Dynamic genome-wide mapping reveals how chromatin context shapes OGG1-mediated repair and related mutagenesis in human cells.

Potassium bromate-induced DNA damage, including 8-oxo-7,8-dihydroguanine (OG) and 2,6-diamino-4-hydroxy-5-formamidopyrimidine (FapyG), are common oxidatively generated DNA lesions with mutagenic potential if not efficiently repaired. While sequencing-based studies have shown that damage formation is influenced by DNA sequence context, secondary structures, and chromatin features, how OGG1-mediated repair is regulated within chromatin remains unclear. Here, we apply CLAPS-seq to generate genome-wide, single-nucleotide resolution maps of OGG1-mediated repair over time in human cells, and systema

The advanced EctoSys electrolysis as an integral part of a ballast water treatment system.

A full-scale 500 m(3)/h ballast water treatment system was tested according to the landbased type approval procedure of the International Maritime Organization (IMO). The system consists of disc filters followed by the advanced EctoSys electrolysis as an integral part for disinfection. The test water quality exceeded by far the minimum requirements for type approval testing. Due to the properties of the special electrodes used together with the striking disinfection effect, the disinfectants assumed to be produced inline by the EctoSys cell in river water were hydroxyl radicals, while in brack

Mechanisms of chemically induced renal carcinogenesis in the laboratory rodent.

Laboratory studies with classical renal carcinogens in the rat and mouse, as well as research investigation with some of the chemicals proving positive for the kidney in National Toxicology Program carcinogenicity bioassays, have demonstrated the existence of a range of diverse mechanisms underlying rodent kidney carcinogenesis. The classical carcinogens used as experimental models for studying renal tumor pathogenesis, such as the nitrosamines, are genotoxic and interact directly with DNA, forming DNA adducts with mutagenic potential. In contrast, potassium bromate and ferric nitrilotriacetat

Quality Structures, Vibrational Frequencies, and Thermochemistry of the Products of Reaction of BrHg(•) with NO2, HO2, ClO, BrO, and IO.

Quantum chemical calculations have been carried out to investigate the structures, vibrational frequencies, and thermochemistry of the products of BrHg(•) reactions with atmospherically abundant radicals Y(•) (Y = NO2, HO2, ClO, BrO, or IO). The coupled cluster method with single and double excitations (CCSD), combined with relativistic effective core potentials, is used to determine the equilibrium geometries and harmonic vibrational frequencies of BrHgY species. The BrHg-Y bond energies are refined using CCSD with a noniterative estimate of the triple excitations (CCSD(T)) combined with core

Silver tungstate/magnesium oxide nanocomposite as a high-performance adsorbent for bromide removal from aqueous systems.

Bromide contamination in water sources poses significant health and environmental concerns due to its transformation into carcinogenic disinfection byproducts (DBPs) such as bromate during water treatment. In this study, a silver tungstate-modified magnesium oxide nanocomposite (Ag2WO4/MgO, denoted as AgW@MgO) was synthesized and applied as an efficient adsorbent for bromide removal from aqueous solutions. The material was thoroughly characterized using X-ray diffraction (XRD), scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), Fourier-transform infrared spectrosco

CuO Promotes the Formation of Halogenated Disinfection Byproducts during Chlorination via an Enhanced Oxidation Pathway.

Previous studies showed that cupric oxide (CuO) can enhance the formation of trihalomethanes (THMs), haloacetic acids, and bromate during chlorination of bromide-containing waters. In this study, the impact of CuO on the formation kinetics and mechanisms of halogenated disinfection byproducts (DBPs) during chlorination was investigated. CuO does not enhance the formation of DBPs (i.e., 1,1,1-trichloropropanone, chloroform, and trichloroacetaldehyde (TCAL) /dichloroacetonitrile) during chlorination of acetone, 3-oxopentanedioic acid (3-OPA), and aspartic acid, respectively. This indicates that

Nephrotoxicity and Renal Pathophysiology: A Contemporary Perspective.

