In view of the coincidence of antiviral and antiparkinsonism activities of amantadine four antiparkinsonism drugs, NorakinR (triperiden), ParkopanR (trihexyphenidyl), AntiparkinR (diethylbenzhydramine) and AkinetonR (biperiden) were tested for antiviral activity in various virus-cell systems. Norakin inhibited the replication of influenza A viruses in chick embryo fibroblast, MDCK and Ehrlich ascites tumour cells. It also inhibited the replication of measles virus in Vero cells, 50% inhibitory concentrations being 2-6 micrograms/ml. The drugs were also active against influenza B virus. Several

We describe a 29-year-old patient who developed acute colitis limited to the sigmoid and left colon with features mimicking ischemic injury after a prolonged administration of trifluoroperazine and levomepromazine, two phenothiazines in association with haloperidol, another neuroleptic, and biperidene, an anticholinergic compound. The discontinuation of these drugs was followed by a prompt and complete recovery, and no other cause of acute colitis was found. The subsequent administration of sultopride, a neuroleptic from the benzamide family and then the readministration of haloperidol were we

A 73-year-old woman, with tuberculosis of the large intestine, developed nausea as a side effect of the antituberculosis drugs. The nausea was treated with metoclopramide. Subsequently she developed severe medication-induced parkinsonism. As her symptoms initially mimicked a depressive disorder, drug-induced parkinsonism was only considered at a later stage. Due to drug-induced impaired function of the liver and kidney the patient had received a toxic dose of metoclopramide. Treatment with biperiden and withdrawal of the metoclopramide resulted in a reduction of the complaints within 3 months,

It was discovered by serendipity that a combined application of chlorpromazine (3.5 mg/kg) and pentobarbitone (20.0 mg/kg) induced intermittent tremor-bursts in four extremities of guinea pigs. This tremor with a prominent extensor activity disappeared after spinalization, indicating a supraspinal process. Biperiden reduced the frequency and amplitude of the tremor, which was further diminished by amantadine. Baclofen was found to be most potent drug in suppressing tremor. It was suggested that the cortico-striato-pallido-thalamo-cortical feedback loop may be impaired by a combined action of c

A Parkinsonian-like tremor can be induced by a combined application of chlorpromazine and pentobarbitone in cats. Interestingly enough, only cats with left-paw preference were found to have a predisposition to this drug-induced tremor. The cats with ambidexterity and right-preference in paw use did not show any visible tremorogenic action of these drugs. Especially the proximal muscles of all four extremities showed oscillations with a frequency of about 12 per second in all animals tested, which completely disappeared after i.v. application of lioresal, atropine and biperiden. It was suggeste

Two weeks after epidemic parotitis a 10-year old boy developed an acute extrapyramidal syndrome together with pyramidal tract signs but without impairment of consciousness. The CSF showed mild pleocytosis. The CT of the brain exhibited a well defined circumscribed and symmetrical swelling and hypodensity of the basal ganglia and of the internal capsule. Only slight and transient dysrhythmia was to be found in the EEG. Therapy with Prednisolone, Biperidene and Tiapride resulted in regression of symptoms. But one year later rigor and pyramidal tract signs remained evident in spite of intensive p

The investigation of the metabolism of the antiparkinson drugs trihexyphenidyl (1), pridinol (2) and biperiden (3) revealed a graduate tendency for hydroxylation in the different structural elements: If alicyclic, saturated heterocyclic and aromatic ring systems are present in one compound like in 1, the alicyclic ring system is attacked predominately. The amount of metabolites with hydroxy-groups in the saturated heterocyclic ring is much lower, and no hydroxylation takes place in the aromatic ring. In drugs without alicyclic ring systems like 2 the saturated heterocyclus is attacked preferen

Acetylcholine is a neurotransmitter that is important for communication between neurons and muscle, is involved in direct neurotransmission in the autonomic parasympathetic nervous system, and has been implicated in cognitive processing, arousal, and attention in the brain. The cholinergic-adrenergic hypothesis of mood disorders states that a given affective state might represent a balance between central cholinergic and adrenergic neurotransmitter activity in those areas of the brain regulating moods. According to this hypothesis, depression would be the clinical manifestation of a state of c

