The role of biofilms in otolaryngologic infections.

PURPOSE OF REVIEW: Bacterial biofilms have recently been shown to be important in diseases of the head and neck. Because the concept of biofilms is novel to most practitioners, it is important to gain a basic understanding of biofilms and to recognize that strategies developed to treat planktonic bacteria are ineffective against bacteria in a biofilm. RECENT FINDINGS: Bacteria preferentially exist in complex, surface-attached organizations known as biofilms. Bacteria in biofilms express a different set of genes than their planktonic counterparts and have markedly different phenotypes. Biofilm

Candida biofilms and their role in infection.

Pathogenic fungi in the genus Candida can cause both superficial and serious systemic disease, and are now recognized as major agents of hospital-acquired infection. Many Candida infections involve the formation of biofilms on implanted devices such as indwelling catheters or prosthetic heart valves. Biofilms of Candida albicans formed in vitro on catheter material consist of matrix-enclosed microcolonies of yeasts and hyphae, arranged in a bilayer structure. The biofilms are resistant to a range of antifungal agents currently in clinical use, including amphotericin B and fluconazole, and ther

Dental unit waterlines: biofilms, disinfection and recurrence.

BACKGROUND: Transmission of microbial pathogens to patients from biofilm within dental unit waterlines, or DUWLs, is a concern. To reduce the risk of toxicity to dental patients when water coolants are used, numerous chemical agents have been tested. In a series of trials, the authors investigated the recurrence of microbial growth after treating DUWLs with sodium hypochlorite (bleach), or B; glutaraldehyde, or G; or isopropanol 15.3 percent, or I. METHODS: The authors excised tubing sections from dental units in a general clinic. The tubing sections were evaluated at baseline and after overn

Blockage of urethral catheters by bacterial biofilms.

We report the case of a 63-year-old woman whose indwelling urethral catheter became blocked regularly at 4-5 day intervals over a period of 10 weeks. 'Worm-like' structures 25-30 cm in length were found either in the catheter, completely occluding the lumen, or in the drainage tube thereby blocking the valve of the drainage bag. Electron microscopy showed that these structures were composed of bacteria, while culture revealed them to be mixed communities of Pseudomonas aeruginosa, Escherichia coli, Enterococcus faecalis and Proteus mirabilis. Following treatment with ciprofloxacin, catheter dr

Combining Essential Oils with Each Other and with Clotrimazole Prevents the Formation of Candida Biofilms and Eradicates Mature Biofilms.

Fungal infections by Candida spp. are opportunistic and most often occur in individuals with some predisposing factor. Essential oils (EO) have anti-Candida potential, being a therapeutic alternative to be explored, especially for superficial and mucosal candidiasis. The objective was to analyze the synergistic potential between the EO of Citrus limon, Cupressus sempervirens, Litsea cubeba and Melaleuca alternifolia, and each of them with clotrimazole, to inhibit in vitro the formation and eradication of Candida spp. biofilms. Added to this, the survival of Caenorhabditis elegans was evaluated

Forging biofilms: metal-induced microbial responses in biofilm formation.

Biofilms are a major contributor to antibiotic resistance and persistence in hospital environments. These bacterial communities form protective structures that shield microbes from various stressors, such as oxidative stress, pH fluctuations, osmotic pressure, and nutrient deprivation. As our understanding of biofilms has grown, it has become evident that metals play a crucial role in several aspects of biofilm biology. Metals are involved in regulating biofilm formation, facilitating communication and competition among bacteria, and supporting the structural integrity and adherence of bacteri

Fungal Biofilms: In Vivo Models for Discovery of Anti-Biofilm Drugs.

During infection, fungi frequently transition to a biofilm lifestyle, proliferating as communities of surface-adherent aggregates of cells. Phenotypically, cells in a biofilm are distinct from free-floating cells. Their high tolerance of antifungals and ability to withstand host defenses are two characteristics that foster resilience. Biofilm infections are particularly difficult to eradicate, and most available antifungals have minimal activity. Therefore, the discovery of novel compounds and innovative strategies to treat fungal biofilms is of great interest. Although many fungi have been ob

Sporothrix spp. Biofilms Impact in the Zoonotic Transmission Route: Feline Claws Associated Biofilms, Itraconazole Tolerance, and Potential Repurposing for Miltefosine.

Sporotrichosis is the most prevalent subcutaneous mycosis globally, and it is typically caused by direct inoculation of the soil saprophytic fungus Sporothrix spp. into the patients' skin. However, sporotrichosis has an important zoonotic transmission route between cats and humans in hot-spot endemic areas such as Brazil. Antifungal itraconazole is the first-line treatment; however, it is frequently associated with recurrence after withdrawal, mainly on cats. Biofilms are important resistance structures related to the environmental persistence of most microorganisms. In the present work, we ev

Porcupine-Quill-Inspired Shape-Morphing Biointerfaces Disassemble Biofilms.

Mature biofilms are difficult to eradicate because the extracellular polymeric substance (EPS) matrix shields embedded bacteria from chemical and mechanical attack. Here, we report a porcupine-spine-inspired photo-responsive dynamic offensive bio-interface that dismantles mature biofilms through electrically actuated mechanical penetration and light-driven oxidative stress. A high-aspect-ratio nano-dagger array undergoes reversible nanoscale deformation (∼110 nm) under an alternating electric field, while a photo-responsive heterostructure generates a surface photovoltage (∼364 mV) that enhanc

Biochemical characterization of cellulose isolated from mycobacterial biofilms.

