Albumin domain mutants with enhanced Aβ binding capacity identified by phage display analysis for application in various peripheral Aβ elimination approaches of Alzheimer's disease treatment.

Deposition of amyloid protein, particularly Aβ1-42 , is a major contributor to the onset of Alzheimer's disease (AD). However, almost no deposition of Aβ in the peripheral tissues could be found. Human serum albumin (HSA), the most abundant protein in the blood, has been reported to inhibit amyloid formation through binding Aβ, which is believed to play an important role in the peripheral clearance of Aβ. We identified the Aβ binding site on HSA and developed HSA mutants with high binding capacities for Aβ using a phage display method. HSA fragment 187-385 (Domain II) was found to exhibit the

Childhood ALL and second neoplasms.

Diabetes mellitus affects over 589 million adults globally, with projections exceeding 900 million by 2050. Conventional antidiabetic therapies are limited by adverse effects, high cost, and single-target pharmacology. Endophytic microorganisms, fungi and bacteria residing asymptomatically within plant tissues, represent an underexplored yet structurally diverse reservoir of bioactive metabolites developed through co-evolutionary adaptation with their hosts. This scoping review aimed to systematically map the breadth and depth of available evidence on: (1) the antidiabetic biochemical mechanis

Long-term primary culture of secretory cells of Bothrops jararaca venom gland for venom production in vitro.

This protocol details the optimal conditions to establish a long-term primary culture of secretory cells from the venom gland of the Bothrops jararaca snake. Furthermore, these conditions allow the production and secretion of venom into the culture medium. Snake venom is a rich source of active molecules and has been used for bioprospection studies. However, obtaining enough venom from snakes is a major obstacle. Secretory cells of venom glands are capable of producing active toxins. Therefore, a culture of secretory cells is a good in vitro system to acquire the venom of snakes without captur

Design and implementation of aerobic and ambient CO2-reduction as an entry-point for enhanced carbon fixation.

The direct reduction of CO2 into one-carbon molecules is key to highly efficient biological CO2-fixation. However, this strategy is currently restricted to anaerobic organisms and low redox potentials. In this study, we introduce the CORE cycle, a synthetic metabolic pathway that converts CO2 to formate at aerobic conditions and ambient CO2 levels, using only NADPH as a reductant. Combining theoretical pathway design and analysis, enzyme bioprospecting and high-throughput screening, modular assembly and adaptive laboratory evolution, we realize the CORE cycle in vivo and demonstrate that the c

Paving the way towards effective plant-based inhibitors of hyaluronidase and tyrosinase: a critical review on a structure-activity relationship.

Human has used plants to treat many civilisation diseases for thousands of years. Examples include reserpine (hypertension therapy), digoxin (myocardial diseases), vinblastine and vincristine (cancers), and opioids (palliative treatment). Plants are a rich source of natural metabolites with multiple biological activities, and the use of modern approaches and tools allowed finally for more effective bioprospecting. The new phytochemicals are hyaluronidase (Hyal) inhibitors, which could serve as anti-cancer drugs, male contraceptives, and an antidote against venoms. In turn, tyrosinase inhibitor

Exploring the influence of lactoferrin on wound healing and related signaling pathways: a systematic review.

This systematic review evaluates the therapeutic efficacy and biological mechanisms of Lactoferrin (Lf) in the wound healing process. The objective was to synthesize evidence regarding its multifunctional role in cellular regeneration, gene modulation, and antimicrobial protection. From an initial screening of 1846 records, 11 in vitro studies (2001-2023) met the eligibility criteria. The synthesized data demonstrate that Lf significantly accelerates the migration and proliferation of human keratinocytes and fibroblasts. At the molecular level, Lf upregulates critical regenerative genes, speci

Effects of Cape Cobra (Naja nivea) Venom and Its Isolated Protein on the Modulation of Platelet Activation.

The Cape cobra (Naja nivea), one of Africa's most lethal snakes, can cause rapid, life-threatening paralysis. However, the impact of this venom on platelet function and blood coagulation remains poorly understood. To address this gap, we investigated the enzymatic profiles and the impacts of N. nivea venom on multiple aspects of haemostasis using human whole blood. Our results illustrate that Cape cobra venom significantly increases clotting time in rotational thromboelastometry without affecting other coagulation parameters. This venom significantly inhibits platelet aggregation and activatio

Utilizing CO2 in industrial off-gas for microalgae cultivation: considerations and solutions.

The utilization of microalgae to treat carbon dioxide (CO2)-rich industrial off-gas has been suggested as both beneficial for emissions reduction and economically favorable for the production of microalgal products. Common sources of off-gases include coal combustion (2-15% CO2), cement production (8-15% CO2), coke production (18-23% CO2), and ore smelting (6-7% CO2). However, industrial off-gas also commonly contains other acid gas components [typically nitrogen oxides (NOX) and sulfur dioxide (SO2)] and metals that could inhibit microalgae growth and productivity. To utilize industrial off-g

Metabolic engineering strategies to produce medium-chain oleochemicals via acyl-ACP:CoA transacylase activity.

