Effect of infectious bursal disease (IBD) vaccine on Salmonella Enteritidis infected chickens.

BACKGROUND: Chickens infected with both infectious bursal disease virus (IBDV) and Salmonella had higher mortality. In this work, we investigated the effect of IBDV vaccine (modified live-virus bursal disease vaccine, Nobilis strain 228E®) on experimentally infected chickens with Salmonella Enteritidis (SE). METHODS: Four experimental groups were included in this study, negative control group, 228E®group, 228E®+SE infected group, and SE infected group. Chickens were ocularly administrated 228E® at 12days of age and orally infected with S. Enteritidis at 13days of age. Sera, intestinal fluid,

Efficacy of HVT-IBD vector vaccine compared to attenuated live vaccine using in-ovo vaccination against a Korean very virulent IBDV in commercial broiler chickens.

The production performance, efficacy, and safety of two types of vaccines for infectious bursal disease virus (IBDV) were compared with in-ovo vaccination of Cobb 500 broiler chickens for gross and microscopic examination of the bursa of Fabricius, bursa/body weight (b/B) ratio, flow cytometry, and serologic response to Newcastle disease virus (NDV) vaccination. One vaccine was a recombinant HVT-IBD vector vaccine (HVT as for herpesvirus of turkeys) and the other was an intermediate plus live IBDV vaccine. A significant difference was detected at 21 d. Eight of 10 chickens that received the IB

Mice as potential carriers of infectious bursal disease virus in chickens.

To investigate the role of mice as potential carriers of infectious bursal disease virus (IBDV), three mice were inoculated with a very virulent strain of IBDV and allowed to have contact with three uninoculated mice. Faeces, intestine and pooled liver and spleen collected from inoculated mice 12 and 24 h post-inoculation were positive for IBDV by reverse transcriptase-polymerase chain reaction (PCR)-nested PCR (RT-PCR-nPCR). IBDV was detected by RT-PCR-nPCR in 3/3 samples of intestine and 2/3 samples of pooled liver and spleen from uninoculated in-contact mice at 24 h after exposure. Chickens

Evaluating how infectious bursal disease virus influences influenza A virus transmission in chickens.

Infectious bursal disease virus (IBDV) is a major cause of immunosuppression in chickens that has been shown to affect influenza A virus (avian influenza virus (AIV)) pathogenicity and shedding patterns. In this study, we compared the shedding of a low passage duck origin AIV in two groups of specific- pathogen free chickens: one group that was IBDV-inoculated and one sham-inoculated group. At 3 weeks of age, the birds were divided and intranasally given one of three doses (102, 103, 105) of A/mallard/OH/421/1987 (H7N8) low pathogenicity AIV. Although the bursae of the IBDV-infected chicks had

Pathogenesis and Host Response to A3B4 and A3B2 Infectious Bursal Disease Virus Strains in Chickens.

Recently, the identification of novel reassortants of infectious bursal disease (IBD) virus (IBDV) was reported. Interestingly, these strains competed with the previously dominant A3B2 lineage, becoming the prevalent lineage in Poland and disseminating to other countries in Europe. However, the specific virulence properties that facilitate their spreading were not elucidated. In this study, we compared the pathogenesis of the A3B4 Bug/03 and A3B2 75/11 IBDV strains in specific-pathogen-free (SPF) chickens, integrating viral replication dynamics, tissue tropism, immunosuppressive effects and ho

Infectious bursal disease virus induces GSDME-dependent pyroptosis via the mitochondrial caspase-9/3 pathway in DF-1 cells.

Infectious bursal disease virus (IBDV) is an important avian pathogen that damages the bursa of Fabricius and continues to threaten the poultry industry. Pyroptosis is an inflammatory form of programmed cell death, but its contribution to IBDV-induced cellular injury remains incompletely defined. Here, using DF-1 cells, we found that IBDV infection was associated with caspase-3 activation, cleavage of gasdermin E (GSDME), generation of the pore-forming N-terminal fragment (GSDME-N), increased lactate dehydrogenase release, plasma membrane damage, and enhanced inflammatory cytokine-associated r

Infection Dynamics and Host Responses to Two IPNV Isolates in Liver of Atlantic Salmon (Salmo salar).