The kidney consists of numerous cell types organized into the nephron, which is the basic functional unit of the kidney. Any stimuli that induce loss of these cells can induce kidney damage and renal failure. The cause of renal failure can be intrinsic or extrinsic. Extrinsic causes include cardiovascular disease, obesity, diabetes, sepsis, and lung and liver failure. Intrinsic causes include glomerular nephritis, polycystic kidney disease, renal fibrosis, tubular cell death, and stones. The kidney plays a prominent role in mediating the toxicity of numerous drugs, environmental pollutants and

The cross talk between pathways in the repair of 8-oxo-7,8-dihydroguanine in mouse and human cells.

Although oxidatively damaged DNA is repaired primarily via the base excision repair (BER) pathway, it is now evident that multiple subpathways are needed. Yet, their relative contributions and coordination are still unclear. Here, mouse embryo fibroblasts (MEFs) from selected nucleotide excision repair (NER) and/or BER mouse mutants with severe (Csb(m/m)/Xpa(-/-) and Csb(m/m)/Xpc(-/-)), mild (Csb(m/m)), or no progeria (Xpa(-/-), Xpc(-/-), Ogg1(-/-), Csb(m/m)/Ogg1(-/-)) or wild-type phenotype were exposed to an oxidizing agent, potassium bromate, and genomic 8-oxo-7,8-dihydroguanine (8-oxoGua)

A sensitive spectrophotometric method for the determination of propranolol HCl based on oxidation bromination reactions.

Three new, simple, sensitive, rapid and economical spectrophotometric methods (A, B and C) have been developed for the determination of propranolol hydrochloride (PRO) in bulk drug and dosage forms. These methods are based on oxidation-bromination reaction of PRO by bromine, generated in situ by the action of acid on a bromate-bromide mixture, followed by determination of unreacted bromine by three different reaction schemes. In method A, the determination of the residual bromine is based on its ability to bleach the indigo carmine dye and by measuring the absorbance at 610 nm. The residual br

Integration of hydrodynamic cavitation and preozonation: a physicochemical strategy for effectively addressing intracellular odorants from filamentous cyanobacteria.

This study introduces hydrodynamic cavitation-integrated preozonation (HCPO), a novel physicochemical strategy that synergistically combines hydrodynamic cavitation (HC) with preozonation to eliminate intracellular taste-and-odor compounds, like 2-methylisoborneol (2-MIB) and geosmin (GSM), from filamentous cyanobacteria. Unlike conventional peroxone preozonation, which fails to degrade cell-bound odorants and requires unstable hydrogen peroxide handling, HCPO employs a Venturi-based HC unit to physically enhance the disruption of cyanobacterial cells while upgrading ozone dispersion and radic

The carcinogenic response of Tsc2 mutant Long-Evans (Eker) rats to a mixture of drinking water disinfection by-products was less than additive.

Cancer risk assessment methods for chemical mixtures in drinking water are not well defined. Current default risk assessments for chemical mixtures assume additivity of carcinogenic effects, but this may not represent the actual biological response. A rodent model of hereditary renal cancer (Eker rat) was used to evaluate the carcinogenicity of mixtures of water disinfection by-products (DBPs). Male and female Eker rats were treated with individual DBPs or a mixture of DBPs for 4 or 10 months. Induction of poly(ADP-ribosyl)ation in the kidney after in vivo application of renal carcinogens.

Dichlorovinylcysteine, the key metabolite thought to be responsible for the nephrocarcinogenicity of trichloroethene and dichloroacetylene, induces DNA double-strand breaks followed by increased poly(ADP-ribosyl)ation of nuclear proteins in cultured renal cells (Vamvakas et al., 1992, Biochem. Pharmacol. 44, 1131-1138). Poly(ADP-ribosyl)ation represents a post-translational modification of nuclear proteins involved in DNA repair, DNA replication, and modulation of gene expression. The present study investigates the induction of DNA double-strand breaks and poly(ADP-ribosyl)ation in the renal c

Silver nanoparticles/activated carbon composite as a facile SERS substrate for highly sensitive detection of endogenous formaldehyde in human urine by catalytic reaction.

