Bioassay and Scanning Electron Microscopic Observations Reveal High Virulence of Entomopathogenic Fungus, Beauveria bassiana, on the Onion Maggot (Diptera: Anthomyiidae) Adults.

When flies were dipped in 1 × 108 conidia/ml conidia suspensions and then kept in the incubator (22 ± 1 °C, 70 ± 5% RH), scanning electron microscope observations revealed that, at 2 h, the majority of adhering Beauveria bassiana conidia were attached to either the wing surface or the interstitial area between the macrochaetae on the thorax and abdomen of the onion maggot adults. Germ tubes were being produced and had oriented toward the cuticle by 18 h. Penetration of the insect cuticle had occurred by 36 h, and by 48 h, germ tubes had completely penetrated the cuticle. Fungal mycelia had eme

The entomopathogenic fungus Beauveria bassiana reduces instantaneous blood feeding in wild multi-insecticide-resistant Culex quinquefasciatus mosquitoes in Benin, West Africa.

BACKGROUND: Mosquito-borne diseases are still a major health risk in many developing countries, and the emergence of multi-insecticide-resistant mosquitoes is threatening the future of vector control. Therefore, new tools that can manage resistant mosquitoes are required. Laboratory studies show that entomopathogenic fungi can kill insecticide-resistant malaria vectors but this needs to be verified in the field. METHODS: The present study investigated whether these fungi will be effective at infecting, killing and/or modifying the behaviour of wild multi-insecticide-resistant West African mos

Divergent immune strategies of Colorado potato beetle larvae against the entomopathogenic fungi Metarhizium robertsii and Beauveria bassiana: A comparative transcriptomic analysis.

The entomopathogenic fungi Beauveria bassiana and Metarhizium robertsii are natural pathogens of the Colorado potato beetle (CPB) and hold promise for biocontrol. However, the insect's immune responses to fungal infections remain poorly understood. In this study, we performed transcriptomic analyses of haemocytes and fat body tissue from CPB larvae topically infected with B. bassiana and M. robertsii at the stage of pathogen encapsulation. Quantitative PCR validation of gene expression, along with complementary physiological and biochemical assays, was also conducted. Reference gene annotation

Evaluation of the Entomopathogenic Fungus Beauveria bassiana on Different Stages of Phlebotomus papatasi (Diptera: Psychodidae), Vector of Zoonotic Cutaneous Leishmaniasis in Iran.

BACKGROUND: Zoonotic cutaneous leishmaniasis is a major public health problem in Iran with the main vector of Phlebotomus papatasi. The use of entomopathogenic fungi for biological control of the vector is a potential substitute for the current methods which are being used. The purpose of the current study was to assess the virulence of two local isolates of Beauveria bassiana (OZ2 and TV) against Ph. papatasi. METHODS: To perform the bioassay test, fungal suspensions were applied for every stage of the sand fly life cycle. The mortality rate, longevity, and number of eggs laid were determine

A p53-like transcription factor, BbTFO1, contributes to virulence and oxidative and thermal stress tolerances in the insect pathogenic fungus, Beauveria bassiana.

The p53-like transcription factor (TF) NDT80 plays a vital role in the regulation of pathogenic mechanisms and meiosis in certain fungi. However, the effects of NDT80 on entomopathogenic fungi are still unknown. In this paper, the NDT80 orthologue BbTFO1 was examined in Beauveria bassiana, a filamentous entomopathogenic fungus, to explore the role of an NDT80-like protein for fungal pest control potential. Disruption of BbTFO1 resulted in impaired resistance to oxidative stress (OS) in a growth assay under OS and a 50% minimum inhibitory concentration experiment. Intriguingly, the oxidation re

N-acetylglucosamine sensor, Ngs1 contributes to Beauveria bassiana vegetative growth, oxidative phosphorylation, fungal development, and cell wall integrity during entomopathogen-insect interaction.

N-acetylglucosamine (GlcNAc), a key component of fungal cell walls and insect cuticles, is an important signal to activate fungal response during entomopathogen-insect interactions. Research on Ngs1, the only identified GlcNAc sensor and transducer, has been primarily restricted to Candida species. Although our previous work identified an Ngs1 homology in Beauveria bassiana, its physiological functions in entomopathogenic fungi remain largely unexplored. In this study, we unveiled the sub-localization of Ngs1 in the nucleolus. Further transcriptomic analysis revealed that Ngs1 plays a crucial

The Thm1 Zn(II)2 Cys6 transcription factor contributes to heat, membrane integrity and virulence in the insect pathogenic fungus Beauveria bassiana.

The ability to withstand heat and cell wall stress is critical for successful infection of target hosts by many pathogenic fungi. We report on the characterization of BbThm1 (transcription factor for heat and membrane integrity), a Zn(II)2 Cys6 (Gal4-like) family member, which regulates resistance to heat (32°C) and specific detergents in the insect pathogenic fungus, Beauveria bassiana. BbThm1 gene knock-out mutants showed severely impaired growth/resistance at 32°C and to SDS, but mild to moderate phenotypic responses to other stresses including oxidative, osmotic and cell-wall targeting com

Bioefficacy of Indian Beauveria bassiana and Lecanicillium lecanii isolates against Lipaphis erysimi (Hemiptera: Aphididae) and their safety to Apis cerana (Hymenoptera: Apidae).

Environmental issues associated with the excessive use of chemical substances have received increasing attention globally, particularly regarding their effects on non-target/beneficial insects such as pollinators. To overcome such drawbacks, alternative control methods are being increasingly evaluated. A study was conducted to assess the effectiveness of two Indian biopesticidal isolates against the mustard aphid, Lipaphis erysimi, on rapeseed crops and to investigate their potential risks to the pollinator honeybee, Apis cerana. Laboratory bioassays showed that Beauveria bassiana AS-4220 was

Multifaceted transcriptional regulation of siderophore biosynthesis by the bZIP transcription factor HapX in the entomopathogenic fungus Beauveria bassiana under the iron-deficiency conditions.