The infectious pancreatic necrosis virus (IPNV) used to be one of the largest loss factors in Atlantic salmon farming. Since 2009, marker-assisted selection for resistance to IPN, targeting a single major quantitative trait locus (QTL), has led to a ten-fold decrease in the number of IPN outbreaks in Norway. However, some IPN-related problems remain, due to isolates of the virus which seem to bypass the resistance mechanism of the QTL. We comparatively characterized a classical isolate affected by the IPN-QTL (cIPNV) and an isolate that circumvents the QTL-based protection (rIPNV). Using both

Mosquito-infecting virus Espirito Santo virus inhibits replication and spread of dengue virus.

The primary vector of dengue virus (DENV) is Aedes aegypti. The mosquito-infecting virus, Espirito Santo virus (ESV), does not infect Vero (mammalian) cells and grows in mosquito (C6/36) cells without cytopathic effects. Effects of ESV infection on replication of DENV were explored in vitro and in vivo, analyzing protein, RNA genome expression, and plaque formation. ESV and DENV simultaneous coinfection did not block protein synthesis from either virus but did result in inhibition of DENV replication in mosquito cells. Furthermore, ESV superinfected with DENV resulted in inhibition of DENV rep

Rapid Generation of Attenuated Infectious Bursal Disease Virus from Dual-Promoter Plasmids by Reduction of Viral Ribonucleoprotein Activity.

Infectious bursal disease virus (IBDV) of the Birnaviridae family leads to immunosuppression of young chickens by destroying B cells in the bursa of Fabricius (BFs). Given the increasing number of variant IBDV strains, we urgently require a method to produce attenuated virus for vaccine development. To accomplish this goal, the dual-promoter plasmids in which the RNA polymerase II and RNA polymerase I (Pol I) promoters were placed upstream of the IBDV genomic sequence, which was followed by mouse Pol I terminator and a synthetic polyadenylation signal, were developed for rapid generation of IB

A new recombinant avian metapneumovirus vaccine candidate provides complete protection against the novel variant infectious bursal disease virus in chickens.

Infectious bursal disease (IBD) is an acute, highly contagious, and immunosuppressive condition in chickens, caused by the infectious bursal disease virus (IBDV). The recent emergence of novel variant IBDV (nVarIBDV) poses a significant threat to the global poultry industry. However, currently available vaccines provide only limited protection against nVarIBDV strains. In this study, a recombinant avian metapneumovirus subtype B (aMPV/B) expressing the nVarIBDV VP2 gene (rLN16A-nVarVP2) was successfully rescued by inserting the gene between the G and L genes of the attenuated aMPV/B strain LN1

Comparative pathogenicity of very virulent and novel variant infectious bursal disease virus Egyptian strains in chickens.

Infectious bursal disease (IBD) is an important viral disease that causing severe immunosuppression in young chickens. This current study compared the pathogenicity of very virulent IBDV (vvIBDV) and Chinese novel variant IBDV (nVarIBDV) strains in Specific Pathogen-Free (SPF) chickens. Chickens infected with the nVarIBDV strain exhibited no clinical signs or mortality, with mild edema and swelling in bursas during early infection, followed by progressive atrophy by the seventh- and fifteenth-days post-infection (dpi). In contrast, vvIBDV displayed severe systemic disease, with early onset ruf

Preparation and immunogenicity studies of NvIBDV VP2-ferritin nanoparticles.

BACKGROUND: Infectious bursal disease (IBD), caused by infectious bursal disease virus (IBDV), is a highly contagious disease that is prevalent worldwide and poses a significant threat to the poultry industry. While commercially available vaccines are used for prevention, IBD outbreaks remain frequent. OBJECTIVE: The continuous mutation of virulent strains and their ability to evade traditional vaccine protection complicate IBD control, which necessitates the development of novel vaccines and a deeper understanding of viral mutation mechanisms. METHOD: Utilizing the self-assembly capability

Very virulent infectious bursal disease virus infection triggered microscopic changes, apoptosis, and inflammatory cytokines imbalance in chicken spleen and thymus.