Most of efforts have been made to prepare high performance surface-enhanced Raman scattering (SERS) substrate for amplifying Raman signals. It still remains a grand challenging task in building a simple, conventional and low-cost SERS substrate with highly dense hotspots for improved sensitivity of the target analytes. Here, we report a very dexterous strategy to fabricate a distinctive SERS substrate with high density hotspots, using common adsorbent activated carbon (AC) as template to assemble silver nanoparticles (Ag NPs). It can be estimated that the enhancement effect of Ag NPs/AC compos

Reactivity of BrCl, Br₂, BrOCl, Br₂O, and HOBr toward dimethenamid in solutions of bromide + aqueous free chlorine.

HOBr, formed via oxidation of bromide by free available chlorine (FAC), is frequently assumed to be the sole species responsible for generating brominated disinfection byproducts (DBPs). Our studies reveal that BrCl, Br(2), BrOCl, and Br(2)O can also serve as brominating agents of the herbicide dimethenamid in solutions of bromide to which FAC was added. Conditions affecting bromine speciation (pH, total free bromine concentration ([HOBr](T)), [Cl(-)], and [FAC](o)) were systematically varied, and rates of dimethenamid bromination were measured. Reaction orders in [HOBr](T) ranged from 1.09 (±

Analysis of preneoplastic and neoplastic renal lesions in Tsc2 mutant Long-Evans (Eker) rats following exposure to a mixture of drinking water disinfection by-products.

Disinfection of surface water for human consumption results in the generation of a complex mixture of chemicals in potable water. Cancer risk assessment methodology assumes additivity of carcinogenic effects in the regulation of mixtures. A rodent model of hereditary renal cancer was used to investigate the carcinogenic response to a mixture of drinking water disinfection by-products (DBPs). Rats carrying a mutation in the Tsc2 tumor suppressor gene (Eker rats) readily develop renal preneoplastic and neoplastic lesions, and are highly susceptible to the effects of renal carcinogens. A hybrid fluidized-bed reactor (HFBR) based on arrayed ceramic membranes (ACMs) coupled with powdered activated carbon (PAC) for efficient catalytic ozonation: A comprehensive study on a pilot scale.

Taking advantage of the high mass transfer in the bulk solution of fluidized-bed reactor (FBR), and the benefits of simultaneous particle separation and ozone catalysis on ceramic membranes, we proposed a hybrid fluidized-bed reactor (HFBR) based on arrayed ceramic membranes (ACMs) coupled with powdered activated carbon (PAC) for efficient catalytic ozonation. The optimum HFBR performance on a pilot scale was found at PAC addition of 3 g/L, ozone dosage of 25 mg/L, hydraulic retention time of 60 min and auxiliary aeration strength of 5 m3/h. GW0742 activates peroxisome proliferator-activated receptor δ to reduce free radicals and alleviate cardiac hypertrophy induced by hyperglycemia in cultured H9c2 cells.

Peroxisome proliferator-activated receptor δ (PPARδ), the predominant PPAR subtype in the heart, is known to regulate cardiac function. PPARδ activation may inhibit cardiac hypertrophy in H9c2 cells while the potential mechanism has not been elucidated. Then, H9c2 cells incubated with high glucose to induce hypertrophy were used to investigate using GW0742 to activate PPARδ. The fluorescence assays were applied to determine the changes in cell size, cellular calcium levels, and free radicals. Comparison of base-line and chemical-induced transcriptomic responses in HepaRG and RPTEC/TERT1 cells using TempO-Seq.

The utilisation of genome-wide transcriptomics has played a pivotal role in advancing the field of toxicology, allowing the mapping of transcriptional signatures to chemical exposures. These activities have uncovered several transcriptionally regulated pathways that can be utilised for assessing the perturbation impact of a chemical and also the identification of toxic mode of action. However, current transcriptomic platforms are not very amenable to high-throughput workflows due to, high cost, complexities in sample preparation and relatively complex bioinformatic analysis. Thus, transcriptom
