Bone-Seeking Matrix Metalloproteinase Inhibitors for the Treatment of Skeletal Malignancy.

Matrix metalloproteinases (MMPs) are a family of enzymes involved at different stages of cancer progression and metastasis. We previously identified a novel class of bisphosphonic inhibitors, selective for MMPs crucial for bone remodeling, such as MMP-2. Due to the increasing relevance of specific MMPs at various stages of tumor malignancy, we focused on improving potency towards certain isoforms. Here, we tackled MMP-9 because of its confirmed role in tumor invasion, metastasis, angiogenesis, and immuno-response, making it an ideal target for cancer therapy. Using a computational analysis, we

The contribution of the pericanalicular matrix to mineral content in human osteonal bone.

The osteocyte lacunar-canalicular network (LCN) penetrates bone and houses the osteocytes and their processes. Despite its rather low volume fraction, the LCN represents an outstanding large surface that is possibly used by the osteocytes to interact with the surrounding mineralized bone matrix thereby contributing to mineral homeostasis. The aim of this study was to quantitatively describe such contributions by spatially correlating the local density of the LCN with the mineral content at the same location in micrometer-sized volume elements in human osteons. For this purpose, 65 osteons from

Ethanol and formaldehyde fixation irreversibly alter bones' organic matrix.

INTRODUCTION: Biomechanical tests on bones are frequently accomplished in anatomically fixed tissues. The effects of ethanol or formaldehyde based fixation in bone material properties are subject to controversial discussions, regarding their appropriateness and usability to answer clinical questioning or biomechanical issues. We hypothesized that ethanol and formaldehyde irreversibly change bone material properties, and that this effect is mainly related to the bone's organic matrix. MATERIAL AND METHODS: Fixation related alterations in material properties were investigated in six fresh and t

An evaluation of bone healing in cavities in the jaws implanted with a collagen matrix.

Bony defects in the jaws after surgery may show poor healing especially in the maxilla and the area may never completely be replaced by bone. 50 patients with pathological bony cavities in the jaws were used to assess the efficacy of a collagen matrix implanted into the treated bony defect and 50 patients in whom no material was implanted were used as controls. The results were assessed radiographically at 3, 6 and 12 months. At 3 months, bone deposition in the control group was in advance of the implanted group, but after 6 and 12 months healing appeared to be similar in both groups. No adver

Impacts of Nonenzymatic Glycation Crosslinks in Bone Matrix on the Mechanosensitivity of Osteocyte.

The products of nonenzymatic glycation (NEG) are a key contributor to various types of osteoporosis, as they affect both the physical properties of bone matrix and the function of osteoblasts and osteoclasts. However, their impact on the mechanosensitivity of osteocytes remains poorly understood. Osteocytes are embedded in lacunar-canalicular system (LCS), and their processes have abundant connections with canalicular matrix to amplify their membrane strain. Thus, we mainly studied the effects of NEG crosslinks in bone matrix on the connections between bone matrix and osteocyte processes, as w

A prospective evaluation of cellular bone matrix for posterolateral lumbar fusion.

INTRODUCTION: Autograft bone is the gold standard graft used in posterolateral lumbar intertransverse fusion (PLF) but can be limited by supply and donor site morbidity. Cadaveric cellular allografts represent an alternative source of graft material that avoids these limitations. OBJECTIVE: To assess the safety and efficacy of ViviGenRCellular Bone Matrix in comparison to autograft bone in PLF. METHODS: We performed a prospective study of 20 subjects who underwent PLF including 55 segmental levels. Each patient received ViviGen over the decorticated posterolateral gutter on one side an autog

Role of cartilage and bone matrix regulation in early equine osteochondrosis.

The objective of this study is to better understand the pathogenesis of early equine osteochondrosis (OC) by identifying differences in gene and protein expression of extracellular matrix components and regulators in normal and diseased cartilage and bone, focusing on the osteochondral junction and cells surrounding the cartilage canals. We expected to find an upregulation of matrix metalloproteinases and a decrease in extracellular matrix constituent expression along the osteochondral junction and cells surrounding the cartilage canals in OC samples. Paraffin-embedded osteochondral samples (6

Mineral tessellation in bone and the stenciling principle for extracellular matrix mineralization.

We review here the Stenciling Principle for extracellular matrix mineralization that describes a double-negative process (inhibition of inhibitors) that promotes mineralization in bone and other mineralized tissues, whereas the default condition of inhibition alone prevents mineralization elsewhere in soft connective tissues. The stenciling principle acts across multiple levels from the macroscale (skeleton/dentition vs soft connective tissues), to the microscale (for example, entheses, and the tooth attachment complex where the soft periodontal ligament is situated between mineralized tooth c

The Bone Extracellular Matrix as an Ideal Milieu for Cancer Cell Metastases.

Bone is a preferential site for cancer metastases, including multiple myeloma, prostate, and breast cancers. The composition of bone, especially the extracellular matrix (ECM), make it an attractive site for cancer cell colonization and survival. The bone ECM is composed of living cells embedded within a matrix composed of both organic and inorganic components. Among the organic components, type I collagen provides the tensile strength of bone. Inorganic components, including hydroxyapatite crystals, are an integral component of bone and provide bone with its rigidity. Under normal circumstance

Function of matrix IGF-1 in coupling bone resorption and formation.

Balancing bone resorption and formation is the quintessential component for the prevention of osteoporosis. Signals that determine the recruitment, replication, differentiation, function, and apoptosis of osteoblasts and osteoclasts direct bone remodeling and determine whether bone tissue is gained, lost, or balanced. Therefore, understanding the signaling pathways involved in the coupling process will help develop further targets for osteoporosis therapy, by blocking bone resorption or enhancing bone formation in a space- and time-dependent manner. Insulin-like growth factor type 1 (IGF-1) ha

Matrix metalloproteinases as master regulators of the vicious cycle of bone metastasis.

