The plant defense hormone, salicylic acid (SA), plays essential roles in immunity and systemic acquired resistance. Salicylic acid induced by the pathogen is perceived by the receptor nonexpressor of pathogenesis-related genes 1 (NPR1), which is recruited by TGA transcription factors to induce the expression of pathogenesis-related (PR) genes. However, the mechanism by which post-translational modifications affect TGA's transcriptional activity by salicylic acid signaling/pathogen infection is not well-established. Here, we report that the loss-of-function mutant of brassinosteroid insensitive

Excessive exposure to high light can lead to photoinhibition, which impairs photosynthetic efficiency and causes oxidative damage in plants, such as sunburn in grapevines. This study investigates the role of resveratrol (Res), a stilbenoid with antioxidant properties, in protecting plants from high light damage. We found that exposure to high light increased reactive oxygen species (ROS) accumulation and induced photoinhibition in grapevine leaves. In response, Res biosynthesis was upregulated, along with an increase in stilbene synthase (VvSTS) expression. Application of exogenous Res allevia

Pathogen-associated molecular pattern (PAMP)-trigged immunity (PTI) is the first defensive line of plant innate immunity and is mediated by pattern recognition receptors. Here, we show that a mutation in BR-SIGNALING KINASE1 (BSK1), a substrate of the brassinosteroid (BR) receptor BRASSINOSTEROID INSENSITIVE1, suppressed the powdery mildew resistance caused by a mutation in ENHANCED DISEASE RESISTANCE2, which negatively regulates powdery mildew resistance and programmed cell death, in Arabidopsis thaliana. A loss-of-function bsk1 mutant displayed enhanced susceptibility to virulent and avirule

Multicellularity arose independently in plants and animals, but invariably requires a robust determination and maintenance of cell fate that is adaptive to the environment. This is exemplified by the highly specialized water- and nutrient-conducting cells of the plant vasculature, the organization of which is already prepatterned close to the stem-cell niche, but can be modified according to extrinsic cues. Here, we show that the hormone receptor BRASSINOSTEROID INSENSITIVE 1 (BRI1) is required for root vascular cell-fate maintenance, as BRI1 mutants show ectopic xylem in procambial position.

Vernalization, a low-temperature-induced transition from vegetative to reproductive growth, is essential for flowering in many Brassica crops, including Wucai (Brassica rapa ssp. chinensis var. rosularis). To elucidate the molecular basis of vernalization in Wucai, we performed an integrative multi-omics analysis of shoot apices from cold-treated plants. Twenty-eight vernalization pathway-related genes, including homologs of FLC, FLM, VIN3, VRN, SOC1, SVP, and VIL2, exhibited conserved cold-responsive expression patterns. Transcription factors from the MYB, bHLH, bZIP, and NAC families showed

Grapevine downy mildew, caused by Plasmopara viticola, poses significant threats to viticulture, necessitating sustainable alternatives to chemical fungicides. This study investigates the efficacy of exogenous 24-epibrassinolide (EBR), a brassinosteroid, in enhancing disease resistance across two grape cultivars: susceptible Vitis vinifera Cabernet Sauvignon and resistant Ecolly. EBR application reduced P. viticola colonization in both cultivars, suppressing hyphal growth and sporulation. In Cabernet Sauvignon, EBR delayed pathogen establishment by modulating stomatal closure and boosting anti

Plant architecture strongly influences planting density, canopy light interception, and mechanical harvesting efficiency in upland cotton (Gossypium hirsutum L.). Fruiting branch angle (FBA) is an important architectural trait, but its molecular regulation remains poorly understood. Here, we compared the MAPK/ERK kinase kinase (MEKK) subfamily across four upland cotton reference genomes and identified 44 MEKK members in each genome. Genomic positions and coding sequences were broadly conserved, whereas motif analysis distinguished 26 motif-conserved from 18 motif-variable homologous groups. In

The genus Renanthera is valued for its fiery-red floral coloration and distinctive spider-like floral architecture; however, the molecular mechanisms underlying floral organ specialization and perianth color differentiation remain poorly understood. In this study, using integrated morphological, transcriptomic, and metabolomic analyses of R. coccinea, we found that the cell types of the dorsal and lateral perianth organs were broadly similar but clearly distinct from those of the lip and the column. Developmentally, the organ-specific expression patterns of AGAMOUS-LIKE6-1/2 (AGL6-1/2), APETAL

Sterols are essential components of eukaryotic cell membranes. However, studies on sterol biosynthesis in bryophytes are limited. This study analyzed the sterol profiles in the bryophyte model plant Marchantia polymorpha L. The thalli contained typical phytosterols such as campesterol, sitosterol and stigmasterol. BLASTX analysis of the M. polymorpha genome against the Arabidopsis thaliana sterol biosynthetic genes confirmed the presence of all the enzymes responsible for sterol biosynthesis in M. polymorpha. We further focused on characterizing two genes, MpDWF5A and MpDWF5B, which showed hig

Plant height and grain size are two important agronomic traits that are closely related to crop yield. Numerous dwarf and grain-shape mutants have been studied to identify genes that can be used to increase crop yield and improve breeding programs. In this study, we characterized a dominant mutant, dwarf and round grain 1 (drg1-D), in bread wheat (Triticum aestivum L.). drg1-D plants exhibit multiple phenotypic changes, including dwarfism, round grains, and insensitivity to brassinosteroids (BR). Cell structure observation in drg1-D mutant plants showed that the reduced organ size is due to ir