Infectious bursal disease virus (IBDV) can cause a highly contagious disease, resulting in severe damage to the immune system that causes immunosuppression in young chickens. Both spleen and thymus are important immune organs, which play a key role in eliciting protective immune responses. However, the effects of very virulent IBDV (vvIBDV) strain LJ-5 infection on chicken spleen and thymus are still unknown. In the present study, 3-week-old specific pathogen-free chickens were infected with vvIBDV for 1-5 days. The vvIBDV infection significantly increased the spleen index and decreased the th

Live IBD vaccine exacerbates disease and pathological effects of Asian lineage H9N2 LPAIV in chickens.

Avian influenza H9N2 is one of the most commonly circulating viruses in numerous Egyptian poultry farms. The Asian lineage H9N2 exhibits an immunosuppressive effect, and its pathogenicity is amplified when it co-infects with other pathogens, especially with the immunosuppressive infectious bursal disease virus (IBDV), resulting in increased mortality rates. Both vaccines and field infection can exacerbate the pathogenicity of H9N2, particularly in the bursa of Fabricius, causing more significant lymphoid depletion. To comprehend the impact of the IBD vaccine on the viral and pathogenic effect

Infectious bursal disease virus tissue tropism and pathogenesis of the infection in chickens by application of in situ PCR, immunoperoxase and HE staining.

Infectious bursal disease is one of an OIE list of notifiable diseases. Chicken is the only host that manifests clinical signs and its pathogenicity is correlated with the distribution of antigens in organs. This study was conducted to determine disease pathogenesis and virus tissue tropism by in situ PCR, immunoperoxidase staining (IPS), and HE staining. Twenty four chickens were infected with very virulent Infectious Bursal Disease Virus (vvIBDV). Fifteen chickens were kept as a control group. Infected chickens were sacrificed at hrs 2, 4, 6, 12, days 1, 2, 4, and 6 post-inoculation (pi). Wh

A recombinant avian antibody against VP2 of infectious bursal disease virus protects chicken from viral infection.

A stable cell-line was established that expressed the recombinant avian antibody (rAb) against the infectious bursal disease virus (IBDV). rAb exhibited neutralization activity to IBDV-B87 strain in DF1 cells. The minimum rAb concentration required for inhibition of the cytopathic effect (CPE) was 1.563μg/mL. To test the efficacy of rAb, a 168-h cohabitation challenge experiment was performed to transmit the disease from the chickens challenged with vvIBDV (HLJ0504 strain) to three test groups of chickens, i.e. (1) chickens treated with rAb, (2) chickens treated with yolk antibody, and (3) non

Effects of cortisol on the intestinal mucosal immune response during cohabitant challenge with IPNV in Atlantic salmon (Salmo salar).

Infectious pancreatic necrosis virus (IPNV) causes high incidence of disease in salmonids during the first period after SW transfer. During this period as well as during periods of stress, cortisol levels increase and indications of a relationship between IPNV susceptibility and cortisol have been suggested. The intestine is an entry route and a target tissue for IPNV displaying severe enteritis and sloughing of the mucosa in infected fish. The mechanisms behind effects of the virus on the intestinal tissue and the impact of cortisol on the effect remain unclear. In the present study, Atlantic

Flow cytometry assay for intracellular detection of Infectious Pancreatic Necrosis virus (IPNV) in Atlantic salmon (Salmo salar L.) leucocytes.

Infectious Pancreatic Necrosis virus (IPNV) is traditionally detected in adherent leucocytes using immunofluorescence labelled specific antibodies, PCR or by further cultivation of infected cell material in cell lines. We present a flow cytometry (FCM) assay for detection of intracellular IPNV in salmon leucocytes, where each single cell is analysed for presence of virus. The method is established using in vitro challenge of salmon leucocytes and CHSE-214 cells. For detection of intracellular virus antigen the Cytofix/Cytoperm kit from BD is optimal compared with paraformaldehyde or acetone/me

Temporal modulation of proteasome and ubiquitin-mediated responses in rainbow trout kidneys during infectious pancreatic necrosis virus infection.

Infectious pancreatic necrosis virus (IPNV) poses a significant economic threat to aquaculture, yet understanding of the host-virus interaction remains limited. This study investigates the impact of IPNV on proteasomal response within rainbow trout kidneys during both subclinical (5 days post-infection, dpi) and clinical (10 dpi) stages of infection. Our results reveal a biphasic response: an initial stimulation of proteasomal activity at 5 dpi followed by inhibition at 10 dpi. Transcript analysis of proteasomal subunits in individual fish shows predominantly a correlation between their expres