Mycobacterium tuberculosis (Mtb) forms cellulose-enriched biofilms. These biofilms play a crucial role in the tuberculosis pathogenesis and drug tolerance exhibited by Mtb in vivo. However, the biophysical characteristics of the cellulose remain poorly explored. In the current study, the Updegraff method was employed to enrich acid-hydrolysis-resistant polysaccharides containing cellulose from three distinct models of biofilms: pellicle, submerged, and macrocolony biofilms of slow-growing Mtb and fast-growing M. smegmatis. The isolated material was subjected to Elemental analysis, Fourier tran

A chemostat-based model for growing bacterial biofilms.

Biofilms are groups of microbes that live together in dense communities, often attached to a surface. They play an outsized role in all aspects of microbial life, from chronic infections to biofouling to dental decay. In recent decades, appreciation for the diversity of roles that biofilms play in the environment has grown. Yet, most bacterial studies still rely upon approaches developed in the 19th century and center on planktonic populations alone. Here, we present a chemostat-based experimental platform to investigate not only biofilms themselves but how they interact with their surrounding

Archaea influence composition of endoscopically visible ileocolonic biofilms.

The gut microbiota has been implicated as a driver of irritable bowel syndrome (IBS) and inflammatory bowel disease (IBD). Recently we described, mucosal biofilms, signifying alterations in microbiota composition and bile acid (BA) metabolism in IBS and ulcerative colitis (UC). Luminal oxygen concentration is a key factor in the gastrointestinal (GI) ecosystem and might be increased in IBS and UC. Here we analyzed the role of archaea as a marker for hypoxia in mucosal biofilms and GI homeostasis. The effects of archaea on microbiome composition and metabolites were analyzed via amplicon sequen

Substrate Specificity of Biofilms Proximate to Historic Shipwrecks.

The number of built structures on the seabed, such as shipwrecks, energy platforms, and pipelines, is increasing in coastal and offshore regions. These structures, typically composed of steel or wood, are substrates for microbial attachment and biofilm formation. The success of biofilm growth depends on substrate characteristics and local environmental conditions, though it is unclear which feature is dominant in shaping biofilm microbiomes. The goal of this study was to understand the substrate- and site-specific impacts of built structures on short-term biofilm composition and functional pot

Photocatalytic synergistic biofilms enhance tetracycline degradation and conversion.

Tetracyclines are refractory pollutants that cause persistent harm to the environment and human health. Therefore, it is urgently necessary to develop methods to promote the efficient degradation and conversion of tetracyclines in wastewater. This report proposes a photobiocatalytic synergistic system involving the coupling of GeO2/Zn-doped phosphotungstic acid hydrate/TiO2 (GeO2/Zn-HPW/TiO2)-loaded photocatalytic optical hollow fibers (POHFs) and an algal-bacterial biofilm. The GeO2/Zn-HPW/TiO2 photocatalyst exhibits a broad absorption edge extending to 1000 nm, as well as high-efficiency pho

The Presence of Biofilms in Instrumented Spinal Fusions.

Study Design: Prospective observational cohort study. Objective: To determine whether biofilms exist on spinal instrumentation recovered during revision surgery in which microbial cultures were negative. Background: Biofilm bacteria are extremely difficult to detect by conventional culture methods used in the standard hospital setting. Chronic infections in which bacteria form biofilms have been demonstrated to slow healing and prevent bony fusion. These slime encased microbial communities serve to isolate the bacteria from the body's immune responses, while simultaneously providing metabolic

Unsaturated fatty acid salts remove biofilms on dentures.

Candidiasis-causing Candida sp. forms biofilms with various oral bacteria in the dentures of the elderly, making it harder to kill and remove the microorganism due to the extracellular polymeric substances. We found that biofilms on dentures can effectively be removed by immersion in an unsaturated fatty acid salt solution. Using optical coherence tomography to observe the progression of biofilm removal by the fatty acid salt solution, we were able to determine that the removal was accompanied by the production of gaps at the interface between the biofilm and denture resin. Furthermore, micros

Growing and Characterizing Biofilms Formed by Streptococcus pneumoniae.

It is estimated that over 80% of bacterial infections are associated with biofilm formation. Biofilms are organized bacterial communities formed on abiotic surfaces, such as implanted or inserted medical devices, or on biological surfaces, such as epithelial linings and mucosal surfaces. Biofilm growth is advantageous for the bacterial organism as it protects the bacteria from antimicrobial host factors and allows the bacteria to reside in the host without causing excessive inflammation. Like many other opportunistic pathogens of the respiratory tract, Streptococcus pneumoniae forms biofilms d

Thermostable xylanase inhibits and disassembles Pseudomonas aeruginosa biofilms.

Pseudomonas aeruginosa biofilms are problematic and play a critical role in the persistence of chronic infections because of their ability to tolerate antimicrobial agents. In this study, various cell-wall degrading enzymes were investigated for their ability to inhibit biofilm formation of two P. aeruginosa strains, PAO1 and PA14. Xylanase markedly inhibited and detached P. aeruginosa biofilms without affecting planktonic growth. Xylanase treatment broke down extracellular polymeric substances and decreased the viscosity of P. aeruginosa strains. However, xylanase treatment did not change the
