Bifidobacterium longum with fructo-oligosaccharide (FOS) treatment in minimal hepatic encephalopathy: a randomized, double-blind, placebo-controlled study.

Minimal hepatic encephalopathy (MHE) describes patients with chronic liver disease or cirrhosis who have no clinical symptoms of brain dysfunction but perform worse on psychometric tests compared with healthy subjects. The pathogenesis of hepatic encephalopathy is controversial although ammonia has been found to induce cerebral dysfunction. Increased intestinal ammonia production is due to bacterial urease activity and the production of other toxin methabolities, such as mercaptans, thioles. This study assesses the clinical efficacy of Bifidobacterium longum plus fructo-oligosaccharides (FOS)

Effects of Bifidobacterium longum 1714 on low mood and related symptoms: A randomized, double-blind, placebo-controlled trial.

Psychobiotics have been suggested as an alternative or adjunctive intervention for depressive symptoms. Bifidobacterium longum (B. longum) 1714 has previously been shown to influence stress response and sleep quality. This double-blind, placebo-controlled trial investigated whether B. longum 1714 (1 × 10¹⁰ colony-forming units/day) affects severity of depressive symptoms in 168 adults (aged 18-70) with mild to moderate depression over 8 weeks. The primary outcome was the change in Beck's Depression Inventory-II scores. Secondary outcomes included changes in sleep quality and mental wellness, m

Bifidobacterium longum supplementation prevents intestinal inflammation and cognitive impairment in sleep-deprived mice by modulating clock gene expression.

Probiotics have shown potential for alleviating circadian disruption, but efficient screening models for identifying effective strains are lacking, and the mechanisms by which probiotics modulate circadian disruption and related diseases require further investigation. In this research, we developed an in vitro screening model targeting clock gene expression and identified Bifidobacterium longum CCFM1238 as an effective strain for alleviating circadian disruption in sleep-deprived mice. B. longum CCFM1238 exhibited a rhythm-regulating effect similar to melatonin in vitro and alleviated both int

A 90-day oral toxicity study of heat-treated Bifidobacterium longum CECT 7347 (ES1 HT) Postbiotic in rats.

Probiotic organisms have a long history of safe consumption in foods and dietary supplements. Probiotics can be inactivated using a method such as heat treatment, creating more shelf stable, non-viable biotics, termed postbiotics. Heat-treated Bifidobacterium longum CECT 7347 (ES1 HT), a postbiotic derived from the probiotic strain B. longum CECT 7347 was tested in a 90-day toxicology study with 10 Wistar rats/sex/group using a study design adapted from the OECD test guideline for the 90-day repeated dose toxicology test in rodents (OECD TG 408). Heat-treated B. longum CECT 7347 (ES1 HT) was t

Alleviating effects of 2'-fucosyllactose combined with Bifidobacterium longum BB536 on loperamide-induced constipation and depressive-like behavior.

Constipation as a public health issue impairs human health and quality of life, while current strategies against constipation often lead to adverse effects. This study evaluated the effects and underlying mechanism of 2'-fucosyllactose (2'-FL) and Bifidobacterium longum BB536 (BB536) on relieving loperamide-induced constipation and depressive-like behavior. 2'-FL + BB536 can significantly elevate fecal water content, shorten defecation time and enhance the intestinal transit rate in constipated mice. 2'-FL + BB536 increased the contents of substance P and 5-hydroxytryptamine, while reducing th

From Identification to Enzymatic Activity Profiling: Comprehensive Characterization of BloCAβ, a Probiotic β-Carbonic Anhydrase From Bifidobacterium longum.

The microbiota comprises communities of microorganisms that establish complex relationships with the host and play key roles in physiology, metabolism, and immunity. Increasing evidence highlights microbiota alterations in several diseases, particularly those driven by oxidative stress-induced inflammation. Thus, identifying probiotic enzymes implicated in essential metabolic pathways is highly relevant to the development of future strategies targeting dysbiosis and related diseases. Herein, we report the first biochemical characterization of β-class carbonic anhydrase (CA) from Bifidobacteriu

Bifidobacterium longum S3 alleviates loperamide-induced constipation by modulating intestinal acetic acid and stearic acid levels in mice.

Constipation is a major gastrointestinal (GI) symptom worldwide, with diverse causes of formation, and requires effective and safe therapeutic measures. In the present study, we used loperamide hydrochloride to establish a constipation model and assessed the effect of Bifidobacterium on constipation and its possible mechanism of relief. The results showed that B. longum S3 exerted a constipation-relieving effect primarily by improving the gut microbiota, enriching genera including Lactobacillus, Alistipes, and Ruminococcaceae UCG-007, and decreasing the bacteria Lachnospiraceae NK4B4 group. Inhibitory Potential of Bifidobacterium longum FB1-1 Cell-Free Supernatant against Carbapenem-Resistant Klebsiella pneumoniae Drug Resistance Spread.

The widespread dissemination of carbapenem-resistant Klebsiella pneumoniae (CRKP) and its drug resistance transfer poses a global public health threat. While previous studies outlined CRKP's drug resistance mechanism, there is limited research on strategies inhibiting CRKP drug resistance spread. This study investigates the potential of Bifidobacterium longum (B. longum) FB1-1, a probiotic, in curbing the spread of drug resistance among CRKP by evaluating its cell-free supernatant (CFS) for antibacterial activity. Evaluating the inhibitory effect of FB1-1 CFS on CRKP drug resistance spread inv

Lactobacillus mucosae and Bifidobacterium longum Synergistically Alleviate Immobilization Stress-Induced Anxiety/Depression in Mice by Suppressing Gut Dysbiosis.

