Placental DNA methylation at thyroid hormone-related genes mediates the association between prenatal exposure to bisphenol analogues and increased offspring obesity risk.

Prenatal exposure to bisphenol analogues (BPs) has been suggested to impair fetal growth and increase obesity risk in future life. However, the mechanisms remain unclear. Based on the Shanghai-Minhang Birth Cohort Study, this study aimed to investigate the associations between prenatal exposure to BPs and placental DNA methylation (DNAm) at thyroid hormone (TH)-related genes and explore whether these epigenetic modifications mediated the effects of BPs on offspring adiposity. The participants were recruited between April and December 2012 from the Minhang Maternal and Child Health Hospital in

Bisphenol S toxicity in zebrafish: Endocrine, neurobehavioural, and metabolic disruptions.

Bisphenol S (BPS), increasingly used as a substitute for Bisphenol A, has raised toxicological concern due to its environmental persistence and widespread exposure. Zebrafish (Danio rerio) provides a valuable model for assessing BPS toxicity because of conserved endocrine and neural pathways, rapid development, and well-established behavioural assays. This review synthesizes current evidence on the effects of BPS exposure in zebrafish. Available studies indicate that BPS is associated with alterations in multiple endocrine axes, including the hypothalamic-pituitary-gonadal, thyroid, and interr

Bisphenol S and female reproductive toxicity: a scoping review of human studies.

BACKGROUND: Bisphenol S (BPS) is a chemical analogue of Bisphenol A (BPA) used in the production of hard plastics, textiles, and thermal papers. As the use of BPA has declined, BPS human exposure has increased and is now widespread. BPS has endocrine-disrupting properties and growing evidence indicates BPS negatively impacts female reproductive health. OBJECTIVE: We conducted a scoping review of primary research articles related to BPS exposure and female reproductive toxicity in humans. METHODS: We searched five biomedical databases through January 2025 for studies that assessed the effects

Simplified QuEChERS-based UPLC-MS/MS for determination of neonicotinoids, bisphenols, and parabens in fresh and processed meat.

This study developed and validated a simplified QuEChERS-based isotope dilution ultra-performance liquid chromatography-tandem mass spectrometry (ID-UPLC-MS/MS) method for the simultaneous quantification of neonicotinoids (NEOs), parabens (PBs), and bisphenols (BPs) in meat products. Twenty-seven fresh and thirty processed meat samples collected in Taiwan were analyzed by the validated ID-UPLC-MS/MS method with isotope dilution to determine seven NEOs, nine PBs, and five BPs, achieving low limits of detection (0.00015-0.194 ng/g ww) and acceptable recoveries (83-105 %). Because of high sensiti

Prenatal and childhood exposure to bisphenols and bone mineral density in 7-year-old children from the Odense Child Cohort.

BACKGROUND: Bisphenol A (BPA) is a well-known endocrine disrupter used in several consumer products. Restricted use of BPA has led to increased use of bisphenol F (BPF) and bisphenol S (BPS). While previous studies found no associations between prenatal BPA and BPF exposure and bone mineral density (BMD), two recent cohort studies found that prenatal BPS exposure was negatively associated with bone mineral density in the offspring. AIM: To determine possible associations between maternal and child urinary bisphenol concentrations, BMD and bone mineral content (BMC) in 7-year-old healthy child

Chemical Identity and Mechanism of Action and Formation of a Cell Growth Inhibitory Compound from Polycarbonate Flasks.

This paper reports the chemical identity and mechanism of action and formation of a cell growth inhibitory compound leached from some single-use Erlenmeyer polycarbonate shaker flasks under routine cell culture conditions. Single-use cell culture vessels have been increasingly used for the production of biopharmaceuticals; however, they often suffer from issues associated with leachables that may interfere with cell growth and protein stability. Here, high-performance liquid-chromatography preparations and cell proliferation assays led to identification of a compound from the water extracts of

Perinatal bisphenols combined exposure caused working memory impairment by disturbing the ventral hippocampus-medial prefrontal cortex circuit.

Bisphenols (BPs), ubiquitous in food packaging, medical, and personal care products, are pervasive environmental contaminants. As bisphenol A (BPA) is increasingly replaced by its alternatives, the risk of mixed exposure and its impact on critical cognitive functions, such as working memory, remains a pressing but understudied concern. Here, we modeled developmental exposure to BPA and a bisphenol mixture (BPmix, BPA: BPS: BPF = 9:0.5:0.5) by administering the compounds to dams via drinking water during gestation and lactation. Behavioral analyses revealed that BPA and BPmix exposure significa

Evaluation of thyroid-disrupting effects of bisphenol F and bisphenol S on zebrafish (Danio rerio) using anti-transthyretin monoclonal antibody-based immunoassays.