Acute dystonia is commonly associated with high-potency antipsychotics. Some cases of acute dystonia had been reported to be associated with antidepressant. However, only few reported cases are related to bupropion. As reported herein, the patient with major depression suffered from acute dystonia twice, which resulted from abrupt bupropion discontinuation. The first episode occurred when medicament was shifted from bupropion to duloxetine abruptly. The patient was requested with nothing per mouth (NPO) due to panendoscopic examination. Therefore, the second one emerged after the patient was s

Exposure to high doses of organophosphorus nerve agents such as soman, even with carbamate pretreatment, produces a variety of toxic cholinergic signs, including secretions, convulsions and death. Evidence suggests that soman-induced convulsions may be associated with postexposure brain neuropathology. The purpose of this study was to investigate the pharmacologic mechanism of action of soman-induced convulsions and of anticonvulsant drugs. Various classes of compounds were evaluated for their efficacy in preventing soman-induced convulsions in rats pretreated with the oxime HI-6 to increase s

A case report of parkinsonism secondary to chronic abuse of amineptine (3 gr/day) and bromazepam (35 mg/day) in a patient diagnosed of borderline personality disorder is presented. The patient did not take any other drugs and he was not recently on neuroleptic treatment; he recognized the abuse of amineptine, as a stimulant, and the bromazepam abuse, looking for a relief to the excessive anxiety secondary to amineptine. The parkinsonism improved after removing both drugs and taking biperiden and diazepam; lastly the patient was discharged without any medication. The patient did not suffer from

A suddenly occurring episode characterized by perceptual alteration (SEPA), mainly of visual and/or auditory modalities, which repeatedly occurred in three schizophrenic patients on long-term neuroleptic medication, is described. Perceptual alteration showed some distinct features that were different from acute symptoms of schizophrenia, and was accompanied by mood changes such as severe anxiety and agitation and, in one of the patients, also by extrapyramidal symptoms. Perceptual alteration, as well as mood changes and extrapyramidal symptoms, responded well to an anticholinergic drug, biperi

OBJECTIVE: To report a case of an acute dystonic episode in a patient with Tourette's disorder (TD) treated with the partial dopamine agonist aripiprazole.
CASE SUMMARY: An 18-year-old male with TD was prescribed aripiprazole 10 mg orally daily, which produced a significant improvement in his symptoms. However, after 3 days of treatment, he experienced an acute episode of dystonia with facial muscle spasm, oculogyric crisis, and torticolis. All symptoms resolved after a single intramuscular injection of biperidine 5 mg. The Naranjo probability scale indicated that the adverse events were prob

Data is presented to support a physiologic vasoconstrictor function of the parasympathetic nervous system in the anterior uvea of the eye. Studies in rabbits, cats and monkeys demonstrate that stimulation of the parasympathetic nerve fibers running with the oculomotor nerve to the eye induces a reduction in blood flow of the iris and in rabbits in the ciliary processes in addition. Pharmacologic blocking experiments indicate that most of this phenomenon is based on a cholinergic component. Since at least in rabbits mechanical factors such as stretching of the iridial tissue, a steeling phenome

Dystonic reactions to neuroleptic and antiemetic medications are commonly seen in the outpatient setting. Despite initially successful treatment in the emergency department, symptoms may later recur. We have recently seen several cases of recurrent dystonic reactions; four representative cases are reported. Three of the patients experienced no further dystonic reactions when treated appropriately as outpatients. The fourth patient was lost to follow up. Based on this experience in the pharmacology of the drugs involved in producing and treating dystonic reactions, it is recommended that patien

A 42-year old-man was found lying in his bed having seizures. Later he became unconscious and hypotonic developing mydriasis as well as rigidity. The body core temperature (rectal temperature) was above 42 °C. Blood pH was decreased during treatment, and his general condition deteriorated. The patient developed gasping respiration, ventricular fibrillation, and died. During autopsy and histological investigation cerebral and pulmonary edema were noted together with general congestion of the internal organs. Further observations included contraction bands of myocytes, a contracted spleen, fibro
