of IL-34 on colitis was dependent on macrophages was validated by depletion of macrophages in a murine model. The association be

The Appendix Orchestrates T-Cell Mediated Immunosurveillance in Colitis-Associated Cancer.

BACKGROUND & AIMS: Although appendectomy may reduce colorectal inflammation in patients with ulcerative colitis (UC), this surgical procedure has been suggested to be associated with an increased risk of colitis-associated cancer (CAC). Our aim was to explore the mechanism underlying the appendectomy-associated increased risk of CAC. METHODS: Five-week-old male BALB/c mice underwent appendectomy, appendicitis induction, or sham laparotomy. They were then exposed to azoxymethane/dextran sodium sulfate (AOM/DSS) to induce CAC. Mice were killed 12 weeks later, and colons were taken for pathologi

The effects of parathyroid hormone fragments on bone formation and their lack of effects on the initiation of colon carcinogenesis in rats as indicated by preneoplastic aberrant crypt formation.

The parathyroid hormone (PTH) and some of its fragments and analogs stimulate bone growth in various animal models and humans and one of them (hPTH-(1-34)) has been approved by the USFDA for treating osteoporosis. However, there are reports that PTH can stimulate the PI-3 kinase/mitogen-activated protein kinases-mediated proliferation of rat enterocytes and that primary hyperparathyroidism in humans is associated with an increased incidence of colon cancer. Here we have investigated the ability of two PTH fragments, hPTH-(1-34)NH(2) and [Leu(27)]cyclo(Glu(22)-Lys(26))hPTH-(1-31)NH(2) to initia

The lipid transfer protein STARD7 controls intestinal tumor development in a context-dependent manner.

The role of phosphatidylcholine transporters such as Stard7 in intestinal cancer development is unknown. To explore this issue, we generated a mouse model lacking Stard7 in intestinal epithelial cells (IECs). Loss of Stard7 impaired mitochondrial Complex I activity, led to a severe metabolic and lipid reprogramming, enhanced mitochondrial ROS production and potentiated an mTORC1/ATF4 signature. As a result, levels of enzymes involved in serine biosynthesis were enhanced in Stard7-deficient IECs. We next assessed the consequences of Stard7 deficiency in both Wnt-dependent tumor initiation and i

ACSS2 is required for colorectal cancer progression and a druggable target for colorectal cancer treatment.

Acyl-CoA synthetase short chain family member 2 (ACSS2) catalyzes the conversion of acetate to acetyl-CoA. Here we show that ACSS2 expression is markedly elevated in all stages of human colorectal cancer (CRC), and Acss2 silencing or genetic ablation leads to a marked reduction in CRC tumor load in allograft and xenograft models and CRC models induced by epithelial Apc deletion or azoxymethane/dextran sodium sulfate treatment. Tumors with ACSS2 depletion exhibit robust DNA damage, excessive apoptosis, and increased recruitment of macrophages and CD8+ T cells to the tumor microenvironment. Trea

Astragalin Suppresses Colorectal Carcinogenesis by Targeting Proliferation, Oxidative Stress, and Inflammation.

BACKGROUND: Colorectal cancer (CRC) is a leading cause of cancer-related death worldwide. This study evaluated astragalin's (AST) anti-proliferative and chemopreventive effects on CRC. METHODS: AST's effects were assessed in vitro on Caco-2 and HT-29 cells and in vivo on azoxymethane (AOM)-induced aberrant crypt foci (ACF) in rats. Rats received AST (50 or 100 mg/kg, 60 days) or 5-fluorouracil (5-FU) (35 mg/kg, 5 days). RESULTS: AST inhibited Caco-2 and HT-29 cell growth (IC50: 20.15-34.50 µg/mL). In AOM-induced rats, AST reduced ACF formation, improved colon histology, downregulated β-caten

Shinan Sea Salt Intake Ameliorates Colorectal Cancer in AOM/DSS with High Fat Diet-Induced C57BL/6N Mice.