The thyroid disrupting chemicals (TDCs) have raised great concerns due to their adverse impacts on thyroid hormones (THs). In this study, we investigated the thyroid-disrupting effects of bisphenol F (BPF) and bisphenol S (BPS), two major BPA substitutes, on adult zebrafish (Danio rerio). Firstly, anti-transthyretin (TTR) monoclonal antibody (anti-TTR mAb) was prepared and used to establish an indirect ELISA, which had a working range of 15.6∼1000 ng/mL of a detection limit of 6.1 ng/mL. The immunoassays based on anti-TTR mAb showed that exposure to BPF (10 and 100 μg/L) and BPS (100 μg/L) sig

Melatonin and Vitamins as Protectors against the Reproductive Toxicity of Bisphenols: Which Is the Most Effective? A Systematic Review and Meta-Analysis.

Bisphenols such as bisphenol A (BPA), S (BPS), C (BPC), F (BPF), AF (BPAF), tetrabromobisphenol, nonylphenol, and octylphenol are plasticizers used worldwide to manufacture daily-use articles. Exposure to these compounds is related to many pathologies of public health importance, such as infertility. Using a protector compound against the reproductive toxicological effects of bisphenols is of scientific interest. Melatonin and vitamins have been tested, but the results are not conclusive. To this end, this systematic review and meta-analysis compared the response of reproductive variables to m

Paternal BPA and BPS exposure induce testicular dysfunction in pubertal male offspring: roles of OCTN2 carnitine transporter disruption and exacerbated oxidative stress/apoptosis.

Although numerous studies have reported the male reproductive toxicity of bisphenol A (BPA) and its substitute bisphenol S (BPS), the impacts of paternal exposure to these chemicals on the male reproductive system of offspring and the underlying molecular mechanism remain inadequately explored. In this study, we investigated male reproductive toxicity in pubertal offspring resulting from paternal exposure to environmentally relevant doses of BPA (0.45 μg/kg body weight [bw]/day) or BPS (0.15 μg/kg bw/day). Our results showed that paternal exposure to BPA or BPS reduced pubertal offspring testo

Effects of bisphenol A and bisphenol S on human fallopian tube contractions: An in vitro and in silico study.

Bisphenol A (BPA), a widely used industrial compound, and its structural analogue Bisphenol S (BPS) are known to exert reproductive toxicity. However, their direct impact on human fallopian tube contractility remains unexplored. This study aimed to investigate the effects of BPA and BPS on spontaneous smooth muscle contractions of the human fallopian tube. Fallopian tube samples from the proliferative phase were used for in vitro contractility assays. The effects of BPA and BPS (1-20 µM) on maximum contractile strength (MCS), basal tone (BT), and contraction frequency (CF) were recorded and an

Assessment of the exposure of Spanish children and adolescents to bisphenols and parabens using dietary and human biomonitoring data.

Bisphenols A and S (BPA, BPS) and methylparaben (MetPB) are considered endocrine disrupting chemicals (EDCs). The development of approaches for more realistic estimation of exposure to these EDCs is crucial for understanding the extent of the actual burden in the population. Dietary exposure to BPA, BPS, and MetPB in Spanish children (3-12 years old) was estimated using 3 different approaches. The 24-h dietary recall (R24h) and food frequency questionnaire (FFQ) were used, together with reverse dosimetry applied to human biomonitoring (HBM) data to calculate daily intake (DI). Based on R24h an

Multidimensional mechanistic analysis elucidates spermatogenesis disruption induced by bisphenol S.

Bisphenol S (BPS), a primary substitute for bisphenol A, has led to widespread environmental exposure globally. However, the effects of BPS on spermatogenesis disruption (SD) remain contentious, and the underlying mechanisms responsible for SD induced by BPS are not fully understood. In this study, we employed a multidimensional computational strategy, integrating network toxicology, bioinformatics, systematic machine learning, single-cell analysis, and biomolecular modeling, to comprehensively elucidate the molecular mechanisms by which BPS induces SD. Our findings suggest that BPS may disrup

Thyroid Disruption by Bisphenol S Analogues via Thyroid Hormone Receptor β: in Vitro, in Vivo, and Molecular Dynamics Simulation Study.

Bisphenol S (4-hydroxyphenyl sulfone, BPS) is increasingly used as a bisphenol A (BPA) alternative. The global usage of BPS and its analogues (BPSs) resulted in the frequent detection of their residues in multiple environmental media. We investigated their potential endocrine-disrupting effects toward thyroid hormone receptor (TR) β. The molecular interaction of BPSs toward TRβ ligand binding domain (LBD) was probed by fluorescence spectroscopy and molecular dynamics (MD) simulations. BPSs caused the static fluorescence quenching of TRβ LBD. The 100 ns MD simulations revealed that the binding