Microbial lipid metabolism is an attractive route for producing oleochemicals. The predominant strategy centers on heterologous thioesterases to synthesize desired chain-length fatty acids. To convert acids to oleochemicals (e.g., fatty alcohols, ketones), the narrowed fatty acid pool needs to be reactivated as coenzyme A thioesters at cost of one ATP per reactivation - an expense that could be saved if the acyl-chain was directly transferred from ACP- to CoA-thioester. Here, we demonstrate such an alternative acyl-transferase strategy by heterologous expression of PhaG, an enzyme first identi

Xenobiotic dynamics in mangroves and peatlands: Microbial mechanisms for nature-based mitigation.

Peatlands and mangroves provide substantial ecosystem services, but are increasingly threatened by xenobiotic contamination, posing escalating ecological risks. Here, we present the first integrative synthesis of the xenobiotic dynamics in these systems, mapping contaminant distributions and identifying emerging research priorities. Over time, research has shifted from oil spills and heavy metal pollution towards persistent organic pollutants, microplastics and climate risks. Peatlands remain substantially understudied compared to mangroves, especially for emerging contaminant classes. Xenobio

Next-generation brewing yeasts for non-alcoholic beers.

Non‑alcoholic beers (NABs) are a rapidly expanding market. However, producing high‑quality products remains challenging because ethanol strongly shapes beer flavor and mouthfeel. Biological production methods, which restrict alcohol formation during fermentation, offer advantages over physical methods, which remove alcohol after fermentation. However, they are limited by the performance and diversity of available yeasts. Recent advances in bioprospecting, breeding, hybridization, adaptive evolution, and genetic engineering reveal how different yeast lineages and trait-development strategies ca

A Versatile Transcription Factor Biosensor System Responsive to Multiple Aromatic and Indole Inducers.

Allosteric transcription factor (aTF) biosensors are valuable tools for engineering microbes toward a multitude of applications in metabolic engineering, biotechnology, and synthetic biology. One of the challenges toward constructing functional and diverse biosensors in engineered microbes is the limited toolbox of identified and characterized aTFs. To overcome this, extensive bioprospecting of aTFs from sequencing databases, as well as aTF ligand-specificity engineering are essential in order to realize their full potential as biosensors for novel applications. In this work, using the TetR-fa

Microorganisms as a Potential Source of Molecules to Control Trypanosomatid Diseases.

Trypanosomatids are the causative agents of leishmaniasis and trypanosomiasis, which affect about 20 million people in the world's poorest countries, leading to 95,000 deaths per year. They are often associated with malnutrition, weak immune systems, low quality housing, and population migration. They are generally recognized as neglected tropical diseases. New drugs against these parasitic protozoa are urgently needed to counteract drug resistance, toxicity, and the high cost of commercially available drugs. Microbial bioprospecting for new molecules may play a crucial role in developing a ne

Cutaneous nocardiosis by a new pathogenic species: Nocardia grenadensis.

Nocardiosis is a rare, predominantly opportunistic, suppurative disease caused by bacteria of the order Actinomycetales. There are currently more than 100 species of Nocardia described, less than half are pathogenic to humans. Cutaneous nocardiosis can be caused by direct inoculation from a contaminated material or by secondary dissemination. The authors present a 70-year-old man with an autoimmune haemolytic anaemia treated with prednisolone and azathioprine. The patient presented multiple erythematous tender nodules with linear distribution and proximal progression along the left upper limb

Plants as antimalarial agents in Sub-Saharan Africa.

Although the burden of malaria is decreasing, parasite resistance to current antimalarial drugs and resistance to insecticides by vector mosquitoes threaten the prospects of malaria elimination in endemic areas. Corollary, there is a scientific departure to discover new antimalarial agents from nature. Because the two antimalarial drugs quinine and artemisinin were discovered through improved understanding of the indigenous knowledge of plants, bioprospecting Sub-Saharan Africa's enormous plant biodiversity may be a source of new and better drugs to treat malaria. This review analyses the medi

Protocol for Differential Biopanning of P. falciparum Phage Display cDNA Library to Identify Parasite Targets of Protective Antibodies.

Malaria remains a significant global health burden, killing hundreds of thousands of children annually (WHO, The world malaria report. WHO, Geneva, 2019). Despite decades of effort, no broadly effective vaccine exists. Differential screening of parasite phage display libraries is a promising approach to identify the targets of human antibodies expressed by resistant but not by susceptible individuals (Raj et al., Nature, 582, 104-108, 2020; Science, 344, 871-877, 2014). Our whole proteome differential screening (WPDS) approach consists of positive selection to capture phage that bind antibodie

GC-MS analysis of the fixed oil from Sus scrofa domesticus Linneaus (1758) and antimicrobial activity against bacteria with veterinary interest.

The bioprospection of zootherapeutic products can be a source of new drugs and to the creation of new strategies of natural resources conservation and management of endangered species. This fact is supported by ethnobiological studies indicating that the usage of zootherapeutic products can be replaced by the use of natural products isolated from plants and domestic animals. The emergence of antibiotic-resistant bacteria has increased the need for research for new active principles. Ethnoveterinary studies in Brazil have shown that Sus scrofa domesticus fat is used for diseases associated with
