Effects of bacterial colonization on mucosal structure of the large bowel of neonatal rats.

Micro-anatomical changes in colonic and caecal epithelia of infant rats between birth and weaning appear to be the outcome of the effects of bacterial colonization, superimposed on the natural postnatal ontogeny of the mucous membranes. Conventional rat pups between 15 and 22 days of age showed development of subepithelial spaces in both caecum and colon, rupture of the overlying epithelium and loss of whole enterocyte plaques. Antibiotic-treated animals retained the large-bowel mucosal morphology of the 10-day-old rat, without any of the lesions described in conventional infants. Mucosal lesi

Spatiotemporal Contamination Characteristics, Bacterial Community Structure, and Airflow-Mediated Transport of Bacterial Aerosols in a Pediatric Emergency Consulting Room.

Pediatric emergency consulting rooms are characterized by high occupant density and frequent close-range interactions, creating potential hotspots for microbial aerosol exposure and cross-transmission in hospital built environments. However, microbial contamination and its spatial heterogeneity across different environmental media in pediatric emergency consulting rooms under routine clinical conditions remain insufficiently characterized. This study investigated a pediatric internal medicine emergency consulting room in a children's hospital to clarify bacterial contamination characteristics

Programming structure and properties of bacterial cellulose via fermentation time and strain: a multivariate process-structure-property approach.

Bacterial cellulose (BC) is a versatile biopolymer whose properties can be influenced by fermentation conditions. Here, the effects of fermentation time (4-10 days) and microorganism (Komagataeibacter xylinus and K. hansenii) on BC were systematically evaluated. BC membranes were produced by static fermentation in Hestrin-Schramm medium at 30 °C for 4, 6, 8, or 10 days and characterized in terms of production, water absorption, structure, and mechanical properties. K. xylinus exhibited higher productivity, reaching 0.40 g L-1 day-1 during the early fermentation period, and produced membranes w

Greenhouse gas emissions in a microbial fuel cell-assisted composting system: Microbial insights into bacterial structures and genetic functions.

Greenhouse gas emissions, including methane (CH4), carbon dioxide (CO2), and nitrous oxide (N2O), have significant environmental impacts during the composting process. Herein, a microbial fuel cell (MFC)-assisted composting system was employed to investigate the emission profiles of these gases. CH4 and CO2 emissions were peaked during the temperature-heating phase, while N2O emission showed maximum levels during the temperature-heating and maturation phases. Compared to the control (without MFC), MFC increased the abundance of Proteobacteria, Bacteroidota, and Gemmatimonadota in the compostin

Crystal structures of a bioactive 6'-hydroxy variant of sisomicin bound to the bacterial and protozoal ribosomal decoding sites.

Parasitic infections recognized as neglected tropical diseases are a source of concern for several regions of the world. Aminoglycosides are potent antimicrobial agents that have been extensively studied by biochemical and structural studies in prokaryotes. However, the molecular mechanism of their potential antiprotozoal activity is less well understood. In the present study, we have examined the in vitro inhibitory activities of some aminoglycosides with a 6'-hydroxy group on ring I and highlight that one of them, 6'-hydroxysisomicin, exhibits promising activity against a broad range of prot

Distribution of bacterial community structures and spread of antibiotic resistome at industrially polluted sites of Mini River, Vadodara, Gujarat, India.

The influence of anthropogenic pollution on the distribution of bacterial diversity, antibiotic-resistant bacteria (ARBs), and antibiotic resistance genes (ARGs) was mapped at various geo-tagged sites of Mini River, Vadodara, Gujarat, India. The high-throughput 16S rRNA gene amplicon sequencing analysis revealed a higher relative abundance of Planctomycetota at the polluted sites, compared to the pristine site. Moreover, the relative abundance of Actinobacteriota increased, whereas Chloroflexi decreased in the water samples of polluted sites than the pristine site. The annotation of functional

Structures of RecBCD in complex with phage-encoded inhibitor proteins reveal distinctive strategies for evasion of a bacterial immunity hub.

Following infection of bacterial cells, bacteriophage modulate double-stranded DNA break repair pathways to protect themselves from host immunity systems and prioritise their own recombinases. Here, we present biochemical and structural analysis of two phage proteins, gp5.9 and Abc2, which target the DNA break resection complex RecBCD. These exemplify two contrasting mechanisms for control of DNA break repair in which the RecBCD complex is either inhibited or co-opted for the benefit of the invading phage. Gp5.9 completely inhibits RecBCD by preventing it from binding to DNA. The RecBCD-gp5.9

Excitonic Structure and Primary Charge Separation in Purple Bacterial and Heliobacterial Reaction Centers.

Photosynthetic reaction centers (RCs) are highly optimized pigment-protein complexes that drive the primary charge separation essential for solar energy conversion. Despite deep evolutionary divergence, many RCs share a conserved two-branch structural architecture, raising fundamental questions about how physical symmetry, excitonic interactions, and protein environments dictate electron-transfer directionality and efficiency. This review provides a comparative analysis of primary charge separation in two paradigmatic systems-the pseudo-symmetric purple bacterial reaction center (PbRC, Type II

Structural and dynamic basis of Ssp4-mediated DNA protection in foodborne bacterial spores.