Hormones are fundamental architects of plant reproduction, orchestrating the transition from pollination to fruit set. Recent advances have underscored the central roles of auxin and gibberellins in ovary growth, seed development, and parthenocarpy, while their intricate interplay with cytokinin, ethylene, abscisic acid, brassinosteroids, jasmonic acid, and salicylic acid fine-tunes early fruit development. A dynamic regulatory network involving transcription factors, miRNAs, and hormone-responsive genes modulates these processes, ensuring coordinated cellular events across diverse fruit types

Brassinosteroid signaling kinases (BSKs) act as core signal transducers downstream of Brassinosteroid (BR) perception and integrate plant growth regulation with broad-spectrum biotic and abiotic stress tolerance. Despite well-established functional characterizations of BSK gene families in Arabidopsis thaliana and rice, comprehensive genome-wide profiling and salt response analysis of BSK homologs remain lacking in wheat. In this study, we systematically identified 18 TaBSK family members. Phylogenetic analysis separated wheat TaBSKs into three distinct evolutionary subgroups. The 18 TaBSK loc

Phytohormones play crucial roles in regulating many aspects of plant development. Although much has been learned about the effects of individual hormones, cross-talk between and integration of different hormonal signals are still not well understood. We present a study of MINI ZINC FINGER 1 (MIF1), a putative zinc finger protein from Arabidopsis, and suggest that it may be involved in integrating signals from multiple hormones. MIF1 homologs are highly conserved among seed plants, each characterized by a very short sequence containing a central putative zinc finger domain. Constitutive overexp

Brassinosteroids (BR) are an essential steroid phytohormone found throughout the plant kingdom. Synthesis of BR occurs at the endoplasmic reticulum, and the mechanism of transport through the membrane to the apoplast is not well understood. The ABCB1 and ABCB19 transporters enhance BR transport through the plasma membrane, but the available cryo-EM structures do not provide a clear understanding of the steroid's diffusion process. Sphingolipid and sterol enriched domains improve the translocation of BR through localization of ABCB1 and ABCB19. The similarity of BR to sterols suggests that BR l

From seed dormancy to pathogen attacks and senescence, many events in a plant's life are critically controlled by low-molecular-weight signaling molecules, called plant hormones. These master regulators include abscisic acid, auxins, brassinosteroids, cytokinins, ethylene, gibberellins, jasmonates, salicylates, and strigolactones. Years of research have unveiled multiple aspects of their biosynthesis, transport, signaling, functions, and interactions. With this knowledge, complex molecular networks have been established and refined over time in an attempt to explain how plant hormones collecti

Endoplasmic reticulum-associated degradation (ERAD) is a key cellular process for degrading misfolded proteins. It was well known that an asparagine (N)-linked glycan containing a free α1,6-mannose residue is a critical ERAD signal created by Homologous to α-mannosidase 1 (Htm1) in yeast and ER-Degradation Enhancing α-Mannosidase-like proteins (EDEMs) in mammals. An earlier study suggested that two Arabidopsis homologs of Htm1/EDEMs function redundantly in generating such a conserved N-glycan signal. Here we report that the Arabidopsis irb1 (reversal of bri1) mutants accumulate brassinosteroid

Secondary growth of the woody tree stem is governed by meristematic cell division and differentiation in the vascular cambium. Multiple hormonal signals and endogenous developmental programs regulate vascular cambium activity. Brassinosteroids (BRs) significantly promote secondary stem growth and wood formation in poplar trees. However, the underlying regulatory mechanisms of BRs within the vascular tissue remain unclear. Genetic and anatomical approaches were used here to elucidate the role of PagDET2, the rate-limiting enzyme for BRs biosynthesis, in regulating secondary vascular cambium act

Since the discovery of auxins as cell division factors a century ago, impressive scientific advances related to phytohormones have revolutionized plant sciences and human progress. This review examines the key features of the ten hormonal groups that operate in plants, here referred to as the 'classical ten': auxins, gibberellins (GAs), cytokinins (CKs), abscisic acid (ABA), ethylene (ETH), salicylates (SAs), jasmonates (JAs), brassinosteroids (BRs), peptide hormones (PEPs), and strigolactones (SLs). By leveraging historical data and sharpening the essentials of each hormonal group, their majo

Phytohormones are physiologically important small molecules that play essential roles in intricate signaling networks that regulate diverse processes in plants. We present a method for the simultaneous targeted profiling of 101 phytohormone-related analytes from minute amounts of fresh plant material (less than 20 mg). Rapid and nonselective extraction, fast one-step sample purification, and extremely sensitive ultra-high-performance liquid chromatography-tandem mass spectrometry enable concurrent quantification of the main phytohormone classes: cytokinins, auxins, brassinosteroids, gibberelli

Due to climate change, drought-prone areas have increased, and soil salinity is progressively increasing, leading to a greater number of regions with highly saline soils. Salt stress represents one of the most detrimental factors in modern agriculture, prompting the scientific community to seek effective solutions. Among the proposed alternatives, the use of bioactive chemical compounds to enhance plant tolerance and counteract saline stress has emerged as a promising approach. Since salt stress tolerance could be conferred by increasing the endocytic trafficking, the use of the synthetic comp