We isolated Lactobacillus mucosae NK41 and Bifidobacterium longum NK46 from human feces, which induced BDNF expression in corticosterone-stimulated SH-SY5Y cells, and examined their anti-depressive effects in mice. NK41, NK46, and their (1:1) mixture significantly mitigated immobilization stress (IS)-induced anxiety-like/depressive behaviors, hippocampal NF-κB activation, BDNF expression, Iba1+ cell population, and blood corticosterone, TNF-α, IL- 6, and lipopolysaccharide levels. Furthermore, they inhibited colitis marker NF-κB activation, and TNF-α expression in mice with IS-induced anxiety/

Isolation and Characterization of Bifidobacterium longum A1, an Isolate from Faeces of an Exclusively Breast-fed Infant with Probiotic Potential.

UNLABELLED: Microbial colonization of the newborn (primocolonization) plays a crucial role in the development of digestive, metabolic, and immune systems. Disruptions may impair intestinal barrier function and raise the risk of chronic diseases like inflammatory bowel disease, allergies, and obesity. Recent evidence suggests probiotic supplementation in infants may help counter or prevent these effects. This study isolates and characterizes potential probiotic candidates from the gut microbiota of exclusively breastfed infants. Bifidobacterium and Lacticaseibacillus strains were selectively is

Faecal hsa-miR-7704 inhibits the growth and adhesion of Bifidobacterium longum by suppressing ProB and aggravates hepatic encephalopathy.

Both gut microbiome and microRNAs (miRNAs) play a role in the development of hepatic encephalopathy (HE). However, the functional link between the microbiome and host-derived miRNAs in faeces remains poorly understood. In the present study, patients with HE had an altered gut microbiome and faecal miRNAs compared with patients with chronic hepatitis B. Transferring faeces and faecal miRNAs from patients with HE to the recipient mice aggravated thioacetamide-induced HE. Oral gavage of hsa-miR-7704, a host-derived miRNA highly enriched in faeces from patients with HE, aggravated HE in mice in a

The Anti-Constipation Effect of Bifidobacterium Longum W11 Is Likely Due to a Key Genetic Factor Governing Arabinan Utilization.

Recent investigations have highlighted, both experimentally and clinically, that probiotic strains equipped with arabinofuranosidase, in particular abfA and abfB, favor regular intestinal motility, thus counteracting constipation. By analyzing the gene expression and the proliferative response in the presence of arabinan of the probiotic B. longum W11, a strain previously validated as an anti-constipation probiotic, we have speculated that its response mechanism to arabinan can effectively explain its clinical action. Our approach could be used in the future to select probiotics endowed with a

Human milk-derived Bifidobacterium longum subsp. infantis CCFM1269 alleviates food allergy by modulating gut microbiota and restoring intestinal barrier.

Food allergy (FA) is an immune-mediated disorder increasingly linked to intestinal dysbiosis and epithelial barrier dysfunction. This study evaluated the protective effects of Bifidobacterium longum subsp. infantis CCFM1269, a human milk-derived probiotic, using a β-lactoglobulin-induced FA mouse model. Oral administration of CCFM1269 significantly reduced allergic symptoms, including poor weight gain, diarrhea, and elevated allergy scores. The probiotic restored immune balance by downregulating IL-4 and IL-17 A and upregulating IL-10 and IFN-γ in both serum and jejunal tissues. It also allevi

Bifidobacterium longum alleviation of metabolic dysfunction-associated steatotic liver disease: A multi-omics landscape of microbiota and metabolome reconfiguration.

The gut microbiome-host metabolism axis plays a critical role in the pathogenesis of metabolic dysfunction-associated steatotic liver disease (MASLD). Although the probiotic Bifidobacterium longum (B. longum) shows promise in ameliorating metabolic disorders, its functional impact on the microbiome-metabolome interplay in MASLD remains elusive. Herein, we established a MASLD mouse model using a high-fat, high-fructose (HFHF) diet and conducted integrated multi-omics analyses, including liver transcriptomics, gut metagenomics, and serum metabolomics, following B. longum intervention. B. longum

Dose-response studies of methylated and nonmethylated CpG ODNs from Bifidobacterium longum subsp. infantis for optimizing Treg cell stimulation.

Background: Allergen immunotherapy (AIT) is the most effective treatment for atopic allergic diseases, aiming to induce regulatory T cells (Treg) that modify the immune response to specific allergens, which leads to long-term tolerance and reduced symptoms. Enhancing Treg activity is crucial for improving immunotherapy outcomes. In a previous murine model study, we examined the effects of a synthetic methylated DNA oligodeoxynucleotide (ODN) from the Bl-T2 m5C motif of Bifidobacterium longum subsp. infantis. The ODN that contains the methylated BI-T2 m5C motif (methylated ODNA) sequence conjug

Methylated CpG ODNs from Bifidobacterium longum subsp. infantis Modulate Treg Induction and Suppress Allergic Response in a Murine Model.

In our previous studies, methylated CpG oligodeoxynucleotides (ODN) derived from Bifidobacterium longum subsp. infantis have demonstrated immunomodulatory effects through the induction of regulatory T cells (Tregs). To define the structural determinants underlying this effect, we synthesized four CpG ODNs varying in methylation degree, CpG motif placement, and backbone length. These include (1) ODN-A (2m-V1), a 20-nucleotide CpG oligodeoxynucleotide incorporating two 5-methylcytosines at positions 4 and 12 within centrally placed CpG motifs; (2) ODN-B (um-V2), a 20-nucleotide CpG oligodeoxynuc