Clostridium perfringens forms metabolically dormant endospores that withstand extreme environmental conditions. Small acid-soluble proteins (SASPs) are ubiquitous DNA-binding proteins in endospores that promote resistance. While their protective role has been previously characterized, we aimed to provide further biophysical insight into the nature of these interactions, focusing on variant-specific structural dynamics through novel single-molecule and NMR approaches. Here, we characterize the DNA-binding properties and structural features of two Ssp4 variants using single-molecule fluorescence

Structure-based design of tirabrutinib derivatives as inhibitors of bacterial tryptophanyl-tRNA synthetase.

The escalating threat of antibiotic resistance poses a critical challenge to global public health, necessitating the urgent development of novel therapeutic agents with distinct mechanisms of action and unique structural scaffolds. Tryptophanyl-tRNA synthetase (TrpRS) has emerged as a promising antibacterial target. Our previous study demonstrated that the clinically utilized Bruton's tyrosine kinase (BTK) inhibitor tirabrutinib, along with several of its analogues, can simultaneously occupy both the substrates l-Trp and tRNATrp A76 binding sites of Escherichia coli tryptophanyl-tRNA synthetas

Structure and activity of the septal peptidoglycan hydrolysis machinery crucial for bacterial cell division.

The peptidoglycan (PG) layer is a critical component of the bacterial cell wall and serves as an important target for antibiotics in both gram-negative and gram-positive bacteria. The hydrolysis of septal PG (sPG) is a crucial step of bacterial cell division, facilitated by FtsEX through an amidase activation system. In this study, we present the cryo-EM structures of Escherichia coli FtsEX and FtsEX-EnvC in the ATP-bound state at resolutions of 3.05 Å and 3.11 Å, respectively. Our PG degradation assays in E. coli reveal that the ATP-bound conformation of FtsEX activates sPG hydrolysis of EnvC

Priority effects dictate community structure and alter virulence of fungal-bacterial biofilms.

Polymicrobial biofilms are a hallmark of chronic wound infection. The forces governing assembly and maturation of these microbial ecosystems are largely unexplored but the consequences on host response and clinical outcome can be significant. In the context of wound healing, formation of a biofilm and a stable microbial community structure is associated with impaired tissue repair resulting in a non-healing chronic wound. These types of wounds can persist for years simmering below the threshold of classically defined clinical infection (which includes heat, pain, redness, and swelling) and cyc

The role of dalbavancin in the treatment of acute bacterial skin and skin structure infections (ABSSSIs).

Introduction: Acute bacterial skin and skin-structure infections (ABSSSI) are a subgroup of skin and soft tissue infections and are a common source of morbidity in both the community and the hospital setting. The most common cause of ABSSSI is Staphylococcus aureus, which also includes methicillin-resistant S. aureus (MRSA), together with beta-hemolytic streptococci, enterococci, and Gram-negative bacteria. Since the emergence of MRSA, the management of ABSSSI has become more challenging. Novel therapies alternative to teicoplanin and vancomycin, intravenous agents commonly used against MRSA a

Bacterial community structure and response to nitrogen amendments in Lake Shenandoah (VA, USA).

Microbial processes are critical to the function of freshwater ecosystems, yet we still do not fully understand the factors that shape freshwater microbial communities. Furthermore, freshwater ecosystems are particularly susceptible to effects of environmental change, including influx of exogenous nutrients such as nitrogen and phosphorus. To evaluate the impact of nitrogen loading on the microbial community structure of shallow freshwater lakes, water samples collected from Lake Shenandoah (Virginia, USA) were incubated with two concentrations of either ammonium, nitrate, or urea as a nitroge

Is perturbation in the quaternary structure of bacterial CysE, another regulatory mechanism for cysteine synthesis?

Drug resistance to almost all antibiotics of Shigella flexneri, a major cause of shigellosis in developing countries, necessitates continuous discovery of novel therapeutics. This study reports a structure-function analysis of a potential drug target serine acetyltransferase (CysE), an enzyme of de novo cysteine biosynthesis pathway that is absent in humans. Analysis of CysE sequences of S. flexneri species and serotypes displayed only two variants that differed by a single amino acid substitution at position 241. Structural inspection of the available crystal structure disclosed this site to

A structural basis for sustained bacterial adhesion: biomechanical properties of CFA/I pili.

Enterotoxigenic Escherichia coli (ETEC) are a major cause of diarrheal disease worldwide. Adhesion pili (or fimbriae), such as the CFA/I (colonization factor antigen I) organelles that enable ETEC to attach efficiently to the host intestinal tract epithelium, are critical virulence factors for initiation of infection. We characterized the intrinsic biomechanical properties and kinetics of individual CFA/I pili at the single-organelle level, demonstrating that weak external forces (7.5 pN) are sufficient to unwind the intact helical filament of this prototypical ETEC pilus and that it quickly r