Bone remodeling is a delicate balancing act between the bone matrix synthesizing osteoblasts and bone resorbing osteoclasts. Active bone metastases typically subvert this process to generate lesions that are comprised of extensive areas of pathological osteogenesis and osteolysis. The resultant increase in bone matrix remodeling enhances cytokine/growth factor bioavailability thus creating a vicious cycle that stimulates tumor progression. Given the extent of matrix remodeling occurring in the tumor-bone microenvironment, the expression of matrix metalloproteinases (MMPs) would be expected, si

A comparison of commercially available demineralized bone matrix for spinal fusion.

In an effort to augment the available grafting material as well as to increase spinal fusion rates, the utilization of a demineralized bone matrix (DBM) as a graft extender or replacement is common. There are several commercially available DBM substances available for use in spinal surgery, each with different amounts of DBM containing osteoinductive proteins. Each product may have different osteoinductivity potential due to different methods of preparation, storage, and donor specifications. The purpose of this study is to prospectively compare the osteoinductive potential of three different

Low Bone Turnover and Decreased Bone Matrix Mineralization at Diagnosis in Children with Acute Lymphoblastic Leukemia.

In children with acute lymphoblastic leukemia (ALL), skeletal complications are well documented, but their exact origin remains unclear. In the present study we investigated bone mineralization density distribution by quantitative back-scattered electron imaging (qBEI) and bone histomorphometry by light microscopy in pediatric ALL patients at diagnosis, before any cancer treatment. In our one-center LELU study, we obtained bone marrow biopsies from the posterior iliac crest in 11 boys and 15 girls (median age 4.3; range 2.6–15.8 years) in connection with ALL diagnostic sampling. Blood samples

Bone organic matrix quality indices associate with fragility fracture occurrence independently of bone mineral density values.

UNLABELLED: Bone mineral density measures are not the sole determinant whether a patient suffers fragility fractures. We tested the hypothesis that changes in bone organic matrix quality indices strongly associate with Fx occurrence. Fx-suffering patients have altered quantitative and qualitative organic matrix attributes, offering new insights into the pathogenesis of Fx. Fragility fractures present a considerable socioeconomic burden. We tested the hypothesis that organic matrix attributes associate with fragility fracture occurrence independent of BMD. We used Raman microspectroscopy to ana

Photobiomodulation Therapy Improves Repair of Bone Defects Filled by Inorganic Bone Matrix and Fibrin Heterologous Biopolymer.

Biomaterials are used extensively in graft procedures to correct bone defects, interacting with the body without causing adverse reactions. The aim of this pre-clinical study was to analyze the effects of photobiomodulation therapy (PBM) with the use of a low-level laser in the repair process of bone defects filled with inorganic matrix (IM) associated with heterologous fibrin biopolymer (FB). A circular osteotomy of 4 mm in the left tibia was performed in 30 Wistar male adult rats who were randomly divided into three groups: G1 = IM + PBM, G2 = IM + FB and G3 = IM + FB + PBM. PBM was applied

Overexpression of Dentin matrix protein 1 in Nestin+ cells causes bone loss in mouse long bone.

The well-known matrix protein Dentin matrix protein 1 (DMP1) is expressed by osteoblasts and osteocytes in bone, and it controls bone mineralization. Recently, it has been found that DMP1 is also expressed in other cell types, such as chondrocytes. Nestin+ cells are one important type of progenitor cell in bone marrow and are associated with bone remodeling. In our preliminary experiment, DMP1 could also be detected in Nestin+ cells in bone marrow. This study was designed to explore the effect on bone of DMP1 in Nestin+ cells. A transgenic mouse model with DMP1 expression driven by the Nestin

The Role of GPR39 in Regulating Osteoblast Function, Bone Matrix Quality, and Gender-Specific Bone Homeostasis.

GPR39, a zinc-sensing receptor, is essential for bone homeostasis in male mice through regulation of osteoblast function and matrix composition. This study examined the effects of GPR39 deficiency in female mice using both global and osteoblast lineage-specific GPR39 knockout models (Gpr39Ob-/Ob-). In vivo, GPR39-deficient female mice exhibited reduced bone mass, increased mineralization rates, and significantly lower and more variable serum levels of pro-collagen type I N-propeptide (PINP), indicating impaired collagen synthesis and matrix remodeling. OVX models further demonstrated that GPR3

Exchange hybrid cranioplasty using particulate bone graft and demineralized bone matrix: the best of both worlds.

Reconstruction of craniofacial defects in children presents several challenges that are not encountered in the adult population. Autologous bone grafts have long been the criterion standard for repairing these defects. Recently, several new materials and techniques have expanded our arsenal of reconstructive options. In this clinical report, we describe the use of both particulate bone grafting and demineralized bone matrix together to repair craniofacial defects encountered in pediatric patients.

Bone Matrix Levels of Dickkopf and Sclerostin are Positively Correlated with Bone Mass and Strength in Postmenopausal Osteoporosis.

Wnt signaling plays a pivotal role in maintaining bone mass. Secreted pathway modulators such as sclerostin (SOST) and Dickkopfs (DKKs) may influence bone mass inhibiting the canonical Wnt pathway. We evaluated whether bone protein content of secreted Wnt antagonists is related to age, bone mass, and strength in postmenopausal osteoporosis. We measured cortical and trabecular bone contents of SOST and Dickkopf-1 (DKK1) in combined extracts obtained after ethylenediaminetetraacetic acid and guanidine hydrochloride extraction in 56 postmenopausal women aged 47-74 (mean, 63) yr with a previous di