Iron acquisition is important for fungal metabolism, growth, development, and pathogenicity. In response to iron limitation/starvation, filamentous fungi excrete siderophores to chelate iron ions from environment. As an iron responsive transcription factor, HapX was found to be required for the siderophore production in the entomopathogenic fungus Beauveria bassiana. Comparative transcriptomic analysis indicated that the HapX-mediated transcriptome was significantly associated with metabolism, energy generation, replication and repair, and membrane transport. Significantly, HapX was required f

Potential biological control of the pupal stage of the European grapevine moth Lobesia botrana by the entomopathogenic fungus Beauveria pseudobassiana in the winter season in Chile.

OBJECTIVE: Lobesia botrana, the European grapevine moth, affects Vitis vinifera L. and other species of economic importance in a number of countries through damage caused by its larvae in berries and associated secondary diseases such as Botrytis cinerea. Control of the moth in urban areas is difficult due to poor chemical management of infested plants in houses. Additionally, in winter, L. botrana is in its pupal stage covered with a cocoon that prevents the penetration of chemical pesticides. For this reason, the objective of this work was to control the pupal stage with a formulation based

The fungal biocontrol agent Beauveria bassiana expresses a HsbA domain-containing protein to postpone the rapid cuticle penetration outwards from the insect cadavers.

The insect biocontrol fungi (IBF) 'hidden' within the dead insect bodies come out of the insect cadavers for dissemination until the suitable environment conditions arising. However, details of the IBF balancing fungal cells 'hidden' within the cadavers and penetration of the cuticle outwards for growth remain limited. Here, a HsbA domain-containing protein, BbHsbA, was found to express during the later stages of insect colonization in an IBF, Beauveria bassiana, which distributed in cytoplasm and cell wall. BbHsbA disruption did not significantly affect fungal virulence, while the mutant pene

Gut Microbial Diversity Reveals Differences in Pathogenicity between Metarhizium rileyi and Beauveria bassiana during the Early Stage of Infection in Spodoptera litura Larvae.

Beauveria bassiana and Metarhizium rileyi are extensively utilized to investigate fungal pathogenic mechanisms and to develop biological control agents. Notwithstanding, notable distinctions exist in their pathogenicity against the same host insect. This study aimed to elucidate the pathogenic differences between M. rileyi and B. bassiana by examining the impact of various ratios of B. bassiana strain AJS91881 and M. rileyi strain SXBN200920 on fifth instar larvae of Spodoptera litura, focusing on early infection stages and intestinal microbial community structure. Control of Hyalomma lusitanicum (Acari: Ixodidade) Ticks Infesting Oryctolagus cuniculus (Lagomorpha: Leporidae) Using the Entomopathogenic Fungus Beauveria bassiana (Hyocreales: Clavicipitaceae) in Field Conditions.

Entomopathogenic fungi are widely used to control arthropods not just in agricultural settings but also in Veterinary Medicine and Public Health. These products have been employed to control tick populations and tick-borne diseases. The effectiveness of these control measures not only depends on the fungi, but also on the tick species and environmental conditions. In Mesomediterranean areas, tick species are adapted to extreme climatic conditions and it is therefore especially important to develop suitable tick control strategies. The aim of this study was to evaluate the effectiveness of a ne

Repellent activity of desiccant dusts and conidia of the entomopathogenic fungus Beauveria bassiana when tested against poultry red mites (Dermanyssus gallinae) in laboratory experiments.

Desiccant dusts and entomopathogenic fungi have previously been found to hold potential against the poultry red mite, which is an important pest in egg production and notoriously difficult to control. Both control agents may cause repellence in other arthropods and potentially also influence control levels adversely when used against the poultry red mite. Five desiccant dust products with good efficacy against the poultry red mite Dermanyssus gallinae caused avoidance behavior in mites when tested in bioassays. The repellent activity was correlated with efficacy, which was found to depend on b

Lesser mealworm Alphitobius diaperinus Panzer (Coleoptera: Tenebrionidae) larvae are differentially susceptible to Beauveria bassiana (Bals.-Criv.) Vuill. (Hypocreales: Clavicipitaceae) under moderate and extreme high temperature stress.

Lesser mealworm larvae topically treated with Beauveria bassiana, under different temperature regimens demonstrated complex interactions with the pathogen. Larvae treated with B. bassiana and incubated at 32.5 °C were less susceptible to B. bassiana than those treated in the same way but then incubated at 25 °C. Larvae incubated at 35 °C for two days after B. bassiana treatment showed limited susceptibility to disease, but those incubated at 35 °C two days prior to treatment and then incubated at 25 °C immediately after B. bassiana treatment were more susceptible than those maintained at 25 °C

N-acetylglucosamine kinase (BbHxk1) has pleiotropic effects on vegetative growth, cell wall integrity, morphological transition, cuticle infection, and metabolic modulation in the biological pesticide Beauveria bassiana.

Beauveria bassiana is a popular and eco-friendly biopesticide. During its pathogen-pest interaction, both N-acetylglucosamine (GlcNAc) catabolism and anabolism are crucial for nutrient supply and cell-wall construction. The initiation of GlcNAc metabolism relies on the catalysis of GlcNAc kinase, which has been extensively studied in the human pathogen Candida albicans. However, the physiological function of GlcNAc kinase remains poorly understood in entomopathogenic fungi. In the present study, a GlcNAc kinase homolog was identified and designated as BbHxk1 in B. bassiana. Deletion of BbHxk1