The anticancer effects of Shinan (Shinan-South Korea) sea salts on azoxymethane (AOM)/dextran sodium sulfate (DSS) with high fat diet (HFD)-induced colon cancer and obesity in C57BL/6N mice were studied. We prepared three types of sea salt: generally manufactured sea salt (GS), generally manufactured after filtering seawater (FS), and manufactured with only new seawater (NS). Sea salt intake increased colon length and reduced colon length/weight ratio, tumor number, and progression of colon cancer in colon tissue. The differently prepared sea salts also ameliorated liver injury. In addition, t

Pinto bean consumption mitigates colorectal tumorigenesis through reshaping gut microbiota and metabolic signatures.

Dietary intervention is increasingly recognized as a vital strategy for the prevention of colorectal cancer (CRC). To evaluate the preventive potential of whole foods, we utilized an azoxymethane/dextran sodium sulfate (AOM/DSS)-induced mouse model to compare the systemic effects of a 5% pinto bean (PB) diet against a control diet. Results demonstrated that PB intervention significantly mitigated tumor burden and intestinal inflammation. Integrated multi-omics analysis revealed that PB consumption reshaped the gut ecosystem, specifically enriching Verrucomicrobiota (Akkermansia muciniphila) an

The role of mitochondrial regulation in macrophage polarization by Ganoderma lucidum polysaccharide for the treatment of colitis-associated colorectal cancer.

Colorectal cancer (CRC) is a common malignant tumor of the digestive tract. Ganoderma lucidum polysaccharide (GLP) has shown significant potential in regulating macrophage polarization and treating CRC. However, the role of mitochondrial regulation in GLP-mediated macrophage polarization in azoxymethane/dextran sulfate sodium (AOM/DSS)-induced colitis-associated CRC remains unclear. Therefore, we performed cellular and animal experiments to explore how GLP influences mitochondrial dynamics and function, macrophage polarization, and exerts its therapeutic effect against AOM/DSS-induced colitis-

Deficiency of BTB and CNC homology 1 promotes colon tumorigenesis by enhancing intestinal epithelial cell proliferation in mice.

Intestinal epithelial cells serve as the frontline of host defense and function as a physical barrier. BTB and CNC homology 1 (Bach1) is a transcriptional repressor involved in gastrointestinal physiology, but its role in epithelial proliferation and colorectal tumorigenesis remains unclear. This study demonstrates that Bach1 deficiency promotes colonic epithelial proliferation and enhances colorectal tumorigenesis. In Bach1-deficient (Bach1-/-) mice, structural analysis revealed elongation of villi in the small intestine and pronounced changes in the colon, with increased Ki67+ and BrdU+ cell

Intestinal microbiota affects the progression of colorectal cancer by participating in the host intestinal arginine catabolism.

Arginine plays a critical role in colorectal cancer (CRC) progression. We find that arginine catabolism is reduced in the intestinal microbiota of patients with CRC but increased in tumor tissue. We further verify that Escherichia coli can consume arginine via the arginine succinyltransferase (AST) pathway, and gavaging mice with the AST-deficient E. coli Nissle 1917 (ΔacEcN) can inhibit arginine catabolism of the intestinal microbiota, thereby increasing the arginine concentration in the colon. In the azoxymethane (AOM)/dextran sulfate sodium (DSS)-induced CRC mouse model, reduced arginine ca

CLMP is a tumor suppressor that determines all-trans retinoic acid response in colorectal cancer.

CAR-like membrane protein (CLMP) is a tight junction-associated protein whose mutation is associated with congenital short bowel syndrome (CSBS), but its functions in colorectal cancer (CRC) remain unknown. Here, we demonstrate that CLMP is rarely mutated but significantly decreased in CRC patients, and its deficiency accelerates CRC tumorigenesis, growth, and resistance to all-trans retinoic acid (ATRA). Mechanistically, CLMP recruits β-catenin to cell membrane, independent of cadherin proteins. CLMP-mediated β-catenin translocation inactivates Wnt(Wingless and INT-1)/β-catenin signaling, the
